3. 沉默促癌基因 Yapl 联合丹参酮 IIA 对肝癌细胞 Huh - 7 的影响.

- Abstract
To investigate the effect of the Yapl gene and tanshinone IIA on the proliferation, migration, and invasion abilities of Huh - 7 hepatoma cells. Methods A total of 10 pairs of human hepatocellular carcinoma (HCC) samples and adjacent tissue samples were collected in Zhongnan Hospital of Wuhan University from June 1 to December 1, 2019. Quantitative real -time PCR and Western blotting were used to measure the expression of the Yapl gene and phenotype - related molecules. MTI cell proliferation detection reagent was used to measure the inhibition rate of cell proliferation after the treatment with different concentrations of tanshinone II A. Western blotting was used to measure the changes in the expression of apoptosis - and migration - related markers after different interventions. Flow cytometry and Transwell assay were used to measure apoptosis and cell migration and invasion abilities. The data of 375 cases of liver cancer and 50 cases of relatively normal liver tissue samples were downloaded from The Cancer Genome Atlas, including clinicopathological information. The t - test was us ed for comparison of continuous data between two groups, and the chi - square test was us ed for comparison of categorical data between two groups. Results In 8 of the 10 pairs of HCC samples and adjacent tissue samples, HCC samples had significantly higher expression of Yapl than the adjacent tissue samples. Compared with the normal human liver epithelial cells L02, the Huh -7 and HCCL - M3 hepatoma cells had a significant increase in the expression of Y apl . The silencing efficiency of si - Y apl - 3 transfection reached 87. 004% at the protein level. MTI results showed that tanshinone II A effectively inhibited the proliferation of Huh - 7 cells, with a half inhibitory concentration of 8. 683 µ.mol/L. After the cells were treated with si - Yapl - 3 and tanshinone II A, there was an increase in the expression of the downstream marker for proliferation and migration E - cadherin and a reduction in the expression of vimentin, and the results of Transwell assay showed that compared with the si - NC group, the tanshinone II A + si - Yapl - 3 group had significant reductions in the migration and invasion abilities of Huh - 7 cells (migration: 43. 19 ± 2. 88 vs 132. 20 ± 10. 03, t = 8. 527, P = 0. 001 ; invasion: 53. 95 ± 4. 20 vs 179. 10 ± 11. 11, t = 4. 484, P = 0. 011). The group treated with si - Y apl - 3 and tanshinone II A had an increase in the expression of the apoptosis - related marker Bax and a reduction in the expression of Bel - 2, as well as a significantly higher early apoptosis rate than the si - NC group ( 25. 98% vs 9. 21 %, x2 = 4. 078, P < O. 05) . Conclusion Oncogene Yapl silencing combined with tanshinone IIA can promote the apoptosis of Huh - 7 hepatoma cells and inhibit their migration and invasion, which can provide certain guiding significance for clinical medication. 5. 假基因DUXAP8在肝癌中的表达及其临床意义.

- Abstract
Objective To investigate the expression,clinical significance,and potential mechanism of action of DUXAP8 in hepatocellular carcinoma. Methods The expression data of liver cancer and related clinical data published up to June 2019 were collected from TCGA database to screen out the differentially expressed long non-coding RNAs( lncRNAs) between liver cancer tissue and adjacent tissue which affected the prognosis of patients. The association of the expression level of DUXAP8 with clinicopathological features and prognosis was analyzed. Gene ontology( GO) and KEGG pathway enrichment analysis was used to investigate the biological functions and biological processes of DUXAP8-related genes. The STRING database and Cytoscape were used to analyze protein-protein interactions and screen out key genes,and then the expression levels of key genes and prognosis were analyzed and a literature search analysis was performed. The Wilcoxon signed-rank test and the Wilcoxon rank-sum test were used to compare the expression of DUXAP8 between liver cancer tissue and adjacent tissue,and the Wilcoxon rank-sum test was used to compare the expression of DUXAP8 between the patients with different clinicopathological features. The Kaplan-Meier method was used to plot survival curves and the log-rank test was used for survival comparison between groups; a Cox regression analysis was used to identify the influencing factors for prognosis. A Pearson correlation analysis was used to analyze DUXAP8-related genes. Results As for the matched and unmatched samples,there was a significant difference in the expression of DUXAP8 between liver cancer tissue and adjacent tissue( P < 0. 001),and the expression of DUXAP8 in liver cancer tissue was significantly higher than that in adjacent tissue. There was a significant difference in the expression of DUXAP8 between the patients with different ages or T stages( P < 0. 05),and the patients with an age of ≥60 years or a T stage of T3 or T4 tended to have higher expression of DUXAP8. The univariate analysis showed that clinical stage,T stage,and expression of DUXAP8 were associated with patients’ overall survival( all P <0. 001),and the multivariate Cox regression analysis showed that an advanced clinical stage( hazard ratio [HR]= 1. 648,95% confidence interval [CI]: 1. 330-2. 709,P < 0. 001) and high expression of DUXAP8( HR = 1. 849,95% CI: 1. 262-2. 713,P < 0. 01) were independent risk factors for poor prognosis. The genes involved in the maintenance of tumor cell proliferation and cell cycle were enriched in the samples with high expression of DUXAP8. TOP2 A,KIF2 C,TTK,PLK1,CDCA8,CDC20,NCAPG,BUB1 B,BUB1,and CCNA2 were the Hub genes of DUXAP8-related genes and were mainly involved in the processes such as cell mitosis and cell cycle,and they were the factors for poor prognosis of patients with liver cancer( all P < 0. 05). Conclusion The high expression of DUXAP8 is a risk factor for poor prognosis in patients with liver cancer and DUXAP8 may promote the development and progression of liver cancer by affecting the processes of cell proliferation and cell cycle. 8. 安徽省某市农田土壤与农产品重金属污染评价.

- Abstract
To explore the relationship between soil and agricultural products of heavy metals, total 1933 soil samples and 43 samples of ag ricultural products were collected from contaminated farmland in a city of Anhui Province. Heavy metal contents of Cr,Pb,Cd, Hg and As, spatial distribution characteristics and the relation between heavy metal contents of agricultural products and the properties of soil were an alyzed. The results showed that the contents of Cr, Pb, Cd, As and Hg in farmland were 62.5, 32.6, 0.209, 8.85, 0.080 mg·kg-1, respective ly, 0.05%, 0, 14.59%, 0.67% and 1.97% higher than the standards. According to the single factor pollution index assessment, farmland soil was contaminated in the order of Cd>Hg>As>Pb>Cr. From the Nemero comprehensive pollution index, 78.58% of all samples was in the clean state. The whole study area was basically at the level of slight ecological risk with the evaluation method of the comprehensive ecologi cal risk index. The enrichment coefficients of Cr, Pb, Cd and As were higher in the agricultural products on the acid soil, while Hg had stronger enrichment ability on the alkaline soil. [ABSTRACT FROM AUTHOR]
