Highly diastereoselective palladium catalyzed cinnamylation of N-tert-butanesulfinyl imines with cinnamyl acetates has been established to provide enantioenriched β-aryl homoallylic amines. The synthetic application of this stragety has been successfully demonstrated in the concise total syntheses of antitumor natural products (+)-lycoricidine and (+)-7-deoxypancratistatin.

Solid-state silica-gel-catalyzed opening of aziridine 6 provided phenanthrene 7, whose oxidative cleavage, recyclization, and further elaboration furnished the C-1 aldehyde and carboxylic acid derivatives of 7-deoxypancratistatin for potential analogue synthesis.

A highly efficient total synthesis of 7-deoxypancratistatin is described. The synthesis features the ready preparation of the phenanthridone skeleton by a Stille-IMDAF cycloaddition cascade. The resulting cycloadduct is converted into a key aldehydic intermediate, which is then induced to undergo a stereospecific decarbonylation reaction using Wilkinson’s catalyst to set the trans B–C ring junction of the target molecule.

Biocatalytic approaches have yielded efficient total syntheses of the major Amaryllidaceae alkaloids, all based on the key enzymatic dioxygenation of suitable aromatic precursors. This paper discusses the logic of general synthetic design for lycoricidine, narciclasine, pancratistatin, and 7-deoxypancratistatin. Experimental details are provided for the recently accomplished syntheses of narciclasine, ent-7-deoxypancratistatin, and 10b-epi-deoxypancratistatin via a new and selective opening of a cyclic sulfate over aziridines followed by aza-Payne rearrangement. The structural core of 7-deoxypancratistatin has also been degraded to a series of intermediates in which the amino inositol unit is cleaved and deoxygenated in a homologous fashion. These truncated derivatives and the compounds from the synthesis of the unnatural derivatives have been tested against six important human cancer cell lines in an effort to further develop the understanding of the mode of action for the most active congener in this group, pancratistatin. The results of the biological activity testing as well as experimental, spectral, and analytical data are provided in this manuscript for all relevant compounds.

The development of an approach leading to the total synthesis of 7-deoxypancratistatin is described. The key features of the approach include a 6-exo radical cyclization reaction between a benzylic radical and an O-benzyloxime ether to establish the C4a−C10b bond. The stereochemistry of this reaction was examined in both acyclic radical precursors and ones in which the aryl moiety was tethered to the C1 oxygen substituent. It was found that the use of such a tether was essential to obtain the stereochemical result required for the synthesis. The correct absolute and relative configurations at the hydroxylated carbons C1−C4 were obtained using d-gulonolactone as the source of C10b−C4a.

The first total synthesis of the title alkaloid has been accomplished in fourteen steps form 1-bromo-4-iodobenzene or 1-fluoro-4-iodobenzene via chemoenzymatic production of the antipodal cis-diene diols 4.

The sedative alkaloid paliurine F 7 is a pentapeptide bridged by an arene. Gwilherm Evano of the Université de Versailles took advantage of this in his synthesis (Angew. Chem. Int. Ed. 2007, 46, 572) of 7, although it was necessary to prepare, from serine, one of the amino acid derivatives, the protected 3-hydroxyprolinol 2. The key step in the synthesis was the Cu-catalyzed intramolecular coupling of 5 to give the macrolactam 6. Deprotection and acylation then gave paliurine F 7. Lepadiformine 14, isolated from the tunicate Clavelina lepadiformis, shows moderate cytotoxicity, and is also a K+ channel blocker. The synthesis of 14 (Angew. Chem. Int. Ed. 2007, 46, 2631) by Donald Craig of Imperial College started with the aziridine 8, prepared from the corresponding epoxide. Opening of the protected aziridine with the anion of methyl phenyl sulfone set the stage for condensation of the dianion derived from 9 with the aldehyde 10, to give, with high diastereocontrol, the amine 11. Deprotection followed by cyclization then led to the activated ether 12. While the opening of 12 with an alkyl Grignard reagent proceeded with undesired inversion at the reacting center, opening with the alkynyl Grignard delivered mainly the desired 13. Reduction followed by oxidation, epimerization and reduction then gave lepadiformine 14. The Amaryllidaceae alkaloid 7-deoxypancratistatin 21 has potent antiviral activity. A challenge in the assembly of 21 is that the ring fusion is trans, less stable than the corresponding cis diastereomer. The synthesis of 21 (J. Org. Chem. 2007, 72, 2570) by Albert Padwa of Emory University started with 17, the preparation of which by the combination 15 and 16 he had previously reported in the course of his synthesis of lycoricidine (OHL December 11, 2006). Ester 17 had the desired trans ring fusion, but with an angular ester substituent that had to be removed. While it would be expected from the mechanism that Rh-mediated decarbonylation of an aldehyde would proceed with retention of absolute configuration, and this had been confirmed experimentally, this reaction had not been applied to such a challenging substrate. In the event, the transformation proceeded smoothly, to give the desired trans 19. Dehydration and dihydroxylation of 19 led to the cyclic sulfate 20, selective SN2 opening of which delivered 7-deoxypancratistatin 21.

The stereocontrolled synthesis of (+)-7-deoxypancratistatin is described. The convergent synthesis has been achieved by two different strategies, both of which commence from a pentose and piperonal. The latter is converted into allylic bromide 7, which is then coupled with a protected methyl 5-deoxy-5-iodo-D-ribofuranoside in the presence of zinc metal. The first strategy involves a total of only 13 steps from D-ribose and piperonal, but suffers from a low yield in the zinc-mediated reaction between ribofuranoside 9, benzylamine, and bromide 7. The second strategy involves a total of 18 steps from D-xylose and piperonal. The former is converted into ribofuranoside 28, which is coupled with bromide 7 in the presence of zinc, and this is followed by ring-closing olefin metathesis. Subsequent Overman rearrangement, dihydroxylation, and deprotection then affords the natural product.
