In order to explore the biological characteristics and functions of GGPPS gene in Prunella vulgaris, the specific primers were designed based on the sequencing of P. vulgaris transcriptome. The full-length nucleotide sequence of GGPPS gene was obtained in P. vulgaris by reverse transcription PCR technology, and the bioinformatics analysis was done. qPCR was used to analyze the expression of PvGGPPS gene in ear induced by different exogenous substances and in different organs in P. vulgaris. The results showed that the PvGGPPS gene had an open reading frame of 1 092 bp and encoded 363 amino acids, with a theoretical molecular weight of 38 815.68 D and an isoelectric point of 5.69. PvGGPPS protein had the characteristic domain of isopentenyl pyrophosphate synthase family. Phylogenetic tree showed that PvGGPPS protein was closely related to Salvia miltiorrhiza and Coleus forskohlii GGPPS protein. qPCR analysis showed that the expression of PvGGPPS gene in leaves was higher than that in ears and stems. The expression level of PvGGPPS gene was increased in the ear after GA3 treatment for 24 h, one of seven exogenous substances treated. The expression of PvGGPPS gene in different tissues of Prunella vulgaris was quite different and was induced by exogenous substances treatment. The results of this study lay a foundation for further study on the function and expression regulation of PvGGPPS gene in the synthesis pathway of terpenoids from P. vulgaris. [ABSTRACT FROM AUTHOR]

Specific primers were designed on the basis of transcriptoine sequencing of Prunella vulgaris. The full-length nucleotide sequence of PvDXS was obtained by reverse transcription PCR and the bioinformatics analysis of the gene was conducted. The expression levels of PvDXS in different tissues and exogenous substances were detected by real-lime quantitative PCK. The results showed the eDNA sequence of PvDXS contained the open reading frame which had 2 181 hp and encoded a predicted protein of 726 amino acids with a theoretical molecular weight of 78 040.47 I) and a isoelectric point of 6.75. The protein had Transketolase C domain and Transket_pyr domain. Phylogenetie tree results showed that PvDXS protein was closely related to DXS (SmDXS2, CrDXS2) from Salvia miltiorrhiza and Catharanthus ro.scw.s-, anil il was inferred thai PvDXS belonged to the Class 11 DXS protein type. Tissue expression pattern analysis revealed that PvDXS gene in leaves was higher than that in ears and stems. After treated with seven exogenous substances for 24 h, the expression of the gene increased in GA3 treatment group and decreased alter treatment with the others. The expression level of the gene decreased significantly alter CaQ2, S1NP and SA treatments. The expressions of PvDXS were variant in different tissues and varied greatly after treatment of exogenous substances, which laid a foundation for further study on the function and expression regulation of PvDXS in the synthesis pathway of terpenoid components of Prunella vulgaris.
