BACKGROUND: At present, the biological functions and molecular changes of bone marrow mesenchymal stem cells in the tumor microenvironment of acute myeloid leukemia are still unclear. OBJECTIVE: To explore the changes in the biological function of bone marrow mesenchymal stem cells in acute myeloid leukemia and the role of acute myeloid leukemia conditioned medium by bioinformatics and experiment. METHODS: Differential genes were screened from GEO data sets, and enrichment analysis was performed. The protein-protein interaction network was constructed and the Hub gene was obtained. Bone marrow mesenchymal stem cells from patients with acute myeloid leukemia and healthy donors were cultured. Bone marrow mesenchymal stem cells from healthy donors were treated with acute myeloid leukemia conditioned culture solution. Each group was subjected to the adipogenic differentiation, osteogenic differentiation, staining of β-galactosidase, detection of the cell cycle, and validation of Hub genes. RESULTS AND CONCLUSION: (1) Gene expression data of bone marrow mesenchymal stem cells from acute myeloid leukemia patients and healthy donors were obtained from GSE84881, and 184 up-regulated genes and 140 down-regulated genes were screened. (2) The biological functions of enrichment mainly include cell cycle, adipocyte differentiation, cell metabolism, and MYC pathway. According to the Degree algorithm, 10 up-regulated Hub genes and 10 downregulated Hub genes were selected. (3) The cell in vitro experiment found that: compared with the control group, the surface antigen of acute myeloid leukemia mesenchymal stem cells did not change, but it showed enhanced lipid differentiation ability, weakened osteogenic differentiation ability, increased β-galactosidase positive cell number, altered cell morphology, arrested cell cycle, increased LGALS3 expression, and decreased MYC expression. Mesenchymal stem cells from healthy donors showed similar changes after being cultured in acute myeloid leukemia conditioned medium. (4) The results show that biological function of mesenchymal stem cells is altered in the acute myeloid leukemia microenvironment, which provides new insights into the interaction between mesenchymal stem cells and tumor cells. Global climate models and emerging artificial intelligence models generate big climate forecasts data for catchment hydrological forecasting at daily, sub-seasonal and seasonal timescales. The utilization of global climate forecasts to drive catchment hydrological models are confronted with the technical issues of climate forecast data retrieval, hydrological forecasting model set-up and verification of hydro-climatic forecasts. Starting with international collaborative research projects on global climate forecasting, this paper conducts a survey of short- term weather forecasts for the next 1 day to 2 weeks, sub- seasonal climate forecasts for the next 1 to 60 days, seasonal climate forecasts for the next 1 to 12 months and artificial intelligence-based climate forecasts. Furthermore, the processes of catchment hydrological forecasting driven by global climate forecasts are illustrated by detailing the technical aspects on the calibration of climate forecasts, the setting- up of hydrological models and the verification of predictive performance. By generating real-time and retrospective catchment hydrological forecasts from global climate forecasts, the efficacy of forecasting models can be quantitatively examined by verifying forecast skill at different lead times, laying a solid basis for practical forecasts- based operations of hydraulic infrastructure. The mixture fraction is a conserved scalar characterizing the fuel-air mixing. As a key reference scalar for turbulent combustion modelling, its spatial distribution is usually obtained through 3D numerical simulation, which are, however, time-consuming and costly for combustors with complex geometries. To overcome such low efficiency in the iterative designing process, a low-order model was developed based on the Gaussian plume function to compute the mixture fraction field in the swirl combustor to accelerate the evaluation of the fuel-air mixing strategy and the parameterized design process. Compared with the conventional formulation, the derived new Gaussian plume function includes the effects of convection and corrections due to swirl flows. A mirror image reflection model was further developed to simulate the wall-plume interactions, together with the relevant corrections to ensure mass conservation. This newly derived Gaussian plume model was applied to the low-older prediction of the mixture fraction field in a methane swirl combustor. Based on the database generated through 3D numerical simulations, the model parameters were optimized with the least square method first. The prediction accuracy under broad working conditions was demonstrated. This study not only provides a novel approach for quick predictions of mixture fractions in swirl combustors, but also sets an instance for further development and application of the Gaussian plume model. With the prevalence of the COVID-19 and the improvement of personal health protection awareness the development of excellent wearable personal protective materials is of great practical significance. The traditional mask filter element is made of micron polypropylene melt-blown nonwoven fabric which has the disadvantages of low filtering efficiency for small particles in the air and poor protection against bacteria and viruses. In recent years the research of electrospun nanofiber membranes in the field of air filtration and personal protection has attracted great attention due to the characteristics of large surface area high porosity interconnected pore structure and controllable diameter. However most of the filter element membranes developed at present have undiversified functions and do not have antibacterial and comfortable properties so they cannot meet people s requirements for the versatility and skin-friendly comfort of filter materials. To improve the antibacterial property wearing comfort and protection ability against small inhalable particulate matter PM 2. 5 of the filter element membrane a bilayer hydrophilic-hydrophobic antibacterial nanofiber filtration membrane was constructed by electrospinning technology through silk protein SF with biocompatibility oxygen and air permeability and polyvinylidene fluoride-hexafluoropropylene copolymer PVDF-HFP with excellent mechanical properties as raw materials. Firstly PVDF-HFP nanofiber hydrophobic membranes were prepared by electrospinning with the concentration of PVDF-HFP solution in the range of 18% and 24% and the suitable concentration of spinning solution was selected. Secondly AgNO3 solution with different contents was mixed with SF solution and the SF nanofiber membrane loaded with silver ion was prepared by electrospinning. After UV reduction SF-AgNPs nanofiber hydrophilic membranes loaded with silver nanoparticles AgNPs were obtained. Finally a series of hydrophilic-hydrophobic bilayer composite nanofiber membranes with a certain thickness ratio were obtained by electrospinning SF nanofiber membranes with different thicknesses on the surface of PVDF-HFP electrospun fiber membranes with fixed thickness the hydrophobic base layer. The effects of spinning solution concentration on surface morphology fiber diameter distribution and hydrophilicity hydrophobicity of nanofiber membranes were characterized by SEM and water contact angle tests. The reduction of silver nanoparticles was successfully demonstrated by TEM EDS and XRD. The effect of silver nanoparticles on the structure of silk fibroin nanofibers was characterized by FTIR and XRD. In addition we tested the water vapor transmission rare filtration properties and antibacterial properties of the hydrophilic and hydrophobic bilayer composite nanofiber membranes prepared with different thickness ratios and different contents of AgNO3 in spinning solution. The results show that when the membrane thickness is in the range of 120 μm and 145 μm the hydrophobic layer thickness of PVDF-HFP is 35 ± 5 μm the filtration efficiency of PM 2. 5 reaches 99. 00% the bacteriostatic rate of Staphylococcus aureus reaches 99. 76% and the water vapor transmission rare reaches 7.80 mg/ (cm². h) which is higher than the transmission value of YY0469-2011 common medical masks. The SF-AgNPs/ PVDF-HFP bilayer nanofiber membranes prepared by electrospinning technology have improved the filtration performance of small particulate matter (PM 2. 5). By UV reduction silver nanoparticles were loaded on SF nanofibers in situ which endowed the composite fiber membranes with excellent antibacterial properties. Based on the hydrophobicity of PVDF-HFP nanofiber membranes and the hydrophilicity of SF-AgNPs nanofiber membranes the hydrophilic-hydrophobic bilayer composite membranes have one-way water vapor transmission function which improves the comfort of the filter membranes. This study extends the application of silk protein in medical filtration membranes and directional moisture removal fabrics paving the way for the preparation of fiber materials with high efficiency air filtration high antibacterial activity and high water vapor transmission rare. Reservoir impoundment has changed the temporal and spatial distribution of water temperature and caused ecological problems, such as ecosystem disorder and low- temperature underwater discharge in reservoir areas. This resulted in adverse effects on the growth and reproduction of indigenous fish. The 3-D water temperature model was established using the Delft3D model. The dynamic change process of water temperature in the transverse, longitudinal, and vertical dimensions of the Nuozhadu Reservoir under typical meteorological and hydrological conditions was simulated. Considering this context, and combined with the ecological water temperature demand of downstream fish, a layered water intake scheme of stacked beam doors with ecological water temperature as the goal was formulated. The most suitable location and time for releasing indigenous fish ( Tor sinensis) in Nuozhadu Reservoir were determined by combining the requirements of the species involving breeding and release. It was shown in the results that there are differences in the layered water intake scheme of stop logs in different years and the suitable location and time for proliferation and release. The universal layered water intake rules for stop logs were formulated based on the water temperature demand in typical years and the engineering conditions of stop logs. The most suitable location for placing Chinese snakehead fry was the Xiaoheijiang tributary, preferably from August to September. A scientific basis for developing a reservoir ecological dispatching strategy to alleviate the water temperature imbalance may be provided by the results of this study. [ABSTRACT FROM AUTHOR]
