【目的】研究布托啡诺在临床使用剂量范围内通过抑制ABCB1的转运功能增加白血病耐药细胞的化 疗药物敏感性。【方法】采用HL60及其ABCB1过表达的耐药细胞株HL60/VCR、K562及其ABCB1过表达的耐药 细胞株K562/ADR 均在培养环境中加入含胎牛血清的DMEM 或RPMI-1640的培养液中进行培养。采用MTT法 检测布托啡诺的细胞毒性,通过流式细胞仪检测阿霉素(DOX)在HL60/VCR细胞内的累积程度;加入布托啡诺 培养72 h后用免疫荧光观察ABCB1在细胞膜表面的形态和分布变化以及采用Western Blotting检测ABCB1的表达 水平。【结果】布托啡诺能将K562/ADR细胞的阿霉素的抵抗倍数由48.1倍下降到9.6倍,长春新碱(VCR)的抵抗倍 数由108.4倍下降到20.7倍,HL60/VCR的DOX的抵抗倍数由37.8倍下降到7.6倍,长春新碱抵抗倍数由23.4下降 到4.8倍,差异均有统计学意义。ABCB1的表达水平及其在细胞膜上的定位没有变化。【结论】布托啡诺明显降低 阿霉素和长春新碱在ABCB1过表达的白血病耐药细胞株中的IC50,有效增加阿霉素在HL60/VCR细胞内的累积水 平。布托啡诺不改变HL60/VCR和K562/ADR细胞ABCB1的表达水平和ABCB1在细胞膜表面的形态和定位。

4. 川西北中三叠统雷口坡组三段储层特征及成因机理.

- Abstract
In recent years, multiple exploration breakthroughs have been made in the Third Member of the Leikoupo Formation in the Middle Triassic of Northwest Sichuan Basin. Currently, the overall exploration level is low. The characteristics and formation mechanism of the reservoir are unclear. Techniques such as core observation, pore permeability testing, conventional and cast thin section identification, scanning electron microscopy, mercury intrusion and nuclear magnetic resonance were used to clarify the sedimentary and reservoir characteristics of the 2nd section of the Maokou Formation in the front of the Longmen Mountains in western Sichuan and to explore the main controlling factors of high-quality reservoirs. Research shows that the reservoir rock types are mainly granular dolomite, microbial dolomite and icro-crystalline dolomite. The storage spaces are mainly composed of secondary dissolved pores such as intergranular dissolved pores, microbial “frame” dissolved pores and intercrystalline dissolved pores. The overall physical properties are relatively dense, belonging to medium-low porosity and low permeability reservoirs, with an average porosity of 2. 78% and an average permeability of 0. 76 mD. Sedimentary facies, diagenesis, paleogeomorphology and tectogenesis jointly control the development of the T2 l3 member reservoir in northwestern Sichuan basin. The development of reservoirs is controlled by sedimentary facies, ancient landforms, and diagenesis. The material basis for the development of favorable reservoirs is formed by the deposition of high-energy sand and debris shoals, biogenic debris shoals. The surface dissolution effect of karst slopes in ancient landforms is the best. The quasi contemporaneous dolomitization, recrystallization, and multi-stage karstification are the key factors for reservoir development. Structural fractures and faults are favorable conditions for reservoir improvement. The Zhongba area and the southwestern part of the Shuangyushi area are located at the intersection of high energy beaches and karst slopes with micro ancient landforms. The thickness of the dolomite is large, with developed faults and fractures, strong dissolution, and the development of Class I and II reservoirs. - Abstract
Liquid atomization is widely used in important scenarios such as internal combustion engines, gas turbines, aero-engines, seawater desalination, heat exchange equipment, and medical equipment. Due to the large difference between the application conditions and the physical properties of the injection object, it is difficult to abstract a general flow field distribution and droplet breakup model. Aiming at the spray “ circling” layer and droplet breakup form in the experiment, the Euler-Lagrange multiphase flow model method was proposed to simulate the flow field in the nozzle and the breakup process after injection, to investigate the characteristics of the flow field in the nozzle during the movement of nozzle-needle valve, and to explore the relationship between the velocity distribution of the spray droplets and the flow in the nozzle. The coupled model and the experimental phenomenon have good compatibility in terms of spray characteristics and liquid core structure, as verified by the visualized spray experimental platform built in a low spray pressure environment. The analysis concludes that the internal disturbance of the nozzle plays an important role in the breakup of the spray, and the intensification of the disturbance leads to the more suitable use of the discrete droplet model after the injection. 8. 联合电休克治疗对抑郁症患者 复发及认知功能影响的5年随访研究.

- Abstract
Objective: To investigate the effects of modified electroconvulsive therapy (MECT) in combination with medication and medication alone on relapse and cognitive functioning in patients with major depressive disorder (MDD). Methods: Totally 324 patients meeting the diagnostic criteria ofthe DSM-5 for MDD were selected and divided into combined MECT group (n = 117) and non-combined MECT group (n=207). At baseline and 5 years of follow-up, both groups completed the General Information Questionnaire, Montreal Cognitive Assessment (MOCA), and Hamilton Depression Scale (HAMD) to analyze associations between cognitive functioning and multiple variables. Results: At baseline, there were no significant differences between the two groups on general information, MoCA, and HAMD scores (Ps >0.05). After 5 years of follow-up, the relapse rate was lower in the combined MECT group than in the un-combined MECT group [(37.6% vs. 72.0%), P<0.001]. MECT was a protective factor against recurrence of MDD [RR(95% CI) = 0.52 (0.40-0.66)], and the MoCA scores were lower in the combined MECT group than in the un-combined MECT group [(20.9±3.5) vs. (23.6 ±4. 2), β = – 1.93, P <0.001]. Conclusion: Combined MECT is a protective factor against MDD recurrence. Its effects on the cognitive functioning of MDD patients may have long-term effects. [ABSTRACT FROM AUTHOR]

BACKGROUND: This review explores the current research status and frontier hot spots of functional magnetic resonance imaging (fMRI) in the field of traditional Chinese medicine (TCM) for the treatment of ischemic stroke, and attempts to grasp future research trends, with a view to providing a reference for subsequent research in this field. OBJECTIVE: To visualize and analyze the hotspots and frontiers in the TCM treatment of ischemic stroke based on fMRI using CiteSpace knowledge mapping combined with binary logistic regression equations, in order to grasp the future research trends and further explore the distribution of brain regions with abnormal neural activity related to the types of post-stroke dysfunction. METHODS: CNKI, WanFang, VIP, Chinese Biomedical Literature Database and Web of Science core set database were searched. CiteSpace was used to plot keyword co-occurrence, keyword clustering timeline, burst term detection, co-cited literature mapping to analyze hotpots and frontiers in this field. Binary logistic regression analysis fitted the distribution of brain regions with abnormal neural activity associated with different dysfunction after ischemic stroke. RESULTS AND CONCLUSION: A total of 354 articles were included for CiteSpace knowledge mapping analysis. The number of annual publications showed that the research popularity has been raised from 2000 to 2022 with a good development prospect, but the core strength is mainly concentrated in China. Keywords co-occurrence and clustering time line analysis showed that aphasia, hemiplegia and cognitive impairment are the hot poststroke dysfunction types. Electroacupuncture, acupuncture and head acupuncture are hotspot intervention measures. Functional connectivity is a hotspot analysis method, and resting fMRI is a hotspot scanning technology. The time span of each research hotspot is long, indicating that it has a certain research value and the relevant research is gradually deepening, promoting the research progress in this field. However, acupuncture is the main intervention measure, and there is a lack of research on traditional Chinese medicine, Chinese patent medicine, acupuncture and medicine combination and other TCM therapy. Burst term detection results showed that functional connectivity, graph theory, degree centrality, default mode network, randomized controlled trials have great influence and strong explosive power. They are the current and future frontier hot spots in this field, suggesting that future research should focus on the brain network information integration and strengthen the scientific and rigorous clinical trial design. The results of co-cited literature analysis showed that the epidemiological investigation of ischemic stroke, the safety and effectiveness of acupuncture in the treatment of stroke, the brain activation patterns under different tasks, and the neuropathological mechanism of brain network dysfunction after stroke are the theoretical basis of this field. Future research direction in this field is to explore TCM-targeted brain regions and neural networks to reveal the brain effect mechanism of TCM promoting neural remodeling after stroke. A total of 255 articles were included for binary Logistic regression analysis. The results showed that sensorimotor cortex and premotor area dysfunction are positively correlated with the incidence of motor dysfunction after stroke; hippocampus, cerebellum posterior lobe, precuneus, inferior temporal gyrus and anterior cingulate nerve dysfunction are positively correlated with the incidence of cognitive impairment after stroke; cuneus, angular gyrus and prefrontal lobe neural dysfunction were positively correlated with the incidence of affective disorder after stroke; anterior cingulate, cerebellum posterior lobe neural dysfunction are positively correlated with the incidence of swallowing disorder after stroke. The above brain regions are the core brain regions of the sensorimotor network, default mode network and reward loop, suggesting that functional abnormalities within or between brain networks related to dysfunction may be potential target areas for TCM intervention, but the specific changes in neural activity activation or inhibition still need to be refined. As a cheap and abundant renewable raw material, lignocellulosic biomass can be converted into cellulosic fuel ethanol through pretreatment, enzymatic hydrolysis and microbial fermentation, etc., which has widely attracted attention in recent decades. Poplar is a kind of fast-growing hardwood that is widely artificially cultivated and is mainly used in papermaking industry. Poplar is considered as an excellent lignocellulosic material for the production of cellulosic ethanol due to its high contents in cellulose and hemicellulose components. The application of poplar in cellulosic ethanol production was focused on in this paper, including briefly introducing the composition and structural characteristics of poplar, and importantly overviewing the research progress of poplar in pretreatment technology, enzymatic hydrolysis of pretreatment raw materials and microbial fermentation and so on. Finally, the technical obstacles and difficulties restricting the application of poplar in cellulosic ethanol industry were summarized, and the corresponding countermeasures were analyzed and the application were prospected. - Abstract
D-S (Dempster-Shafer) theory of evidence is an effective method to deal with uncertain information, but there is still a problem of conflict between evidence. Maxwell’s electromagnetic force theory was introduced to convert the problem of conflict and fusion between different evidences in evidence theory into the problem of electromagnetic repulsion and electromagnetic gravity under electromagnetic force theory, and this conversion proposed a new method for quantifying the distance between evidences. Among them, the evidence distance factor in the traditional conflict coefficient was introduced and a new method was proposed for representing the conflict of evidence, which improved the sensitivity of the conflict coefficient. The simulation results show that the recognition rate of the method in fault diagnosis is higher than the traditional method, which prove the correctness and effectiveness of the proposed method. - Abstract
Objective To investigate the relationships between serum phosphorylated Tau-181 (pTau181), lipopro⁃ tein-associated phospholipase A2 (Lp-PLA2) and vascular dementia (VD) in patients with acute cerebral infarction (ACI) . Methods Totally 163 patients with ACI were divided into the VD group (60 cases) and non-VD group (103 cases) accord⁃ ing to whether VD occurred or not. In the VD group, 15 patients had mild cognitive impairment, 32 patients had moderate cognitive impairment, and 13 patients had severe cognitive impairment. On the next day of enrollment, we collected all sub⁃ jects' fasting elbow vein blood in the morning, and centrifuged the upper serum. The serum pTau181 was detected by ELISA, and serum Lp-PLA2 was detected by up-forwarding light method. Multivariate Logistic regression model was used to analyze the risk factors of VD in patients with ACI, and the predictive value of serum pTau181 and Lp-PLA2 levels for patients with ACI complicated with VD was analyzed using the receiver operating characteristic (ROC) curve. Results The serum pTau181 and Lp-PLA2 levels in the VD group were significantly higher than those in the non-VD group (both P<0. 05) . With the aggravation of cognitive impairment, the serum pTau181 and Lp-PLA2 levels in patients with ACI complicated with VD gradually increased (both P<0. 05) . Multivariate Logistic regression analysis showed that type 2 diabetes mellitus, pTau181 and Lp-PLA2 were all risk factors of VD in ACI patients (all P<0. 05) . The ROC curve analysis showed that the area under the curve of serum pTau181 combined with Lp-PLA2 in predicting VD in patients with ACI was significantly higher than that of serum pTau181 and Lp-PLA2 levels alone (both P<0. 05) . - Abstract
Objective To observe the effects of butyphthalide on the differentiation and autophagy agonism in the differentiation of adipose-derived stromal cells (ADSC)into neuronal-like cells. Methods ADSC was divided into the β-mercaptoethanol group, β-mercaptoethanol + butylphthalide group, and β-mercaptoethanol + rapamycin group after ADSC was pre-induced for 24 h by 1 mmol/L β -mercaptoethanol. Cells in the β -mercaptoethanol group were added with 5 mmol /L β -mercaptoethanol, β -mercaptoethanol + butyphthalide group with 5 mmol/L β -mercaptoethanol and 10 μmol/L butylphthalide, and β-mercaptoethanol + rapamycin group with 5 mmol/L β-mercaptoethanol and 200 μg/L rapamycin. In the pre-induction, and at 1, 3, 5, and 8 h of induction, cell morphology changes were observed using inverted phase contrast microscopy, and the cell differentiation rate was calculated. At 1 and 5 h of induction, the ultrastructure of cells was observed under transmission electron microscopy, the numbers of autophagosomes and autophagy lysosomes were counted. At 1, 3, 5, and 8 h of induction, the cell survival rate was detected by MTT. At 5 h of induction, neuronspecific enolase (NSE) and neuorfliamne-200 (NF-200)were detected by immunocytochemistry in each group. At 1, 3, 5, and 8 h of induction, the LC3 protein in each group was detected by immunocytochemistry. Results The morphology of cells was similar in the three groups, the differentiated cells showed a typical neuron-like cell morphology. The nuclei became round and big, the staining was shallow, and they had a large amount of euchromatin but little heterochromatin. The cytoplasm of some neuronal-like cells had a large number of outstretched endoplasmic reticulum, mitochondrial edema, volume increasing and cristae rupture; and double-membrane autophagosome appeared. Over the course of induction, the differentiation rate and expression rate of LC3-positive cells in each group increased significantly (both P<0. 05)and reached a peak at 5 h of induction. Compared with the β -mercaptoethanol group, the β -mercaptoethanol + rapamycin group and the β-mercaptoethanol + butyphthalide group had higher differentiation rates and expression rates of LC3-positive cells (all P<0. 05). Compared with the β-mercaptoethanol + rapamycin group, the cell differentiation rates and the positive expression rates of LC3 significantly increased at 1 and 3 h of induction, and significantly decreased at 8 h of induction in the β-mercaptoethanol + butylphthalatecline group (all P<0. 05). The numbers of autophagosomes and autophagic lysosomes in each group at 5 h were larger than those at 1 h of induction (all P<0. 05). The numbers of autophagosomes autophagic lysosomes in the β-mercaptoethanol + butyphthalide group and β-mercaptoethanol + rapamycin group were larger than those in the β-mercaptoethanol group (all P<0. 05). Over the course of induction, the cell viability of each group decreased significantly (P<0. 05). Compared with the β-mercaptoethanol group, the cell differentiation rates of the β-mercaptoethanol + rapamycin group and the β-mercaptoethanol + butylphthalide group increased significantly (P< 0. 05). Compared with the β-mercaptoethanol + rapamycin group, the cell differentiation rates of the β-mercaptoethanol + butyphthalide group were significantly reduced at 1, 5 and 8 h of induction (all P<0. 05). Compared with the β-mercaptoethanol group, the positive expression rates of NSE and NF-200 in the β-mercaptoethanol + rapamycin group and β-mercaptoethanol + butylphthalide group significantly increased (both P<0. 05). Compared with the β-mercaptoethanol + rapamycin group, the positive expression rate of NSE in the β-mercaptoethanol + butyphthalide group significantly increased (P<0. 05). Conclusion In the differentiation of ADSC into neuronal-like cells, butyphthalide promotes cell differentiation and improves cell survival by increasing autophagic agonism such as increasing the number of autophagosomes and autophagic lysosomes. At present, in the research of Chinese text emotion analysis, the method based on semantic rules and emotion dictionary usually needs to set the emotional threshold manually. However, the method based on deep learning can’t fully extract emotional features because it fails to use language knowledge such as semantic rules and emotional dictionary. As to shortcomings of two methods, a text emotion analysis method combining language knowledge and deep learning was proposed. Firstly, the key emotional segments in the text were extracted according to the semantic rules. Secondly, more explicit emotion words were extracted from the key emotional segments according to the emotional dictionary to construct the emotion set. Thirdly, the deep level features were extracted from the original text, key emotional segments and emotional set by using the deep learning model. Finally, the features were weighted and fused, and the classifier was used to judge the emotional polarity. The experimental results show that compared with the deep learning model without language knowledge, this method has significantly improved the ability of emotional polarity classification. 目的 观察经肛门内镜微创手术 (TEM) 后病理分期pT1期高危和pT2期直肠癌追加同步放化疗 (CCRT) 的临床效果. 方法 接受TEM治疗且术后病理分期为pT1期高危或pT2期的直肠癌患者69例, 根据TEM术后是否 追加CCRT治疗分为TEM + CCRT组23例, TEM组46例. TEM + CCRT组患者在TEM术后追加CCRT治疗, TEM组 患者在 TEM 术后不进行治疗. 两组患者均随访至今或死亡, 随访时间 8~201个月, 观察两组患者的复发率, 死亡 率, 无病生存期 (DFS), 总生存期 (OS). 结果 TEM + CCRT组死亡率为0, TEM组死亡率为15. 21%, 两组死亡率相 比, P>0. 05. TEM + CCRT组复发率为4. 34%, TEM组复发率为34. 78%, 两组复发率相比, P<0. 05. TEM + CCRT组 DFS为 93 (64. 0, 139. 0) 个月, TEM 组 DFS为 44. 5 (23. 5, 83. 2) 个月, 两组 DFS相比, P<0. 05. In order to make the rapid and large-scale propagation of fast-growing varieties of Pinus elliottii, the induction and proliferation of embryogenic callus were studied. The immature zygotic embryos (with endosperm) of ten P. elliottii families were used to systematically study the effects of different factors on the induction efficiency of embryogenic callus, including genotype, the developmental stage of zygotic embryos, basic medium, types and concentrations of plant growth regulator ( PGR) . At the same time, the proliferation conditions of embryogenic callus were discussed. The esults were as follows: The genotype, the developmental stage of zygotic embryos, basic medium and types and concentrations of plant growth regulator (PGR) all had different degrees of influence on the induction of embryogenic callus. Embryogenic callus were induced in all ten genotypes, of which the induction rate of Genotype 2 was the highest, reaching 25.37%; In the stage of zygotic embryo development, the induction rate of cones at the multiple embryo stage was the highest; Among the four basic media, the induction effect on DCR basic medium was the best; The induction rate of 2,4-D 2.0 mg L+KT 2.5 mg L-1 was the highest, reached 27.78%. The proliferation of embryogenie callus for P. elliottii was achieved on DCR medium, supplemented with 2,4-D (0.5 mg L-1), KT (1.0 mg - L-1), CH (500 mg L) and Gin (300 mg L-1).
