The condition known as scrotal elephantiasis is a debilitating rarity where the scrotum undergoes significant swelling, typically arising from chronic obstructive lymphedema. A case is presented of a 60-year-old man who had suffered severe and persistent enlargement of his scrotum for multiple years. This affliction greatly affected both his ability to function effectively and caused detrimental psychosocial consequences.After a clinical evaluation, the patient was diagnosed with scrotal elephantiasis.A comprehensive strategy was adopted, utilizing scrotal volume reduction surgery to yield a significant enhancement in the quality of life following the operation.

This study investigates the hydrochemical formation mechanism of shallow groundwater in the Upper Kebir upstream sub-basin (Northeastern Algeria). The objective is to evaluate water quality suitability for domestic purposes through the application of water quality index (WQI). A total of 24 water points (wells and borewells) evenly distributed in the basin were collected and analyzed in the laboratory for determining the major ions and other geochemical parameters in the groundwater. The groundwater hydrochemical types were identified as Cl–Na and Cl–HCO3 –Na, with the dominant major ions were found in the order of Na+ > Ca2+ > Mg2+ for cations, and Cl− > SO42− > HCO3– > NO3− for anions. Results suggest that weathering, dissolution of carbonate, sulfate, salt rocks, and anthropogenic activities were the major contributors to ion content in the groundwater. The Water Quality Index (WQI) was calculated to assess the water quality of potable water. Approximately 50% of the sampled sites exhibited good water quality. However, the study highlights significant NO3 contamination in the study area, with 50% of samples exceeding permissible limits. Therefore, effective treatment measures are crucial for the safe consumption of groundwater.

We prove the existence and uniqueness of renormalized solution in anisotropic Sobolev spaces with data belonging to L1 -data, based on the properties of the renormalized solutions and the generalized monotonicity method in the functional spaces.

Résumé : Certains textes littéraires de langue française –surtout contemporain- (ou francophones comme préfèrent les qualifier certains critiques et lecteurs avertis) échappent incontestablement aux codes standards régissant l’univers romanesque. Et les outils littéraires conventionnels ne permettent pas vraiment l’analyse et/ou l’interprétation de ces œuvres insaisissables dans le temps et même dans l’espace. Le recours donc aux « neurosciences », tout particulièrement à certains concepts et notions, s’avère incontournable pour mieux lire et bien comprendre ces nouvelles formes de création littéraire. Les tous derniers romans de Kaouther Adimi comme « Au vent mauvais » illustrent parfaitement cette littérature de l’extrême contemporain, cette littérature innovante pas du tout facile à autopsier suivant un protocole de lecture dit traditionnel. Abstract Mycobacterium tuberculosis (MTB) is a pathogenic bacterium accountable for 10.6 million new infections with tuberculosis (TB) in 2021. The fact that the genetic sequences of M. tuberculosis vary widely provides a basis for understanding how this bacterium causes disease, how the immune system responds to it, how it has evolved over time, and how it is distributed geographically. However, despite extensive research efforts, the evolution and transmission of MTB in Africa remain poorly understood. In this study, we used 17,641 strains from 26 countries to create the first curated African Mycobacterium tuberculosis (MTB) classification and resistance dataset, containing 13,753 strains. We identified 157 mutations in 12 genes associated with resistance and additional new mutations potentially associated with resistance. The resistance profile was used to classify strains. We also performed a phylogenetic classification of each isolate and prepared the data in a format that can be used for phylogenetic and comparative analysis of tuberculosis worldwide. These genomic data will extend current information for comparative genomic studies to understand the mechanisms and evolution of MTB drug resistance.

RIPK1 is a key regulator of innate immune signalling pathways. To ensure an optimal inflammatory response, RIPK1 is regulated post-translationally by well-characterized ubiquitylation and phosphorylation events, as well as by caspase-8-mediated cleavage1-7. The physiological relevance of this cleavage event remains unclear, although it is thought to inhibit activation of RIPK3 and necroptosis8. Here we show that the heterozygous missense mutations D324N, D324H and D324Y prevent caspase cleavage of RIPK1 in humans and result in an early-onset periodic fever syndrome and severe intermittent lymphadenopathy-a condition we term 'cleavage-resistant RIPK1-induced autoinflammatory syndrome'. To define the mechanism for this disease, we generated a cleavage-resistant Ripk1D325A mutant mouse strain. Whereas Ripk1-/- mice died postnatally from systemic inflammation, Ripk1D325A/D325A mice died during embryogenesis. Embryonic lethality was completely prevented by the combined loss of Casp8 and Ripk3, but not by loss of Ripk3 or Mlkl alone. Loss of RIPK1 kinase activity also prevented Ripk1D325A/D325A embryonic lethality, although the mice died before weaning from multi-organ inflammation in a RIPK3-dependent manner. Consistently, Ripk1D325A/D325A and Ripk1D325A/+ cells were hypersensitive to RIPK3-dependent TNF-induced apoptosis and necroptosis. Heterozygous Ripk1D325A/+ mice were viable and grossly normal, but were hyper-responsive to inflammatory stimuli in vivo. Our results demonstrate the importance of caspase-mediated RIPK1 cleavage during embryonic development and show that caspase cleavage of RIPK1 not only inhibits necroptosis but also maintains inflammatory homeostasis throughout life.

Abstract Cell death mechanisms in T lymphocytes vary according to their developmental stage, cell subset and activation status. The cell death control mechanisms of mucosal-associated invariant T (MAIT) cells, a specialized T cell population, are largely unknown. Here we report that MAIT cells express key necroptotic machinery; receptor-interacting protein kinase 3 (RIPK3) and mixed lineage kinase domain-like (MLKL) protein, in abundance. Despite this, we discovered that the loss of RIPK3, but not necroptotic effector MLKL or apoptotic caspase-8, specifically increased MAIT cell abundance at steady-state in the thymus, spleen, liver and lungs, in a cell-intrinsic manner. In contrast, over the course of infection with Francisella tularensis, RIPK3 deficiency did not impact the magnitude of the expansion nor contraction of MAIT cell pools. These findings suggest that, distinct from conventional T cells, the accumulation of MAIT cells is restrained by RIPK3 signalling, likely prior to thymic egress, in a manner independent of canonical apoptotic and necroptotic cell death pathways.

Background: Wilson Disease (WD) is an autosomal recessive inherited metabolic disease caused by mutations in the ATP7B gene. WD is characterized by heterogeneous clinical presentations expressed by hepatic and neuropsychiatric phenotypes. The disease is difficult to diagnose, and misdiagnosed cases are commonly seen. Methods: In this study, the presented symptoms of WD, the biochemical parameters as well as its natural history are described based on cases collected in Mohammed VI Hospital University of Marrakech (Morocco). We screened and sequenced 21 exons of ATP7B gene from 12 WD patients that confirmed through biochemical diagnosis. Results: Mutational assessment of the ATP7B gene showed six homozygous mutations in 12 individuals however, 2 patients had no evidence of any mutation in promoter and exonic regions. All mutations are pathogenic and most were missense mutations. c.2507G > A (p.G836E), c.3694A > C (p.T1232P) and c.3310 T > C (p.C1104R) that were identified in 4 patients. The other mutations were a non-sense mutation (c.865C > T (p.C1104R)) detected in 2 patients, a splice mutation (c.51 + 4A > T) detected in 2 patients and a frameshift mutation (c.1746 dup (p.E583Rfs*25) detected in 2 patients. Conclusion: Our study is the first molecular analysis in Moroccan patients with Wilson's disease, the ATP7B mutational spectrum in the Moroccan population is diverse and still unexplored.

The fennel is a typical plant of the Mediterranean region used in traditional pharmacopoeia and as an aromatic spice. This study was interested in an evaluation of the physical, physicochemical and biochemical criteria and the biological activities of the seeds and seeds oils of local (beldi) fennel produced at four stages of maturity in four terroirs of Essaouira province (Central West, Morocco), compared to those of a local fennel produced in a neighboring terroir and an imported fennel. Seeds of different maturity stages and geographic origins were clearly differentiated into homogeneous groups according to studied criteria. The water activity of these seeds is between 0.43 and 0.51 with an average ash content of 6.24 g/100 g of dry matter (DM) containing fairly high contents of potassium, calcium, sodium and baryum. Seed total polyphenols recorded interesting levels ranging between 623.64 and 959.24 mg gallic acid equivalent/100 g DM. The antioxidant activity is estimated between 23.53 and 43.83 mg ascorbic acid equivalent/g DM. Yields of vegetable and essential oils varied from 8.55 to 16.28 % and from 1.4 to 2.7 %, respectively. However, the high values recorded for free acidity, peroxide index and specific extinction of vegetable oil indicate that it cannot be considered as edible oil. Concerning the essential oil, significant increases in the aroma compounds s-limonene and fenchone were noted during maturity. Besides, the antibacterial activity tests have shown that Staphylococcus aureus is sensitive to the action of this essential oil. Moreover, the quality criteria of the studied fennel seeds and seed oils showed the influence of the geographical origin. These results would make it possible to propose ways of valorizing, quality control and labeling of beldi fennel under a distinctive sign of origin and quality.

The tribological performance of the tungsten carbide substrate (WC‐Co), improved by ceramic coatings, is still being reported in new studies that have been carried out to date. It has become a hot research topic that are widely applied in hard material research, especially in the tools manufacturing fields. This study was conducted to investigate the wear characteristics of a commercial cemented carbide tool (WC‐Co) coated with a physical vapor deposition chromium‐vanadium nitride film (CrVN), followed by a boriding process as a final thermochemical treatment. Tested in dry sliding contact against an alumina ball as a static partner, the tribological responses of the specimen were analyzed and compared with an uncoated specimen. Friction coefficients, calculated from volume loss, were around.58 for all specimens except the uncoated specimen at 10 N of applied load. Wear scar analyses revealed the occurrence of several wear mechanisms that is polishing, oxidation, wear debris formation, surface binder removal, grain fragmenting, and grain pull‐out. Summary: Pathogen recognition and TNF receptors signal via receptor interacting serine/threonine kinase-3 (RIPK3) to cause cell death, including MLKL-mediated necroptosis and caspase-8-dependent apoptosis. However, the post-translational control of RIPK3 is not fully understood. Using mass-spectrometry, we identified that RIPK3 is ubiquitylated on K469. The expression of mutant RIPK3 K469R demonstrated that RIPK3 ubiquitylation can limit both RIPK3-mediated apoptosis and necroptosis. The enhanced cell death of overexpressed RIPK3 K469R and activated endogenous RIPK3 correlated with an overall increase in RIPK3 ubiquitylation. Ripk3K469R/K469R mice challenged with Salmonella displayed enhanced bacterial loads and reduced serum IFNγ. However, Ripk3K469R/K469R macrophages and dermal fibroblasts were not sensitized to RIPK3-mediated apoptotic or necroptotic signaling suggesting that, in these cells, there is functional redundancy with alternate RIPK3 ubiquitin-modified sites. Consistent with this idea, the mutation of other ubiquitylated RIPK3 residues also increased RIPK3 hyper-ubiquitylation and cell death. Therefore, the targeted ubiquitylation of RIPK3 may act as either a brake or accelerator of RIPK3-dependent killing.

This study aims to estimate the solute transport parameters in saturated porous media using a hybrid algorithm. In this study, the Physical Non-Equilibrium (PNE) model was used to describe the transport of solutes in porous media. A numerical solution for the PNE model is obtained using the Finite Volume Method (FVM) based on the Ttri-Diagonal Matrix Algorithm (TDMA). The developed program, written in Matlab, is capable to solve the advection-dispersion (ADE) and the PNE equations for the mobile -immobile (MIM)model with linear sorption isotherm. The Solute transport parameters, (immobile water content, mass transfer coefficient, and dispersion coefficient), are estimated using different algorithms such as the Levenberg-Marquardt algorithm (LM), genetic algorithm (GA), simulated annealing algorithm (SA). To overcome the limitations of deterministic optimization models which are rather unstable and divergent around a local minimum, a hybrid algorithm (GA+LM, SA+LM) permits to estimate of the solute transport parameters. Numerical solutions are verified using the experiments conducted by Semra (2003) which are about the transport of toluene through a column composed of impregnated Chromosorb grains at ambient temperature (20 °C) for three flow rates (1, 2 and 5ml/min). The results show that the hybrid algorithm (GA+LM, SA+LM) is more accurate than others (GA, SA, and LM). Comparing to the ADE model, The PNE with linear isotherm model gives a better description to the BeakThrough Curves (BTCs) with higher values of determination coefficient (R2) and lower values of Root Mean Square Error (RMSE). Also, the solute transport parameters tended to vary with the flow rate.

In this paper, we propose and analyze a mathematical model with stochastic perturbation of the issue of unemployment by considering three main variables. Namely graduates or individuals who have reached the age of employment, salaried individuals and employees, and then individuals who have been made redundant or who have resigned from work. After the description of the proposed model, we study the existence and the uniqueness of the nonnegative solution. Furthermore, we investigate the asymptotic behavior of this solution by analyzing the stability of the system at the equilibrium points. Abstract Necroptosis is a pro-inflammatory cell death program executed by the terminal effector, mixed lineage kinase domain-like (MLKL). Previous studies suggested a role for the necroptotic machinery in platelets, where loss of MLKL or its upstream regulator, RIPK3 kinase, impacted thrombosis and haemostasis. However, it remains unknown whether necroptosis operates within megakaryocytes, the progenitors of platelets, and whether necroptotic cell death might contribute to or diminish platelet production. Here, we demonstrate that megakaryocytes possess a functional necroptosis signalling cascade. Necroptosis activation leads to phosphorylation of MLKL, loss of viability and cell swelling. Analyses at steady state and post antibody-mediated thrombocytopenia revealed that platelet production was normal in the absence of MLKL, however, platelet activation and haemostasis were impaired with prolonged tail re-bleeding times. We conclude that MLKL plays a role in regulating platelet function and haemostasis and that necroptosis signalling in megakaryocytes is dispensable for platelet production.

Mixed lineage kinase domain-like (MLKL) is the terminal protein in the pro-inflammatory necroptotic cell death program. Here the authors show that MLKL trafficking and plasma membrane accumulation are crucial necroptosis checkpoints, and that accumulation of phosphorylated MLKL at intercellular junctions promotes necroptosis.
