Resistance against anti-tubercular drugs is a significant problem. This elucidates the need for novel drug targets. Altering and targeting the enzymes involved in cell wall synthesis led to fatal damage to the bacterial cell. Mycolic acids are critically responsible for the virulence of Mycobacterium Tuberculosis. This pathway represents an essential reservoir of novel targets for developing new TB drugs. The study aims to identify phytochemicals with the capacity to bind with enzymes of mycolic acid synthesis pathways. This study shows the interaction between phytochemicals and proteins responsible for mycolic acid synthesis is shown through bioinformatics & molecular docking tools. Docking showed binding affinity between protein molecules of the mycolic acid synthesis pathway and ligand molecules in the study. PKS13 (polyketide synthase) interacts with the ligand beta-amyrin acetate with a vina score of -7.1 Kcal/mol. At the same time, its binding energy with Piperine is -6.8 Kcal/mol. DprE1 (Decaprenylphosphoryl-bet-D-ribose-2-epimerase), the other protein docked with beta-amyrin acetate, showed a vina score of -9.7 Kcal/mol binding energy. Piperine with DprE1 exhibits interaction with a score of -8.3 Kcal/mol. Beta-amyrin acetate is docked with a score of -6.9 Kcal/mol against KasA (Beta-ketoacyl-acyl carrier protein synthase). On the other hand, Piperine with KasA gave a result of -7.0 Kcal/mol. Piperine and Beta-amyrin acetate binds to PKS13, DprE1 & KasA protein/enzymes responsible for mycolic acid biosynthesis.

The toxicity and irresponsive nature of the available drugs for lung cancer treatment demands an alternative approach.Methods: In this study, four known compounds namely, cirsimaritin (4′,5, -dihydroxy-6,7-dimethoxyflavone) (1), eupatorin (5,3′-dihydroxy-6,7,4′-trimethoxyflavone) (2), betulin (Lup-20 (29)-ene-3, 28-diol) (3), and β-amyrin acetate (12-Oleanen-3yl acetate) (4) have been isolated from the leaves extract of Quercus incana. Preliminary screening of these natural compounds (1–4) was performed against non-small cell lung carcinoma (NCI-H460) and normal mouse fibroblast (NIH-3T3) cell lines.Results: The compounds were found to be antiproliferative against cancer cells with wide therapeutic index in comparison to the normal cells. Effects of betulin (3) on cell migration, invasion, apoptosis, and expression of important apoptosis- and metastasis-related markers were observed at different concentrations. The results showed significant dose-dependent induction of apoptosis after the treatment with betulin (3) followed by increased expression of the caspases family (ie, caspase-3, -6, and -9), proapoptotic genes (BAX and BAK), and inhibiting anti-apoptotic genes (BCL-2L1 and p53). Furthermore, wound healing and transwell invasion assays suggested that betulin (3) could also regulate metastasis by inhibiting MMP-2/-9. Osteopontin, a central regulator of apoptosis and metastasis was also inhibited in a dose-dependent manner.Conclusion: The present findings suggest that betulin (3) can be an attractive chemotherapeutic target for treating resistant lung cancers. The aim of this study was to conduct a histochemical analysis to localize lipids, terpenes, essential oil, and iridoids in the trichomes of the L. album subsp. album corolla. Morphometric examinations of individual trichome types were performed. Light and scanning electron microscopy techniques were used to show the micromorphology and localization of lipophilic compounds and iridoids in secretory trichomes with the use of histochemical tests. Additionally, the content of essential oil and its components were determined using gas chromatography-mass spectrometry (GC-MS). Qualitative analyses of triterpenes carried out using high-performance thin-layer chromatography (HPTLC) coupled with densitometric detection, and the iridoid content expressed as aucubin was examined with spectrophotometric techniques. We showed the presence of iridoids and different lipophilic compounds in papillae and glandular and non-glandular trichomes. On average, the flowers of L. album subsp. album yielded 0.04 mL/kg of essential oil, which was dominated by aldehydes, sesquiterpenes, and alkanes. The extract of the L. album subsp. album corolla contained 1.5 × 10−3 ± 4.3 × 10−4 mg/mL of iridoid aucubin and three triterpenes: oleanolic acid, β-amyrin, and β-amyrin acetate. Aucubin and β-amyrin acetate were detected for the first time. We suggest the use of L. album subsp. album flowers as supplements in human nutrition.

Thirteen pentacyclic triterpenes, methyl 3-oxours-12-en-23-oate, marsformosanone, taraxerone, β-amyrenone, α-amyrenone, lupenone, 24-methylencycloartan-3-one, moretenol acetate, β-amyrin acetate, germanicol acetate, 24-methylencycloartanyl acetate, β-amyrin, and α-amyrin were identified in a chloroform-methanol propolis extract from Melipona beecheii. Additionally, were identified in this propolis, hexadecanoic acid, methyl ester, octadecanoic acid, methyl ester and 1-triacontanol. The purification of the propolis extract was carried out using different chromatographic techniques, including vacuum liquid chromatography, gravity column chromatography and gel filtration chromatography Sephadex LH-20. Abstract: The ethanolic extract and fractions of Wrightia tomentosa Roem. & Schult (Apocynaceae) leaves were tested in vivo for their antidyslipidemic activity in high fat diet (HFD) induced dyslipidemic hamsters. Activity guided isolation resulted in identification of antidyslipidemic compounds β-AA and β-AP. Compounds β-AA and β-AP decrease the levels of LDL by 36% and 44%, and increase the HDL-C/TC ratio by 49% and 28%, respectively, at a dose of 10mg/kg. In addition, the isolated compounds β-AA and β-AP showed significant HMG-CoA-reductase inhibition, which was further established by docking studies. [Copyright &y& Elsevier]

The phytochemical investigation of the ethyl acetate soluble fraction of the dried floral buds of Syringa patula palibiniana Nakai (Oleaceae) has led to the isolation of six compounds: β-amyrin acetate ( 1), β-sitosterol ( 2), nortropin ( 3), eugenol ( 4), syringaresinol ( 5), and oleoside 11-methyl ester ( 6). The structures were elucidated by spectroscopic and chemical means. Investigation of the biological activities indicated that 5 and 6 possessed a strong cytotoxic activity against Hep-G2 cells; 1 and 5 remarkably increased the macrophage proliferation index; 1 and 5 exhibited high anti-inflammatory activities as indicated by inhibited LPS-induced NO generation; and only 5 showed a strong scavenging activity against DPPH. Taken together, results indicate that 5 is a multipotent compound active as an anti-cancer, immunoproliferative, anti-inflammatory, and antioxidant agent. [ABSTRACT FROM AUTHOR]

The microbial transformation of β-amyrin acetate (compound 1) occurred in the culture of Rhodobacter sphaeroides producing three metabolites. The metabolites were purified and identified as β-amyrin (compound 2), (3β)-olean-12-ene-3, 23-diol (compound 3), and erythrodiol (compound 4). Objective: The aim of this work is to identify and purify bioactive compounds from 70℅ aqueous methanolic extract (AME) of B. spectabilis leaves collected in Cairo, Egypt and assessed their rheumatoid arthritis activity., Methods: The methanolic extract of B. spectabilis leaves was fractionated and subjected to different chromatographic techniques to isolate pure new compounds which were identified by one dimensional and two dimensional nuclear magnetic resonance (NMR) spectroscopic analyses and mass spectrometric methods. The isolated compounds were evaluated for their anti-inflammatory activity on adjuvant induced chronic rheumatoid arthritis., Results & Discussion: Seven bioactive compounds were purified and identified for the first time from the methanolic extract of Bougainvillea spectabilis leaves; secologanin dimethyl acetal, α- and β-amyrin, α- and β-amyrin acetate, Kaempferol and kaempferol-3-O-rhamnoside.
