Abstract Background The choroid plexus (ChP) helps maintain the homeostasis of the brain by forming the blood-CSF barrier via tight junctions (TJ) at the choroid plexus epithelial cells, and subsequently preventing neuroinflammation by restricting immune cells infiltration into the central nervous system. However, whether chronic cerebral hypoperfusion causes ChP structural damage and blood-CSF barrier impairment remains understudied. Methods The bilateral carotid stenosis (BCAS) model in adult male C57BL/6 J mice was used to induce cerebral hypoperfusion, a model for vascular contributions to cognitive impairment and dementia (VCID). BCAS-mediated changes of the blood-CSF barrier TJ proteins, apical secretory Na+-K+-Cl− cotransporter isoform 1 (NKCC1) protein and regulatory serine-threonine kinases SPAK, and brain infiltration of myeloid-derived immune cells were assessed. Results BCAS triggered dynamic changes of TJ proteins (claudin 1, claudin 5) accompanied with stimulation of SPAK-NKCC1 complex and NF-κB in the ChP epithelial cells. These changes impacted the integrity of the blood-CSF barrier, as evidenced by ChP infiltration of macrophages/microglia, neutrophils and T cells. Importantly, pharmacological blockade of SPAK with its potent inhibitor ZT1a in BCAS mice attenuated brain immune cell infiltration and improved cognitive neurological function. Conclusions BCAS causes chronic ChP blood-CSF damage and immune cell infiltration. Our study sheds light on the SPAK-NKCC1 complex as a therapeutic target in neuroinflammation.

Space habitats typically have elevated CO2 and NASA is considering growing leafy greens in space to supplement astronauts’ diets. Dragoon’ and ‘Outredgeous’ lettuce, ‘Amara’ mustard, ‘Extra Dwarf’ pak choi, shungiku, ‘Red Russian’ kale, ‘Toscano’ kale, and ‘Barese’ Swiss chard were grown for 4 weeks at 400, 1500, 3000, and 6000 ppm CO2. Shoot fresh mass at 28 days was greater for one of more elevated CO2 levels for all species/cultivars except ‘Toscano’ kale. Fresh mass varied by species/cultivar, with pak choi and ‘Dragoon’ lettuce showing the greatest yields. Super-elevated CO2 (6000 ppm) reduced shoot mass for both lettuce cvs. compared to 3000 ppm. Elevated CO2 increased K levels for most species/cultivars but decreased Mg for some species/cultivars. CO2 affected Vitamin B1 and Vitamin C content but had no effect on Vitamin K. ‘Toscano’ and ‘Red Russian’ kale, and Amara mustard had the highest mineral and vitamin content.

More than 3.5 billion people depend on rice for more than 20% of their daily calories. Globally, Bangladesh is the third largest rice producer. With 171 million people, Bangladesh is also among the top consumers. Local rice production not only affects the country’s food security but also influences the global rice trade. A large yield gap has been reported due to weeds. Traditional hand weeding is very costly because of labor shortages resulting from industrialization. Limited data showed a higher yield and profits when using herbicides. However, quantitative data on various aspects of weed management and associated issues representing the country’s variable rice ecosystem, which is characterized by 30 agroecological zones, are lacking. We collected data on weed management practices from 865 farmers and 69 agrochemical shops covering all 30 agro-ecological zones (AEZs) through a structured survey. We observed a significant regional variation among various parameters. Approximately 82% of farmers use herbicides, and few rely solely on either manual weeding or herbicides. Pre-emergence herbicides are the predominant. Application procedures are almost the same across the country. Although 40% of farmers had secondary and higher-level education, most depend upon local sellers’ suggestions rather than reading the product label regarding the dose. Few farmers consider herbicides hazardous, and respondents rarely perceive any environmental impact. Pyrazosulfuron ethyl (35%) and acetochlor-containing bensulfuron methyl (27%) are the most-used chemical species. Approximately 45% of farmers observed that herbicides suppress early seedling growth. Additional fertilizer is required to compensate for this. Multiple weed species that are difficult to control through presently used herbicides were noted in all AEZs. Around 64% of farmers observed that herbicide application contributes to higher yields as a function of timely weeding. Cost comparisons showed that high labor prices will make rice cropping unprofitable in most parts of the country if herbicides are eliminated. Clear adverse effects of pre-emergence herbicides on early crop growth implied the potential benefits of broad-spectrum herbicide-tolerant genetically engineered (GE) rice to sustain the country’s food security. Additionally, such GE rice could incentivize the adoption of alternate wet and dry irrigation methods, leading to water and cost savings.

Abstract Mechanical stress can directly activate chemical reactions by reducing the reaction energy barrier. A possible mechanism of such mechanochemical activation is structural deformation of the reactant species. However, the effect of deformation on the reaction energetics is unclear, especially, for shear stress-driven reactions. Here, we investigated shear stress-driven oligomerization reactions of cyclohexene on silica using a combination of reactive molecular dynamics simulations and ball-on-flat tribometer experiments. Both simulations and experiments captured an exponential increase in reaction yield with shear stress. Elemental analysis of ball-on-flat reaction products revealed the presence of oxygen in the polymers, a trend corroborated by the simulations, highlighting the critical role of surface oxygen atoms in oligomerization reactions. Structural analysis of the reacting molecules in simulations indicated the reactants were deformed just before a reaction occurred. Quantitative evidence of shear-induced deformation was established by comparing bond lengths in cyclohexene molecules in equilibrium and prior to reactions. Nudged elastic band calculations showed that the deformation had a small effect on the transition state energy but notably increased the reactant state energy, ultimately leading to a reduction in the energy barrier. Finally, a quantitative relationship was developed between molecular deformation and energy barrier reduction by mechanical stress.

In this study, we synthesized novel α,β-unsaturated 2-cyanoacetamide derivatives (1–5) using microwave-assisted Knoevenagel condensation. Characterization of these compounds was carried out using FTIR and 1H NMR spectroscopy. We then evaluated their in vitro antibacterial activity against both gram-positive and gram-negative pathogenic bacteria. Additionally, we employed in silico methods, including ADMET prediction and density functional theory (DFT) calculations of molecular orbital properties, to investigate these cyanoacetamide derivatives (1–5). Molecular docking was used to assess the binding interactions of these derivatives (1–5) with seven target proteins (5MM8, 4NZZ, 7FEQ, 5NIJ, ITM2, 6SE1, and 5GVZ) and compared them to the reference standard tyrphostin AG99. Notably, derivative 5 exhibited the most favorable binding affinity, with a binding energy of -7.7 kcal mol−1 when interacting with the staphylococcus aureus (PDB:5MM8), while also meeting all drug-likeness criteria. Additionally, molecular dynamics simulations were carried out to evaluate the stability of the interaction between the protein and ligand, utilizing parameters such as Root-Mean-Square Deviation (RMSD), Root-Mean-Square Fluctuation (RMSF), Radius of Gyration (Rg), and Principal Component Analysis (PCA). A 50 nanosecond molecular dynamics (MD) simulation was performed to investigate stability further, incorporating RMSD and RMSF analyses on compound 5 within the active binding site of the modeled protein across different temperatures (300, 305, 310, and 320 K). Among these temperatures, compound 5 exhibited an RMSD value ranging from approximately 0.2 to 0.3 nm at 310 K (body temperature) with the 5MM8 target, which differed from the other temperature conditions. The in silico results suggest that compound 5 maintained significant conformational stability throughout the 50 ns simulation period. It is consistent with its low docking energy and in vitro findings concerning α,β-unsaturated cyanoacetamides.Key insights from this study include: • The creation of innovative α,β-unsaturated 2-cyanoacetamide derivatives (1–5) employing cost-effective, licensed, versatile, and efficient software for both in silico and in vitro assessment of antibacterial activity. • Utilization of FTIR and NMR techniques for characterizing compounds 1–5.

Cerebrovascular diseases and their sequalae, such as ischemic stroke, chronic cerebral hypoperfusion, and vascular dementia are significant contributors to adult disability and cognitive impairment in the modern world. Astrocytes are an integral part of the neurovascular unit in the CNS and play a pivotal role in CNS homeostasis, including ionic and pH balance, neurotransmission, cerebral blood flow, and metabolism. Astrocytes respond to cerebral insults, inflammation, and diseases through unique molecular, morphological, and functional changes, collectively known as reactive astrogliosis. The function of reactive astrocytes has been a subject of debate. Initially, astrocytes were thought to primarily play a supportive role in maintaining the structure and function of the nervous system. However, recent studies suggest that reactive astrocytes may have both beneficial and detrimental effects. For example, in chronic cerebral hypoperfusion, reactive astrocytes can cause oligodendrocyte death and demyelination. In this review, we will summarize the (1) roles of ion transporter cascade in reactive astrogliosis, (2) role of reactive astrocytes in vascular dementia and related dementias, and (3) potential therapeutic approaches for dementing disorders targeting reactive astrocytes. Understanding the relationship between ion transporter cascade, reactive astrogliosis, and cerebrovascular diseases may reveal mechanisms and targets for the development of therapies for brain diseases associated with reactive astrogliosis.

Abstract Plasma polymerized (PP) methyl acrylate (MA) and vinyl acetate (VA) composite thin films were deposited onto glass substrate varying MA and VA monomer concentrations. Thickness of the composite polymers is observed to vary on the MA and VA monomer ratios, where MA is found more reactive. The FESEM images of the composite polymers show better surface morphology compared to those of the homopolymers. Appearance of broad absorption bands in the FTIR spectra of polymer indicates the structural changes compared to monomer during polymerization. Thermogravimetric analysis and differential scanning calorimetry indicate that composite films are thermally more stable (up to 617 K) compared to homopolymer thin films (563 K). The current density versus voltage (J–V) characteristics of PP(MA-VA) composite films (sandwiched between aluminum electrodes) with different MA and VA ratios showed that the J values of the composite films gradually increase with elevating VA monomer and also with temperature (298–373 K). On the other hand, this value increases with decreasing the thickness of the composite films, which complies with the other studies. The conduction of the thickness-dependent composite films showed Ohmic in nature in the lower voltage region ( 10 V) operating over the entire range of temperature. The activation energy at room temperature was found to be ~ 0.019 eV in the Ohmic region and 0.260 eV in the non-Ohmic region.

Genetic disruption of glycosyltransferases has provided clear information on the roles of their reaction products in the body. Our group has studied the function of glycosphingolipids by genetic engineering of glycosyltransferases in cell culture and in mice, which has demonstrated both expected and unexpected results. Among these findings, aspermatogenesis in ganglioside GM2/GD2 synthase knockout mice was one of the most surprising and intriguing results. There were no sperms in testis, and multinuclear giant cells were detected instead of spermatids. Although serum levels of testosterone in the male mice were extremely low, testosterone accumulated in the interstitial tissues, including Leydig cells, and seemed not to be transferred into the seminiferous tubules or vascular cavity from Leydig cells. This was considered to be the cause of aspermatogenesis and low serum levels of testosterone. Patients with a mutant GM2/GD2 synthase gene (SPG26) showed similar clinical signs, not only in terms of the neurological aspects, but also in the male reproductive system. The mechanisms for testosterone transport by gangliosides are discussed here based on our own results and reports from other laboratories.

The major industrial cities of Bangladesh are experiencing significant air-pollution-related problems due to the increased trend of particulate matter (PM2.5) and other pollutants. This paper aimed to investigate and understand the relationship between PM2.5 and land use and climatic variables to identify the riskiest areas and population groups using a geographic information system and regression analysis. The results show that about 41% of PM2.5 concentration (μg/m3) increased within 19 years (2002–2021) in the study area, while the highest concentration of PM2.5 was found from 2012 to 2021. The concentrations of PM2.5 were higher over barren lands, forests, croplands, and urban areas. From 2002–2021, the concentration increased by about 64%, 62.7%, 57%, and 55% (μg/m3) annually over barren lands, forests, cropland, and urban regions. The highest concentration level of PM2.5 (84 μg/m3) among other land use classes was found in urban areas in 2021. The regression analysis shows that air pressure (hPa) (r2 = −0.26), evaporation (kg m−2) (r2 = −0.01), humidity (kg m−2) (r2 = −0.22), rainfall (mm/h) (r2 = −0.20), and water vapor (kg m−2) (r2 = −0.03) were negatively correlated with PM2.5. On the other hand, air temperature (k) (r2 = 0.24), ground heat (W m−2) (r2 = 0.60), and wind speed (m s−1) (r2 = 0.34) were positively correlated with PM2.5. More than 60 Upazilas were included in the most polluted areas, with a total population of 11,260,162 in the high-risk/hotspot zone (1,948,029 aged 0–5, 485,407 aged 50–69). Governmental departments along with policymakers, stainable development practitioners, academicians, and others may use the main results of the paper for integrated air pollution mitigation and management in Bangladesh as well as in other geographical settings worldwide.

Taking the work conducted by the global navigation satellite system (GNSS) software-defined radio (SDR) working group during the last decade as a seed, this contribution summarizes, for the first time, the history of GNSS SDR development. This report highlights selected SDR implementations and achievements that are available to the public or that influenced the general development of SDR. Aspects related to the standardization process of intermediate-frequency sample data and metadata are discussed, and an update of the Institute of Navigation SDR Standard is proposed. This work focuses on GNSS SDR implementations in general-purpose processors and leaves aside developments conducted on field programmable gate array and application-specific integrated circuit platforms. Data collection systems (i.e., front-ends) have always been of paramount importance for GNSS SDRs and are thus partly covered in this work. This report represents the knowledge of the authors but is not meant as a complete description of SDR history.

Abstract Background Infection Prevention and Control (IPC) is critical in controlling the COVID-19 pandemic and is one of the pillars of the WHO COVID-19 Strategic Preparedness and Response Plan 2020. We conducted an Intra-Action Review (IAR) of IPC response efforts to the COVID-19 pandemic in Cox's Bazar, Bangladesh, to identify best practices, challenges, and recommendations for improvement of the current and future responses. Methods We conducted two meetings with 54 participants purposively selected from different organizations and agencies involved in the frontline implementation of IPC in Cox's Bazar district, Bangladesh. We used the IPC trigger questions from the WHO country COVID-19 IAR: trigger question database to guide the discussions. Meeting notes and transcripts were then analyzed manually using content analysis, and results were presented in text and quotes. Results Best practices included: assessments, a response plan, a working group, trainings, early case identification and isolation, hand hygiene in Health Facilities (HFs), monitoring and feedback, general masking in HFs, supportive supervision, design, infrastructure and environmental controls in Severe Acute Respiratory Infection Isolation and Treatment Centers (SARI ITCs) and HFs and waste management. Challenges included: frequent breakdown of incinerators, limited PPE supply, inconsistent adherence to IPC, lack of availability of uniforms for health workers, in particular cultural and gender appropriate uniforms and Personal Protective Equipment (PPE). Recommendations from the IAR were: (1) to promote the institutionalization of IPC, programs in HFs (2) establishment of IPC monitoring mechanisms in all HCFs, (3) strengthening IPC education and training in health care facilities, and (4) strengthen public health and social measures in communities. Conclusion Establishing IPC programmes that include monitoring and continuous training are critical in promoting consistent and adaptive IPC practices. Response to a pandemic crisis combined with concurrent emergencies, such as protracted displacement of populations with many diverse actors, can only be successful with highly coordinated planning, leadership, resource mobilization, and close supervision.

Abstract Positioning, Navigation and Timing (PNT) are services generally provided by Global Navigation Satellite Systems (GNSS). GNSS reside in medium to high orbital altitudes from the Earth’s surface, resulting in weak signal reception. However, user applications are increasingly in need of higher power signal strength or alternative PNT solutions. An influx of satellites in Low Earth Orbit (LEO) are driving such innovation in PNT technology. Improved signal strength compared to GNSS can be obtained from LEO satellites merely due to their proximity to Earth. Therefore, even communication satellite transmissions are becoming appealing to navigation, as so-called Signals of OPportunity (SOP). In order to benefit user applications, the receiver architectures for LEO-SOP, as well as potential LEO-PNT signals are explored.

The Global Navigation Satellite System (GNSS) software-defined receivers offer greater flexibility, cost-effectiveness, customization, and integration capabilities compared to traditional hardware-based receivers, making them essential for a wide range of applications. The continuous evolution of GNSS research and the availability of new features require these software-defined receivers to upgrade continuously to facilitate the latest requirements. The Finnish Geospatial Research Institute (FGI) has been supporting the GNSS research community with its open-source implementations, such as a MATLAB-based GNSS software-defined receiver `FGI-GSRx’ and a Python-based implementation `FGI-OSNMA’ for utilizing Galileo’s Open Service Navigation Message Authentication (OSNMA). In this context, longer datasets are crucial for GNSS software-defined receivers to support adaptation, optimization, and facilitate testing to investigate and develop future-proof receiver capabilities. In this paper, we present an updated version of FGI-GSRx, namely, FGI-GSRx-v2.0.0, which is also available as an open-source resource for the research community. FGI-GSRx-v2.0.0 offers improved performance as compared to its previous version, especially for the execution of long datasets. This is carried out by optimizing the receiver’s functionality and offering a newly added parallel processing feature to ensure faster capabilities to process the raw GNSS data. This paper also presents an analysis of some key design aspects of previous and current versions of FGI-GSRx for a better insight into the receiver’s functionalities. The results show that FGI-GSRx-v2.0.0 offers about a 40% run time execution improvement over FGI-GSRx-v1.0.0 in the case of the sequential processing mode and about a 59% improvement in the case of the parallel processing mode, with 17 GNSS satellites from GPS and Galileo. In addition, an attempt is made to execute v2.0.0 with MATLAB’s own parallel computing toolbox. A detailed performance comparison reveals an improvement of about 43% in execution time over the v2.0.0 parallel processing mode for the same GNSS scenario.

Neuroinflammation is a pathological event associated with many neurological disorders, including dementia and stroke. The choroid plexus (ChP) is a key structure in the ventricles of the brain that secretes cerebrospinal fluid (CSF), forms a blood-CSF barrier, and responds to disease conditions by recruiting immune cells and maintaining an immune microenvironment in the brain. Despite these critical roles, the exact structural and functional changes to the ChP over post-stroke time remain to be elucidated. We induced ischemic stroke in C57BL/6J mice via transient middle cerebral artery occlusion which led to reduction of cerebral blood flow and infarct stroke. At 1–7 days post-stroke, we detected time-dependent increase in the ChP blood-CSF barrier permeability to albumin, tight-junction damage, and dynamic changes of SPAK-NKCC1 protein complex, a key ion transport regulatory system for CSF production and clearance. A transient loss of SPAK protein complex but increased phosphorylation of the SPAK-NKCC1 complex was observed in both lateral ventricle ChPs. Most interestingly, stroke also triggered elevation of proinflammatory Lcn2 mRNA and its protein as well as infiltration of anti-inflammatory myeloid cells in ChP at day 5 post-stroke. These findings demonstrate that ischemic strokes cause significant damage to the ChP blood-CSF barrier, contributing to neuroinflammation in the subacute stage.

To determine head pose within an MR scanner using measurements of gradient-induced voltage from a multiple coil. A practical implementation of this approach would involve measuring the position-sensitive voltages induced by the imaging gradients rather than additional gradient pulses. This would speed up the model formation phase and increase the temporal resolution of position monitoring. We propose a novel method to measure the gradient-induced voltages in a set of five coils to estimate the position and orientations (POS) into MR scanner. A training set of measurements was first made and then Principal Component Analysis (PCA) was applied to the POS data estimated by using Statistical Parametric Mapping (SPM8), and the principal components were collected into a design matrix. Experimental data were collected and then the coefficients derived from the training set were used to estimate the POS changes and the results compared to movement parameters found using image co-registration. The changes in POS estimated from SPM8 co-registration and from the measured voltages for the phantom and the subject. The differences in the positions were estimated by using SPM8 and the model relating the change in induced voltages to position. The range and root-mean-square (RMS) amplitude of these differences for the phantom/subject data. The results suggest that it is possible to estimate the position and orientation with 0.22mm and 0.24° root-mean-square error using this set-up. The new approach could be used for prospective or retrospective motion correction.

Abstract Background The mechanisms underlying dysfunction of choroid plexus (ChP) blood–cerebrospinal fluid (CSF) barrier and lymphocyte invasion in neuroinflammatory responses to stroke are not well understood. In this study, we investigated whether stroke damaged the blood–CSF barrier integrity due to dysregulation of major ChP ion transport system, Na+–K+–Cl− cotransporter 1 (NKCC1), and regulatory Ste20-related proline-alanine-rich kinase (SPAK). Methods Sham or ischemic stroke was induced in C57Bl/6J mice. Changes on the SPAK–NKCC1 complex and tight junction proteins (TJs) in the ChP were quantified by immunofluorescence staining and immunoblotting. Immune cell infiltration in the ChP was assessed by flow cytometry and immunostaining. Cultured ChP epithelium cells (CPECs) and cortical neurons were used to evaluate H2O2-mediated oxidative stress in stimulating the SPAK–NKCC1 complex and cellular damage. In vivo or in vitro pharmacological blockade of the ChP SPAK–NKCC1 cascade with SPAK inhibitor ZT-1a or NKCC1 inhibitor bumetanide were examined. Results Ischemic stroke stimulated activation of the CPECs apical membrane SPAK–NKCC1 complex, NF-κB, and MMP9, which was associated with loss of the blood–CSF barrier integrity and increased immune cell infiltration into the ChP. Oxidative stress directly activated the SPAK–NKCC1 pathway and resulted in apoptosis, neurodegeneration, and NKCC1-mediated ion influx. Pharmacological blockade of the SPAK–NKCC1 pathway protected the ChP barrier integrity, attenuated ChP immune cell infiltration or neuronal death. Conclusion Stroke-induced pathological stimulation of the SPAK–NKCC1 cascade caused CPECs damage and disruption of TJs at the blood–CSF barrier. The ChP SPAK–NKCC1 complex emerged as a therapeutic target for attenuating ChP dysfunction and lymphocyte invasion after stroke.

More and more satellites are populating the sky nowadays in the Low Earth orbits (LEO). Most of the targeted applications are related to broadband and narrowband communications, Earth observation, synthetic aperture radar, and internet-of-Things (IoT) connectivity. In addition to these targeted applications, there is yet-to-be-harnessed potential for LEO and positioning, navigation, and timing (PNT) systems, or what is nowadays referred to as LEO-PNT. No commercial LEO-PNT solutions currently exist and there is no unified research on LEO-PNT concepts. Our survey aims to fill the gaps in knowledge regarding what a LEO-PNT system entails, its technical design steps and challenges, what physical layer parameters are viable solutions, what tools can be used for a LEO-PNT design (e.g., optimisation steps, hardware and software simulators, etc.), the existing models of wireless channels for satellite-to-ground and ground-to-satellite propagation, and the commercial prospects of a future LEO-PNT system. A comprehensive and multidisciplinary survey is provided by a team of authors with complementary expertise in wireless communications, signal processing, navigation and tracking, physics, machine learning, Earth observation, remote sensing, digital economy, and business models.

Nonsteroidal anti-inflammatory drugs (NSAIDs) are commonly used as analgesics, antipyretics, anti-clotting, and anti-inflammatory agents. One such NSAID, ketoprofen (KTP), is prescribed to treat pain, sprains, menstrual irregularities, osteoarthritis (OA), and rheumatoid arthritis (RA). By inhibiting cyclooxygenase, it halts prostaglandin synthesis. However, it exhibits significant side effects in humans and other living things. Herein, the chemical structure of KTP has been modified to search for improved chemical and medicinal effects. Density functional theory (DFT) has been employed for geometry optimization. Molecular docking and molecular dynamics (MD) simulations were conducted to investigate the binding affinity and stability of the protein-drug complex with prostaglandin synthase protein. ADMET predictions were carried out to calculate and compare the pharmacokinetic behavior of KTP and its new analogs. Spectral and chemical results support the structural conformation of the new analogs, all of which exhibit improved physicochemical, and binding properties compared to the parent drug. After a 100 ns MD simulation, the drugs remained tightly bound to the binding site of the receptor protein, showing significant interactions with shorter distances among the new analogs. The ADMET predictions confirmed better pharmacokinetic properties of novel analogs compared to KTP. The study compares several KTP metabolites using thermo-chemical, spectral, biological, and toxicological analysis. Results showed the reactivity and stability, with spectral data evaluating all KTP analogs. ADMET predicts that all KTP derivatives are less toxic and have better pharmacokinetic features than KTP. Most KTP analogs exhibit excellent oral bioavailability and significant analgesic, antipyretic, and anti-inflammatory functions. These findings support the potential use of these drugs in the future drugs.

Apyrase (APY) is a nucleoside triphosphate (NTP) diphosphohydrolase (NTPDase) which is a member of the superfamily of guanosine diphosphatase 1 (GDA1)-cluster of differentiation 39 (CD39) nucleoside phosphatase. Under various circumstances like stress, cell growth, the extracellular adenosine triphosphate (eATP) level increases, causing a detrimental influence on cells such as cell growth retardation, ROS production, NO burst, and apoptosis. Apyrase hydrolyses eATP accumulated in the extracellular membrane during stress, wounds, into adenosine diphosphate (ADP) and adenosine monophosphate (AMP) and regulates the stress-responsive pathway in plants. This study was designed for the identification, characterization, and for analysis of APY gene expression in Oryza sativa. This investigation discovered nine APYs in rice, including both endo- and ecto-apyrase. According to duplication event analysis, in the evolution of OsAPYs, a significant role is performed by segmental duplication. Their role in stress control, hormonal responsiveness, and the development of cells is supported by the corresponding cis-elements present in their promoter regions. According to expression profiling by RNA-seq data, the genes were expressed in various tissues. Upon exposure to a variety of biotic as well as abiotic stimuli, including anoxia, drought, submergence, alkali, heat, dehydration, salt, and cold, they showed a differential expression pattern. The expression analysis from the RT-qPCR data also showed expression under various abiotic stress conditions, comprising cold, salinity, cadmium, drought, submergence, and especially heat stress. This finding will pave the way for future in-vivo analysis, unveil the molecular mechanisms of APY genes in stress response, and contribute to the development of stress-tolerant rice varieties.

Abstract Background Chronic cerebral hypoperfusion (CCH) causes white matter damage and cognitive impairment, in which astrogliosis is the major pathology. However, underlying cellular mechanisms are not well defined. Activation of Na+/H+ exchanger-1 (NHE1) in reactive astrocytes causes astrocytic hypertrophy and swelling. In this study, we examined the role of NHE1 protein in astrogliosis, white matter demyelination, and cognitive function in a murine CCH model with bilateral carotid artery stenosis (BCAS). Methods Sham, BCAS, or BCAS mice receiving vehicle or a selective NHE1 inhibitor HOE642 were monitored for changes of the regional cerebral blood flow and behavioral performance for 28 days. Ex vivo MRI-DTI was subsequently conducted to detect brain injury and demyelination. Astrogliosis and demyelination were further examined by immunofluorescence staining. Astrocytic transcriptional profiles were analyzed with bulk RNA-sequencing and RT-qPCR. Results Chronic cerebral blood flow reduction and spatial working memory deficits were detected in the BCAS mice, along with significantly reduced mean fractional anisotropy (FA) values in the corpus callosum, external capsule, and hippocampus in MRI DTI analysis. Compared with the sham control mice, the BCAS mice displayed demyelination and axonal damage and increased GFAP+ astrocytes and Iba1+ microglia. Pharmacological inhibition of NHE1 protein with its inhibitor HOE642 prevented the BCAS-induced gliosis, damage of white matter tracts and hippocampus, and significantly improved cognitive performance. Transcriptome and immunostaining analysis further revealed that NHE1 inhibition specifically attenuated pro-inflammatory pathways and NADPH oxidase activation. Conclusion Our study demonstrates that NHE1 protein is involved in astrogliosis with pro-inflammatory transformation induced by CCH, and its blockade has potentials for reducing astrogliosis, demyelination, and cognitive impairment.

The properties of cyclohexane (CHex) and plasma polymerized cyclohexane (PPCHex) thin films are studied, and the results are compared with those obtained using a computing model (under the density functional theory framework). Quantum chemical calculations are performed to calculate their molecular electrostatic potential (MEP), highest occupied molecular orbital (HOMO) and lowest unoccupied molecular orbital (LUMO) gaps, thermodynamic stability, chemical reactivity, and vibrational properties. The HOMO–LUMO energy gap sharply decreased in the case of the PPCHex thin film compared to that of CHex. Gibbs free energy shows a similar phenomenon where the enthalpy of PPCHex (formation) is almost six times higher than that of CHex. The computed FTIR spectra indicate good agreement with the experimental results. RMS roughness shows a smooth surface, and it is compared with the FESEM image. Moreover, the dielectric constant and electrical conductivity of the PPCHex thin films are also studied.

It is essential to develop high salt-tolerant rice varieties in order to cultivate the salt-affected lands. In this study, Na+/H+ exchanger 1 (NHX1) gene isolated from Vigna radiata L. Wilczek was transferred in Bangladesh Rice Research Institute (BRRI) developed two indica rice genotypes BRRI Dhan28 and BRRI Dhan29 using in-planta approach for improvement of salinity tolerance. Embryonic axes of matured dehusked rice seeds were injured and co-cultivated with Agrobacterium strain harboring VrNHX1 gene and finally regenerated. GUS histochemical assay and PCR amplification of GUS-a and VrNHX1 were performed to confirm the transformation. Expression confirmation was done by semi-quantitative RT-PCR. Under salinity stress, transgenic lines showed higher chlorophyll, relative water content and decreased electrolyte leakage, proline content, lipid peroxidation level, and catalase enzyme activity which represent the better physiology than control plants. Moreover, under salinity stress (150 mM), transgenic lines exhibited superior growth and salt tolerant than non-transgenic plants.

Background: Over 700,000 Myanmar nationals known as the ‘Rohingyas’ fled into Cox’s Bazar, Bangladesh, in late 2017. Due to this huge displacement into unhygienic areas, these people became vulnerable to communicable diseases including cholera. Assessing the risk, the Government of Bangladesh (GoB), with the help of the International Centre for Diarrhoeal Disease Research, Bangladesh (ICDDR,B) and other international partners, decided to take preventive measures, one of which is the execution of oral cholera vaccination (OCV) campaigns. This paper describes the implementation and delivery of OCV campaigns during humanitarian crises in Bangladesh. Methods: Seven rounds of OCV campaigns were conducted between October 2017 and December 2021. The OCV campaigns were conducted by applying different strategies. Results: Approximately 900,000 Rohingya Myanmar nationals (RMNs) and the host population (amounting to 528,297) received OCV across seven campaigns. In total, 4,661,187 doses of OCVs were administered, which included 765,499 doses for RMNs, and 895,688 doses for the host community. The vaccine was well accepted, and as a result, a high level of coverage was achieved, ranging from 87% to 108% in different campaigns. Conclusions: After successful pre-emptive campaigns in Cox’s Bazar humanitarian camps, no cholera outbreaks were detected either in the RMN or host communities.

Abstract In this work, a fully integrated active microfluidic device transforming a conventional 96-well kit into point-of-care testing (POCT) device was implemented to improve the performance of traditional enzyme-linked immunosorbent assay (ELISA). ELISA test by the conventional method often requires the collection of 96 samples for its operation as well as longer incubation time from hours to overnight, whereas our proposed device conducts ELISA immediately individualizing a 96-well for individual patients. To do that, a programmable and disposable on-chip pump and valve were integrated on the device for precise control and actuation of microfluidic reagents, which regulated a reaction time and reagent volume to support the optimized protocols of ELISA. Due to the on-chip pump and valve, ELISA could be executed with reduced consumption of reagents and shortening the assay time, which are crucial for conventional ELISA using 96-well microplate. To demonstrate highly sensitive detection and easy-to-use operation, this unconventional device was successfully applied for the quantification of cardiac troponin I (cTnI) of 4.88 pg/mL using a minimum sample volume of 30 µL with a shorter assay time of 15 min for each ELISA step. The limit of detection (LOD) thus obtained was significantly improved than the conventional 96-well platform.

Datura inoxia is a subshrub plant known for its toxicity which results from the presence of the tropane alkaloid scopolamine and hyoscyamine. Saccharides are one of the most important elicitors that can alter physiological and biochemical responses in plants. This study targeted to increase the production of secondary metabolites in Datura inoxia avoiding genetic alteration and processes using different nontoxic and biodegradable compounds, utilizing calli induced from Datura inoxia stems to observe the effects of mannitol and sorbitol on the production of the two major secondary metabolites, scopolamine and hyoscyamine. Methanolic extract of the whole plant and callus was used to examine the production of two secondary metabolites in Datura inoxia using HPLC in a qualitative and quantitative manner which revealed the increased production of scopolamine and hyoscyamine in calli. The addition of mannitol and sorbitol in the media had a negative effect on both the fresh and dry weight of the calli but production of scopolamine and hyoscyamine increased significantly. In-vitro anti-microbial assay of hyoscyamine against Escherichia coli ATCC25922 and Candida albicans resulted in total inhibition of both the microbes in concentrations as low as 200 µg/ml.

Carbon capture and storage (CCS) is an inevitable action to achieve CO2 emission reduction targets including becoming net-zero by 2050. Increased efforts are therefore required to identify suitable locations for large-scale CO2 storage. In addition to large aquifers, shut down oil and gas fields in the North Sea are logical candidates for offshore large-scale CO2 storage because of their proven storage capacity, reliable caprock integrity, established infrastructure, and public acceptance. However, in some cases, old and legacy wells are subject to high uncertainties in their integrity, and they can compromise CO2 containment in such reservoirs. On the Norwegian Continental Shelf (NCS), such wells are numerous even outside of oil and gas production areas, i.e., legacy wells affecting aquifers. Therefore, there is a clear need for reliable and cost-effective technologies for well integrity evaluation and remediation. This paper discusses a workflow for screening, monitoring, and remediation of legacy wells. In a first stage, the screening of the Horda Platform areas suggested the need for integrity investigation for the exploration well 32/4-1 T2, drilled into the Alpha structure of the Smeaheia fault block if CO2 is stored in the structure. Our initial well screening of drilling documentation indicates that the well is not suitable to be reused for CO2 injection and geophysical monitoring is recommended. In a second stage, a numerical representation of the well architecture is built including realistic geological setting. We evaluate the sensitivity of non-invasive low-frequency electromagnetic monitoring to corrosion levels in the casing. Numerical end-member simulations of assuming casing corrosion of different degrees by changing material conductivity are performed. Results comparing different corrosion scenarios with a base case (no corrosion) give an above noise signal at receiver locations enabling to separate the different cases. Comparison of the gained electrical fields at seafloor suggests that well casing corrosion monitoring should be possible. Finally, the electrochemical deposition potential of the Sognefjord Formation water is analyzed, revealing depositional potential for portlandite, which might be useful for cement remediation. We recommend such an analysis for all legacy wells penetrating candidate reservoirs for future CO2 or hydrogen storage.

Abstract In soybean breeding program, continuous selection pressure on traits response to yield created a genetic bottleneck for improvements of soybean through hybridization breeding technique. Therefore an initiative was taken to developed high yielding soybean variety applying mutation breeding techniques at Plant Breeding Division, Bangladesh Institute of Nuclear Agriculture (BINA), Bangladesh. Locally available popular cultivar BARI Soybean-5 was used as a parent material and subjected to five different doses of Gamma ray using Co60. In respect to seed yield and yield attributing characters, twelve true breed mutants were selected from M4 generation. High values of heritability and genetic advance with high genotypic coefficient of variance (GCV) for plant height, branch number and pod number were considered as favorable attributes for soybean improvement that ensure expected yield. The mutant SBM-18 obtained from 250Gy provided stable yield performance at diversified environments. It provided maximum seed yield of 3056 kg ha-1 with highest number of pods plant-1 (56). The National Seed Board of Bangladesh (NSB) eventually approved SBM-18 and registered it as a new soybean variety named ‘Binasoybean-5’ for large-scale planting because of its superior stability in various agro-ecological zones and consistent yield performance.

Deep learning-based automatic classification of breast tumors using parametric imaging techniques from ultrasound (US) B-mode images is still an exciting research area. The Rician inverse Gaussian (RiIG) distribution is currently emerging as an appropriate example of statistical modeling. This study presents a new approach of correlated-weighted contourlet-transformed RiIG (CWCtr-RiIG) and curvelet-transformed RiIG (CWCrv-RiIG) image-based deep convolutional neural network (CNN) architecture for breast tumor classification from B-mode ultrasound images. A comparative study with other statistical models, such as Nakagami and normal inverse Gaussian (NIG) distributions, is also experienced here. The weighted entitled here is for weighting the contourlet and curvelet sub-band coefficient images by correlation with their corresponding RiIG statistically modeled images. By taking into account three freely accessible datasets (Mendeley, UDIAT, and BUSI), it is demonstrated that the proposed approach can provide more than 98 percent accuracy, sensitivity, specificity, NPV, and PPV values using the CWCtr-RiIG images. On the same datasets, the suggested method offers superior classification performance to several other existing strategies.

Laboratory tests were performed on the Draupne shale formation, which may serve as a seal over CO2 storage sites. Different techniques were used to assess the integrity and mechanical properties of the shale, with the main objective of investigating whether exposure to CO2 would in any manner alter these properties. The laboratory methods used encompass traditional triaxial tests; however, with fluid substitution prior to increasing axial stress to failure. These tests were conducted on smaller cylindrical plugs than standard, taking advantage of the finer grained nature of the shale. Another set of experiments used the low-frequency technique, whereby small amplitude, cyclic axial strains are applied on the specimen, allowing a direct measurement of stiffness. Long exposure, with change of fluid from brine to CO2, allowed for quantifying small changes in stiffness, thanks to the many repeated cycles of non-destructive testing. In a final experimental technique, the punch test, shear strength of the same material was obtained by cutting a central disk from a larger intact shale disk, while measuring the shear force needed to perform the cut.

Structural, electronic, optical, and thermal properties of undoped and metal, M (Ni, Cu, and Zn), doped cadmium iodide (CdI2) were studied using a generalized gradient approximation of density functional theory. Lattice constants found from the theoretical studies of the structure of the undoped and doped CdI2 are in good agreement with those found in the available experimental and theoretical investigations. A strong mixing of I 5p and M 3d states is found after the doping of 3d M in CdI2, which alters the bandgap from positive to negative. Among all M (Ni, Cu, and Zn), Ni doped CdI2 with a narrow negative bandgap evolve the quantum dot level. Due to interactions between the Cd 4d and M 3d states, the measured optical and thermal properties of the doped system assessed with pure CdI2 indicate unusual behaviors, which suggest that the material can be used in different nano-electronic and electrochemical applications and in biological levels such as detection of COVID-19 pathogens.
