目的 探讨miR-146a在肺鳞癌发生、发展中的作用。方法 采用qRT-PCR法检测70例份肺鳞癌组织 及其配对的癌旁正常组织、20例份正常肺组织miR-146a表达，并分析miR-146a表达与肺鳞癌患者临床病理参数 的关系。筛选人肺鳞癌细胞SK-MES-1、NCI-H520、NCI-H2170、NCI-H226及正常肺组织上皮细胞（BEAS-2B)中 miR-146a表达最低的NCI-H520细胞进行后续实验。采用脂质体介导法转染miR-146a mimics或阴性对照 mimics-NC，Transwell小室法检测转染miR-146a mimics和阴性对照mimics-NC的NCI-H520细胞迁移和侵袭能力。 结果肺鳞癌组织miR-146a表达明显低于癌旁正常组织和正常肺组织(P均<0.01)。肺鳞癌组织miR-146a表达 与肿瘤组织分化程度、临床分期、淋巴结转移有关（P均<0.01)。转染miR-146a mimics的NCI-H520细胞迁移和 侵袭细胞数目较转染mimics-NC的NCI-H520细胞明显降低(P <0.05或<0.01)。 结论肺鳞癌组织miR-146a表 达降低，其表达变化与肿瘤的恶性生物学行为有关;miR-146a能够明显抑制肺鳞癌细胞的迁移和侵袭。 [ABSTRACT FROM AUTHOR]

Objective To investigate the effects of saxagliptin on the proliferation，differentiation，and apoptosis of osteoblasts in high glucose environment，so as to provide the theoretical basis for the selection of rational hypoglycemic agents for patients with diabetic osteoporosis. Methods Osteoblasts MC3T3-E1 were cultured and passaged in a-MEM medium. The cells of 3 generations in the logarithmic phase were randomly divided into the normal control group，high glucose group，and high glucose i saxagliptin group. The a-MEM medium was immediately replaced with 5. 5 mmol/L glucose medium in the normal control group，the a-MEM medium was immediately replaced with 30. 0 mmol/L glucose medium in the high glucose group，and the a-MEM medium was immediately replaced with 3 0 .0 mmol/L glucose +1 μmol/L saxagliptin medium in the high glucose + saxagliptin group. MC3T3-E1 osteoblasts were then cultured in different culture mediums for 48 h. CCK-8 method and flow cytometry were used to detect the proliferation ability and apoptosis rate，ELISA assay was used to detect alkaline phosphatase ( ALP ) activity，and qRT-PCR was used to detect the mRNA expression of COL-I， Runx2，OCN and OPN. Results Compared with the normal control group，the proliferation，ALP activity and expression of COL-I mRNA，Runx2 mRNA， OCN mRNA， and OPN mRNA were significantly decreased， and the apoptosis rate was significantly increased in the high glucose group and high glucose + saxagliptin group ( all P < 0 .05 ). Compared with the high glucose group， the proliferation， ALP activity and expression of COL-I mRNA， Runx2 m RNA， OCN mRNA， and OPN mRNA were significantly increased，and the apoptosis rate was significantly decreased in the high glucose + saxagliptin group ( all P <0.05 ). Conclusion Saxagliptin can reverse the effects of high glucose environment on proliferation, differentiation，and apoptosis of MC3T3-E1 osteoblasts to a certain extent，which can be used for hypoglycemic therapy for patients with diabetic osteoporosis.
