【目的】用RT-PCR 法获得人类神经元性芳香族氨基酸脱羧酶(AADC)基因全序列, 并建立优化方法。【方法】 从人嗜铬细胞瘤中提取组织总RNA , 自行设计引物, 采用两步RT-PCR 法扩增出长约1 550 bp 的基因片断, 凝胶回收后与高 效克隆PCR 产物的新型载体T-载体连接, 并转化入大肠杆菌, 通过蓝白斑反应筛选出重组体, 质粒提取后进行重组体的酶切 鉴定及基因测序。【结果】BamH Ⅰ 和Hin dⅢ 酶切证实了目的基因同克隆载体进行了有效的连接, 基因测序证实了所克隆的 片断为人AADC 基因。【结论】本实验通过对引物, Taq 酶的选择及反应条件的优化, 保证了扩增的有效性和特异性。新型 的克隆载体明显提高了克隆效率。基因测序证实了AADC 基因扩增的成功。本研究为AADC 基因治疗帕金森病等多巴胺能 缺乏疾病的研究打下了基础。

This article uses the Silk Road as the research backdrop to explore the inheritance of tea culture in Kazakhstan and its integration and impact in contemporary society. It aims to reveal the role of tea as a cultural symbol in promoting crosscultural exchange, deepening social cohesion, and economic development. Through methods such as literature review and participatory observation, it thoroughly examines the spread, inheritance and development of tea culture in Kazakhstan, including the social functions, symbolic meanings, and integration with local life customs across different historical periods. It covers the status of tea in Kazakh culture, discusses the multifaceted functions of tea in health, culture, and economy, and analyzes the practices of tea culture inheritance and innovation during modernization. The findings indicate that tea culture, as an essential part of the lives of the people of Kazakhstan, not only fosters cultural inheritance and social cohesion but also positively impacts local economic development. With the large -scale ships and the complexity of the dynamic environment in the berthing waters, when usingappendix F of the current JTS 144-1—2010 Load Code for Harbor Engineering to calculate the ship flow force, it is found thatthere are many deficiencies in the formula, mainly as follows: 1) the applicable range of relative water depth is narrow; 2) Thecalculation methods for flow direction angle sections are not unified; 3) the action points of longitudinal and transverse flowcomponents are not clear; 4) the introduction of relevant data defects, etc. Seriously restrict the promotion and application ofstandards and specifications. The cast-in -place wave wall of the west artificial island of the Shenzhen -Zhongshan Link adopts a ladder-like structure with complex section forms. The structural foundation is replaced with filling foundation and there are differences along the cross section direction. Based on the considerations such as controlling differential settlement of the wave walls, improving construction quality of wave walls, and enhancing architectural landscape effects, an optimization research was carried out on the structural design and construction technology of cast-in-place wave walls. The practice has proved that the optimized structural design is scientific and reliable, and the construction process is safe and efficient. The structure of the wave wall after implementation has no obvious difference settlement, the structure line is smooth, and the landscape effect is good, which verifies the scientific and feasibility of this optimization scheme, and can provide relevant reference for similar projects. The ore (rock) bodies between the 570 m and 630 m levels in Jiaojia Gold Mine are characterized by well-developed joints and fissures, prone to forming wedge-shaped unstable structures, leading to rock mass col- lapse. Geological drilling and structural surface condition surveys indicate that the rock mass in Jiaojia Gold Mine is relatively fractured, with well-developed joints and a wide range of joint occurrence distributions. This study investi- gates and summarizes the convergence deformation patterns of roadways under mining disturbances. Radar testing and analysis reveal that the damage depth of the roadways in Jiaojia Gold Mine generally remains at about 2 m, with many weak structural surfaces present, necessitating timely reinforcement measures. This research provides a rational plan for the support of the ore body and guides the reasonable selection of stope structural parameters to ensure safe production in this mining section. Based on the data of Kenya's four national population censuses, exploratory spatial data analysis, standard deviation ellipse, and population shift-share model were used to analyze the spatial pattern evolution characteristics of the population distribution of 47 administrative units in Kenya from 1989 to 2019. Then, this paper employed geographic detector to further analyze the influencing factors. The results show that: ① During the study period, the distribution pattern of population density in Kenya is relatively stable, showing high southwest and low northeast. ② There are obvious spatial differences in population offset growth in different cities, mainly positive offset growth and negative offset growth types. And the population distribution center is northwest of the capital Nairobi and continues to migrate southeast. ③ Natural and socio-economic factors jointly influence the spatial evolution pattern of population distribution in Kenya. Under the influence of elevation, climate, precipitation, economic development level and transportation infrastructure construction, the population is concentrated in a few areas along the southwest and southeast coasts of Kenya. Kenya has roughly formed a high-density population cluster centered on Nairobi, the national administrative center, Kisumu on the shores of Lake Victoria, and Mombasa, the largest port on the coast.
