【目的】观察海洋甾体YC-1 对低钾(5 mmol/L, low K+)诱导小脑颗粒神经元凋亡的影响, 并对其机制进行初步探 讨。【方法】小脑颗粒神经元原代培养;采用二乙酸荧光素(fluorescein diacetate , FDA)和Hoechst 33258 DNA 染色法观察神经元 存活率及形态学特征;用琼脂糖凝胶电泳分析神经元死亡的生化特征。【结果】低钾引起小脑颗粒神经元凋亡。Hoechst 33258 DNA 染色表现出染色体固缩, DNA 凝胶电泳表现出由大小不一的断裂DNA 片段形成的“ 梯形” 条带。YC-1(1.25～ 20 μmol/ L) 呈时间和剂量依赖性地抑制上述现象的出现。蛋白质合成抑制剂放线菌酮(cycloheximide)不能改变由不同孵育时间造成的 YC-1 对神经元不同保护效果的差异。【结论】YC-1 抑制低钾诱导小脑颗粒神经元凋亡, YC-1 的这一作用可能属于一非基因 效应。

Objective To investigate the effects of Huoxueqiwei granules on the expressions of eNOS, nNOS and Cx43 in the rats with VC and ED, and explore its mechanism in the treatment of VC and ED. Methods A total of 120 SD rats were divided into the normal group, the sham group, the model group, and low-, medium-, and high-dose Huoxueqiwei granule groups. A rat model of VC and ED was established through Turner’s method and APO test. APO tests were performed to detect erectile function on the 1st, 3rd, and 8th weekend; the diameter of the left spermatic vein was measured before and after intervention; Levels of serum FSH, LH, FT, and TT were detected by ELISA; The ultrastructure of penile tissue was observed using a transmission electron microscope; The mRNA expressions of nNOS and eNOS in penile were analyzed by RT-PCR; The expression of Cx43 in penile was detected by western blot. Results After modeling, the width of the left spermatic vein in the model group and each treatment group was increased significantly（P＜0.01). After intervention, the vein width in each treatment group was decreased（P＜0.05). For the first APO test, there was no difference in the erectile function of rats in each group（P＞0.05). For the second APO test, the erectile function of rats in the model group and each treatment group was decreased significantly（P＜0.01). For the third APO test, the erectile function of rats in each treatment group was recovered（P＜0.05). The levels of FSH and LH in the model group were increased（P＜0.05), while the levels of TT and FT were decreased（P＜0.05). Levels of serum FSH and LH in the medium and high dose groups were all decreased（P＜0.05), while levels of TT and FT were increased（P＜0.05). The mRNA expressions of eNOS and nNOS in penis were significantly downregulated（P＜0.01) in the model group. The mRNA expressions of eNOS and nNOS in the penis of each treatment group were significantly upregulated（P＜0.01). Cx43 protein level was significantly decreased（P＜0.01) in the model group, and its level in each treatment group was significantly increased（P＜0.01). The structure of CCSMC in each treatment group was intact, and in a small number of fields, the collagen fiber structure and mitochondria were restored, the number of endoplasmic reticulum was increased. Conclusions Huoxueqiwei granules not only improve varicocele and sex hormone levels, but also upregulate eNOS, nNOS and Cx43 expressions in the corpus cavernosum of the penis, which thereby enhances the erectile function of rats. OBJECTIVE: The rabbit model of steroid-induced osteonecrosis of femoral head is the most commonly used animal model of femoral head necrosis. The pathological changes of the femoral head are close to clinical practice, however, the conditions, methods and evaluation standards of animal models reported in and outside China are not uniform, which leads to the low scientific value of animal models and is difficult to popularize. This study aimed to clarify the influence of different mold-making conditions on the establishment of steroid-induced osteonecrosis of femoral head rabbit model and analyze the appropriate conditions for the successful model establishment. METHODS: We searched the CNKI, WanFang, VIP, CBM, WoS, PubMed and EMbsae databases for the literature on the modeling of steroid-induced osteonecrosis of femoral head rabbits up to April 1, 2022, completed the screening of the literature according to the inclusion and exclusion criteria and literature quality evaluation, and extracted the outcome index data in the literature. RevMan Stata and ADDIS statistical software were used to conduct a metaanalysis of the included data. RESULTS: (1) A total of 82 articles with 1 366 rabbits were included in the study. The steroid-induced osteonecrosis of femoral head modeling methods were divided into three types: steroid-alone method, steroid combined lipopolysaccharide method and steroid combined serum method. Among these, 33 articles used steroid-alone method; 20 articles used steroid combined lipopolysaccharide method; 29 articles used steroid combined serum method. (2) Meta-analysis results showed that the three modeling methods significantly increased the rate of empty bone lacunae in the femoral head of steroid-induced osteonecrosis of femoral head rabbits (P < 0.001), and significantly decreased the ratio of the trabecular bone area in the femoral head of steroid-induced osteonecrosis of femoral head rabbits (P < 0.001). The order of empty bone lacunae rate of each modeling method was: steroid combined with lipopolysaccharide method > steroid-alone method > steroid combined with serum method > normal group, and the order of trabecular bone area rate of each modeling method was: normal group > steroid combined with serum method > steroid-alone method > steroid combined with lipopolysaccharide method. (3) The results of subgroup analysis suggested that the rate of empty bone lacunae in the rabbit model induced by steroid alone might be related to the rabbit variety and the type of steroid used for modeling (difference between groups P < 0.05), in which the combined effect amount of New Zealand white rabbits was higher than that of Chinese white rabbits (P < 0.05) and Japanese white rabbits, and the combined effect amount of dexamethasone was higher than that of other steroids. The rate of empty bone lacunae induced by steroid combined with lipopolysaccharide was related to the administration mode of lipopolysaccharide and the type of steroid (P < 0.05), among which the combined effect of methylprednisolone sodium succinate was significantly higher than that of other steroids (P < 0.05), and the combined effect of prednisolone was significantly lower than that of other steroids (P < 0.05). The combined effect of lipopolysaccharide 100 μg/kg × twice was significantly lower than 10 μg/kg × twice and 50 μg/kg × twice (P < 0.05). The rate of empty bone lacunae in the model induced by steroid combined with serum was related to serum dose and steroid type (P < 0.05), among which the combined effect amount of dexamethasone sodium phosphate was significantly higher than other steroid types (P < 0.05), and the combined effect amount of dexamethasone was significantly lower than other steroid types (P < 0.05); the combined effect amount of serum “10 mL/kg+6 mL/kg” combined dose was lower than other serum doses (P < 0.05). CONCLUSION: (1) With the rate of empty bone lacunae and the ratio of trabecular bone area as the judgment standard for the successful establishment of the model, the three modeling methods can successfully construct the rabbit steroid-induced osteonecrosis of femoral head model, of which the steroid combined with lipopolysaccharide method is the best. (2) New Zealand white rabbits and dexamethasone are recommended when selecting the steroid-alone method. Methylprednisolone sodium succinate and low-dose lipopolysaccharide are recommended when selecting the steroid combined with lipopolysaccharide method. Objective: To analyze the relationship between serum fibroblast growth factor 23（FGF23）, alkaline phosphatase（ALP）and fetuin-A（FA） levels and coronary artery calcification（CAC） in patients with coronary heart disease（CHD） and to explore their predictive value for CAC. Methods: 165 patients with CHD who were admitted to our hospital from February 2021 to February 2022 were selected, and they were divided into CAC group（n=73） and no-CAC group（n=92） according to whether they were accompanied by CAC or not. Clinical data of the patients were collected, and serum FGF23, ALP and FA levels were measured by enzyme-linked immunosorbent assay（ELISA）. The influencing factors of CAC in patients with CHD were analyzed by multivariate Logistic regression, and the predictive value of serum FGF23, ALP and FA levels on CAC in patients with CHD was drawed by receiver operating characteristic（ROC）curves. Results: Serum FGF23 and ALP levels in the CAC group were higher than in the no-CAC group, and FA level was lower than thae in the no-CAC group（all P<0.01）. Multivariate Logistic regression analysis showed that that age（older）（OR=1.220, 95% CI:1.087～1.369）, hypertension（OR=1.461, 95%CI:1.062～2.010）, blood calcium（higher）（OR=1.532, 95%CI:1.042～2.251）, blood phosphorus（higher）（OR=1.209, 95%CI:1.097～1.333）, FGF23（higher）（OR=1.012, 95%CI: 1.007～1.018）, ALP（higher）（OR=1.046,95%CI:1.023～1.070） were independent risk factors for CAC in patients with CHD, FA（higher）（OR=0.827, 95%CI:0.750～0.912） was independent protective factor（all P<0.05）. ROC curve analysis showed that the area under the curve（AUC） of serum FGF23, ALP and FA were 0.790, 0.773, 0.786 and 0.915, respectively, and the AUC of serum FGF23, ALP and FA in combination with CHD was greater than that of each indicator alone. Conclusion: The serum FGF23 and ALP levels increased and serum FA level decreased are closely related to the occurrence of CAC in patients with CHD, which can be used as auxiliary predictors of THE occurrence of CAC in patients with CHD, and the combined prediction value of the three indicators is high in the risk of CAC in patients with CHD. Objective To investigate the effects of miR-422a targeting kallikinase 4 (KLK4) on proliferation, migration and invasion of cervical cancer cells. Methods The expression levels of miR-422a in human cervical cancer cells (HeLa, SiHa) and human normal cervical epithelial cells (H8) were quantified by quantitative real-time PCR. Cervical cancer HeLa cells were divided into the blank group, the miR-NC group, the miR-422a group, the mut miR-422a group, the miR-422a+ pcDNA group and the miR-422a+pcDNA-KLK4 group. CCK-8 assay and transwell assay were used to detect the proliferation, migration and invasion of HeLa cells in each group. TargetScan software was used to predict the binding target genes. Double luciferase activity test was used to verify the targeting relationship. Western blot assay was used to detect the KLK4 protein expression in each group. Results Compared with normal human cervical epithelial cells H8, the expression levels of miR-422a were low in both cervical cancer HeLa cells and SiHa cell lines (P＜0.01), and cervical cancer HeLa cells were selected for the subsequent experiments. Compared with that in the miR-NC group and blank group, the OD450 of the miR-422a group decreased after cells were cultured for 4, 6 d, the numbers of invasion and migration cells decreased in the miR-422a group after cells were cultured for 6 and 12 h (P＜0.05). Compared with miR-422a+pcDNA group, OD450 of the miR-422a+pcDNA-KLK4 group was significantly increased at 4 and 6 d after culturing, and the number of cell migration and invasion was increased at 6 and 12 h after culturing (P＜0.01). TargetScan software predicted that there was a binding site between miR-422a and KLK4, which was verified by double luciferase reporter gene experiment. Compared with the miR-NC group, the expression level of KLK4 protein was decreased in the miR-422a group, and the expression level of KLK4 protein was increased in the mut miR-422a group (P＜0.01). Compared with the miR-422a+pcDNA group, the expression level of KLK4 protein increased in the miR-422a+pc DNA-KLK4 group (P＜0.01). Objective: To explore the application value of apolipoprotein E (ApoE) gene polymorphism detection in lipid-lowering therapy of patients with acute coronary syndrome (ACS). Methods: 180 patients with ACS from February 2019 to June 2020 were selected, and they were divided into groups A, B and C by using random number table method, with 60 cases in each group. ApoE gene polymorphism was tested among all patients. According to the ApoE gene phenotypes (E2, E3, E4 phenotypes) determined by Sanger sequencing, group A was given oral administration of rosuvastatin (10 mg/d), and group B was given oral administration of rosuvastatin (20 mg/d), and group C was given oral administration of rosuvastatin (10 mg/d) + ezetimibe (10 mg/d). After 1 month of continuous treatment, the blood lipids [total cholesterol (TC), triacylglycerol (TG), low-density lipoprotein cholesterol (LDL-C)] and LDL-C compliance rate were evaluated among the three groups of phenotypes, and the medication side effects were recorded. All patients were followed up for 1 month, and the occurrence of major adverse cardiovascular events (MACE) were counted. Results: There were no statistically significant differences in the composition ratios of E2, E3, and E4 phenotypes of ApoE gene among the three groups (P>0.05). After treatment, the levels of TC, TG and LDL-C of different ApoE gene phenotypes in each group were decreased compared with those before treatment, and there were statistically significant differences in the change rates (P<0.05), manifested as E2 phenotype>E3 phenotype>E4 phenotype (P<0.05). There were no statistically significant differences in the change rates of TC, TG and LDL-C levels of E2 phenotype among the three groups (P>0.05). The change rates of TC, TG and LDL-C levels of E3 phenotype patients in groups B and C were significantly higher than those in group A(P<0.05), but there was no significant difference in the change rate of each index between group B and group C(P>0.05). There were statistically significant differences in the change rates of TC, TG and LDL-C levels of E4 phenotype patients among the three groups (P<0.05), showing group C>group B>group A (P<0.05). After treatment, the LDL-C compliance rate in group A was significantly lower than that in group B and group C (61.67% vs 85.00% vs 90.00%) (P<0.05). There was no statistically significant difference in the LDL-C compliance rate of E2 phenotype patients among the three groups (P>0.05). The LDL-C compliance rate of E3 phenotype patients in group A was significantly lower than that in groups B and C (P<0.05), and the LDL-C compliance rate of E4 phenotype patients in group A and group B was lower than that in group C (P<0.05). During treatment, only one case in group B was with ALT exceeding 3 times of the upper limit of normal value, and the patient could return to normal after drug withdrawal. There was a statistically significant difference in the incidence rate of MACE among the three groups (P<0.05). The incidence rate in group A was significantly higher than that in group B and group C (18.33% vs 5.00% vs 3.33%) (P<0.05), but there was no significant difference in the incidence rate of MACE between E2 phenotype patients and E4 phenotype patients in the three groups (P>0.05), and the incidence rate of MACE of E3 phenotype patients in group A was higher than that in groups B and C (P<0.05). Conclusion: The lipid-lowering efficacy in patients with ACS is related to the ApoE gene phenotypes. Rosuvastatin alone can achieve a good lipid-lowering effect for the E2 phenotype, and rosuvastatin intensive treatment or combination of ezetimibe for E3 phenotype can better improve the lipid-lowering effect than rosuvastatin alone, and combination of ezetimibe for E4 phenotype has better lipid-lowering effect than rosuvastatin alone or intensive treatment [ABSTRACT FROM AUTHOR]
