The mutations of survival motor neuron (SMN) gene result in spinal muscular atrophy (SMA), a common neurodegenerative disease. Some of the motor neurons undergoing cell death is the predominant characteristic in SMA pathology. However, the viability and sensitivity to stresses of other cell types also need to be determined. In this article, we established HeLa stable cell line with inducible SMN knockdown to study its viability and sensitivity to oxidative stress. SMN knockdown in the HeLa stable cell line was induced by doxycycline. The proliferative and survival rates of SMN knockdown cells with or without hydrogen peroxide (H2O2) treatment were determined. Our results showed that the proliferative rate of SMN knockdown cells decreased only slightly compared with that of the cells without doxycycline treatment. In contrast, after H2O2 reached certain concentrations, the survival rate of SMN knockdown cells decreased significantly. Our data indicate that SMN knockdown alone is not critical to cell viability. However, when SMN knockdown cells are under stress, such as oxidative stress, their survival rate may significantly decrease. Our results will be helpful to prevent the detrimental effect caused by the cell death of non-motor neurons under stress in SMA patients. In addition, the cell model we established can be used to study the mechanism and screen drugs to prevent the detrimental effects in cases of SMA disease.

The traditional maintenance support capability system is difficult to meet the dynamic and complex maintenance support needs. By analyzing the factors and relationships generated by the actual combat maintenance and support capability of aerospace equipment, the idea of constructing a capability system based on the decomposition of maintenance support activities is proposed, and the improved gray correlation analysis method is used to optimize, and the optimized maintenance support capability system is obtained. 6. 葡萄耐铝毒基因 STOP1 的克隆与表达分析.

[Objective] STOP1 is an important gene for tolerance to aluminum (Al) -toxicity in plants. The purpose of the present study was to explore the relationship between the structural characteristics, expression level of STOP1 gene and its tolerance to Al in grape. [Method] Leaf cDNA of Vitis amurensis with weak tolerance to Al and V. sinocinerea with strong tolerance to Al were used as reference materials. Sequences of two genes VaSTOP1 and VsSTOP1 were obtained by PCR and TA cloning technology, and then bioinformatics analysis and expression analysis under Al stress were carried out. The membrane lipid peroxidation indices under Al stress were determined. Finally, the relationship between the expression of STOP1 genes, the changes of membrane lipid peroxidation of two species and tolerance to Al were compared. [Result] The sequences of VaSTOP1 and VsSTOP1 were 1782 and 1800 bp respectively, encoding 594 and 600 amino acids. Their relative molecular weights were 67. 03 and 67. 48 kD, and the theoretical isoelectric points were 5. 21and6. 42, respectively. The secondary structure in two species were the law of irregular curl > α-helix > folding extension chain, and the tertiary spatial structure were composed of zinc protein, α-helix, β-folded sheet and β-rotation angle. The phylogenetic tree analysis found that the homology between VaSTOP1 and VsSTOP1 was as high as 96%, and the homology between them and CcSTOP1, PnSTOP1, NtSTOP1 and ErSTOP1 was more than 80%. The expression of VaSTOP1 gene increased first and then decreased within 28 days under Al stress, but the peak value appeared earlier, whereas the VsSTOP1 gene expression increased steadily, which was basically consistent with the change law of membrane lipid peroxidation. [Conclusion] The tolerance to Al of Vitis was positively correlated with the expression of STOP1 gene, which provided a reference for further exploring the molecular regulation mechanism of Vitis tolerance to Al. 14. 铁、钼营养对毛苕子苗期根瘤及植株生长生理的影响.

- Abstract
[Objective] The puipose of this study was to explore the effect of ferrum, molybdenum nutrition on root nodule and plant physiological growth of Vicici villosa in seedling stage. [Method] A local cultivar, Vicia villosa from Qujing city, Yunnan province was used as experiment materials using the method of no soil cultivation, adding 0.01 mmol/L sodium molybdate and 0.2 mmol/L ferrous sulfate respectively or simultaneously in the Hoaglands nutrient solution of removal ferrum and molybdenum salt. The effects of ferrum, molybdenum nutrition on root nodule and growth physiology of Vicia villosa seedling stage were studied. [Result] Ferrum and molybdenum significantly increased root nodule number and weight, and the effect of separate molybdenum treatment was the best, in the 105 days after sowing the root nodule number and weight of separate molybdenum treatment were higher than that of control nearly 100 % and 150 %. Ferrum and molybdenum nutritions made the stem length and biomass of Vicia villosa had varying degrees of decreased, and root length and root/shoot ratio increased significantly, stem length with ferrum treatment and composite treatment reduced to a greater extent than that with molybdenum treatment, and root length with molybdenum treatment increased maximum. After the application of the ferrum and molybdenum fertilizer, the contents of ferrum and molybdenum significantly improved, and the content of nitrogen, soluble protein and free amino acid also had varying degrees of increased, the separate application of ferrum increased greatly, and phosphorus content had different degrees of decreased, the biggest drop was single ferrum fertilization treatment in Vicia villosa. Apply ferrum and molybdenum fertilizer after 90 days, the soluble sugar content decreased significantly, and appling molybdenum fertilizer reduced the starch content, and application ferrum fertilizer increased the starch content. In addition, the study also found that the application of ferrum, molybdenum fertilizer decreased the SOD activity and enhanced the POD activity in Vida villosa. [Conclusion] The application offerrum, molybdenum fertilizer could promote the root nodule and plant growth. [ABSTRACT FROM AUTHOR]

18. 基于叶片解剖特征分析三种无患子科果树的亲缘关系.

- Abstract
Ten germplasms of three Sapindaceae fruit trees were analyzed by their leaf transection and epidermal structure,using paraffin method and tissue segregation procedure. On this basis, genetic relationship among these 10 germplasms was examined preliminarily by cluster analysis in SPSS. The results showed that the leaves of 10 accessions were bifacial type,and their leaf transection structures all consists of epidermis, mesophyll and vein. The midvein thickness had significantly different as longan was the greatest in three fruit trees, longli in the middle and litchi was the least. Midvein transverse of litchi was rounded triangular, longan approximate semicircle, and longli was approximately flat circular. Among the difference materials, the range of leaf thickness, upper and lower epidermis, palisade tissue and sponge tissue were 175.23-318.84, 11.18-25.13, 7.49-20.43, 50.01-124.59 and 84-173.64 μm, respectively. Palisade tissue cell had 2-3 layers in all materials. In addition, VDP, P/S, RT and RL were 2.65-5.77, 0.52-0.82, 28.89%-39.95% and 44.89%-55.57%, respectively. Epidermal cell of litchi was small, polygon, anticlinal wall arc, lower epidermal without trichomes, and stoma was long oval; epidermal cell of longan was big, irregular, anticlinal wall sinuate, lower epidermal with trichomes, and stoma was oval or suborbicular; epidermal cell morphology of longli was similar with longan, but its anticlinal wall was corrugated. The cluster analysis found that 10 accessions were firstly divided into 2 categories, which are Litchi and Dimocarpus. And then the Sanyuehong and longli were separated as subgroup in each category.
