The genus Genista L. (Family: Fabaceae) is a plant having several traditional uses for treating common ailments such as diabetes, ulcer, and respiratory diseases. In this current study, the composition of essential oil and the biological activities of Genista sagittalis L. (Fabaceae) from Kocaeli: Yuvacık Dam Basin have been studied. A total of fourteen components were identified in the essential oil. The identified compounds belonged to straight-chain alkane, aromatic ether, and terpenoid derivatives. The antibacterial activity analyses demonstrated that G. sagittalis flower extract only had low activity against P. mirabilis and P. aeruginosa with MICs 1 to 2 mg/mL, as the peduncle extract showed strong anti-QS activity at 1.3 mg/mL. To the best of our knowledge, the current work is the first to report the antimicrobial and anti-quorum sensing activity of G. sagittalis growing in Turkey. Double-stranded DNA binding affinity investigations of the flower and peduncle ethanol extracts indicate that there are interactions with double-stranded DNA and related binding constants (Kb) were found as 1.97×103±0.37 and 3.68×102±0.44 for the flower and peduncle extract, respectively.

Bu tezde, kanallardaki zamandan bağımsız, laminer, tam gelişmiş, noktasal manyetikkaynak veya düzgün manyetik alan etkisi altınndaki akışın karşılıklı sınır elemanları metodu (DRBEM) kullanılarak simülasyonu yapılmıştır. Navier-Stokesve enerji denklemleri hız, basınç ve sıcaklık cinsinden çözülmüş ve böylece tümorijinal problem bilinmeyenleri elde edilmiştir. Fiziksel olarak kayıp basınç denklemielde edilmiş ve sonlu fark yöntemi ile DRBEM koordinat matrisi kullanılarakbasınç için sınır koşulları üretilmiştir. Lineer olmayan kismi türevli denklemler,Laplace denkleminin temel çözümü kullanılarak sınır integral denklemlerinedönüştürülmüştür. Laplace dışındaki terimlere bir dizi radyal baz fonksiyonu ileyaklaşılmıştır. Temel denklemlerdeki lineer olmayan terimler DRBEM koordinatmatrisi ile kolaylıkla ele alınmıştır. ilk olarak, noktasal manyetik kaynağın Ferrohidrodinamik(FHD) Stokes, sıkıştırılamayan ve zorlanmış konveksiyon akışlarıüzerindeki etkisi incelenmiştir. Kaldırma kuvveti, manyetik kuvvet ve viskoz disipasyonarasındaki etkileşim tartışılmıştır. Daha sonra, birden fazla noktasal manyetik kaynağın FHD sıkıştırılamaz akış üzerindeki etkisi çalışılmıştır. Duvarlardakayma koşulu olması durumunda, Magnetohidrodinamik (MHD) kanalakışının ve paralel sonsuz plakalar arasındaki akışın DRBEM ile simülasyonu dayapılmıştır. Momentum ve manyetik indüksiyon denklemlerinden oluşan sistem,birleştirilmiş ve iterasyon olmadan çözülmüştür. Bu yöntem problemin tanımbölgesinde ve sınırında noktasal çözümlerin bir seferde elde edilmesine olanaksağlamıştır. In this thesis, steady, laminar, fully developed flows in pipes subjected to apoint magnetic source or uniform magnetic field are simulated by the dual reciprocityboundary element method (DRBEM). The Navier-Stokes and energyequations are solved in terms of the velocity, pressure and the temperature ofthe fluid which are all of the original variables of the problem. The missingpressure equation is derived and pressure boundary conditions are generatedby a finite difference approximation and the DRBEM coordinate matrix. Fundamentalsolution of Laplace equation is made use of to convert the nonlinearpartial differential equations into the boundary integral equations. The termsother than Laplacian are approximated by a series of radial basis functions. Thenonlinearities in the governing equations are easily handled by the use of theDRBEM coordinate matrix. The influence of the point source magnetic fieldon the Ferrohydrodynamics (FHD) Stokes, incompressible, and forced convectionflows are investigated first. The interaction between the buoyancy force, magnetization force and the viscous dissipation is discussed. Then, the effectof multiple point magnetic sources on the FHD incompressible flow is studied.DRBEM simulations of Magnetohydrodynamics (MHD) pipe flow and the flowbetween parallel infinite plates with slip velocity conditions are also presented.The coupled momentum and magnetic induction equations are combined andsolved without an iteration. This process provides the nodal solutions in onestroke both on the boundary and inside the problem domain. The importanceof the thesis study is in the fact that it is the first DRBEM application to FHDflow under point magnetic source and MHD flow with slip walls. 223

Bu çalışmada, metal iyonu olarak bakır(II) iyonu, anyonik ligant olarak bir amino asit olan L-triptofan (L-Trp), nötral ligant olarak ise düzlemsel bir yapıya sahip olan fenantrolin türevlerinden; 1,10-fenantrolin (phen), 4-metil-1,10-fenantrolin (4-mphen) ve 4,7-dimetil-1,10-fenantrolin (dmphen) seçildi. Çalışmada öncelikle; bakır(II) iyonunun phen, 4-mphen ve dmphen (A) ve L-trp (B) ile oluşturduğu ikili (MA, MB) ve karışık ligant (MAB) komplekslerine ait denge tepkimeleri 0,1 M KCl iyonik ortamında ve üç farklı sıcaklıkta (25°C, 35°C ve 45°C) potansiyometrik yöntem ile incelendi ve komplekslerin oluşum tepkimelerine ait termodinamik parametreler (∆H, ∆S ve ∆G) hesaplandı. Ligantların protonlanma sabitleri ve komplekslerin kararlılık (oluşum) sabitleri potansiyometrik titrasyon verilerinin BEST bilgisayar programına uygulanması ile hesaplandı. Ayrıca, çözeltide oluşan türlerin dağılımını belirlemek için ise SPE bilgisayar programı kullanıldı. İkinci olarak; seçilen ligantların bakır(II) iyonu ile oluşturduğu karışık ligant kompleksleri sentezlendi ve sentezlenen komplekslerin yapıları, elementel analiz, elektronik absorbsiyon spektroskopisi, IR spektroskopisi ve tek kristal X-ışını kırınım yöntemi ile aydınlatıldı. Son olarak ise; sentezlenen komplekslerin CT-DNA ile etkileşimleri ve antikanser aktiviteleri araştırıldı. Komplekslerin CT-DNA'ya bağlanma özellikleri; termal denatürasyon, elektronik absorbsiyon ve floresans spektroskopisi yöntemleri ile CT-DNA'yı bölme özellikleri ise agaroz jel elektroforez yöntemi ile incelendi. Antikanser aktivite çalışmalarında ise; komplekslerin akciğer, meme ve kolon kanser hücre hatlarında ve bir sağlıklı hücre hattında XTT testi ile IC50 değerleri hesaplandı ve sitotoksisiteleri belirlendi. In this study, copper(II) ion, L-tryptophan (L-trp) as an anionic ligand and 1,10-phenanthroline (phen), 4- methyl -1,10- phenanthroline (4-mphen) ve 4,7- dimethyl -1,10- phenanthroline (dmphen) as neutral ligand which have a planar structure were choosen.In the study firstly; the equilibrium reactions of binary (MA, MB) and mixed ligand (MAB) complexes formed with phen, 4-mphen and dmphen (A), L-trp (B) of copper (II) ion were examined at 25°C, 35°C and 45°C and in 0,1 M ionic medium and thermodynamic parameters (ΔH, ΔS and ΔG) via formation reactions of the complexes were calculated. The protonation constants of the ligands and the stability constants of the complexes were calculated with BEST computer program using potentiometric titration data. Therefore, the SPE computer program was used to determine the distrubition of the species formed in solution.Secondly; the mixed ligand complexes forming with copper(II) ion with the selected ligands were synthesized and the structures of the synthesized complexes were elucidated by elemental analysis, electronic absorption spectroscopy, IR spectroscopy and single crystal X-ray diffraction method.Finally, the interactions with CT-DNA of the synthesized complexes and anticancer activities were investigated. The binding properties with CT-DNA of the synthesized complexes were investigated by thermal denaturation, electronic absorption and fluorescence spectroscopy. Additionally, the cleavage activities with DNA of the synthesized complexes were investigated by agarose gel electrophoresis. In anticancer activity studies; IC50 values for the complexes were calculated by XTT test in lung, breast and colon cancer cell lines and in a healthy cell line of the complexes and their cytotoxicities were determined. 154
