4. 粪肠球菌在体外细胞模型和乳鼠轮状病毒感染模型中对轮状 病毒SA11 株复制的抑制作用.

- Abstract
Objective:To explore the replication of rotavirus SA11 strain in vivo and in vitro after adding Enterococcus faecalis( E. faecalis),and to provide the evidence for clarifying the effect of E. faecalis on rotavirus infection. Methods:In in vitro experiment, 108-1012 CFU ·mL-1 E. faecalis were diluted with 10 times concentration gradient and co-cultured with Caco-2 cells for 12, 18, 24, 30,and 48 h,and control group (Caco-2 cells infacted with rotavius without the addttion of E. faecalis) was set up at the same time. The survival rates of Caco-2 cells in various groups after treatment of different concentrations of E. faecalis were determined by CCK-8 method. One hour after the Caco-2 cells were treated with rotavirus with multiplicity of infection ( MIO ) value of 0. 1,they were cultured with 108-1012 CFU·mL-1 E. faecalis for 18 h. The rotavirus VP6 gene copy numbers of Caco-2 cells in various groups were detected by real-time fluorescence quantitative PCR(RT-qPCR) method and the rotavirus titers of Caco-2 cells in various groups were detected by immunofluorescence focus method. In in vivo experniment, 18 litters of 4- 5 d old SPF Kunming suckling mice were randomly divided into control group, antibiotic treatment group and E. faecalis transplantation group. The suckling mice in antibiotic treatment group and E. faecalis transplantation group were gavaged with quadruple antibiotics for 3 d to deplete their intestinal flora, and the suckling mice in three groups received 107 FFU·mL-1 rotavirus 100 μL by gavage daily for 8 d, and the suckling mice in E. faecalis transplantation group were given 1010 CFU·mL-1 E. faecalis 100 μL at the same time. The feces were collected on the 2nd,4th,6th,and 8th days after virus infection, and the copy numbers of rotavirus in feces were measured by RT-qPCR method. Results:In in vitro experiment,the CCK-8 method results showed that compared with control group,the survival rates of Caco-2 cells in various groups in 48 h of treatment of different concentrations of E. faecalis were increased (P<0. 05), indicating that E. faecalis was non-toxic to the Caco-2 cells within this concentration range. The RT-qPCR results showed that compared with control group,the number of rotavirus gene copies in the Caco-2 cells in 1010, 1011,and 1012 CFU·mL-1 E. faecalis groups were decreased by 75. 8%, 99. 9%, and 99. 9%, respectively(P<0. 05). The results of immunofluorescence focus detection showed that compared with control group,the rotavirus titer in 108,1010,and 1012 CFU·mL-1 E. faecalis groups were decreased by 13%,77%, and 100%, respectively (P<0. 05). In in vivo experiment, compared with control group, the copy number of fecal virus gene of the suckling mice in E. faecalis transplantation group was significantly decreased(P<0. 05). Conclusion:In in vitro cell model and rotavirus infection model of suckling mice, E. faecalis can inhibit the replication of rotavirus. [ABSTRACT FROM AUTHOR]

8. 水分亏缺条件下褪黑素对春小麦光合特性及产量性状的影响.

- Abstract
[Objective] To clarify the regulating effect of melatonin treatment on spring wheat yield under water deficit conditions. [Methods] In the experiment, the spring wheat irrigated varieties Nongmai2 and Yongliang4 were used as test materials. Using randomized complete block design, the effects of exogenous melatonin on the main phenotypic traits of spring wheat, such as chlorophyll content, photosynthetic rate, biomass per plant, spike number, grain number per spike, thousand kernel weight and grain yield, were studied under well-watered and water deficit treatments. [Results] Compared with well-watered treatment, water deficit treatment reduced the chlorophyll content and photosynthetic rate of functional leaves of spring wheat, hindered the accumulation of biomass per plant, reduced spike number, grain number per spike, and thousand kernel weight, reduced yield by 19.01%-20.35%. Under water deficit conditions, melatonin treatment could alleviate the damage to leaves caused by water deficit, maintain high chlorophyll content and photosynthetic rate, promote the accumulation of biomass per plant, inhibit the decrease of spike number, grain number per spike and thousand kernel weight, and increase yield by 13.19%-17.27%. [Conclusion] As a green growth regulator, melatonin played an important role in enhancing the adaptability of spring wheat and coping with frequent water deficit. [ABSTRACT FROM AUTHOR]

14. 高血压肾损害患者IL-6-174 基因多态性及其与贝那普利治疗反应的相关性研究.

- Abstract
To study the relationship between the genetic polymorphism of interleukine-6 (IL-6)-174 and the response to benazepril treatment in patients with hypertensive renal damage. Methods Two hundred and eighty-four patients with hypertension were enrolled in this study. The hypertensive renal damage was defined by the measurement of urinary albumin excretion rate (UAER). One hundred and sixty healthy subjects were enrolled simultaneously as control group. Blood samples were obtained from all the subjects, and plasma levels of IL-6 and the genotype of gene IL-6-174 were detected. The patients with hypertensive renal damage were treated with benazepril for 16 weeks. The responses were evaluated by the changes of UAER level to benazepril in different genotypes. Results Genotype CC was the most common of the gene IL-6-174 in patients with hypertension, followed by GG and GC successively, with the G/C allele frequency of 47% and 53% (P<0.05), while in patients with hypertensive renal damage, GG was the most common genotype of the gene IL-6-174, followed by GC and CC successively, with the G/C allele frequency of 68% and 32% (P<0.05). After benazepril treatment, the UAER was decreased most in patients with genotype CC, followed by GC and GG successively (P<0.05). Conclusion The G allele frequency of the gene IL-6-174 is related with hypertensive renal damage in patients in Ningxia, with GG as the most common genotype. The patients with CC genotype have the best response to benazepril treatment, with most decreased UAER. [ABSTRACT FROM AUTHOR]
