BACKGROUND: Research has shown that fatty acid metabolism genes are closely related to the development of rheumatoid arthritis. Therefore, exploring the progression of rheumatoid arthritis based on fatty acid metabolism genes is of clinical significance. OBJECTIVE: To investigate whether fatty acid metabolism genes can serve as reliable biomarkers for predicting the progression of rheumatoid arthritis. METHODS: Gene data related to synovial tissue were downloaded from the Gene Expression Comprehensive Database (GEO). STRING was used to construct the protein-protein interaction network analysis. Cytoscape was utilized for biological annotation (gene ontology) and signaling pathway enrichment analysis (Kyoto Encyclopedia of Genes and Genomes). Fatty acid metabolism related genes were screened from the molecular feature database (MSigDB). Least absolute shrinkage and selection operator and support vector machine recursive feature elimination feature were used to screen for potential biomarkers. Immune cell infiltration levels in normal individuals and rheumatoid arthritis patients were assessed using the CIBERSORT algorithm. Finally, the expression levels of fatty acid metabolism related genes were verified using the receiver operating characteristic curve in GSE77298. RESULTS AND CONCLUSION: 361 differentially expressed genes in rheumatoid arthritis were identified, of which 13 overlapped with the reported fatty acid metabolism related genes. Based on machine learning algorithms, five genes were selected, and the receiver operating characteristic curve showed that five genes (PCK1, PDK1, PTGS2, PLA2G2D, and DPEP2) could predict the development of rheumatoid arthritis. The CIBERSORT algorithm results showed that five genes were associated with activated mast cells, neutrophils, resting mast cells, and memory resting CD4+ T cells. The receiver operating characteristic curve showed that PLA2G2D and PCK1 have high diagnostic value. To conclude, the expression characteristics of fatty acid metabolism related genes can serve as potential biomarkers for predicting clinical outcomes, which can further improve the accuracy of prediction in RA patients. [Objective] The study aimed to refine the types of cultivated land use, explore the spatial and temporal evolution patterns of cultivated land use types and their driving mechanisms, and provide a reference for optimizing the utilization of cultivated land resources and ensuring national food security. [Method] Based on the state of cultivated land use, farmland in Guanglong province was categorized into three types: paddy fields, dry land and croplund abandonment. Employing multiple methods such as the dynamic degree of land use, standard ellipse deviation model and geographical detectors, the study examined the spatiotemporal changes of long-term cultivated land use types in Guangdong province from three perspectives: time, space and structure. It explored the spatiotemporal evolution patterns and the underlying driving mechanisms, aiming to clarify the changing churacteristics of different types of cultivated land use. [Result] (i) Over the past 30 years, the types of cultivated land use in Guangdong province underwent significant changes in their spatiotemporal distribution and internal structure. Paddy fielda, dry land and cropland abandonment lemonstrated apatial agglomeration characteristics, primarily concentrated in areas with extensive cultivated land distribution but less developed economies, such as the Leizhou Peninsula hilly platform in western Guangdong, the southwestern Guangdong hills, and the central and southern Guangdong hills. The centroid of cultivated land exhibited a U-shaped migration trajectory, moving southwestward overall. (ii) The most significant changes were the conversion of dry land to paddy fields and dry land to cropland abandonment. These changes predominantly clustered in the southwestern Guangdong hills and the Leizhou Peninsula hilly platform, regions with extensive cultivated land distribution but underdeveloped economies. (iii) The area changes in cultivated land use types in Guangdong province were based on the natural environment and influenced by social, economic, and agricultural development factors. The primary driving factors for the area changes of the three types of cultivated land use were the amount of chemical fertilizer applied, the amount of pesticide used, and the labor force in the primary industry. Economic and agricultural factors such as agricultural gross output value, agricultural gross output value per capita, added value of the primary industry, cultivated land area per capita, grain yield, and total power of agricultural machinery have shown an increasing drive for changes in the areas of paddy fields, dry land, and fallow land year by year. (Conclusion] The spatiotemporal distribution, land type conversion, and driving mechanisms of paddy fields, dryland, and cropland abandonment in Guangdong province have formed distinct evolutionary patterns. Objective: To study the relationship between the angiomotin (Amot) and Yes-associated protein 1 (YAP1) expression and clinicopathological parameters and prognosis in type I endometrial cancer (EC) tissues. Methods: 75 type I EC patients who were diagnosed and treated in Affiliated Hospital of Xuzhou Medical University from January 2014 to June 2017 were selected as the study object (EC group), 30 patients with pathologically confirmed atypical hyperplasia endometrial tissue specimens diagnosed and treated during the same period were selected as the atypical hyperplasia endometrial group, and 30 patients with pathologically confirmed normal endometrial tissue specimens during the same period were selected as the normal endometrial group. The Amot and YAP1 protein expressions in each group were detected by immunohistochemistry. Spearman rank correlation was used to analyze the correlation between Amot and YAP1 protein expression in type I EC cancer tissues. The differences of Amot and YAP1 protein expression in cancer tissues of type I EC patients with different clinicopathological characteristics were compared. Kaplan-Meier analysis (Log-rank test) was used to analyze the relationship between the expression of Amot and YAP1 protein and the survival prognosis of type I EC patients. Univariate and multivariate COX regression analysis was used to analyze the factors affecting the survival prognosis of type I EC patients. Results: The positive rates of Amot and YAP1 protein expression in type I EC tissues were 80.00%(60/75) and 85.33%(64/75), respectively, which were significantly higher than those in atypical hyperplasia endometrial tissue 43.33% (13/30) and 46.67% (14/30), and the difference was statistically significant (all P＜0.05). The positive rates of Amot and YAP1 protein in atypical hyperplasia endometrial tissue were significantly higher than those in normal endometrial tissue with 16.67% (5/30) and 20.00% (6/30), and the difference was statistically significant (all P＜0.05). There was a significant positive correlation between Amot and YAP1 protein expression in type I EC cancer tissues(P＜0.05). The positive rates of Amot and YAP1 protein expression in EC type I patients with different International Federation of Gynecology and Obstetrics (FIGO) stages, histological grades and lymph node metastasis were statistically significant (P＜0.05). The cumulative survival time of Amot-positive expression typeⅠEC patients was significantly lower than that of Amot-negative expression typeⅠEC patients (P＜0.05). The cumulative survival time of patients with YAP1-positive expression of typeⅠEC was significantly lower than that of patients with YAP1-negative expression of typeⅠEC (P＜0.05). Amot-positive expression, YAP1 positive expression, FIGO stage Ⅲ and lymph node metastasis were independent risk factors for poor prognosis in patients with EC type I (P＜0.05). Conclusion: The Amot and YAP1 protein positive expression increased in type I EC cancer tissues, which are related to FIGO stage, histological grade and lymph node metastasis. Objective To investigate the expression and clinical significance of silent information regulator 2 (SIRT2) form tissue of urothelial bladder cancer(UBC) and the effects of down-regulation of SIRT2 on the proliferation, invasion and migration of bladder cancer cells. Methods The expression of SIRT2 protein was detected in 95 samples of urothelial bladder cancer tissues and 39 paracancerous tissues using the immunohistochemical EnVision method. Its relationship with clinicopathological parameters was analyzed. The expression of SIRT2 mRNA in bladder cancer cell lines was detected by RT-qPCR, and the expression levels of SIRT2 protein in bladder cancer cell lines, 12 pairs of fresh bladder cancer tissues were detected by Western blot. Lentiviral infection was used to stably downregulate the SIRT2 gene in bladder cancer cell line T24. CCK8 and cell colony assays were performed to detect changes in cell proliferation ability and Transwell assay to detect changes in cell invasion and migration activity. Results The positive expression rate of SIRT2 in urothelial bladder cancer tissues was 84%(80/95), which was higher than that in normal uroepithelial tissues (5%, 2/39) (P<0.05). The expression rate of SIRT2 was higher in the high-grade uroepithelial cancer group (95%, 38/40) than in the low-grade group (76%, 42/55) (P<0.05); The expression rate was higher in the muscle-infiltrated group (96%, 24/25) than in the non-muscle-infiltrated group (80%, 56/70) (P<0.05). Expression of SIRT2 protein was higher in fresh bladder cancer tissues than in normal tissues adjacent to the cancer. Expression of mRNA and protein of SIRT2 were also upregulated in bladder cancer cell lines. Knockdown of SIRT2 in T24 cells resulted in a significant decrease in cell activity of proliferation, invasion and migration (P<0.05). Conclusions SIRT2 is highly expressed in bladder uroepithelial carcinoma, and interfers with SIRT2 significantly and so inhibites bladder cancer cell proliferation, invasion and migration. Objective To explore the diagnosis and treatment of complex cerebral arteriovenous malformations by microsurgery. Methods The clinical data of 38 patients with complex cerebral arteriovenous malformations treated by simple microsurgery and combined surgery in the neurosurgery department of the First Affiliated Hospital of Soochow University from January 2011 to January 2021 were analyzed retrospectively, with 24 cases of grade Ⅲ, 9 cases of grade Ⅳ, and 5 cases of grade Ⅴ, according to Spetzler-Martin classification. The mean size of the malformed vascular mass was 5.9 cm (3.5-12.3 cm), located in the functional area in 27 cases and the non-functional area in 11 cases. CTA, DSA, and MRI were performed preoperatively, and CT, CTA or DSA were performed postoperatively. The neurological deficits were evaluated by the Glasgow prognostic score (GOS) at regular follow-up. Results In all cases, 28 underwent combined surgical treatment, and 10 underwent simple microsurgical resection. Postoperative imaging showed total resection in 35 (92.1%) and a significant reduction from a preoperative level in 3 (7.9%) . Postoperative follow-up was 6-79 months, with good recovery in 34 (GOS=4~5 points), moderate disability in 2 (GOS=3 points), severe disability in 1 (GOS=2 points), and death in 1 (GOS=1 point) . Conclusions Microsurgical resection is one of the effective methods for the treatment of complex cerebral arteriovenous malformations. The combination of preoperative imaging, preoperative supply artery embolization, intraoperative navigation, electrophysiological monitoring, and fine intraoperative microscopic manipulation is the key to successful surgery.
