The geochemical characteristics and genesis of selenium are of increasing interest in soil geochemistry research, as there exist fundamental problems in the development of selenium-enriched agricultural products. Therefore, this study performed a comprehensive analysis of data statistics using 43185 surface soil samples obtained in southeast Guizhou. The results showed that the geometric mean of selenium in the soil is 0.49 μg/g; the mean value of selenium in the black shale of cultivated soil is 1.55 μg/g, which is > sandstone (siliceous; 1.22 μg/g) > carbonaceous limestone (0.66 μg/g) > phyllitic slate (0.60 μg/g) > mudstone (0.58 μg/g) > dolomite (0.54 μg/g) > diabase (0.52 μg/g) > granite (0.48 μg/g) > variegated sand (gravel; 0.47 μg/g) > limestone (0.43 μg/g) > slate and metamorphic tuffite (0.42 μg/g) > purple sandstone (0.32μg/g) > mud, gravel, and sand (0.27μg/g). This indicates that the average content of selenium in soil is closely related to the parent rock. Meanwhile, combined with heavy metal elements in the area of an arable land soil pollution risk assessment, the cultivated land with black shale, sandstone (siliceous), carbonaceous limestone, mudstone, and dolomite was divided into high-grade and low-grade selenium-rich potential areas. The germanium content and quality grade characteristics of germanium-rich soil in cultivated land of the Qiandongnan prefecture were determined in this study through the investigation and evaluation of the surface soil geochemistry and the material sources and causes of germanium-rich soil were discussed. The results obtained were as follows: (1) the germanium content in the surface soil of the area was 0.13–9.21 mg/kg, and the arithmetic average was 1.54 mg/kg. (2) The germanium-rich farmland in the area was 1794.30 km², accounting for 40.17%. The environmental comprehensive grade of germanium-rich farmland is mainly for priority protection with an area of 1167.35 km², accounting for 65.06%. The safe use category is second with an area of 593.38 km², accounting for 33.07%. The strictly controlled farmland is less with an area of 33.57 km². (3) The main source of germanium in the soil is hydrothermal action or hot water activity that causes germanium to intrude and accumulate on the Earth’s surface. Ge is positively correlated with the soil Pb and Zn content, negatively correlated with N and organic matter, and weakly and positively correlated with other elements, suggesting that Ge is less affected by man-made materials. The copy number variation sequencing (CNV-Seq) technique can be used to explore the fetal marker chromosome and its possible source in the prenatal diagnosis. With this technique, we found a case of Pallister-Killian syndrome (PKS). We discuss the clinical and genetic characteristics of PKS. High-throughput sequencing of free DNA in the peripheral blood of pregnant woman, followed by amniocentesis, whole -genome copy number variation analysis of fetal DNA, and karyotype analysis of amniotic fluid cells to confirm PKS syndrome. A duplication of the most whole short arm of chromosome 12 was detected by CNV-seq in the fetal. The amniotic fluid karyotype was 47,XY,+i (12) (p10) [58]/46,XX [42]. CNV-seq as a first-line prenatal diagnostic technique can effectively find PKS, when combined with clinical features, karyotyping and other techniques. [ABSTRACT FROM AUTHOR]

Objective: To investigate the gene mutations of seven families suspected with urea cycle disorders (UCD), and to perform prenatal diagnosis for two reproductive families. Methods: Target sequence capture combined with high -throughput next -generation sequencing, and Sanger sequencing/LD-PCR/MLPA (multiplex ligation-dependent probe amplification) technologies were used to investigate the gene mutations for seven families suspected with UCD. After the genotypes of the pedigrees were identified, two amniotic fluid samples from highrisk pregnant women were used for prenatal genetic diagnosis. Results: The diagnosis of UCD was confirmed in seven suspected families. Among of them, four patients were diagnosed as the neonatal intrahepatic cholestasis caused by citrin deficiency (NICCD) and one of them was coexist with spinal muscular atrophy (SMA). One patient was diagnosed with citrullinaemia typeⅠ(CTLN1). And two patients were diagnosed with ornithine transcarbamylase deficiency (OTCD). Prenatal genetic diagnosis of two families show that one fetus had a mutation at a heterozygous mutation of SLC25A13 with paternal; and another one was a proband with normal OTC gene. Conclusions: Genetic diagnostic can make accurate and effective diagnosis of UCD. However, some families with suspected UCD could have normal OTC gene. The superhydrophilic polymer coating was fabricated by using the sol-gel method and introducing silica nanoparticles into modified PAMAM. The characterization was conducted through FT-IR, SEM, AFM and polymer surface tensiometer to investigate the wettability of the modified PAMAM polymer and structure characteristics of the surface. Moreover, the ice prevention performance was studied. The results showed that the modified PAMAM polymer coating had the micro-nano composite structure and the static contact angle was 9.4°, which reached at superhydrophilic state. Super hydrophilic coating based on modified PAMAM displayed favourable anti-icing characteristics owing to amino and molecular cavity of PAMAM, which contributed to form water film on superhydrophilic surface to suppress icing process. At temperature of-5 °C, the quantity of ice was only 1. 19 g within 40 min, and icing delay time reached to 133 swhile that of the hydrophobic coating was only 38 s. The ice adhesion force of ice on the surface was about 0.33 N, which was significantly better than that of ordinary hydrophilic and hydrophobic coatings. For the test of stability, the average variation rate of the static contact angle and the surface adhesion force of ice on the surface was 1.4%and 1.2%, respectively. [ABSTRACT FROM AUTHOR]

BACKGROUND: Inability of degradation makes free cytosolic ß-catenin in tumor cells aggregate and bind to TCF/LEF into the nuclei, which initiates transcription of downstream genes, and eventually induces abnormal changes in Wnt/ß-catenin signaling pathways. Therefore, investigations on the relationship between ß-catenin and ovarian cancer stem cells can provide a new insight into the specific inhibition of ovarian cancer stem cells. OBJECTIVE: To observe the proliferation, migration and invasion abilities of ovarian cancer stem cells after ß-catenin gene silencing. METHODS: CD133 positive tumor stem cells were isolated from human ovarian cancer cell line A2780 by immunomagnetic beads. CD133 positive cells with 70%-80% confluence were selected and transfected with pLKO.1-shRNA-ß-catenin (experimental group) and pLKO.1-shRNA-ctrln (control group), respectively. The expression of ß-catenin, E-cadherin and Vimentin were detected by western blot at 72 hours after transfection. Cell proliferation was detected by MTT, and cell migration and invasion ability was detected by Transwell chamber assay. RESULTS AND CONCLUSION: (1) The expression of ß-catenin protein in the experimental group was significantly lower than that in the control group at 72 hours after transfection (P < 0.05). (2) With the extension of culture time, the proliferation ability of the two groups increased gradually, but the cell proliferation ability of the experimental group was lower than that of the control group at 2 and 3 days after transfection (P < 0.05). (3) After 72 hours of transfection, the cell migration and invasion abilities of the experimental group were significantly lower than those of the control group (P < 0.05). (4) At 72 hours after transfection, the expression of E-cadherin protein in the experimental group was significantly higher than that of the control group (P < 0.05), and the expression of Vimentin protein was lower than that of the control group (P < 0.05). To conclude, the Wnt/ß-catenin signaling pathway is involved in the migration and invasion of ovarian cancer stem cells. ß-catenin silencing can inhibit the proliferation, migration and invasion of ovarian cancer stem cells.
