The separation of a mixture of trinitrotoluenes (TNTs) and their metabolites by reverse phase liquid chromatography using different columns with octadecyl stationary phase is presented. The retention behaviour of a mixture of TNTs and their metabolites was studied under different chromatographic conditions. The mixture of methanol/water was used as a mobile phase in isocratic and gradient modes. The effect of content of organic modifier and temperature of mobile phase has been investigated. Very good RP-HPLC separation of 14 nitroaromatic compounds was obtained using stepped gradient elution. Different properties of extraction procedures for isolation of trinitrotoluenes and their metabolites from soil were tested. Experimental conditions of Soxhlet warm extraction (extractant type, extraction time) suitable for efficient extraction of individual analytes were studied. Recoveries over 80% for all studied compounds and over 90% for almost all (11 compounds) were obtained in short extraction time (1 hour). The formation of precipitate during the Soxhlet extraction was observed and the further treatment of extract was necessary. The amount of precipitate formed from an artificial soil was higher than that from a natural one. THF was found as the best solvent suitable for total dissolution of precipitate. © 2009 Taylor & Francis.

Bakalářská práce je zaměřena na manažerské řízení nákladů a rozpočtů ve společnosti. V teoretické části je provedena rešerše odborné literatury a internetových zdrojů. Jsou zde popsány účetní systémy, úkoly manažerského účetnictví, náklady společnosti a rozpočetnictví. Dále je v práci popsána tvorba manažerského rozhodování a strategické plánování. Analytická část práce je zaměřena na popis vybraného podniku, jeho organizační struktury, výrobní program, konkurenceschopnost, technologické procesy a vzdělávání zaměstnanců ve firmě. Z rozvahy a výkazů zisků a ztrát je zpracována horizontální a vertikální analýza za roky 2012 až 2014. V závěru práce jsou popsány návrhy na zlepšení stávající úrovně manažerského účetnictví a optimalizace nákladů ve společnosti., This bachelor thesis is focused on the managerial cost management in the company. The theoretical part provides literature and internet sources. In this part is described accounting systems, the tasks of management accounting, budgeting and costs of the company. Here is also described the production of managerial decision making and strategic planning. Analytical part is focused on the description of the selected company, its organizational structure, production program, competitiveness, technological processes and education of employees in the company. From the balance sheet and profit and loss account is processed horizontal and vertical analysis of the years 2012 to 2014. In the conclusion is described proposals for improvement of managerial accounting and cost optimization in the company.

Předložená práce je studií chromatografického chování azaarenů v separačním systému vysoceúčinné kapalinové chromatografie s obrácenými fázemi. Hlavním cílem této práce bylo použití základních principů chromatografické separace pro popis a interpretaci výsledků separace azaarénů v různých chromatografických podmínkách včetně efektů jako je nereprodukovatelná retence, chvostování elučních zón či silné zadržení ve stacionární fázi. Práce byla zaměřena na bazické azaarény, často označované jako akridiny, které patří mezi perzistentní organické polutanty s karcinogenní a mutagenní aktivitou. Chromatografické chování bylo studováno na modelové skupině osmi dvou a trojjaderných akridinů, které se běžně vyskytují ve vzorcích životního prostředí znečištěných polycyklickými aromatickými sloučeninami. Separace byly realizovány za podmínek izokratické eluce se třemi oktadecylovými stacionárními fázemi různého původu. Nejprve byly otestovány povrchové vlastnosti zvolených stacionárních fází, hydrofobicita a silanolová aktivita, následovaly separační experimenty. Jejich cílem bylo posouzení vlivu tří rozdílných parametrů na retenční chování akridinů: povrchových vlastností stacionární fáze, složení mobilní fáze a fyzikálně-chemických vlastností akridinů. Zpracování všech výsledků bylo založeno na hodnotách kapacitních poměrů, které poskytují nejvyšší míru reprodukovatelnosti, avšak uvedeny byly také ostatní chromatografické parametry hodnotící termodynamické a kinetické aspekty separace. Na základě provedených experimentů byly nalezeny jednoduché vztahy mezi třemi hodnocenými parametry a charakterem interakcí separačního mechanismu. Praktické využití těchto znalostí znamená zjednodušení optimalizace separačních procesů. Současně také nabízí řešení separačních problémů s akridiny, ale i s ostatními bazickými látkami, které vykazují srovnatelné hodnoty pKa a Mr., Under the framework of this PhD project, a study on chromatographic behaviour of selected azaarenes on octadecylsilica stationary phases during their separation by reversed phase liquid chromatography was developed. The main goal was aimed at an application of the basic RPLC principles on the retention behaviour of azaarenes including the effects such as peak tailing, peak broadening, irreproducible retention or strong retention on a stationary phase. This study was particularly focused on basic azaarenes, also called acridines, which belong to a group of persistent organic pollutants providing mutagenic and/or carcinogenic activity. The retention patterns were studied on a group of eight acridines that are often present in environmental samples polluted with polycyclic aromatic compounds. The mixture of acridines was separated under isocratic elution conditions on three octadecylsilica stationary phases using two different binary mixtures as a mobile phase. Evaluation of the surface properties of the selected stationary phases was firstly performed, followed by the separation experiments. The structure of the experiments was designed to evaluate the effect of three different factors on the retention behaviour of acridines: properties of a stationary phase, composition of a mobile phase and physical-chemical properties of acridines. The treatment of the results was based on the capacity factor values providing the best fitting and repeatability of data, but other parameters that evaluate thermodynamic and kinetic aspects of the separation process were also given. Simple correlations between the three factors and a character of retention mechanism of acridines on the given separation system were found. Application of this knowledge simplifies an optimisation process and helps to solve common separation problems for acridines, but also for other basic analytes displaying similar physical-chemical properties (Mr and pKa).
