The main objective of the National Project for Research and Incidence of Childhood Leukemias is to reduce early mortality rates for these neoplasms in the vulnerable regions of Mexico. This project was conducted in the states of Oaxaca, Puebla, and Tlaxcala. A key strategy of the project is the implementation of an effective roadmap to ensure that leukemia patients are the target of maximum benefit of interdisciplinary collaboration between researchers, clinicians, surveyors, and laboratories. This strategy guarantees the comprehensive management of diagnosis and follow-up samples of pediatric patients with leukemia, centralizing, managing, and analyzing the information collected. Additionally, it allows for a precise diagnosis and monitoring of the disease through immunophenotype and measurable residual disease (MRD) studies, enhancing research and supporting informed clinical decisions for the first time in these regions through a population-based study. This initiative has significantly improved the diagnostic capacity of leukemia in girls, boys, and adolescents in the regions of Oaxaca, Puebla, and Tlaxcala, providing comprehensive, high-quality care with full coverage in the region. Likewise, it has strengthened collaboration between health institutions, researchers, and professionals in the sector, which contributes to reducing the impact of the disease on the community.

Abstract Background The identification of critically ill COVID-19 patients at risk of fatal outcomes remains a challenge. Here, we first validated candidate microRNAs (miRNAs) as biomarkers for clinical decision-making in critically ill patients. Second, we constructed a blood miRNA classifier for the early prediction of adverse outcomes in the ICU. Forests are critical in the terrestrial carbon cycle, and the knowledge of their response to ongoing climate change will be crucial for determining future carbon fluxes and climate trajectories. In areas with contrasting seasons, trees form discrete annual rings that can be assigned to calendar years, allowing to extract valuable information about how trees respond to the environment. The anatomical structure of wood provides highly-resolved information about the reaction and adaptation of trees to climate. Quantitative wood anatomy helps to retrieve this information by measuring wood at the cellular level using high-resolution images of wood micro-sections. However, whereas large advances have been made in identifying cellular structures, obtaining meaningful cellular information is still hampered by the correct annual tree ring delimitation on the images. This is a time-consuming task that requires experienced operators to manually delimit ring boundaries. Classic methods of automatic segmentation based on pixel values are being replaced by new approaches using neural networks which are capable of distinguishing structures, even when demarcations require a high level of expertise. Although neural networks have been used for tree ring segmentation on macroscopic images of wood, the complexity of cell patterns in stained microsections of broadleaved species requires adaptive models to accurately accomplish this task. We present an automatic tree ring boundary delineation using neural networks on stained cross-sectional microsection images from beech cores. We trained a UNETR, a combined neural network of UNET and the attention mechanisms of Visual Transformers, to automatically segment annual ring boundaries. Its accuracy was evaluated considering discrepancies with manual segmentation and the consequences of disparity for the goals of quantitative wood anatomy analyses. In most cases (91.8%), automatic segmentation matched or improved manual segmentation, and the rate of vessels assignment to annual rings was similar between the two categories, even when manual segmentation was considered better. The application of convolutional neural networks-based models outperforms human operator segmentations when confronting ring boundary delimitation using specific parameters for quantitative wood anatomy analysis. Current advances on segmentation models may reduce the cost of massive and accurate data collection for quantitative wood anatomy.

There is a limited understanding of the pathophysiology of postacute pulmonary sequelae in severe COVID-19. The aim of current study was to define the circulating microRNA (miRNA) profiles associated with pulmonary function and radiologic features in survivors of SARS-CoV-2-induced ARDS. The study included patients who developed ARDS secondary to SARS-CoV-2 infection (n = 167) and a group of infected patients who did not develop ARDS (n = 33). Patients were evaluated 3 months after hospital discharge. The follow-up included a complete pulmonary evaluation and chest computed tomography. Plasma miRNA profiling was performed using RT-qPCR. Background: Mesoamerica is one of the most important biodiversity hotspots on the planet. Despite significant efforts made over two centuries to contribute to the floristic knowledge of this region, our understanding of its flora is still scattered and uneven. Questions: What is the magnitude of the vascular plant species richness in the Usumacinta River Basin? Study site and dates: Usumacinta River Basin (Guatemala and Mexico), 1838-2018. Methods: We compiled the checklist by systematizing the floristic information acquired from various sources derived from numerous floristic and ecological studies. Results: We recorded 6,977 species, 1,892 genera and 274 families. The largest numbers of species (5,746) and records (58,859) correspond to the Mexican portion of the Usumacinta River Basin, compared to its Guatemalan counterpart (4,445 species and 19,952 records). The most species-rich families were Orchidaceae (598 species), Fabaceae (512), and Asteraceae (476). The prevalence of these and all other families with significant contributions to the flora varied among three elevation-defined sectors into which the Usumacinta River Basin was subdivided (lower, middle, and upper basin). Conclusions: The Usumacinta River Basin is a strategic region for plant biodiversity conservation as it hosts almost one third of all vascular plant species known for Mesoamerica and ca. 6 % of the entire flora in the Americas. Further botanical exploration should focus on those areas of the basin for which little or no information is available in order to gain a better appreciation of its flora.

Laurel forests are quite relevant for biodiversity conservation and are among the island ecosystems most severely damaged by human activities. In the past, Canary laurel forests have been greatly altered by logging, livestock and agriculture. The remains of laurel forests are currently protected in the Canary Islands (Spain). However, we miss basic information needed for their restoration and adaptive management, such as tree longevity, growth potential and responsiveness to natural and anthropogenic disturbances. Using dendrochronological methods, we studied how forest dynamic is related to land-use change and windstorms in two well-preserved laurel forests on Tenerife Island. Wood cores were collected from over 80 trees per stand at three stands per forest. We used ring-width series to estimate tree ages and calculate annual basal area increments (BAI), cumulative diameter increases, and changes indicative of released and suppressed growth. Twelve tree species were found in all stands, with Laurus novocanariensis, Ilex canariensis and Morella faya being the most common species. Although some individuals were over 100 years old, 61.8%–88.9% of the trees per stand established between 1940 and 1970, coinciding with a post-war period of land abandonment, rural exodus and the onset of a tourism economy. Some trees have shown growth rates larger than 1 ​cm diameter per year and most species have had increasing BAI trends over the past decades. Strong growth releases occurred after windstorms at both sites, but the effects of windstorms were site-dependent, with the 1958 storm affecting mainly the eastern tip of the island (Anaga massif) and the 1991 storm the western tip (Teno massif). Given the great ability of laurel forest trees to establish after land use cessation and to increase growth after local disturbances such as windstorms, passive restoration may be sufficient to regenerate this habitat in currently degraded areas.

IntroductionBronchial aspirates (BAS) obtained during invasive mechanical ventilation (IMV) constitutes a useful tool for molecular phenotyping and decision making.AimTo identify the proteomic determinants associated with disease pathogenesis, all-cause mortality and respiratory sequelae in BAS samples from critically ill patients with SARS-CoV-2-induced ARDSMethodsMulticenter study including 74 critically ill patients with COVID-19 and non-COVID-19 ARDS. BAS were obtained by bronchoaspiration after IMV initiation. Three hundred sixty-four proteins were quantified using proximity extension assay (PEA) technology. Random forest models were used to assess predictor importance.ResultsAfter adjusting for confounding factors, CST5, NADK, SRPK2 and TGF-α were differentially detected in COVID-19 and non-COVID-19 patients. In random forest models for COVID-19, CST5, DPP7, NADK, KYAT1 and TYMP showed the highest variable importance. In COVID-19 patients, reduced levels of ENTPD2 and PTN were observed in nonsurvivors of ICU stay, even after adjustment. AGR2, NQO2, IL-1α, OSM and TRAIL showed the strongest associations with in-ICU mortality and were used to construct a protein-based prediction model. Kaplan-Meier curves revealed a clear separation in mortality risk between subgroups of PTN, ENTPD2 and the prediction model. Cox regression models supported these findings. In survivors, the levels of FCRL1, NTF4 and THOP1 in BAS samples obtained during the ICU stay correlated with lung function (i.e., DLCO levels) 3 months after hospital discharge. Similarly, Flt3L and THOP1 levels were correlated with radiological features (i.e., TSS). These proteins are expressed in immune and nonimmune lung cells. Poor host response to viral infectivity and an inappropriate reparative mechanism seem to be linked with the pathogenesis of the disease and fatal outcomes, respectively.ConclusionBAS proteomics identified novel factors associated with the pathology of SARS-CoV-2-induced ARDS and its adverse outcomes. BAS-based protein testing emerges as a novel tool for risk assessment in the ICU.

BackgroundThe pathophysiology of COVID-19-related critical illness is not completely understood. Here, we analyzed the microRNA (miRNA) profile of bronchial aspirate (BAS) samples from COVID-19 and non-COVID-19 patients admitted to the ICU to identify prognostic biomarkers of fatal outcomes and to define molecular pathways involved in the disease and adverse events.MethodsTwo patient populations were included (n = 89): (i) a study population composed of critically ill COVID-19 and non-COVID-19 patients; (ii) a prospective study cohort composed of COVID-19 survivors and non-survivors among patients assisted by invasive mechanical ventilation (IMV). BAS samples were obtained by bronchoaspiration during the ICU stay. The miRNA profile was analyzed using RT-qPCR. Detailed biomarker and bioinformatics analyses were performed.ResultsThe deregulation in five miRNA ratios (miR-122-5p/miR-199a-5p, miR-125a-5p/miR-133a-3p, miR-155-5p/miR-486-5p, miR-214-3p/miR-222-3p, and miR-221-3p/miR-27a-3p) was observed when COVID-19 and non-COVID-19 patients were compared. In addition, five miRNA ratios segregated between ICU survivors and nonsurvivors (miR-1-3p/miR-124-3p, miR-125b-5p/miR-34a-5p, miR-126-3p/miR-16-5p, miR-199a-5p/miR-9-5p, and miR-221-3p/miR-491-5p). Through multivariable analysis, we constructed a miRNA ratio-based prediction model for ICU mortality that optimized the best combination of miRNA ratios (miR-125b-5p/miR-34a-5p, miR-199a-5p/miR-9-5p, and miR-221-3p/miR-491-5p). The model (AUC 0.85) and the miR-199a-5p/miR-9-5p ratio (AUC 0.80) showed an optimal discrimination value and outperformed the best clinical predictor for ICU mortality (days from first symptoms to IMV initiation, AUC 0.73). The survival analysis confirmed the usefulness of the miRNA ratio model and the individual ratio to identify patients at high risk of fatal outcomes following IMV initiation. Functional enrichment analyses identified pathological mechanisms implicated in fibrosis, coagulation, viral infections, immune responses and inflammation.ConclusionsCOVID-19 induces a specific miRNA signature in BAS from critically ill patients. In addition, specific miRNA ratios in BAS samples hold individual and collective potential to improve risk-based patient stratification following IMV initiation in COVID-19-related critical illness. The biological role of the host miRNA profiles may allow a better understanding of the different pathological axes of the disease.

Las características cambiantes del clima obligan a su estudio de manera sistemática y abierta. El objetivo propuesto radicó en evaluar la percepción al cambio climático en el sector apicultor y agrícola de la comunidad de Quimis, Jipijapa. Para su desarrollo se aplicaron métodos asociados a los procesos lógicos del pensamiento y de carácter empírico, fundamentalmente encuestas y entrevistas. La muestra se conformó con residentes permanentes de dicha comunidad, dedicados de manera sistemática a labores agrícolas y apícolas. Se contrastaron los resultados de la información brindada por los participantes en la investigación con la obtenida mediante registros históricos realizados por instituciones especializadas. Los resultados pusieron de manifiesto que se valoró acertadamente la variabilidad del indicador de temperatura ambiental pero no ocurre lo mismo con la intensidad y volumen de las precipitaciones. CLIMATE CHANGE PERCEPTION OF IN FARMERS AND BEEKEEPERS OF QUIMIS COMMUNITY IN JIPIJAPA ABSTRACT The changing characteristics of the climate force its study in a systematic and open way. The proposed objective is to evaluate the perception of climate change in the beekeeping and agricultural sector of the community of Quimis, Jipijapa. For its development, methods associated to the logical processes of thought and empirical character were applied, mainly surveys and interviews. The sample was made up of permanent residents of said community, systematically dedicated to agricultural and beekeeping work. The results of the information provided by the participants in the research were contrasted with that obtained through historical records made by specialized institutions. The results show that the variability of the environmental temperature indicator is correctly assessed, but the same does not occur with the intensity and volume of rainfall. Abstract Background Lignin is a potential feedstock for microbial conversion into various chemicals. However, the microbial degradation rate of native or technical lignin is low, and chemical depolymerization is needed to obtain reasonable conversion rates. In the current study, nine bacterial strains belonging to the Pseudomonas and Rhodococcus genera were evaluated for their ability to grow on alkaline-treated softwood lignin as a sole carbon source. Results Pseudomonas fluorescens DSM 50090 and Rhodococcus opacus DSM1069 showed the best growth of the tested species on plates with lignin. Further evaluation of P. fluorescens and R. opacus was made in liquid cultivations with depolymerized softwood Kraft lignin (DL) at a concentration of 1 g/L. Size-exclusion chromatography (SEC) showed that R. opacus consumed most of the available lower-molecular weight compounds (approximately 0.1–0.4 kDa) in the DL, but the weight distribution of larger fractions was almost unaffected. Importantly, the consumed compounds included guaiacol—one of the main monomers in the DL. SEC analysis of P. fluorescens culture broth, in contrast, did not show a large conversion of low-molecular weight compounds, and guaiacol remained unconsumed. However, a significant shift in molecular weight distribution towards lower average weights was seen after cultivation with P. fluorescens. Conclusions Rhodococcus opacus and P. fluorescens were identified as two potential microbial candidates for the conversion/consumption of base-catalyzed depolymerized lignin, acting on low- and high-molecular weight lignin fragments, respectively. These findings will be of relevance for designing bioconversion of softwood Kraft lignin.
