2. 缺血性视网膜损伤致细胞凋亡的内质网 应激机制研究.

- Abstract
Objective To analyze the effects of ischemia on the expression of endoplasmic reticulum stress (ERS)- related proteins glucose regulated protein78 (GRP78), C/BEBP homologous protein (CHOP), and cysteinyl aspartate specific proteinase (Caspase)-12 during the apoptotic process of retinal ganglion cells (RGCs), and to investigate the molecular level pathogenesis of ischemic retinal injury (IRI)-induced RGCs apoptosis, to provide a new idea and method for the prevention and treatment of IRI. Methods The anterior chamber high intraocular pressure method was applied to produce a rat right eye IRI model. Forty healthy male SD rats were randomly divided into the normal control group, 30-min IRI group, 60-min IRI group, and 120-min IRI group according to the ischemic time, with 10 rats in each group. Hematoxylin-eosin (HE) staining was applied to observe the changes in retinal histomorphology in different groups of rats; immunohistochemical (IHC) staining was applied to observe the expression and localization of apoptosis-related factors such as B-cell lymphocyte/leukemia-2 (Bcl-2), Bcl-2 associated X protein (Bax), and Caspase-3 in different groups; immunofluorescence (IF) staining was applied to observe the expression and localization of ERS-related proteins such as GRP78, CHOP and Caspase-12 in other groups. Results HE staining results showed that compared with the normal control group, retinal edema, increased thickness of the inner retinal layer, disturbed distribution of retinal cells, decreased number of RGCs, and increased vacuolar tissues were observed in the IRI groups. IHC staining results showed that only a small amount of Bax and Caspase-3 proteins were expressed in the retina of the normal control group, while Bax and Caspase-3 proteins were mainly expressed in the ganglion cell layer and the inner nuclear layer of the retina in the IRI groups. The expression of Bax and Caspase-3 proteins in the retina of the IRI groups was significantly higher than that of the normal control group, and the differences between the IRI groups were also statistically significant. A large amount of Bcl-2 protein was observed in the ganglion cell layer and inner nuclear layer of the retina in the normal control group. Compared to the normal control group, the expression of Bcl-2 protein in the retina of each IRI group was significantly reduced, and the differences between the IRI groups were also statistically significant. The results of IF staining showed that the positive expressions of GRP78, CHOP, and Caspase-12 in the retina of the normal control group were small, while their expressions in the ganglion cell layer and the inner nuclear layer in the retina of the IRI groups were significantly increased compared with the normal control group. Conclusions IRI can induce apoptosis in the retinal cells, and its mechanism may be related to the activation of ERS-related pathways. [ABSTRACT FROM AUTHOR]

Objective To research the effect of centralized procurement of coronary stent on total hospitalization expenses and structural expenses, explore the possible changes in treatment behavior and provide basis for the formulation of relevant operation management focus after the centralized procurement of coronary stent. Methods We collected the data of patients who underwent coronary stent implantation from January 2019 to December 2022 in a 3A hospital, using the interrupted time series analysis, descriptive analysis method, chi - square test and Welch' s ANOVA test to analyze the data at 48 time - observation points before and after centralized procurement. Results Centralized procurement immediately reduced the average hospitalization expenses of coronary stent implantation by 19 954. 38 Yuan (0, = -19 954. 381, 95% CI: - 21 546. 085 ~ - 18 362. 676) and medical materials cost dropped most (侯=-16 410. 418, 95% CI: - 17 548. 630 ~ -15 272. 206), other categories expenses had also varying degrees decrease (P <0.05). After centralized procurement, the average hospitalization expense of coronary stent implantation was gradually rising (艮=315. 605, 95% CI: 198. 061 -433. 148), mainly due to the rise in the expenses of medical examination (角=110. 484, 95% CI: 95.525 -125.443) and materials (民=265.433, 95% CI: 181. 378 - 349. 487), the proportion of medical service fees was gradually decreasing (P < 0. 05). Conclusion The centralized procurement of coronary stent reduces the total hospitalization expenses of related patients. However, the cost of medical examination and materials continues growth after the centralized procurement of coronary stent, so hospital managers should strengthen supervision to avoid unreasonable recovery of expenses. Objective To analyze the influencing factors of hospitalization expenses of high-rate cases paid by Diagnosis Related Groups (DRG), and to provide references for medical institutions to reasonably control expenses and reduce patients' burden of seeking medical treatment. Methods In total 3 444 patients with high rate of DRG in a tertiary hospital were selected as the research subjects. Univariate analysis and multiple linear regression model were used to analyze the factors affecting the hospitalization expenses of high-rate cases, and the structural equation model was used to verify the correlation among the variables. Results The results of univariate analysis showed that sex, age, DRG group, fund type, hospitalization days, and main diagnosis group had significant influence on the hospitalization cost of high-rate cases. The results of multiple linear regression model showed that the top three factors affecting hospitalization expenses were length of stay (β=0.308, P< 0.001), DRG group (β=0.112, P < 0.001), and main diagnostic group (β=0.035, t=8.744, P < 0.001). The modified structural equation model fitted well (RMSEA=0.007, GFI=0.984, NFI=0.904, IFI=0.911, and CFI=0.901). The results of structural equation model showed that the length of stay was the biggest factor affecting hospitalization cost, and the path coefficient was 0.81. The second was DRG grouping (0.41), and the influence of fund type was small (0.09). Other factors had little effect on hospitalization cost. Conclusion The length of stay is the biggest factor affecting the hospitalization cost of high-rate cases. Under the premise of reasonable diagnosis and treatment, we can shorten the days of hospitalization, achieve accurate cost control, and optimize the hospital operation and management mechanism to reduce the high-rate cases. - Abstract
The spatial distribution of soil water content is a key factor affecting agricultural irrigation decisions. In this study, 38 technology was adopted to explore the spatial distribution of soil water content in the irrigation area. With soil water content as the main forecast object, combined with the weather forecast and the growth of planted crops, and based on the field water balance equation as the theoretical basis, the agricultural irrigation water requirement in the irrigation area was estimated by using soil water content in the irrigation area, and its spatial distribution was analyzed. The results could provide scientific basis for rational water use and agricultural water-saving irrigation in irrigated areas, and also provide technical support for scientific and rational water-saving potential assessment. The results show that the predicted water demand of Xiaokaihe irrigation area in late May is 6 217 700 m³, which is consistent with the statistics of the irrigation area. The prediction technology of agricultural irrigation water demand based on 35 can realize the precision irrigation of irrigation area. [ABSTRACT FROM AUTHOR]

- Abstract
Objective To investigate the predictive value of neutrophil to lymphocyte ratio (NLR) combined with highsensitivity cardiac troponin T (hs-cTnT) for atrial fibrillation (AF) after off-pump coronary artery bypass grafting (OPCAB). Methods A total of 108 patients who were received OPCAB treatment in our hospital from January 2017 to June 2019 were selected, and they were divided into AF group (n=32) and non AF group (n=76) according to whether AF occurred after operation. The general data, cardiac function, OPCAB perioperative indexes, NLR and hs-cTnT were collected and compared between the two groups. The risk factors of AF after OPCAB were analyzed by multiple Logistic regression. The predictive values of NLR and hs-cTnT on AF after OPCAB were analyzed by receiver operating characteristic (ROC) curve. Results The age, NYHA cardiac function grade≥grade Ⅲ, left atrial diameter (LAD), NLR, hs-cTnT and mechanical ventilation time of patients were significantly higher in the AF group than those in the non AF group (P<0.05). Multiple Logistic regression analysis showed that the elevated LAD (OR=3.032, 95%CI: 1.614-5.696), NLR (OR=2.186, 95%CI: 1.171-4.081) and hs-cTnT (OR=2.284, 95%CI: 1.313-3.973) were independent risk factors of AF after OPCAB (P<0.05). ROC analysis showed that NLR and hs-cTnT had higher predictive values for AF after OPCAB, and the areas under the curve were 0.781 (95%CI: 0.685-0.877) and 0.764 (95%CI: 0.663-0.866). The cut-off values were 3.52 and 13.08 ng/L respectively. The combined analysis of the two could further improve the predictive values, and the area under the curve was 0.846 (95%CI: 0.767-0.925). Conclusion The combination of NLR and hs-cTnT has a higher clinical value in predicting AF after OPCAB, which has certain clinical application value. [ABSTRACT FROM AUTHOR]

The stress state of surrounding rock in deep roadway is within the limit of rock strength limit, and the surrounding rock will produce large creep deformation, especially the deep disturbance load is more sensitive to the influence of creep deformation of roadway surrounding rock. In order to study the nonlinear creep behavior and creep disturbance effect of deep rock under disturbance load, the self-developed rock triaxial disturbance creep test bench is used to use gneiss rock as rock sample and graded loading method. The uniaxial compression perturbation creep test of rock under static load axial pressure, different disturbance amplitude and different disturbance frequency is obtained, and the influence of disturbance load on rock creep characteristics is obtained. The test results show that the axial com-pression is the dominant factor of creep deformation of rock. With the increase of axial compression, the creep deformation of rock increases gradually. The creep deformation of rock increases with the increase of disturbance amplitude and disturbance frequency. Under the same axial pressure and action time, the disturbance amplitude is more obvious than the disturbance frequency of creep creep deformation; under the same disturbance condition, with the increase of axial pressure, the impact of rock creep damage is more affected. When the axial pressure is greater than 40 MPa, the creep of rock creep is affected by the disturbance. After each disturbance loading, the rock creep strain has a stepped sudden increase, and the creep curve also appears steep, with the disturbance amplitude. And the increase of the disturbance frequency, the larger the mutation value. Based on the analysis of the test results, the nonlinear viscoplastic perturbation creep element and the damage original were introduced, and the composite model of rock nonlinear disturbance creep damage was established by series connection with Burger. The improved nonlinear least squares method based on pattern search is used to invert the creep parameters, and the results of the creep equations are fitted with the experimental results and the effect is good. - Abstract
Objective To design a CT -guided puncture positioning device to shorten puncture time while increase the accuracy. Methods The device realized 3D positioning by the principle of plane geometry, which was composed of a positioner, a fixation mechanism and a direction guiding mechanism for puncture needle. The positioner and direction guiding mechanism were made of transparent plastic, and the fixation mechanism was made of elastic cloth. Results The device gained advantages over the traditional means in puncture time, times of needle insertion and times of CT scanning, and had the puncture success rate in one time being 90%. - Abstract
In order to study the variation of coal mechanical properties under different temperature, this paper conducted the numerical test of the mechanical property of coal influence by using RFPA2D- thermal and with the method of theoretical analysis and numerical test combination. From the viewpoint of thermal effect,the fracture mechanics knowledge is applied in this study. The result shows that the temperature-change stress make cracks of coal easier to produce and develop, the fracture strength factor of internal cracks in coal was enhanced and the crack tip stress field was changed by thermal. The carrying capacity of coal decreased with the temperature growth, and the result further verified the rule of carrying capacity of rock decreased with the temperature growth future from the view of numerical test. With the growth of heterogeneity and the discreteness of mechanical property of the inner coal matrix particles unit, the stress increment of single facula improve significantly at linear stage, and the carrying capacity of coal increased gradually. - Abstract
The stability of gas drainage borehole affects the quality of extraction engineering. In the process of mining,the abutment moves along with the advance of the working face,and loading and unloading stresses act on the borehole in mining influence area.In order to obtain the deformation and failure law of the borehole under the condition of loading-unloading,the authors used a numerical method to calculate the failure of boreholes at different stress change rates. Based on ten calculation schemes,the drilling plastic zone distribution and the hole wall displacement curve under different loading and unloading stress changing rates were obtained. At the same time,according to the relationship between the rate of unloading stress and the deformation of the hole wall was obtained by numerical calculation. The calculation model of shear stress around the borehole was established,which considered the rate of unloading stress change. Research indicates that the loading process,loading rate do not affect the extent of damage surrounding borehole and hole wall displacement. The unloading process around borehole damage range is associated with the initial unloading stress and unloading rate.In the drilling arrangement the stress characteristics of boreholes should be considered. - Abstract
The unconventional gas resource in coal measures strata is the main part of unconventional gas. At present, the understanding of coalbed methane, shale gas and tight sandstone gas in the coal measures strata is relatively limited. Also, the degrees of resources exploration and development are relatively low. According to the experiences of gas reservoir combined mining, the coal measure strata can be considered as a whole, then the cost of exploration and development on coalbed methane, shale gas and tight sandstone gas in the coal measures strata could be reduced. In addition, the efficiency and single well profit of gas well could be improved. In this paper, the coalbed methane, the shale gas and the tight sandstone gas in coal measures strata are referred to as “the three coal gases”. The commingling possibility of “the three coal gases” and the difficulties that commingling urgently needs were analyzed from the formation lithology distribution characteristics of coal measures strata lithology, accumulation mechanisms, reservoir characteristics, exploitation characteristics, etc. [ABSTRACT FROM AUTHOR]

In order to explore the effect of trichoderma on the decomposition and revegetation of coal gangue, the trichoderma with low(0.03%), medium(0.15%), high(0.30%) doses was applied in coal gangue decomposition experiments and alfalfa pot experiments to determine coal gangue decomposition rate, alfalfa fresh weight and root characteristics, as well as the physical, chemical, and microbiological properties of coal gangue matrix. The results showed that trichoderma could increase the root length, root-shoot ratio, root volume and number of lateral roots of alfalfa. Trichoderma could significantly change the mechanical com-position of coal gangue, and fine gravel was the main component of weathering. With the increase of the dose of trichoderma, the fine gravel had a tendency to be further decomposed into coarse sand and medium sand. Trichoderma promoted the fine microscopic structure of coal gangue. Trichoderma could also increase avail-able nitrogen, available phosphorus, available potassium content, microbial biomass carbon, nitrogen, phos-phorus and catalase, urease, phosphatase activity of coal gangue matrix, thereby promoting the decomposing and revegetation effect of coal gangue. In general, the low dose trichoderma did not obviously promote the revegetationeffect of coal gangue, the medium and high dose of trichoderma had the significant effect, which increased the decomposition rate of coal gangue and alfalfa fresh weight by 44.24%, 38.86% and 31.33%, 22.04%, respectively. Medium dose trichoderma had the best effect on the decomposition and revegetation of coal gangue. Therefore, the medium dose of trichoderma can be used for the revegetation of coal gangue. In order to calculate the concrete scale range of coal and rock mass in different rockburst danger degrees, and prevent and control rockburst in coal mine, the concept of dynamic system of coal and rock is put forward in this paper. The model of the relationship between the occurrence of rockburst and dynamic system is built at the same time. The calculation method of dynamic system scale and its evaluation system and quantitative indicators are put forward based on blasting theory and Mises yield strength criterion. According to the source position and energy value of happened rockburst and microseismic events, the concrete scale range of dynamic nuclear area zone, damage zone, injury zone and influence zone are calculated, so that the different rockburst danger degrees in different areas of coal and rock mass could be determined. The accuracy of dynamic system scale calculation method is validated in Laohutai coal mine, and the results show that the calculation method is highly reliable and practical, which could be widely used in forecasting the risk of rockburst. - Abstract
In order to study the methane displacement in coalbed by carbon dioxide with different volume stress and temperature, using self-developed seepage device for triaxial absorbing and desorbing, an experimental study was carried out on methane displacement with the consideration of volume stress and temperature variation under the thermo-mechanical action, and under the pore pressure of equaling to coalbed. The results show that at the same temperature, the adsorption decreases with the increase of volume stress, and the gradient declines obviously;the volume stress and temperature are the main causes of methane displacement, and the formula is fitted that the coalbed methane displacement varies with volume stress change at 20-50 ℃. At the same temperature, the displacement efficiency drops linearly with the increase of volume stress;as temperature increases, the displacement efficiency increases obviously. At the same temperature, the replacement volume ratio expands;with temperature increase, the replacement volume ratio falls, and the gradient remains the same. [ABSTRACT FROM AUTHOR]
