- Abstract
Objective: To study the expressions of IL-33 and its receptor ST2L in peripheral blood T cells and skin lesions of patients with psoriasis. Methods: The expression and distribution of IL-33 and ST2L in psoriasis vulgaris(PV group), psoriasis pustulosa(PP group) and nevus resection]control group(CON group)]were detected by immunohistochemical method. Western blot and RT-PCR were used to detect the expression of IL-33 and IL-33 mRNA in Jurkat T cells and HaCat keratinocytes induced by IL-17A, respectively. The same method was used to detect the expression of ST2L and ST2L mRNA in Jurkat T cells, PV group T cells, PP group T cells and CON group T cells after IL-33 induction. Results: In the skin lesions of patients in PV group and PP group, we observed positive staining of IL-33 and its receptor ST2L protein in epidermis and dermis cells with different densities; a small amount of IL-33 protein was expressed in T cells of CON group, but in psoriasis group, including PV group and PP group, IL-33 protein was significantly expressed in T cells, and the expression level of PP group was higher than that of PV group, the difference was statistically significant(P<0.05); IL-33 induced Jurkat T cells, PV group and PP group T cells to express a certain amount of ST2L protein and mRNA in a dose-dependent manner; the expression of ST2L protein in PV group and PP group were higher than the level of ST2L induced by IL-33 in CON group, and the difference was statistically significant(P<0.05). IL-17A induced Jurkat T cells to produce IL-33 protein and mRNA in a certain amount of effect and aging relationship; IL-17A induced the expression of IL-33 protein and mRNA in HaCat keratinocytes in a dose-dependent manner. IL-33 protein was mainly expressed in the nucleus of HaCat keratinocytes, and some of the cytoplasm had some expression, and the expression was significantly increased after high concentration of IL-17A stimulation. Conclusion: The expression levels of IL-33 and its receptor ST2L in peripheral blood T cells and skin lesions of patients with psoriasis are significantly increased, suggesting that IL-33 may play an important role in the occurrence and development of psoriasis.
