[Objective] The present paper aimed to investigate the inhibition effect of raspberry pulp oil on ultraviolet B (UVB) induced photoaging of human keratinocytes (HaCaT cells) . [Method] The raspberry pulp oil was extracted by supercritical CO2 and its chemical compositions were determined by gas chromatography-mass spectrometry (GC-MS) .The photoaging model was established by UVB, the cell viability of HaCaT cells were determined by CCK-8 method, and the concentrations of matrix metalloproteinase-3 (MMP-3), Collagen type 1 (COL1A1), Interleukin-1β (IL-1β), Interleukin-6 (IL-6), Tumor necrosis factor α (TNF-α), Thioredoxin (Trx) and Superoxide dismutase (SOD) were detected by Enzyme-linked immunosorbent assays (ELISA) and microplate assay. [Result] The analysis of GC-MS showed that raspberry pulp oil consisted of 53 chemical components, mainly including fatty acids, ester of fatty acid, tocopherols, and phytosterols.Oleic acid, linoleic acid, ethyl Z-9-oleate, vitamin E and sterol were closely associated with inhibition effects of raspberry pulp oil on photoaging of UVB-induced HaCaT cells.Compared with the model group, raspberry pulp oil significantly improved the cell viability of photodamaged HaCaT cells induced by UVB (P<0.01) and showed in a does-dependent manner. In addition, raspberry pulp oil significantly increased concentrations of COL1A1 and reduced the concentrations of MMP-3 to promote skin elasticity (P<0.05, P<0.01), reduced the levels of IL-1β, IL-6 and TNF-α to alleviate the inflammatory response (P<0.05, P<0.01), increased the contents of Trx and SOD to improve skin oxidative stability (P<0.05, P<0.01) in the UVB-induced photodamaged HaCaT cells. [Conclusion] Raspberry pulp oil exhibits good anti-photoaging activity probably by alleviating inflammatory reactions, easing the degradation of extracellular matrix components and reducing oxidative stress in the photodamaged HaCaT cells. - Abstract
Objective To investigate the association of the metabolism of intestinal short - chain fatty acids ( SCFAs ) with the development and progression of the disease spectrum of nonalcoholic fatty liver disease ( NAFLD ) by determining the content of fecal SCFAs in patients with different N AFLD diseases and the change in the content of fecal SCF As after treatment in patients at a high risk of nonalcoholic steatohepatitis ( NASH ) . Methods A total of 90 patients who were diagnosed with N AFLD in The Affiliated Hospital of Qinghai University from July 2020 to July 2021 were enrolled and divided into simple nonalcoholic fatty liver ( NAFL) group with 30 patients, NASH group with 30 patients, and nonalcoholic fatty liver fibrosis group with 30 patients, and 40 individuals who underwent physical examination during the same period of time were enrolled as control group . Related case data and fecal SCF As content were collected for the four groups, and related clinical indices and fecal SCFAs content were collected for 10 patients at a high risk of NASH after 3 months of intervention. The analysis of variance was used for comparison of normally distributed continuous data between multiple groups, and the paired samples t - test was used for comparison within each group; the Kruskal - Wallis H test was used for comparison of non - normally distributed continuous data between multiple groups, and the paired samples Wilcoxon signed rank sum test was used for comparison within each group; a Spearman correlation analysis was used to investigate the correlation between variables; the receiver operating characteristic ( ROC) curve analysis was used for diagnostic evaluation. Results Compared with the control group, the nonalcoholic fatty liver fibrosis group had significantly higher contents of valeric acid and caproic acid, and the NAFL group had significantly lower contents of valeric acid and caproic acid (all P < 0. 05); the nonalcoholic fatty liver fibrosis group had significantly higher contents of valeric acid and caproic acid than the N AFL group ( P < 0. 05) ; the nonalcoholic fatty liver fibrosis group had a significantly higher content of valeric acid than the NASH group ( P < 0. 05) ; the NASH group had a significantly higher content of caproic acid than the N AFL group ( P < 0. 05) . After treatment, the high - risk patients in the NASH group had significant reductions in HbAlc, fasting plasma glucose (FPG), triglyceride (TG), total cholesterol (TC), alanine aminotransferase (ALT), aspartate aminotransferase (AST), gamma - glutamyl transpeptidase ( GGT), total bile acid (TBA), prothrombin time (PT), uric acid ( UA), controlled attenuation parameter (CAP), and liver stiffness measurement ( LSM) ( Z = - 2. 805, - 2. 703, - 2. 193, - 2. 599, - 2. 805, - 2. 701, - 2. 803, - 1. 988, - 2. 807, - 2. 803, - 2. 803, and - 2. 668, all P < 0. 05) ; for these patients, the contents of acetic acid and propionic acid after treatment were significantly higher than those before treatment ( Z = - 2. 803 and - 2. 803, both P < 0. 05), and the content of isobutyric acid after treatment was significantly lower than that before treatment ( Z = - 2. 803, P < 0. 05). In the diagnosis of nonalcoholic fatty liver fibrosis, valeric acid had an area under the ROC curve (AUC) of 0. 842, with a sensitivity of 86. 7% and a specificity of 70% at the optimal cut - off value of 141. 42 µ,gig; caproic acid had an AUC of 0. 819, with a sensitivity of 70% and a specificity of 85% at the optimal cut - off value of 6. 93 µ,gig. Conclusion Valerie acid and caproic acid may promote the development of NAFLD disease spectrum. Acetic acid and propionic acid may have a certain protective effect on the liver of NAFLD patients, and isobutyric acid may promote the development and progression of NASH. The protective effect of acetic acid and propionic acid on the liver may further lead to the reductions in HbAl c, FPG, TG, TC, ALT, AST, GGT, TBA, PT, UA, CAP, and LSM. V aleric acid and caproic acid have an inferior diagnostic value to PIIIP N - P and a superior diagnostic value to type IV collagen and hyaluronic acid. V aleric acid with the optimal cut - off value of 141. 42 µ,gig and caproic acid with the optimal cut - off value of 6. 93 µ,gig can be used as the auxiliary diagnostic indicators for the early diagnosis of nonalcoholic fatty liver fibrosis. Objective: To explore the clinical curative effect of Shenmai injection combined with budesonide in treatment of patients with acute alcoholismand aspiration pneumonia. Methods: A total of 112 patients with acute alcoholismand aspiration pneumonia, who were admitted to Langfang People's Hospital of Hebei Province from June 2012 to June 2015, were chosen and randomly divided into observation group (n=57) and control group (n=55). The control group was treated with budesonide on the basis of basic treatment, while the observation group was added Shenmai injection on the basis of control group's therapy. The recovery and disappeared time of symptoms, adverse reactions, liver function indexes and serum inflammatory factors indexes in the two groups before and after treatment were collected and analyzed. Results: The awake time, recovery time of blood pressure, and the incidence of dizziness, headache and electrolyte disorders in the observation group were significantly lower than those in the control group (P<0.05). The levels of aspartate transaminase(AST), alanine aminotransferase(ALT), gamma-glutamyl transferase(GGT) in the two group after treatment significantly decreased than before treatment, and the indexes in the observation group were significantly lower than those in control group (P<0.05). The time of cough stopping, asthma relieving, wheezing and wet rales disappearing time in the observation group were significantly shorter than those in the control group(P<0.05). The levels of serum IL-6, CRP and TNF-αin the two groups after treatment significantly decreased than before treatment, and the indexes in the observation group were significantly lower than those in the control group (P<0. 05). Conclusions: Shenmai injection plus budesonide has a reliable curative effect in the treatment of patients with acute alcoholism and aspiration pneumonia, which can effectively shorten the recovery and disappearing time of symptoms, reduce the incidence of adverse reactions, improve liver function and relieve the inflammatory reaction, which has an important clinical value.
