4. 基于转录组测序分析猪子宫内膜妊娠中的关键非编码 RNA 及其调控通路.

- Abstract
【Objective】The study aims to explore miRNA, lncRNA and target genes regulating endometrial pregnancy in Chuanxiang Black pigs and Tibetan pigs with different litter sizes, and to further reveal the signal pathways affecting the enrichment of non-coding RNA related to endometrial pregnancy in different breeds of pigs.【Method】Transcriptome sequencing was performed on the endometrium of the 2nd and 5th litters Chuanxiang Black pigs and Tibetan pigs with an average litter size of 11.35 and 6.70, respectively. RNA was extracted, transcriptome sequencing was carried out after quality inspection, sequencing data were spliced, miRNA and lncRNA were aligned, and target genes were predicted. GO and KEGG enrichment analysis were performed to screen candidate regulatory genes and signaling pathways regulating different litter sizes of Chuanxiang Black pigs and Tibetan pigs.【Result】 Multiple miRNAs, lncRNAs and target genes were differentially expressed between Chuanxiang Black pigs and Tibetan pigs. The second parity had 1571 miRNAs and 2042 lncRNAs differentially expressed, and the fifth parity had 1042 miRNAs and 2096 lncRNAs differentially expressed. Further GO and KEGG enrichment analysis revealed that Chuanxiang Black pigs had higher enrichment in gonadotropin-releasing hormone (GnRH) secretion, ECM-receptor interaction and Wnt signaling pathway than Tibetan pigs. Functional enrichment was also found in some metabolic pathways, including aldosterone regulation of sodium reabsorption, glutathione metabolism, fructose and mannose metabolism, etc. Besides, key candidate regulatory genes identified included FSHR (Follicle stimulating hormone receptor), DBX1 (Developing brain homeobox 1), ARID2 (AT-rich interaction domain 2), TNC (Tenascin C), NT5C2 (Neuropilin 2), LAMC3 (laminin subunit gamma 3), MADD (MAP kinase activating death domain), etc. 【Conclusion】Chuanxiang Black pigs with high litter size have higher gonadotropin-releasing hormone (GnRH) secretion, ECM-receptor interaction, Wnt signaling pathway enrichment (GO analysis result), and metabolic pathways (KEGG analysis result) during endometrial pregnancy compared with Tibetan pigs. The key regulatory genes involved mainly include FSHR, DBX1, etc.. Overall, the study provides new insights for exploring the function and molecular regulation mechanism of non-coding RNA in the process of porcine endometrial pregnancy. [ABSTRACT FROM AUTHOR]

- Abstract
The penetration ability on warship target by naval gun semi-armor-piercing ammunition is analyzed. LS-DYNA finite element software is used to simulate the ricochet, attacking main deck and ship board by Oto Melara 127 mm and 76 mm naval gun ammunition. It is concluded that the critical ricochet angle of 127 mm and 76 mm ammunition is 9.5°and 29°when striking on 15 mm E36 plate in 300 m/s. Both ammunitions are able to penetrate several layers attacking from main deck and ship board. When attacking warships, it is considerable to delay initiation time properly, so the ammunition can enter central cabin for improving damage effect. [ABSTRACT FROM AUTHOR]

In synchrotron radiation field, the apertures of optical components increase gradually and their surface figure precisions have been demanded to be a nano-radian level. This paper researches the modern surface figure measurement methods in this field, stitching interferometry, to realize the high resolution two-dimensional measurement of optical components. The basic principles of the stitching interferometry are introduced. Then, a series of common optical surface shape measurement instruments in the synchrotron radiation field are overviewed, such as long trace profilers based on laser beams, high precision and auto-collimation measuring machines for nanometer optical components and stitching interferometers. It describes their development history and working characteristics and compares their shortcomings and advantages. Finally, this paper analyzes the main error sources involved in the stitching interferometry, and points out that the application and development trends of the technology are mainly the innovation of the stitching algorithm, the improvement of measuring speeds of interferometers, the commercialization of interferometers and the integration of the interferometry and other science technologies. 漏 2016, Science Press. [Objectives] This study aimed to verify the correlation between the c.-751 A>C in ACSF2(encoded by acyl-CoA synthetase family member 2) promoter and laying performance of Yangzhou goose, and analyze the effect of the SNP on ACSF2 expression. [Methods] We used allele specific PCR(AS-PCR) to analyze the genotype of SNP c.-751 A>C in a 343 Yangzhou geese population, and performed the correlation analysis between genotype and laying performance of Yangzhou goose. Quantitative real-time PCR(qPCR) and luciferase reporter gene vector were used to detect the difference between AA and CC promoter activity. ACSF2 over-expression was performed to analyze its role in the energy metabolism of granulosa cells and laying performance. [Results] Genotyping and association analysis results showed that the egg numbers of geese with AA genotype were significantly higher than those CC ones. Quantitative real-time PCR analysis indicated that AA individuals showed lower ACSF2 expression level in ovary tissues. Moreover, the overexpression experiment indicated that ACSF2 was involved in the energy metabolism of granulosa cells. [Conclusions] We identified a functional mutation in the promoter region of ACSF2 affecting gene expression in Yangzhou goose, and indicated the role of ACSF2 in laying performance by regulating energy metabolism of granulosa cells. Our results provide insight into the improvement of goose laying performance by ACSF2 genetic variation selection. [ABSTRACT FROM AUTHOR]

In order to study the temperature effects on polyacrylamide solution spectrum measurement, the transmission spectra of polyacrylamide solution under non-normal temperature were measured by using the double-beam ultraviolet-visible spectrophotometer, Fourier transform infrared spectrometer and the temperature control device. The effects of temperature on the spectral properties of polyacrylamide solution were analyzed. Research shows that when the polyacrylamide solution at 20~60C°, the ultraviolet and visible light band transmission spectra are less influenced by temperature, the near-infrared transmission spectra are influenced by temperature greatly. The experimental study for the wastewater containing polyacrylamide spectrum detection research has certain reference value. [ABSTRACT FROM AUTHOR]

- Abstract
The damage effect of aluminized explosives on the ship targets is an important evidence of checking whether the explosive formula is reasonable.In order to study the influence of the molar mass of aluminum and oxygen (Al/O ratio) on ship damage,the damage effect of aluminized RDX-based explosive (RDX/Al) on ship is researched by means of numerical simulation.The damage effect is explained from four aspects:displacement,acceleration,stress and strain.Simulated results show that the overall and the local damage effects of a ship increases and then decreases with the increase in Al/O ratio in the near field.The damage effect of explosive on ship is optimal when Al/O ratio is 0.36.It can be seen from the relationship between Al/O ratio and energy release rate that the main reason of ship damage is the shock wave energy in the near field.
