The bridge structure of bridge crane is a typical welded structure, and fatigue cracks often appear in the weld area of the main beam during daily service. In order to ensure the service safety of the crane, a multi-level sub model of the dangerous crack region was established based on the multi-scale finite element analysis method. The thermal structural coupling welding finite element model was established on the secondary sub model, and the whole welding process was simulated and analyzed. The influence of post weld heat treatment on residual stress was simulated. Combined with the multiaxial critical interface method, a fatigue analysis method using the strain amplitude as the multiaxial damage parameter was constructed, and the fatigue performance of the dangerous crack region was evaluated. The results show that the proposed method transforms the overall fatigue problem of the structure into the fatigue problem of the local welding position, which not only increases the calculation efficiency, but also considers more influencing factors of the fatigue details, and provides a certain reference for the fatigue life assessment and life extension of the crane weld. Bromodomain-containing protein 4 (BRD4) is a member of the bromodomain and extraterminal domain family, playing a crucial role in the regulation of gene transcription, cell proliferation, apoptosis, and inflammation. BRD4 has been closely linked to various diseases, including cancer, neurological disorders, and viral infections. Although BRD4 has obtained significant attention in the field of cancer research, limited investigation has been conducted regarding its implications in bone-related diseases, encompassing osteoarthritis, intervertebral disc degeneration, osteoporosis, and bone tumors, among others. Recent research has indicated the involvement of BRD4 in the pathogenesis of bone-related diseases, highlighting its significant role. Therefore, this review summarizes the research results of BRD4 and its inhibitors in bone-related diseases in recent years. By expounding the structure and function of BRD4 and the role of BRD4 and its inhibitors in bone-related diseases, it is suggested that BRD4 may be a potential target for the treatment of bone-related diseases. Logs record the software and hardware running information. By viewing system logs, you can find system faults and causes to ensure system stability and normal running. The purpose of log parsing is to parse semi-structured original logs into readable log templates. The existing parsing methods often only focus on parsing the original logs, but ignore the later template processing. As a result, the accuracy of the results cannot be further improved. Since then, this paper proposed a new log parsing method FMLogs. This method designed regular expressions and adjusted threshold parameters to obtain the best performance. Meanwhile, it used character-level frequency statistics and MinHash methods to merge log templates with the same length and different log templates. FMLogs conducted extensive experiments on seven real datasets, achieving an average analytic accuracy of 0.924 and an F1-score of 0.983. The experimental results show that FMLogs is an effective log parsing method with high accuracy, high efficiency, and stable performance. [ABSTRACT FROM AUTHOR]

Objective To evaluate the role of autophagy in the treatment of neuropathic pain (NP) with hydrogen-rich saline. Methods Forty adult male Sprague-Dawley rats with successful intubation were randomly divided into 5 groups (n= 8) using a random number table: the sham operation group (group S), the neuropathic pain group (group C), the hydrogenrich saline group (group H), the autophagy inhibitor group (group M) and the hydrogen-rich saline + autophagy inhibitor group (group HM). There were 8 rats in each group. The NP model was established by chronic constriction of the sciatic nerve (CCI) in rats. The autophagy inhibitor 3-methyladenine (3-MA) was intraperitoneally injected with 30µg/kg in the group M and the group HM. The hydrogen-rich saline (0.6 mmol/L) was intraperitoneally injected with 10 mL/kg in the group H and the group HM. The other groups were intraperitoneally injected with the same amount of normal saline twice a day for 7 consecutive days. Paw withdrawal threshold to mechanical stimulation (MWT) and paw withdrawal latency to thermal stimulation (TWL) were measured at 1 day before and 1, 3, 5, 7 and 14 days after modeling (T0-T5). After the last measurement of pain threshold, the L4-L6 segment of spinal cord was removed for determination of the expression of autophagy-related proteins microtubule-associated protein light chain 3 (LC3) Ⅱ, Beclin-1 and p62 proteins by Western blot assay. The expression levels of superoxide dismutase (SOD) and malondialdehyde (MDA) in spinal cord tissue were detected. Results Compared with the group S, MWT and TWL were decreased in the group C at T2-5, the expression levels of LC3 Ⅱ, Beclin-1 and p62 were increased, SOD activity was decreased, and MDA content was increased at T5 (P＜0.05). Compared with the group C, MWT and TWL were increased in the group H at T2-5, LC3 Ⅱ and Beclin-1 protein expression levels were increased, p62 protein expression levels were decreased, SOD activity was increased, and MDA content was decreased at T5 (P＜0.05). MWT and TWL were decreased in the group M at T2-5, LC3 Ⅱ and Beclin-1 protein expression levels were decreased, p62 protein expression levels were increased, SOD activity was decreased, and MDA content was increased at T5 (P＜0.05). Compared with the group M, MWT and TWL were increased in the group HM at T2-5, LC3 Ⅱ and Beclin-1 protein expression levels were increased, p62 protein expression levels were decreased, SOD activity was increased, and MDA content was decreased at T5 (P＜0.05). Conclusion Hydrogen-rich saline can alleviate neuropathic pain and inhibit oxidative stress in spinal cord in rats, and the mechanism may be related to the increase of autophagy.
