Objective: To study Efficacy of tenofovir in the treatment of chronic hepatitis B and its effect on serum Hyaluronic acid (HA), laminin (LN), type IV collagen (IV - C), a former Ⅲ type collagen peptide (PC - III). Methods: 101 patients with chronic hepatitis B who were treated in our hospital from January 2018 to January 2020 were selected for this study. They were divided into experimental group (n=51) and control group (n=50) by random number table method. The control group received entecavir treatment and the experimental group received tenofovir treatment. Clinical efficacy, changes in serum HA, LN, IV-C, PC-III, HBV-DNA, hepatitis B surface antigen (HBsAg), alanine aminotransferase (ALT), aspartate aminotransferase (AST) levels and the incidence of adverse drug reactions were compared between the two groups. Results: After treatment, the total effective rate between the two groups was significantly different (P<0.05). Compared with before treatment, there were no significant differences in serum HA, LN, IV-C and PC-III levels between the experimental group and the control group. After treatment, the serum levels of HA, LN, IV-C and PC-III in experimental group and control group decreased with time, and the levels in experimental group were lower than those in control group, the differences were significant (P<0.05). Compared with before treatment, serum HBV-DNA and HBsAg levels of the experimental group and the control group were not significantly different. After treatment, the levels of serum HBV-DNA and HBsAg in experimental group and control group decreased with the passage of time, and the difference was significant (P<0.05). Compared with before treatment, there was no significant difference in serum ALT and AST levels between the experimental group and the control group. After treatment, the serum levels of ALT and AST in experimental group and control group decreased with the passage of time, and the levels in experimental group were lower than those in control group, the difference was significant (P<0.05). The total incidence of ADR in the two groups was 7.84% and 10.00% (P>0.05), respectively. Conclusion: The effect of tenofovir on chronic hepatitis B may be related to its effective improvement of serum THA, LN, IV-C and PC-III levels. [ABSTRACT FROM AUTHOR]

[Objective] The present paper aimed to fully understand the genetic diversity and phylogenetics of Xinjiang indigenous sheep breeds, and provide theoretical basis for breeds, conservation and utilization. [Method] The squences of mitochondrial DNA (mtDNA) Cylb of 13 Xinjiang indigenous sheep breeds were amplified and sequenced. [Result] It was found that these breeds had abundant genetic diversity, except Hotan sheep. Neutral test showed that Hotan sheep had experienced population expansion. The genetic distance between Hotan sheep breeds and Qira black sheep breeds was the closest, and between Hotan sheep breeds and Bachu sheep breeds was the furthest. There were relatively far genetic distance between Bachu sheep breeds and other breeds. 13 indigenous sheep breeds were divided into 3 branches by haplotype network. After AMOVA molecular variation analysis, genetic variation between populations and within populations was 91.33% and 8.67%, respectively. There was no significant genetic differentiation among populations. [Conclusion] The results showed that there were three matrilineal origins in 13 Xinjiang indigenous sheep breeds. The genetic relationship was basically consistent with the breeding history and geographical distribution. AIM: To investigate the synergistic anti-tumor effects of two novel inhibitors, zanubrutinib, a Bruton tyrosine kinase（ BTK） inhibitor, and pracinostat, a histone deacetylase（ HDAC） inhibitor, in diffuse large B-cell lymphoma （DLBCL） cells. METHODS: （1） NU-DUL-1 （ABC type DLBCL cell line） and SU-DHL-6 （GCB type DLBCL cell line） were treated with single-agent zanubrutinib at concentrations of 0, 2. 5, 5, 10, 20, 40, 80 and 160 μmol/L and single-agent pracinostat at concentrations of 0, 15. 63, 31. 25, 62. 5, 125, 250, 500, 1 000 and 2 000 nmol/L for 24, 48, and 72 h. The CellTiter-Glo assay was used to detect the inhibition of cell proliferation, and the half-maximal inhibitory concentration （IC50） was calculated. （2） The NU-DUL-1 and SU-DHL-6 cells were treated with zanubrutinib at concentrations of 0, 5, 7. 5, 10, 12. 5 and 15 μmol/L and pracinostat at concentrations of 0, 15. 62, 31. 25, 62. 5, 125 and 250 nmol/L, alone or in combination, for 48 h. The CellTiter-Glo assay was used to detect the inhibition of cell proliferation, and the synergy index of the combination was calculated through zero interaction potency （ZIP） model on the SynergyFinder （https：//synergyfinder. fimm. fi） website. （3） The NU-DUL-1 cells were treated with zanubrutinib （0 and 20 μmol/L） and pracinostat （0 and 200 nmol/L）, alone or in combination, for 48 h. The SU-DHL-6 cells were treated with zanubrutinib （0 and 20 μmol/L） and pracinostat （0 and 150 nmol/L）, alone or in combination, for 48 h. Flow cytometry was used to detect the cell apoptosis. （4） The NU-DUL-1 cells were treated with zanubrutinib at concentrations of 0, 10 and 20 μmol/L and pracinostat at concentrations of 0, 100 and 200 nmol/L, alone or in combination, for 48 h. The SUDHL-6 cells were treated with zanubrutinib at concentrations of 0, 10 and 20 μmol/L and pracinostat at concentrations of 0, 150 and 300 nmol/L, alone or in combination, for 48 h. Western blot analysis was used to detect the expression of apoptotic proteins, including poly（ADP-ribose） polymerase 1（ PARP1）, cleaved PARP1, caspase-3, cleaved caspase-3, caspase-8, and cleaved caspase-8. RESULTS: （1） Zanubrutinib and pracinostat can inhibit NU-DUL-1 and SU-DHL-6 cell proliferation in vitro, and exhibit time- and dose-dependent effects.（ 2） The combination of zanubrutinib and pracinostat showed significant synergistic effects in NU-DUL-1 cells （synergy index, 19. 553>10） and SU-DHL-6 cells （synergy index, 19. 392>10）.（3） The combination of zanubrutinib and pracinostat can enhance cell apoptosis compared with the control group, the proportion of apoptotic cells in the combination group is significantly increased （P<0. 05） than that in the control and single-drug groups.（ 4） Western blot results showed that zanubrutinib combined with pracinostat increased the expression of apoptotic proteins cleaved caspase-3, cleaved caspase-8 and cleaved PARP1 in NU-DUL-1 and SU-DHL-6 cells. The relative expression of apoptotic protein in combination group was significantly increased compared with control group and monotherapy group （P<0. 05）. CONCLUSION: The combination of zanubrutinib and pracinostat can significantly inhibit the proliferation of NU-DUL-1 and SU-DHL-6 cells and promote cell apoptosis, exerting a synergistic effect by inducing cell apoptosis through increasing the cleavage of apoptotic proteins caspase-3, caspase-8 and PARP1.
