Chinese sturgeon Acipenser sinensis is an anadromous migratory fish species, which inhabits the Chinese coastal continental shelf areas and breeds in the Yangtze River Basin. The Yangtze River Estuary is an important migration passage of Chinese sturgeon. The Chinese sturgeon population is endangered, which has been listed as a top-level protected animal in List of key protected wild animals in China and a critically endangered species in IUCN Red List of Threatened Species. Since 2013, the abnormal interruption of natural spawning behavior of wild Chinese sturgeon has brought great challenges to the protection and restoration of Chinese sturgeon population. Based on the survey data of juvenile Chinese sturgeon resources in the Yangtze River estuary from 2004 to 2020, this study analyzed the population characteristics and migration habits of juvenile Chinese sturgeon in the Yangtze River estuary following the abnormal natural spawning behavior. The results show that:(1) Since natural spawning behavioral interruption of Chinese sturgeon, juvenile Chinese sturgeon was not detected in the Yangtze River estuary in 2014, 2016, 2018, 2019 and 2020. The recruitment of juvenile Chinese sturgeon population in the Yangtze River estuary showed an interannual continuous fracture. (2) The migration habits of juvenile Chinese sturgeon changed significantly, it showed that the first occurrence time was advanced to mid-April, and the final occurrence time was delayed to late October, and the occurrence time span was extended by 118 days. (3) The biological indexes of juvenile Chinese sturgeon were lower than the same period in history; Their minimum body length and minimum body weight were 7. 0 cm and 4. 9 g respectively. To sum up, the juvenile Chinese sturgeon recruitment decreased year by year. The migration habit shows that the earlier the first occurrence time, the later the final occurrence time in the Yangtze River estuary. The results of this study will provide an important scientific reference for the protection of rare and endangered species of Chinese sturgeon. [ABSTRACT FROM AUTHOR]

Background and purpose: In clinical studies, checkpoint inhibitor-related pneumonitis (CIP) ranks first among the causes of death in programmed cell death protein-1 (PD-1)/programmed death ligand-1 (PD-L1) inhibitor immune-related adverse events. Real-world CIP situations lack extensive population reports. This study aimed to understand the incidence of CIP in the real world of lung cancer in China, and to summarize its characteristics, treatment status and outcomes. Methods: This study retrospectively collected the essential clinical information of patients with an initral diagnosis of lung cancer who received treatment with immune checkpoint inhibitors (ICIs) at the First Affiliated Hospital of Guangzhou Medical University and Shanghai Pulmonary Hospital between January 2019 and September 2021. For patients with CIP, we also collected the time of its onset, grade, treatment regimen and outcome. The analyses of incidence, patient's characteristics and the risk factors of CIP in overall and subgroup were carried out. Moreover, we analyzed the outcomes of patients treated with immunosuppressive therapy. Results: A total of 2 031 patients with immunotherapy were enrolled, with a CIP incidence rate of 7.2% (147/2 031), a severe CIP rate of 2.6% (52/2 031) and a mortality rate of 0.4% (9/2 031). The rate of severe grade in the population with CIP was 35.4%, and the mortality rate was 6.1% (9/147). Compared with non-CIP patients, more CIP patients were male, older (>65 years), with combination therapy, and on firstand second-line immunotherapy in advanced treatment. In subgroup analyses, the incidence of CIP was higher in men, the elderly (>65 years), squamous cancer, combination therapy, anti-PD-1 inhibitors, and first- and second-line therapy in advanced treatment. The median onset time of CIP in the real world was 148 days, with a double-peak characteristic, that was, 60-90 days and 150-210 days after immunotherapy were both the peak time periods for CIP onset. The incidence of CIP was also influenced by seasonality, with a high incidence in autumn and winter. All treated patients used corticosteroids as first-line treatment; the immunosuppressive treatment rate of CIP in this study was 76.2%. After treatment, 97.9% of mild CIP patients and 81.2% of severe CIP patients had a good prognosis, and 17.3% of severe CIP patients died due to CIP. Conclusion: In the real world, the incidence of CIP for lung cancer patients was 7.2%, incidence of severe CIP was 2.6%, and mortality rate was 0.4%; the incidence of severe disease in the population with CIP was 35.4%, and mortality rate was 6.1%. The median onset time of CIP was characterized by a double peak, and incidence of CIP was higher in autumn and winter. Men, the elderly, squamous cancer patients, patients on combination therapy, patients who used anti-PD-1 inhibitors, and patients with advanced treatment had higher incidence of CIP. 9. 羟基红花黄色素A 生物合成途径短链还原酶基因的特征及功能研究.

- Abstract
Objective: To explore the function of short-chain dehydrogenases/reductases (SDRs) in safflower flavonoid, especially hydroxysafflower yellow A (HSYA) biosynthesis. Methods SDRs involved in HSYA biosynthesis pathway were screened based on safflower transcriptome database and metabolome database. The expression pattern was analyzed by qRT-PCR. The overexpression vector was constructed by seamless cloning technology, then genetically transformed to the Yunnan Weishan safflower strain by Agrobacterium gv3101. The transgenic T2 generation plants were positively verified, and the gene expression of corolla SDRs was analyzed. The content of secondary metabolites was assayed by UPLC-Q-TOF/MS. Results Three SDRs genes named CtSDR1, CtSDR2 and CtSDR3 involved in HSYA biosynthesis pathway were screened. Their expression in safflower from high to low was corolla > leaf > stem > root. The expression level in corolla increased gradually with corolla development. qRTPCR analysis of corolla with positive verification of genome insertion sequence showed that the transcription level of CtSDR3 in corolla of T2 positive plants increased by 2~ 3 times compared with the blank control group, and the content of secondary metabolite HSYA increased by 7.1%~16.6% (P< 0.05). Conclusion CtSDR3 may be involved in the biosynthesis of flavonoids, especially HSYA, in safflower. It provides the support data for explaining the function of CtSDR3 in HSYA biosynthesis pathway. 11. 红花昼夜节律相关基因CtPRR1 的特征及功能研究.

- Abstract
Objective To explore the effect of circadian rhythm genes on flavonoids biosynthesis in safflower and its molecular mechanism. Methods Based on the transcriptome and metabolomic database of safflower corolla, we screened the circadian rhythm genes that correlate with biosynthesis of flavonoids in safflower. qPCR was used to quantify the expressions of circadian rhythm genes in different flowering stages at different time points in a single day. LC-MS was performed to determine the accumulation of flavonoids. The correlation between them was analyzed as well. Yeast Two-Hybrid experiment was used to verify the interactive proteins of these genes. Results Seven circadian rhythm genes PRR1, PRR2, ELF3, FT, PHYB, GI and ZTL were obtained. PRR1 gene was positively correlated with flavonoids accumulation (r≥0.7). The full length of PRR1 is 3 201 bp, encoding 421 amino acids, which is highly homologous with rice OsPRR73 gene and named as CtPRR1 (GenBank accession number: MW492035). CtPRR1 was mainly expressed in flowers, and the expression level increased in the daytime and declined in the evening gradually. Correspondingly, the content of flavonoids showed an opposite variation. Both of them displayed a circadian rhythm with a negative correlation (r≥−0.7). In addition, 2 heat shock proteins along with 3 AP2 transcription factors interacting with CtPRR1 protein were obtained via Yeast Two-Hybrid experiment. Conclusion CtPRR1 negatively regulated the safflower flavonoids accumulation in a circadian rhythm way, which may be affected by these interacting proteins. [ABSTRACT FROM AUTHOR]

目的 总结移植肾输尿管结石患者经输尿管镜逆行碎石手术的方法与经验。方法 回顾性分析1例移 植肾输尿管结石患者,在经尿道逆行钬激光碎石手术过程中,因导丝滑脱导致逆行插管失败。遂在膀胱镜引导下,采用耻骨上经皮膀胱微穿刺术对移植肾输尿管行逆行插管。结果 成功逆行置入输尿管支架管。操作过程中,穿 刺点处未见出血。术后留置导尿管,穿刺点覆盖无菌敷料。患者术后恢复良好,未见出血和感染,于术后 3 d 出院。 结论 对移植肾输尿管结石患者,采用经皮膀胱微穿刺术经输尿管镜逆行碎石手术对患者损伤小,可以避免对移植 肾的穿刺性操作,该术式成功的关键在于寻找异位输尿管开口和留置导丝。 [ABSTRACT FROM AUTHOR]
