目的　通过观察胆管闭锁患儿肝脏组织中跨膜转运蛋白39A（TMEM39A）的表达变化，初步探讨TMEM39A 与胆管闭锁发病的关系。方法　选取胆管闭锁患儿30例纳入病例组，同期收治的先天性胆总管囊肿患儿30例纳入对 照组。分别采用免疫组化法和Western blotting法检测两组患儿肝脏组织中的TMEM39A蛋白，采用RT-qPCR法检测 TMEM39A mRNA。结果　免疫组化检测结果显示，TMEM39A蛋白主要分布在汇管区周围肝细胞胞质内，病例组 TMEM39A蛋白相对表达量高于对照组（分别为0. 695 ± 0. 137、0. 375 ± 0. 096，P<0. 05）；Western blotting检测结果显 示，病例组TMEM39A 蛋白相对表达量高于对照组（分别为0. 624 ± 0. 082、0. 352 ± 0. 082，P<0. 05）。病例组 TMEM39A mRNA相对表达量高于对照组（分别为5. 724 ± 1. 490、2. 338 ± 0. 984，P<0. 05）。结论　胆管闭锁患儿肝 脏组织中TMEM39A蛋白及mRNA呈高表达，TMEM39A异常表达可能与胆管闭锁的发病有关。 [ABSTRACT FROM AUTHOR]
