Facing the global biodiversity crisis, conservation practitioners and decision-makers seek to catalyze wildlife recoveries in their region. Here we examined social-ecological attributes related to threatened species recovery in Canada. First, we used a retrospective approach to compare the trajectories of the original species assessed by Canada’s species-at-risk committee and found that only eight of 36 species now have decreased extinction risk relative to the past. There were no significant differences in human or financial capacity provided for recovery across species doing better, the same, or worse; the only significant difference was whether the primary cause of decline was alleviated or not. Second, when looking at species assessed at least twice between 2000 and 2019 we found that only eight of 422 (1.9%) experienced both increasing abundance and decreasing extinction risk. The defining characteristic of successful recoveries was first alleviating the original cause of decline, which was most often accomplished through strong regulatory intervention. Once declines were halted, practical interventions were highly species-specific. It is instructive to learn from conservation successes to scale resources appropriately and our results emphasize the importance of threat-specific intervention as a fundamental precursor to the successful restoration of biodiversity in Canada.

Abstract Achieving more effective and equitable environmental conservation practices and policies involves shifting from a human‐centric, top‐down perspective of environmental conservation to a perspective that respects and cares for all living and non‐living beings. Many Indigenous Peoples from around the globe embody approaches to environmental care that are rooted in values such as responsibility, respect and reciprocity (a.k.a. relational values), which, through meaningful engagement and support of Indigenous self‐determination, can guide Western society towards a fundamental shift in perspective, practices and relationships. We conducted interviews and sharing circles with 40 individuals from 12 Indigenous communities across Canada to describe: (1) the values, teachings and customs that are inherent to the way Indigenous Peoples relate to and care for the Land, (2) how these values and practices have changed over time and (3) ways to create environmental initiatives that are rooted in Indigenous values. Generally, participants emphasized the critical link between people and place, and how this leads to environmental practices rooted in values such as respect, reciprocity, humility and responsibility. They also reflected on the negative impacts of colonialism, environmental change and modernization on their connections to the Land and opportunities to practice these values, but highlighted that cultural revitalization efforts have started to restore traditional values and practices. To create environmental initiatives that are rooted in important values, research participants called for building better relationships both with nature and with each other through nature. Ultimately, uplifting Indigenous values systems and, specifically, the ways Indigenous Peoples relate to and care for the natural world stands to heal our relationship with the Land and safeguard it into the future. Long-term ecological research (LTER) projects are considered valuable training grounds for graduate student researchers, yet student voices are largely absent from discussions of LTER merits in the literature. We aimed to identify benefits and challenges encountered by current and former graduate students in conducting graduate research within LTER projects. To explore graduate student experiences and perspectives, we conducted a survey comprising both closed-ended questions (i.e., multiple choice and Likert scale) and open-ended questions. From the responses, we identified emergent categories related to positive and negative experiences using sentiment analysis. We found agreement with purported benefits in areas including networking and access to established field sites and protocols. However, participants also identified data accessibility, authorship decisions, communication, and interpersonal conflicts as significant sources of challenges. We synthesized survey results with existing literature to provide actionable recommendations for principal investigators in four main areas (data, authorship, communication, and management) through an LTER lens. In addition to providing longitudinal data, LTER projects offer graduate students both physical and methodological infrastructure that can serve as the scaffold for new research questions to be developed. However, the likelihood of success of student research, as well as the success of the students themselves, can be improved when the needs of graduate students are prioritized.

Introduction Antivenom is a lifesaving medicine for treating snakebite envenoming, yet there has been a crisis in antivenom supply for many decades. Despite this, substantial quantities of antivenom stocks expire before use. This study has investigated whether expired antivenoms retain preclinical quality and efficacy, with the rationale that they could be used in emergency situations when in-date antivenom is unavailable.Methods Using WHO guidelines and industry test requirements, we examined the in vitro stability and murine in vivo efficacy of eight batches of the sub-Saharan African antivenom, South African Institute for Medical Research polyvalent, that had expired at various times over a period of 30 years.Results We demonstrate modest declines in immunochemical stability, with antivenoms older than 25 years having high levels of turbidity. In vitro preclinical analysis demonstrated all expired antivenoms retained immunological recognition of venom antigens and the ability to inhibit key toxin families. All expired antivenoms retained comparable in vivo preclinical efficacy in preventing the lethal effects of envenoming in mice versus three regionally and medically important venoms.Conclusions This study provides strong rationale for stakeholders, including manufacturers, regulators and health authorities, to explore the use of expired antivenom more broadly, to aid in alleviating critical shortages in antivenom supply in the short term and the extension of antivenom shelf life in the longer term.

Abstract Antivenom is currently the first-choice treatment for snakebite envenoming. However, only a low proportion of antivenom immunoglobulins are specific to venom toxins, resulting in poor dose efficacy and potency. We sought to investigate whether linear venom epitopes displayed on virus like particles can stimulate an antibody response capable of recognising venom toxins from diverse medically important species. Bioinformatically-designed epitopes, corresponding to predicted conserved regions of group I phospholipase A2 and three finger toxins, were engineered for display on the surface of hepatitis B core antigen virus like particles and used to immunise female CD1 mice over a 14 weeks. Antibody responses to all venom epitope virus like particles were detectable by ELISA by the end of the immunisation period, although total antibody and epitope specific antibody titres were variable against the different epitope immunogens. Immunoblots using pooled sera demonstrated recognition of various venom components in a diverse panel of six elapid venoms, representing three continents and four genera. Insufficient antibody yields precluded a thorough assessment of the neutralising ability of the generated antibodies, however we were able to test polyclonal anti-PLA2 IgG from three animals against the PLA 2 activity of Naja nigricollis venom, all of which showed no neutralising ability. This study demonstrates proof-of-principle that virus like particles engineered to display conserved toxin linear epitopes can elicit specific antibody responses in mice which are able to recognise a geographically broad range of elapid venoms.

Birds communicate primarily using vocal signals. These signals undergo strong selection for effective transmission, which is often critical for survival. Passerines are an excellent group for studying vocal communication due to their diverse vocal repertoires, from simple calls to complex songs. However, the study of songbird calls is often overshadowed by studies of more conspicuous songs, which may bias our understanding of the true diversity of bird vocalizations. Here, we detail the vocal communication system of the Rifleman ( Acanthisitta chloris ), a New Zealand Wren (family: Acanthisittidae), a songless species from a species-poor suborder (Acanthisitti), sister to the oscine and suboscine Passerines. We describe nine adult call types and three juvenile call types, providing the most complete vocal repertoire of this species to date. Within these call types, we found variation in the spectral acoustic structure between different behavioral contexts. Furthermore, we also found negligible differences between sexes in contact calls, despite substantial size dimorphism. Using these data, we classify call types and discuss their probable function(s) based on behavioral context, such as foraging, provisioning nests, nuptial feeding, and alarm calling. Collectively, we hope that this study will provide a foundation for understanding the evolution and function of calls in Passerines and encourage more study of calls across taxa.

Traditional food systems based on harvest from the local environment are fundamental to the well-being of many communities, but their security is challenged by rapid socio-ecological change. We synthesized literature and data describing how a fundamental form of biodiversity, animal body size, contributes to the security of traditional food systems through relationships with species availability, accessibility, adequacy, and use. We found larger vertebrate species were more available, accessible, and used on a per kilogram basis, particularly for mammals. Conversely, larger species were no more or less adequate from a combined nutritional, health, and cultural perspective. Larger species represented more biomass, and this biomass required less time to harvest, with greater but more variable mean caloric returns over time. Smaller species provided more consistent caloric returns and were harvested during documented shortages of prey. This reliance on species with a range of body sizes is consistent with optimal foraging theory and the evolutionary value of flexibility, and highlights the importance of a biodiverse pool of species for traditional food security in times of change. Our synthesis of published literature and data highlights the many socio-ecological correlates of species size and how these relate to the security of traditional food systems.

Snakebite envenoming can be a life-threatening medical emergency that requires prompt medical intervention to neutralise the effects of venom toxins. Each year up to 138,000 people die from snakebites and threefold more victims suffer life-altering disabilities. The current treatment of snakebite relies solely on antivenom—polyclonal antibodies isolated from the plasma of hyperimmunised animals—which is associated with numerous deficiencies. The ADDovenom project seeks to deliver a novel snakebite therapy, through the use of an innovative protein-based scaffold as a next-generation antivenom. The ADDomer is a megadalton-sized, thermostable synthetic nanoparticle derived from the adenovirus penton base protein; it has 60 high-avidity binding sites to neutralise venom toxins. Here, we outline our experimental strategies to achieve this goal using state-of-the-art protein engineering, expression technology and mass spectrometry, as well as in vitro and in vivo venom neutralisation assays. We anticipate that the approaches described here will produce antivenom with unparalleled efficacy, safety and affordability.

BackgroundSnakebite is a major public health concern in Eswatini, where treatment relies upon one antivenom-SAIMR Polyvalent. Although effective in treating snakebite, SAIMR Polyvalent is difficult to source outside its manufacturing country (South Africa) and is dauntingly expensive. We compared the preclinical venom-neutralising efficacy of two alternative antivenoms with that of SAIMR Polyvalent against the lethal and tissue-destructive effects of venoms from five species of medically important snakes using in vivo murine assays. The test antivenoms were 'Panafrican' manufactured by Instituto Clodomiro Picado and 'PANAF' manufactured by Premium Serums & Vaccines.Principal findingsIn vivo murine preclinical studies identified both test antivenoms were equally or more effective than SAIMR Polyvalent at neutralising lethal and tissue-destructive effects of Naja mossambica venom. Both test antivenoms were less effective than SAIMR Polyvalent at neutralising the lethal effects of Bitis arietans, Dendroaspis polylepis, Hemachatus haemachatus and Naja annulifera venoms, but similarly effective at neutralising tissue damage induced by B. arietans and H. haemachatus venoms. In vitro immunological assays identified that the titres and toxin-specificities of immunoglobulins (iGs) in the test antivenoms were comparable to that of SAIMR Polyvalent. Plasma clotting disturbances by H. haemachatus and N. mossambica were neutralised by the test antivenoms, whereas SAIMR Polyvalent failed to neutralise this bioactivity of N. mossambica venom. B. arietans SVMP activity was equally reduced by all three antivenoms, and H. haemachatus and N. mossambica PLA2 activities were neutralised by all three antivenoms.ConclusionsWhile both Panafrican and PANAF antivenoms exhibited promising preclinical efficacies, both were less poly-specifically effective than SAIMR Polyvalent in these murine assays. The efficacy of these antivenoms against the lethal and tissue-destructive effects of N. mossambica venom, the most common biting species in Eswatini, identify that Panafrican and PANAF antivenoms offer effective alternatives to SAIMR Polyvalent for the treatment of snakebite in Eswatini, and potentially for neighbouring countries.

Snakebite envenoming affects more than 250,000 people annually in sub-Saharan Africa. Envenoming by Dispholidus typus (boomslang) results in venom-induced consumption coagulopathy (VICC), whereby highly abundant prothrombin-activating snake venom metalloproteinases (SVMPs) consume clotting factors and deplete fibrinogen. The only available treatment for D. typus envenoming is the monovalent SAIMR Boomslang antivenom. Treatment options are urgently required because this antivenom is often difficult to source and, at US$6000/vial, typically unaffordable for most snakebite patients. We therefore investigated the in vitro and in vivo preclinical efficacy of four SVMP inhibitors to neutralise the effects of D. typus venom; the matrix metalloproteinase inhibitors marimastat and prinomastat, and the metal chelators dimercaprol and DMPS. The venom of D. typus exhibited an SVMP-driven procoagulant phenotype in vitro. Marimastat and prinomastat demonstrated equipotent inhibition of the SVMP-mediated procoagulant activity of the venom in vitro, whereas dimercaprol and DMPS showed considerably lower potency. However, when tested in preclinical murine models of envenoming using mixed sex CD1 mice, DMPS and marimastat demonstrated partial protection against venom lethality, demonstrated by prolonged survival times of experimental animals, whereas dimercaprol and prinomastat failed to confer any protection at the doses tested. The preclinical results presented here demonstrate that DMPS and marimastat show potential as novel small molecule-based therapeutics for D. typus snakebite envenoming. These two drugs have been previously shown to be effective against Echis ocellatus VICC in preclinical models, and thus we conclude that marimastat and DMPS should be further explored as potentially valuable early intervention therapeutics to broadly treat VICC following snakebite envenoming in sub-Saharan Africa.

To improve the characterization of snake venom protein profiles, we report the application of a new generation of proteomic methodology to deeply characterize complex protein mixtures. The new approach, combining a synergic multi-enzymatic and a time-limited digestion (MELD), is a versatile and straightforward protocol previously developed by our group. The higher number of overlapping peptides generated during MELD increases the quality of downstream peptide sequencing and of protein identification. In this context, this work aims at applying the MELD strategy to a venomics purpose for the first time, and especially for the characterization of snake venoms. We used four venoms as the test models for this proof of concept: two Elapidae (Dendroaspis polylepis and Naja naja) and two Viperidae (Bitis arietans and Echis ocellatus). Each venom was reduced and alkylated before being submitted to two different protocols: the classical bottom-up proteomics strategy including a digestion step with trypsin only, or MELD, which combines the activities of trypsin, Glu-C and chymotrypsin with a limited digestion approach. The resulting samples were then injected on an M-Class chromatographic system, and hyphenated to a Q-Exactive Mass Spectrometer. Toxins and protein identification were performed by Peaks Studio X+. The results show that MELD considerably improves the number of sequenced (de novo) peptides and identified peptides from protein databases, leading to the unambiguous identification of a greater number of toxins and proteins. For each venom, MELD was successful, not only in terms of the identification of the major toxins (increasing of sequence coverage), but also concerning the less abundant cellular components (identification of new groups of proteins). In light of these results, MELD represents a credible methodology to be applied as the next generation of proteomics approaches dedicated to venomic analysis. It may open new perspectives for the sequencing and inventorying of the venom arsenal and should expand global knowledge about venom composition.

Abiotic and biotic factors drive compositional differences among local species assemblages. Determining the influence of different drivers on beta diversity patterns can provide insights into processes governing community organization. Examining beta diversity patterns along taxonomic, phylogenetic and functional dimensions enables a nuanced understanding of underlying processes that govern community assembly and dynamics. The dynamic and complex riparian habitats in the Himalaya, and the hyper-diverse riverine bird community offer a fascinating setup to examine the role of environmental factors in influencing community structuring. Using a large dataset on river bird communities from field census across multiple drainages in the Indian Himalaya, we aimed at discerning processes that structure these communities through an understanding of pair-wise dissimilarities in species composition across sites. We determined the relative contributions of turnover and nestedness in taxonomic, phylogenetic, and functional beta diversity patterns in the Eastern and Western Himalaya that differ in species richness. Generalized Dissimilarity Modeling was used to examine the relative contributions of climatic, geographic, and anthropogenic factors toward explaining different metrics of beta diversity. The riverine bird communities in the drier and seasonal Western Himalaya were poorer in species richness, more phylogenetically and functionally clustered than that in the Eastern Himalaya. The contribution of the turnover component to the overall beta diversity was higher than the nestedness component in river bird communities, particularly in the Eastern Himalaya. Habitat and climatic factors differentially influenced the beta diversity patterns in both Eastern and Western Himalaya, with river width consistently explaining a large variation in beta diversity in the east and the west. The results show that environmental filtering plays a crucial role in structuring riverine bird communities in the Himalayan headwaters, highlighting the need to ameliorate the threats posed by the slew of hydroelectric projects and forest loss in the region.

Abstract Unbiased population density estimates are critical for ecological research and wildlife management but are often difficult to obtain. Researchers use a variety of sampling and statistical methods to generate estimates of density, but few studies have compared estimates across methods. During 2016–2017, we surveyed Canada lynx (Lynx canadensis) in southwestern Yukon Territory, Canada, using track transect counts, hair snares, camera traps, live traps, and Global Positioning System (GPS) collars. From these data, we estimated lynx density with two linearly scaled count methods, one spatial mark–recapture method, three spatial mark–resight methods, and one cumulative‐time method. We found up to fivefold variation in point density estimates despite adhering to method requirements and assumptions in a manner consistent with other studies. Our results highlight the dependency of density estimates on sampling process and model assumptions and demonstrate the value of careful and unbiased sampling design. Further research is needed to fully assess the accuracy and limitations of the many wildlife density estimation methods that are currently in use so that techniques can be appropriately applied to typical study systems and species.

For decades, Indigenous voices have called for more collaborative and inclusive research practices. Interest in community-collaborative research is consequently growing among university-based researchers in Canada. However, many researchers receive little formal training on how to collaboratively conduct research with Indigenous communities. This is particularly problematic for early-career researchers (ECRs) whose fieldwork often involves interacting with communities. To address this lack of training, two peer-led workshops for Canadian ECRs were organized in 2016 and 2017 with the following objectives: (i) to cultivate awareness about Indigenous cultures, histories, and languages; (ii) to promote sharing of Indigenous and non-Indigenous ways of knowing; and (iii) to foster approaches and explore tools for conducting community-collaborative research. Here we present these peer-led Intercultural Indigenous Workshops and discuss workshop outcomes according to five themes: scope and interdisciplinarity, Indigenous representation, workshop environment, skillful moderation, and workshop outcomes. Although workshops cannot replace the invaluable experience gained through working directly with Indigenous communities, we show that peer-led workshops can be an effective way for ECRs to develop key skills for conducting meaningful collaborative research. Peer-led workshops are therefore an important but insufficient step toward more inclusive research paradigms in Canada.

Abstract Vertebrate scavenging can impact food web dynamics, but our understanding of this process stems predominantly from monitoring herbivore carrion and extrapolating results across carcass types. Recent evidence suggests carnivores may avoid intraguild scavenging to reduce parasite transmission. If this behavior is widespread across diverse ecosystems, estimation of nutrient cycling and community scavenging rates are likely biased to a currently unknown degree. We examined whether the time to initiate scavenging, carcass persistence, or the richness of species scavenging in the boreal forest of Yukon, Canada, differed between carnivore and herbivore carcasses. Vertebrates took longer to initiate scavenging on carnivore carcasses (3.2 d) relative to herbivore carcasses (1.1 d), and carnivore carcasses persisted on the landscape for over a month longer (48.4 d and 5.5 d, respectively). The longer persistence times were due to the reduction in scavenging by carnivores such as Canada lynx (Lynx canadensis). Decreased scavenging was caused by changes in the propensity to consume carnivore carrion, as the number of species detecting a carcass within the first week did not differ between carnivore and herbivore carcasses. These results have ramifications for our understanding of nutrient cycling and food web dynamics in the boreal forest and provide further support that carcass type should be included in future studies.

BackgroundAdverse reactions to antivenom considerably complicate the clinical management of snakebite envenomed patients because it necessitates a temporary suspension of life-saving antivenom, increases costs and can compromise patient outcomes. This study sought to explore the association between cattle-herding occupation and ethnic group and the occurrence of early adverse reactions to antivenom.MethodsThis cross-sectional study was conducted between the 25th April and 11th July 2011 at the Kaltungo General Hospital in north east Nigeria. The exposure variable of cattle-herding occupation showed a strong correlation with the ethnic group variable, thus these were combined into a new variable with three categories (Fulani and herder, either Fulani or herder, and neither Fulani nor herder). The outcome variable was the occurrence of early adverse reactions, defined as any new symptoms occurring within 6 hours of antivenom administration. Odds Ratios were estimated using multivariable logistic regression models controlling for potential confounders.ResultsAmong 231 envenomed snakebite victims, the overall incidence of early adverse reactions was 11.9% (95% confidence intervals: 8.0-16.9%). Patients who were Fulani and herders had a higher incidence of early adverse reactions compared to patients who were neither Fulani nor herders (20% vs 5.7%). After adjusting for age and gender, victims who were Fulani and herders were 5.9 times more likely to have an early adverse reaction, compared to victims who were neither Fulani nor herders (95% CI: 1.88-18.59; p = 0.002).InterpretationTo the best of our knowledge, this is the first study to provide evidence of higher odds of early adverse reactions among patients from a particular occupation and/or ethnic group. We recommend that snake envenomed patients of Fulani origin be especially closely monitored for adverse reactions, that hospitals receiving these patients be appropriately resourced to manage both envenoming and adverse reactions and that premedication with adrenaline should be considered. Our findings provide an argument for speculation on the influence of immunological or lifestyle-related differences on the occurrence of early adverse reactions to antivenom.

Snakebite is a neglected tropical disease that causes high rates of global mortality and morbidity. Although snakebite can cause a variety of pathologies in victims, haemotoxic effects are particularly common and are typically characterised by haemorrhage and/or venom-induced consumption coagulopathy. Despite polyclonal antibody-based antivenoms being the mainstay life-saving therapy for snakebite, they are associated with limited cross-snake species efficacy, as there is often extensive toxin variation between snake venoms, including those used as immunogens for antivenom production. This restricts the therapeutic utility of any antivenom to certain geographical regions. In this study, we explored the feasibility of using recombinantly expressed toxins as immunogens to stimulate focused, pathology-specific, antibodies in order to broadly counteract specific toxins associated with snakebite envenoming. Three snake venom serine proteases (SVSP) toxins, sourced from geographically diverse and medically important viper snake venoms, were successfully expressed in HEK293F mammalian cells and used for murine immunisation. Analyses of the resulting antibody responses revealed that ancrod and RVV-V stimulated the strongest immune responses, and that experimental antivenoms directed against these recombinant SVSP toxins, and a mixture of the three different immunogens, extensively recognised and exhibited immunological binding towards a variety of native snake venoms. While the experimental antivenoms showed some reduction in abnormal clotting parameters stimulated by the toxin immunogens and crude venom, specifically reducing the depletion of fibrinogen levels and prolongation of prothrombin times, fibrinogen degradation experiments revealed that they broadly protected against venom- and toxin-induced fibrinogenolytic functional activities. Overall, our findings further strengthen the case for the use of recombinant venom toxins as supplemental immunogens to stimulate focused and desirable antibody responses capable of neutralising venom-induced pathological effects, and therefore potentially circumventing some of the limitations associated with current snakebite therapies.

BackgroundThe World Health Organization's strategy to halve snakebite mortality and morbidity by 2030 includes an emphasis on a risk-benefit process assessing the preclinical efficacy of antivenoms manufactured for sub-Saharan Africa. To assist this process, we systematically collected, standardised and analysed all publicly available data on the preclinical efficacy of antivenoms designed for sub-Saharan Africa.Methodology/principal findingsUsing a systematic search of publication databases, we focused on publicly available preclinical reports of the efficacy of 16 antivenom products available in sub Saharan Africa. Publications since 1999 reporting the industry standard intravenous pre-incubation method of murine in vivo neutralisation of venom lethality (median effective dose [ED50]) were included. Eighteen publications met the criteria. To permit comparison of the several different reported ED50 values, it was necessary to standardise these to microlitre of antivenom resulting in 50% survival of mice challenged per milligram of venom (μl/mg). We were unable to identify publicly available preclinical data on four antivenoms, whilst data for six polyspecific antivenoms were restricted to a small number of venoms. Only four antivenoms were tested against a wide range of venoms. Examination of these studies for the reporting of key metrics required for interpreting antivenom ED50s were highly variable, as evidenced by eight different units being used for the described ED50 values.Conclusions/significanceThere is a disturbing lack of (i) preclinical efficacy testing of antivenom for sub Saharan Africa, (ii) publicly available reports and (iii) independent scrutiny of this medically important data. Where reports do exist, the methods and metrics used are highly variable. This prevents comprehensive meta-analysis of antivenom preclinical efficacy, and severely reduces the utility of antivenom ED50 results in the decision making of physicians treating patients and of national and international health agencies. Here, we propose the use of a standardised result reporting checklist to resolve this issue. Implementation of these straightforward steps will deliver uniform evaluation of products across laboratories, facilitate meta-analyses, and contribute vital information for designing the clinical trials needed to achieve the WHO target of halving snakebite morbidity and mortality by 2030.

Technological miniaturization is driving a biologging revolution that is producing detailed and sophisticated techniques of assessing individual behavioral responses to environmental conditions. Among the many advancements this revolution has brought is an ability to record behavioral responses of nocturnal, free-ranging species. Here, we combine captive validations of acceleration signatures with acoustic recordings from free-ranging individuals to classify behavior at two resolutions. Combining these classifications with ~2 month-long recordings, we describe winter time budgets, and responses of free-ranging snowshoe hares to changing moonlight. We successfully classified snowshoe hare behavior into four categories (not moving, foraging, hopping, and sprinting) using low frequency accelerometry, with an overall model accuracy of 88%, and acoustic recordings to three categories (silence, hopping, and chewing) with an accuracy of 94%. Broad-scale accelerometer-classified categories were composed of multiple fine-scale behavioral states with the composition varying between individuals and across the day. Time budgets revealed that hares spent ~50% of their time foraging and ~50% not moving, with most foraging and feeding occurring at night. We found that hares adjusted timing of activity in response to moon phase, with a 6% reduction in foraging and 30% reduction in traveling during the night when the moon was full. Hares compensated for this lost foraging time by extending foraging into the morning hours of the following day. Using two biologging technologies to identify behavior, we demonstrate the possibility of combining multiple devices when documenting behavior of cryptic species.

Current drugs to treat African sleeping sickness are inadequate and new therapies are urgently required. As part of a medicinal chemistry programme based upon the simplification of acetogenin-type ether scaffolds, we previously reported the promising trypanocidal activity of compound 1, a bis-tetrahydropyran 1,4-triazole (B-THP-T) inhibitor. This study aims to identify the protein target(s) of this class of compound in Trypanosoma brucei to understand its mode of action and aid further structural optimisation. We used compound 3, a diazirine- and alkyne-containing bi-functional photo-affinity probe analogue of our lead B-THP-T, compound 1, to identify potential targets of our lead compound in the procyclic form T. brucei. Bi-functional compound 3 was UV cross-linked to its target(s) in vivo and biotin affinity or Cy5.5 reporter tags were subsequently appended by Cu(II)-catalysed azide-alkyne cycloaddition. The biotinylated protein adducts were isolated with streptavidin affinity beads and subsequent LC-MSMS identified the FoF1-ATP synthase (mitochondrial complex V) as a potential target. This target identification was confirmed using various different approaches. We show that (i) compound 1 decreases cellular ATP levels (ii) by inhibiting oxidative phosphorylation (iii) at the FoF1-ATP synthase. Furthermore, the use of GFP-PTP-tagged subunits of the FoF1-ATP synthase, shows that our compounds bind specifically to both the α- and β-subunits of the ATP synthase. The FoF1-ATP synthase is a target of our simplified acetogenin-type analogues. This mitochondrial complex is essential in both procyclic and bloodstream forms of T. brucei and its identification as our target will enable further inhibitor optimisation towards future drug discovery. Furthermore, the photo-affinity labeling technique described here can be readily applied to other drugs of unknown targets to identify their modes of action and facilitate more broadly therapeutic drug design in any pathogen or disease model.

In recent years, the Xishuangbanna Tropical Botanic Garden (XTBG) and the Xishuangbanna prefecture government have woken up to the environmental threat that extensive monoculture rubber cultivation poses in this tropical site in southern Yunnan. In response, XTBG has become a centre of knowledge production on how to restore some ethnic minority farmers′ rubber fields to natural forest. As is common globally, the plan relies on mainstream ecology and leaves out farmers′ experience and opinions. Curiously, in the 1980s and 1990s XTBG was a centre of human ecology research on farmers′ knowledge about managing ecosystems and protecting biodiversity. This paper traces the transformation in the values, actors, institutional configurations, and epistemic communities that enabled farmers′ experiences to be central to knowledge production and inscription across surrounding landscapes in the 1980s and 1990s, only to be disparaged and ignored in the production of knowledge and landscapes in the 2000s. Farmers who once taught XTBG scientists the names of species and how to survive on them during the Cultural Revolution are now excluded from determining how to restore those same species to the hills around them. Our analysis suggests possible pathways for ecological knowledge to be generated as well as lost.

Soil-transmitted helminths (STH) infect more than 2 billion humans worldwide, causing significant morbidity in children. There are few data on the epidemiology and risk factors for infection in pre-school children. To investigate risk factors for infection in early childhood, we analysed data prospectively collected in the ECUAVIDA birth cohort in Ecuador.Children were recruited at birth and followed up to 3 years of age with periodic collection of stool samples that were examined microscopically for STH parasites. Data on social, demographic, and environmental risk factors were collected from the mother at time of enrollment. Associations between exposures and detection of STH infections were analysed by multivariable logistic regression. Data were analysed from 1,697 children for whom a stool sample was obtained at 3 years. 42.3% had at least one STH infection in the first 3 years of life and the most common infections were caused by A. lumbricoides (33.2% of children) and T. trichiura (21.2%). Hookworm infection was detected in 0.9% of children. Risk of STH infection was associated with factors indicative of poverty in our study population such as Afro-Ecuadorian ethnicity and low maternal educational level. Maternal STH infections during pregnancy were strong risk factors for any childhood STH infection, infections with either A. lumbricoides or T. trichiura, and early age of first STH infection. Children of mothers with moderate to high infections intensities with A. lumbricoides were most at risk.Our data show high rates of infection with STH parasites during the first 3 years of life in an Ecuadorian birth cohort, an observation that was strongly associated with maternal STH infections during pregnancy. The targeted treatment of women of childbearing age, in particular before pregnancy, with anthelmintic drugs could offer a novel approach to the prevention of STH infections in pre-school children.

Animal personality or temperament refers to individual differences in behaviour that are repeatable over time and across contexts. Personality has been linked to life-history traits, energetic traits and fitness, with implications for the evolution of behaviour. Personality has been quantified for a range of taxa (e.g., fish, songbirds, small mammals) but, so far, there has been little work on personality in bats, despite their diversity and potential as a model taxon for comparative studies. We used a novel environment test to quantify personality in little brown bats (Myotis lucifugus) and assess the short-term repeatability of a range of behaviours. We tested the hypothesis that development influences values of personality traits and predicted that trait values associated with activity would increase between newly volant, pre-weaning young-of-the-year (YOY) and more mature, self-sufficient YOY. We identified personality dimensions that were consistent with past studies of other taxa and found that these traits were repeatable over a 24-hour period. Consistent with our prediction, older YOY captured at a fall swarming site prior to hibernation had higher activity scores than younger YOY bats captured at a maternity colony, suggesting that personality traits vary as development progresses in YOY bats. Thus, we found evidence of short-term consistency of personality within individuals but with the potential for temporal flexibility of traits, depending on age.
