【目的】构建包含丙型肝炎病毒(HCV)C 区、E2 区及NS3 区的嵌合重组载体。【方法】设计合成3 对寡核苷酸引 物, 用PCR 法从HCV 重组质粒pHCVc 、pBE2 和pGMXNS3-5 中特异扩增出编码HCV C 区、E2 区和NS3 区抗原的部分基因 片段(501、603 和900 bp), 分别用Hin dⅢ 、BamH Ⅰ 、EcoRⅠ 和Xho Ⅰ 酶切后, 逐个定向连接到质粒pcDNA3 中, 转化宿主菌 XL1-Blue , 分别用上述内切酶酶切及DNA 测序鉴定重组质粒。【结果】酶切鉴定示所切下的片段大小均与预计相符, 测序结 果与文献报道序列及预计结果一致, 证实符合表达框架。【结论】成功构建了HCV 嵌合重组质粒pcDNA3-C-E2-NS3 。

【目的】探讨病毒性肝炎纤维化的病理改变与血清学指标之间的关系。【方法】在彩色B 超定位下穿刺取活体肝 组织299 例, 作苏木素-伊红(HE)、网织纤维(RT)染色, 显微镜下观察分级、分期。血清标本用放射免疫测定检测透明质酸 (HA)、Ⅲ 型人前胶原(HPC Ⅲ)、IV-型胶原(IV-C)。【结果】慢性肝炎轻度97 例, 血清学纤维化指标升高不明显。慢性肝炎中 度126 例, 血清学纤维化指标均有不同程度升高, 慢性肝炎重度29 例、肝硬化47 例, 血清学纤维化指标均显著升高。299 例 慢性病毒性肝炎病理的炎症活动度和纤维化分期与血清学纤维化程度有密切关系。【结论】慢性病毒性肝炎纤维化的病理改 变和血清学的变化随临床病情的变化而改变。肝组织活检结合血清学指标的检测在临床上具有较高的诊断价值。

【目的】为探索共刺激分子 B7-1 增强 HBsAg 真核表达载体基因免疫的效果,并用于消除乙肝免疫耐受。【方法】 用高保真 PCR 法扩增目的基因片段 B7-1 及带接头和启动子的 IRES-HBs 基因片段,亚克隆到 pBluescript KS+载体中测序,再 克隆到逆转录病毒载体 PLXSN 中。【结果】所扩增的目的基因片段经测序证实, 未发现序列有改变。先将 B7-1 通过 EcoR Ⅰ 、Xho Ⅰ插入 plxsn 中,构建成 plxsn-B7-1, 再用 Xho Ⅰ 、 BamHⅠ将 IRES-HBs 插入, 构建成 plxsn-B7-1-HBs。【结论】成功 构建了共表达 HBsAg 及 B7-1抗原的重组逆转录病毒载体, 可行体外真核基因表达, 研究目的抗原表达的特性, 或行基因免 疫,研究 B7-1 对 HBsAg 免疫反应的影响, 进一步可用于防治乙肝的研究。

Aiming at two problems of existing multi-label feature selection methods: first, ignoring the influence of noise information in the process of learning label correlations; second, neglecting to explore the comprehensive label information of each cluster, the paper proposed a multi-label feature selection method that enhanced label correlation learning. Initially, it clustered the samples and treated each cluster center as a representative instance of the comprehensive semantic information of the samples, while computing its corresponding label vectors which reflected the importance of different labels contained in each cluster. Then, through the label-level self-representation of the original samples and the center of each cluster, it both captured the label correlations in the original label space, and explored the label correlations within each cluster. Finally, the self-representation coefficient matrix was sparse to reduce the effect of noise, and the original sample and the representative instance of each cluster were mapped from the feature space to the reconstructed label space for feature selection. Aiming at the problem of function model solution with parameter constraint equation, a unified solution of constraint equation weighting is suggested, which combines the weighting value of constraint equation with the unified solution of observation equation. In order to verify the reliability and efficiency of this method, the orthogonal triangular decomposition formula and the generalized singular value decomposition formula suitable for the solution of the parameter constraint equation are deduced based on the matrix analysis method, and the three solution algorithms are compared and analyzed through examples. The results show that when a suitable weight value is given to the constraint equation, the parameter estimation values obtained through the unified solution of the constraint equation are consistent with the parameter estimation results of the matrix decomposition algorithm within the required accuracy range. The computational efficiency of the weighted unified solution is close to that of the orthogonal triangular decomposition method, and the computational efficiency of the generalized singular value decomposition method is low. Objective To investigate the expression of myeloid-derived suppressor cells (MDSC), regulatory T cells (Treg), IL-17-producing CD4+ T cells (Th17), and CD8+ T cells (Tc17) in hepatitis B virus-related acute-on-chronic pre-liver failure (pre-ACHBLF), and to provide ideas for the early treatment of acute-on-chronic hepatitis B liver failure (ACHBLF). Methods A total of patients with pre-ACHBLF and 15 patients with ACHBLF who were hospitalized in Shijiazhuang Fifth Hospital, from August 2018 to May 2019 were enrolled as subjects, and 15 patients with chronic hepatitis B (CHB) and 15 healthy controls (HC) who underwent physical examination were enrolled as controls. Flow cytometry was used to measure the expression levels of MDSC and Th17, Treg, and Tc17 cells in peripheral blood; a blood analyzer was used to measure routine blood parameters and calculate neutrophil-to-lymphocyte ratio (NLR), monocyte-to-lymphocyte ratio (MLR), platelet-to-lymphocyte ratio (PLR), and systemic immune-inflammation index (SIRS) to evaluate the degree of inflammation, and the correlation between the expression of immune cells and the degree of inflammation was analyzed. An analysis of variance for independent samples was used for comparison of normally distributed continuous data between multiple groups, and the least significant difference t-test was used for further comparison between two groups; the Kruskal-Wallis H test was used for comparison of non-normally distributed continuous data between multiple groups, and the Nemenyi test was used for further comparison between two groups. A Pearson linear correlation analysis or Spearman’s rank correlation analysis was used to investigate the correlation between variables. Results Compared with the CHB group, the ACHBLF and pre-ACHBLF groups had significant increases in the expression levels of Th17, Treg, and Tc17 cells, and the pre-ACHBLF group also had a significant increase in the expression level of MDSC (all P<0.05). The correlation analysis showed that in pre-ACHBLF patients, MDSC were positively correlated with leukocyte count, neutrophil count, NLR, MLR, and SII (r=0.775, 0.727, 0.571, 0.786, and 0.846, all P<0.05), and Treg cells were only positively correlated with leukocyte count (r=0.618, P=0.043) ; Th17/Treg ratio and Tc17 cells were negatively correlated with the number of lymphocytes (r=-0.790 and-0.795, both P<0.05). Conclusion Cellular immune dysfunction is observed in patients with pre-ACHBLF, and the expression of MDSC is closely associated with the degree of inflammation and should be taken seriously in the early stage. Objective To investigate the expression of ITGBL1 in hepatocellular carcinoma (HCC) and its role in promoting the proliferation, migration, and invasion of HCC cell lines. Methods RT-qPCR and immunohistochemistry were used in investigating ITGBL1 expression in 12 pairs of fresh HCC and adjacent normal liver tissue samples and 160 paraffin HCC specimens. The relationships of ITGBL1 expression level with clinicopathological parameters and prognosis were analyzed. HUH7 cell line with the stable overexpression of ITGBL1 and LM3 cell line with stable downregulated expression of ITGBL1 were constructed. The effects of ITGBL1 on the proliferation, migration, and invasion of HCC cells were examined by CCK8 assay, wound-healing assay, and Transwell invasion assay. Results The expression levels of ITGBL1 gene and protein in tumor tissues were higher than those in surrounding liver tissues. The high expression of ITGBL1 was correlated with serum AFP level, tumor capsular invasion, vascular invasion, tumor differentiation, and clinical stage (P<0.05). The results of Kaplan-Meier analysis showed that patients with high expression of ITGBL1 had shorter DFS. In vitro, increased ITGBL1 expression promoted HCC cell proliferation, migration, and invasion, whereas ITGBL1 knockdown inhibited these processes. Conclusion ITGBL1 expression is highly expressed in HCC tissues, and ITGBL1 can increase the proliferation, migration, and invasion of HCC cells. However, the mechanism needs further study.
