2. 独活寄生汤抑制 NLRP3 炎性小体活化治疗痛风性关节炎.

- Abstract
BACKGROUND: The activation of NOD-like receptor thermal protein domain associated protein 3 (NLRP3) inflammasome has been found to be an important factor in the pathogenesis of gouty arthritis, and the activation of NLPR3 inflammasome can be effectively inhibited by regulating the PTEN-induced kinas 1 (PINK1)/Parkin signaling pathway. OBJECTIVE: To investigate the effect of Duhuo Jisheng Decoction on the PINK1/Parkin/NLRP3 signaling pathway. METHODS: (1) Animal experiment: Male Sprague-Dawley rats were randomly divided into control group, model group, positive control group (Qiushuixian tablets 0.3 mg/kg), and low-, medium-, and high-dose Duhuo Jisheng Decoction groups (6.9, 13.8, and 27.6 g/kg, respectively). A rat gouty arthritis model was constructed by injecting monosodium urate crystal suspension into the right hind ankle joint cavity of rats in all the groups except for the control group. (2) Cell experiment: Human monocytes (THP-1) were divided into blank control group, model group, Duhuo Jisheng Decoction-containing serum group, and PINK1 siRNA+Duhuo Jisheng Decoction-containing serum group. The cells in each group except the blank control group were stimulated with sodium urate to establish an in vitro gouty arthritis model. (3) The swelling degree, joint circumference, gait score, joint inflammation index and degree of pathological grading in the ankle joints of rats were observed. Enzyme-linked immunosorbent assay was used to detect the levels of uric acid, C-reactive protein, NLRP3, and interleukin 1β. Immunoblotting assay was used to detect the levels of proteins related to the PINK1/Parkin/NLRP3 pathway. Tetramethyl azolidine blue assay was used to detect cell activity. Immunofluorescent double staining was performed to detect the level of mitophagy. Immunofluorescence was used to detect the expression of NLRP3 and interleukin 1β. RESULTS AND CONCLUSION: (1) Compared with the control group, rats in the model group showed elevated ankle swelling and ankle circumference at 6-48 hours (P < 0.05), elevated gait scores and joint inflammation indexes at 24 and 48 hours (P < 0.05), elevated serum uric acid and C-reactive protein levels, degree of pathological classification, expression of PINK1, Parkin, and NLRP3 proteins, and interleukin 1β level in synovial tissues (P < 0.05). Compared with the model group, the protein expression levels of PINK1 and Parkin in all the Duhuo Jisheng Decoction groups were elevated, while the levels of the other indexes were decreased (P < 0.05). (2) Compared with the model group, NLRP3 activity, interleukin 1β level and mitochondrial outer membrane translocator protein 20 protein level in the Duhuo Jisheng Decoction-containing serum group were decreased (P < 0.05), while the proliferation inhibition rate, PINK1, Parkin and LC3B protein level were increased (P < 0.05). Compared with the model group, the mitochondrial autophagy level of cells in the Duhuo Jisheng Decoction-containing serum group was elevated (P < 0.05), while NLRP3 and interleukin 1β protein levels were decreased (P < 0.05). (3) Compared with the Duhuo Jisheng Decoction-containing serum group, the mitochondrial autophagy level of cells in the PINK1 siRNA+Duhuo Jisheng Decoction-containing serum group was decreased (P < 0.05), and the expression levels of NLRP3 and interleukin 1β were elevated (P < 0.05). To conclude, Duhuo Jisheng Decoction inhibits the activation of NLRP3 inflammasome by regulating the PINK1/Parkin signaling pathway, thereby exerting a therapeutic effect on gouty arthritis. 风速-功率是风电机组发电性能的重要指标,对风电场的运行管理具有重要意义。风速-功率数据是通过安装在风电场的监视控制与数据采集(SCADA)系统采集得到的,原始数据存在大量噪声,给后续应用研究带来了很大的挑战。基于风速-功率数据的空间分布特征,将风速-功率数据分为3类,并改进了数据预处理方法bin算法,提出了基于分区域(dbin)算法的异常数据识别清洗方法及流程。实验结果表明,dbin算法识别异常数据的效率比传统算法更高,具有较强的通用性。

11. 施用不同有益菌剂对长期连作烤烟根际土壤微生态的影响.

- Abstract
[Objective] The present paper aimed to investigate the impact of different beneficial bacteria on the micro-ecological environment of tobacco rhizosphere soil in long-term continuous cropping of flue-cured tobacco, aiming to provide insights for enhancing continuous cropping soil quality and mitigating obstacles associated with consecutive planting. [Method] The physical and chemical properties, enzyme activities, as well as structure and function of the microbial community in flue-cured tobacco rhizosphere soil were analyzed using pot experiment and high-throughput sequencing technology. The analysis was conducted after long-term continuous cropping of flue-cured tobacco with beneficial bacteria agents including Trichoderma harzianum agent T-22 (T1 treatment), Bacillus subtilis agent (T2 treatment), and a combination of T. harzanum agent T-22 and B. subtilis agent (T3 treatment). [Result] Compared with not applying beneficial bacteria (CK), the application of beneficial bacteria agents could enhance the availability of nutrients and enzyme activities in the rhizosphere soil of long-term continuous cropping flue-cured tobacco. Among the 10 environmental factors examined, available phosphorus (AP) content exhibited the highest contribution (4.97%) to bacterial community distribution in flue-cured tobacco rhizosphere soil. Additionally, Alkali-hydrolyzed nitrogen (AN) content demonstrated the highest contribution (6.91%) to fungal community distribution, while sucrase (SC) activity ranked the second with a contribution of 6.75%; Application of beneficial bacteria agents could increase the diversity index of microbial community structure and the number of unique OTU in long-term continuous cropping tobacco rhizosphere soil; Application of beneficial bacteria agents could increase the abundance, diversity index of microbial community and number of bacteria endemic OTUs in rhizosphere soil of long-term continuous cropping flue-cured tobacco, and change the abundance of indicator species in rhizosphere soil of long-term continuous cropping fluecured tobacco, among them, Pseudomonas and Mortierella were identified as the indicator species for Tl treatment, with Sphingomonas (12. 2%) and Mortierella (25.0%) exhibiting high abundance. The abundance of the main metabolic pathways in the rhizosphere soil of longterm continuous cropping tobacco was decreased by single application of bacteria, but not by single application of fungi. The abundance of bacteria metabolic pathway in rhizosphere soil of flue-cured tobacco could be increased by applying beneficial bacteria agent alone, while beneficial fungal agent along had the opposite effect. The abundance of fungal metabolic pathway in rhizosphere soil of flue-cured tobacco could be increased by applying beneficial fungal agent, beneficial bacteria agent and mixed agents. [Conclusion] The application of beneficial bacteria agents has positive impact on the micro-ecological environment of rhizosphere soil in long-term continuous flue-cured tobacco cultivation. Among them, T. harzianum T-22 demonstrates superior efficacy and can be effectively utilized for the prevention and control of obstacles associated with continuous cropping of flue-cured tobacco. 功能脑网络为理解大脑功能激活模式及大脑信息传递结构提供了一种有效的生物标记,如何更加有效地利用先验信息构建准确的脑网络模型尤为重要。提出了一种基于群体相似性约束的功能脑网络模型,通过引入张量低秩约束,利用张量的核范数,求解一个在组内具有低秩先验的脑网络组。这种方法利用组内的相似性约束,收缩脑网络的解空间,从而有效地构建更优的脑网络模型。将所构建的脑网络用于轻度认知障碍的判别任务,实验结果表明,所提出的基于群体相似性约束的脑网络建模方法,能够构建出更具判别性的脑网络,并得到了与以往研究一致的显著性连边,进一步验证了所提出方法的有效性。

Objectives: The ground-based radar has features such as working all-time in all-weather, providing continuous observations with high spatiotemporal resolutions and can monitor landslides with high precision in near real-time. However, atmospheric effects represent one major limitation of ground-based radar, due to the spatiotemporal variations of the troposphere, especially in mountainous areas. Atmospheric variations are often treated as static signals and corrected by the range function model which can be unreliable, particularly when strong dynamic atmospheric turbulence occurs. Methods: Based on its successful experience in the generic atmospheric correction online service for interferometric synthetic aperture radar (InSAR), the ground based iterative tropospheric decomposition (GBITD) model is utilized to decompose the tropospheric delay into stratification and turbulence components, and perform atmospheric correction. The GBITD model doesn't require global navigation satellite system or any external meteorological observations, which makes it flexible for InSAR atmospheric correction. Results: The application of the GBITD model to the 2018 Baige landslide suggests that the root mean square of two interferograms with temporal baselines of 2 min and 10 min decreased from 1.43 mm and 1.69 mm to 0.21 mm and 0.24 mm, respectively. The maximum cumulative displacement derived from the 778 images acquired during the period from 4 to 10 December 2018 was 1.4 m with the maximum displacement rate of 0.23 m/d. Conclusions: The experimental results show that the GBITD method performes best compared against the homogeneous model and the stratified model in complex mountainous areas. Furthermore, Ground-based radar has obvious unique advantages in landslide monitoring, which can provide strong technical support for landslide emergency monitoring and risk assessment. An accurate and timely monitoring is a high demand for the oestrus behavior of dairy cows in modern dairy farming. In this research, a lightweight recognition was proposed to detect the oestrus behavior of cows using the combined YOLO v5n and channel pruning. The high precision of the network was established before that. Firstly, the model was sparsely trained, according to different sparsity rates. The best sparsity effect of the model was obtained at the sparsity rate of 0.005. Then, the channel pruning was utilized to prune the modules, including the CSPDarknet53 backbone feature extraction network, in order to compress the model structure and parameters for the high detection speed. 2 239 images of cows’ mounting behavior were also collected, including 1 473 images in the daytime, 88 images in the daytime (backlight), and 160 images in the nighttime. Four occlusion situations were considered, including 1 045 images without occlusion, as well as 397, 184, and 95 images with the slight, moderate, and heavy occlusion, respectively. Three target sizes were also considered, including 46, 1 191, and 484 images with the large, medium, and small targets, respectively. The pruned model was veried on the test set, and then to compare with the Faster R-CNN, SSD, YOLOX-Nano, and YOLOv5-Nano. The test results showed that the mean average precision (mAP) of the model after pruning was 97.70%, the Params were 0.72 M, the Floating Point operations (FLOPs) were 0.68 G, and the detection speed was 50.26 fps. Furthermore, the Params and FLOPs were reduced by 59.32%, and 49.63%, respectively, under the condition of constant mAP, and the detection speed increased by 33.71%, compared with the original model YOLOv5-Nano. Consequently, the pruning operation was effectively improved the performance of the model. The mAP of the model was close to the Faster R-CNN, SSD, YOLOX-Nano, but the Params were reduced by 135.97, 22.89, and 0.18 M, respectively, the FLOPs were reduced by 153.69, 86.73, and 0.14 G, respectively, and the detection speed increased by 36.04, 13.22, and 23.02 fps, respectively. In addition, the detection performance of the model was tested in the complex environments, such as different lighting, different occlusion, multi-scale targets, and new environments. The results showed that the mAP was 99.50% in the nighttime environment, the average mAP was 93.53% under the three occlusion conditions, the average mAP was 98.77% under the medium and small targets, the detection rate was 84.62% in the generalization test, and the false detection rate was 7.69%. To sum up, the improved model can fully meet the high requirements for the accurate and real-time monitoring of cows' oestrus behavior under complex breeding environments and all-weather conditions, due to the lightweight, high precision, robust, and high generalization. - Abstract
The motion behavior of dairy cows contains much of health information. The application of information and intelli‐ gent technology will help farms grasp the health status of dairy cows in time and improve breeding efficiency. In this paper, the development trend of intelligent morning technology of cow's motion behavior was mainly analyzed. Firstly, on the basis of ex‐ pounding the significance of monitoring the basic motion (lying, walking, standing), oestrus, breathing, rumination and limping of dairy cows, the necessity of behavior monitoring of dairy cows was introduced. Secondly, the current research status was summarized from contact monitoring methods and non-contact monitoring methods in chronological order. The principle and achievements of related research were introduced in detail and classified. It is found that the current contact monitoring methods mainly rely on acceleration sensors, pedometers and pressure sensors, while the non-contact monitoring methods mainly rely on video images, including traditional video image analysis and video image analysis based on deep learning. Then, the develop‐ ment status of cow behavior monitoring industry was analyzed, and the main businesses and mainstream products of representa‐ tive livestock farm automation equipment suppliers were listed. Industry giants, such as Afimilk and DeLaval, as well as their products such as intelligent collar (AfiCollar), pedometer (AfiActll Tag) and automatic milking equipment (VMS™ V300) were introduced. After that, the problems and challenges of current contact and non-contact monitoring methods of dairy cow motion behavior were put forward. The current intelligent monitoring methods of dairy cows' motion behavior are mainly wearable de‐ vices, but they have some disadvantages, such as bring stress to dairy cows and are difficult to install and maintain. Although the non-contact monitoring methods based on video image analysis technology does not bring stress to dairy cows and is low cost, the relevant research is still in its infancy, and there is still a certain distance from commercial use. Finally, the future de‐ velopment directions of relevant key technologies were prospected, including miniaturization and integration of wearable moni‐ toring equipment, improving the robustness of computer vision technology, multi-target monitoring with limited equipment and promoting technology industrialization. - Abstract
Objective To study the risk factors for admission to intensive care unit (ICU) in patients after hip and knee arthroplasty. Methods Totally 1 490 patients after primary unilateral hip and knee arthroplasty were divided into the admission group (99 cases, requiring ICU admission after the operation) and the non-ICU admission group (1 391 cases, not requiring ICU admission after the operation) according to whether they needed to be admitted to the ICU after the operation. Univariate and multivariate Logistic regression analysis was used to analyze the risk factors of ICU admission in patients after hip and knee arthroplasty. Results Univariate analysis showed that age, American Society of Anaesthesiologists ( ASA) classification, preoperative diabetes, hypertension, pulmonary infection, use of β -blockers and ACEI, blood levels of urea nitrogen (BUN) and creatinine, as well as type of surgery, surgeons' seniority, anesthesia method, and intraoperative RBC infusion were the influencing factors for patients who needed to be admitted to ICU after hip and knee arthroplasty (all P<0. 05). Multivariate Logistic regression analysis showed that age >80 years, ASA Ⅲ and Ⅳ, femoral head replacement, general anesthesia, intraoperative red blood cell infusion and preoperative BUN elevation were the risk factors for patients who needed to be admitted to ICU after hip and knee arthroplasty (all P<0. 05). Conclusion Age >80 years, ASA Ⅲ and Ⅳ, femoral head replacement, general anesthesia, intraoperative red blood cell transfusion and preoperative BUN elevation are risk factors for ICU admission in patients after hip and knee arthroplasty.
