As the knowledge base and importance of mitochondrial physiology to human health expand, the necessity for harmonizing nomenclature concerning mitochondrial respiratory states and rates has become increasingly apparent. Clarity of concept and consistency of nomenclature are key trademarks of a research field. These trademarks facilitate effective transdisciplinary communication, education, and ultimately further discovery. Peter Mitchell’s chemiosmotic theory establishes the link between vectorial and scalar energy transformation and coupling in oxidative phosphorylation. The unifying concept of the protonmotive force provides the framework for developing a consistent theory and nomenclature for mitochondrial physiology and bioenergetics. Herein, we follow IUPAC guidelines on general terms of physical chemistry, extended by considerations on open systems and irreversible thermodynamics. We align the nomenclature and symbols of classical bioenergetics with a concept-driven constructive terminology to express the meaning of each quantity clearly and consistently. In this position statement, in the frame of COST Action MitoEAGLE, we endeavour to provide a balanced view on mitochondrial respiratory control and a critical discussion on reporting data of mitochondrial respiration in terms of metabolic flows and fluxes. Uniform standards for evaluation of respiratory states and rates will ultimately support the development of databases of mitochondrial respiratory function in species, tissues, and cells.

Bakalářská práce se zabývá markery jaterního poškození. První část práce nás seznamuje s anatomií jater, včetně jaterních buněk a funkce jater. Základní funkce jater lze dělit na metabolické, sekreční a biotransformační. Druhá část se věnuje jednotlivým markerům a jejich detekci. V práci je zařazena i kapitola, která se věnuje nově používaným markerům. Biochemická vyšetření enzymových aktivit patří mezi nejvýznamnější ukazatele jaterního poškození. Závěrečná část práce popisuje jaterní poškození a onemocnění jater., My bachelor thesis is concerned with markers of liver injury. The first part of my work describes anatomy of the liver including liver cells and functions of the liver. Basic liver functions can be divided into metabolic, secretory and biotransformation. Second part is about particular markers and their detection. Also a chapter dealing with newly introduced markers is included. Biochemical assessments of enzymatic activities belong to most important markers of liver damage. Glutamátdehydrogenáza (GLDH) je enzym patřící do třídy oxidoreduktáz.Uplatnění nachází pri metabolismu dusíku, kde katalyzuje oxidační deaminaci glutamátu a také hraje důležitou roli v udržení energetiské homeostázy. Glutathionreduktasa (GR) je řazena do skupiny dimerických flavoenzymů, které katalyzují přenos elektronů díky přítomnosti flavinadenindinukleotidu ve své struktuře. Skládá se ze dvou identických podjednotek, na kterých jsou umístěna vazebná místa pro NADPH a také pro GSSG. Hlavní funkcí tohoto enzymu je NADPH-dependentní redukce oxidované formy glutathionu (GSSG) na formu redukovanou (GSH). Aktivita glutathionreduktasy je nejčastěji stanovována spektrofotometrickou metodou, která je založena na měření poklesu absorbance způsobené oxidací NADPH při 340 nm. Rychlost poklesu absorbance je přímo úměrná aktivitě GR. Při tomto stanovení se využívají dva substráty - GSSG a NADPH. Cílem naší práce bylo otestovat vliv různých parametrů metodiky (teplota, pH, stabilita) na stanovení aktivity glutathionreduktasy u purifikované kvasinkové GR a zjistit optimální podmínky stanovení. Pro stanovení aktivity glutathionreduktasy jsme jako optimální pufr určili draselný fosfátový pufr (pH 7,4). Testovali jsme také závislost koncentrace GSSG a NADPH na aktivitě GR, kdy jako optimální koncentrace byly zvoleny 10 mM GSSG a 1,6 mM NADPH. Zjistili jsme, že GR je stabilní po dobu nejméně dvou hodin při laboratorní teplotě (21 °C). P ři dlouhodobém zamražení (-20 °C) a zárove ň opakovaným zamražením/rozmražením došlo k poklesu aktivity. Dále jsme sledovali možný inhibiční vliv některých komerčně dostupných látek (ethakrynová kyselina, N-ethylmaleimid, diethylmaleát, glutathion) na aktivitu GR. V přítomnosti N-ethylmaleimidu překvapivě nedošlo k poklesu aktivity GR. Pouze u ethakrynové kyseliny a glutathionu byla inhibice prokázána. Při použití 500 mM koncentrace ethakrynové kyseliny byla zjištěna téměř 20% inhibice aktivity. Zjistili jsme také, že v přítomnosti 10 mM GSH, což je koncentrace vyskytující se běžně v buňce, došlo k poklesu glutathionreduktasové aktivity o 60 %., Glutathione reductase (GR) belongs to a group of dimeric flavoenzymes, which catalyze the transfer of electrons due to the presence of flavin adenin dinucleotide in their structure. GR consists of two identical subunits on which binding sites for NADPH and GSSG are located. The main function of this enzyme is NADPH-dependent reduction of oxidized glutathione form (GSSG) to reduced form (GSH). Activity of glutathione reductase is mostly determined by a spectrophotometric method, which is based on measurement of the absorbance decrease at 340 nm caused by oxidation of NADPH. The rate of absorbance decrease is directly proportional to activity of glutathione reductase. There are two substrates used - GSSG and NADPH. The aim of our study was to estimate the influence of various parameters of the method (temperature, pH, stability), to determine the activity of purified yeast glutathione reductase and establish the optimal conditions for determination. To determine the GR activity, we evaluated potassium phosphate buffer (pH 7.4) as the optimal buffer. We estimated the concentration dependence of GSSG and NADPH on the activity of glutathione reductase. The concentrations of 10 mM GSSG and 1.6 mM NADPH were chosen as optimal. We found that glutathione reductase is stable for at least two hours at laboratory temperature (21 °C). During prolonged freezing at -20 °C and also repeated freeze/thaw cycles the activity decreased significantly. Furthermore, we investigated the possible inhibitory effect of several commercially available substances (ethacrynic acid, N-ethylmaleimide, diethyl maleate, glutathione) on GR activity. Surprisingly, the decrease of GR activity did not occure in presence of N-ethylmaleimide. The enzyme activity was decreased only in presence of ethacrynic acid and glutathione. In 500 mM ethacrynic acid, we found almost 20% inhibition of the activity. Acetaminophen is one of commonly used analgesics and antipyretics. It is a safe drug at therapeutic doses but in the overdose, it causes liver injury which may lead to acute liver failure. The aim of the present work was to investigate a new mechanism contributing to the toxic acting of acetaminophen especially to explore the possible inhibition of glutathione reductase through an acetaminophen-glutathion conjugate. The acetaminophen-glutathione conjugate was synthesized by the reaction between glutathione and N-acetyl p-benzoquinone imine and purified by column chromatography. The inhibition effect of the conjugate on two types of glutathione reductase (from yeasts and rat hepatocytes) was tested spectrophotometrically. We synthesized the conjugate and found that the enzyme activity was reduced significantly after treatment with different concentrations of acetaminophenglutathione conjugate in both yeast and hepatocyte glutathione reductases (0.014 U/ml); the enzyme activity (from hepatocytes) was decreased to 79±7 %, 67±2 % and 39±7 %, in 0.37, 1.48 and 3.7 mM concentration of the conjugate, respectively. We found that glutathione reductase was dose-dependently inhibited by the conjugate of acetaminophen and glutathione. Our finding results in new consequences in description of acetaminophen toxicity and, in addition, explains previously published findings which have not been fully explainable till now.

Glutathion (gama-glutamylcysteinylglycin) je nízkomolekulární thiol vyskytující se v buňce v koncentraci 0,1-10 mmol/l, a je tak nejhojnějším intracelulárním thiolem. Díky své jedinečné struktuře je glutathion spojený s mnoha důležitými pochody. Působí jako antioxidant a chrání tak buňky před působením volných radikálů a reaktivních sloučenin. Dále hraje významnou roli při detoxikaci potenciálně škodlivých xenobiotik. Glutathion se vyskytuje v redukované (GSH), nebo oxidované formě (GSSG). Za fyziologických podmínek je více než 99 % glutathionu v buňce ve formě GSH. Zvýšená koncentrace GSSG se vyskytuje při oxidačním stresu, se kterým jsou spojena onemocnění, jako je rakovina, neurodegenerativní choroby, kardiovaskulární choroby, aj. Je tedy důležité najít optimální metodu, která přesně a spolehlivě stanoví množství glutathionu. Nejvíce rozšířenými jsou HPLC metody. Jejich velkou výhodou je vysoká specifita, citlivost a reprodukovatelnost. Hlavní nevýhodou je vysoká časová náročnost, a tím nemožnost měření velkého množství vzorků během krátké doby. Další nevýhodou metod HPLC jsou vysoké náklady. Cílem této práce bylo zavedení a optimalizace nové spektrofluorimetrické metody pro stanovení GSSG, jejímž základem je stanovení fluoreskujícího produktu, který vzniká reakcí glutathionu s o ftalaldehydem. Po optimalizaci vlnových délek ( EX = 335 nm, EM = 420 nm), hodnoty pH (pH 6) a teploty (T = 4 °C), byly zjištěny tyto parametry analýzy: pro kalibraci s linearitou 0 100 mol/l GSSG, R2 = 0,9999; intraassay (n = 10), CV(%) = 3,43 %; interassay (n = 10), CV(%) = 8,17 %; REC(%) = 100,95 % a hodnota detekčního limitu 0,30 mol/l GSSG. Korelační koeficient získaný porovnáním hodnot koncentrací vzorků s HPLC/FL byl stanoven r = 0,993. Můžeme tedy říci, že byla zavedena spolehlivá, citlivá a přesná metoda pro stanovení GSSG, jejíž velkou výhodou oproti HPLC je vysoká rychlost analýzy a nízká cenová náročnost., Glutathione (gama-glutamylcysteinylglycine) is a low molecular weight thiol, which is distributed in the cell in concentration 0.1-10 mmol/l, thus it is the most abundant intracellular thiol. Due to unique structural features, glutathione is linked to a number of important functions. Glutathione acts as an antioxidant and protects cells against the toxic effects of radicals and reactive oxygen species. It plays an important role in the detoxification of potentially harmful xenobiotics. Glutathione can be present as the reduced (GSH) and as the oxidized (GSSG) form. At physiological conditions, 99 % of glutathione is in reduced form. Higher amount of GSSG is observed during oxidative stress and is linked to cancer, neurodegerative and cardiovascular diseases. Regarding above described, it is important to find an optimal assay, which determines amount of both forms of glutathione precisely and reliably. HPLC is the mostly used method. The major advantage of HPLC is high specificity, sensitivity and reproducibility. However, it is not possible to measure a lot of samples in short time and another disadvantage is high cost. The aim of our work was optimization of GSSG assay. It is based on assay of fluorescent product, which is formed by reaction of glutathione and o phthalaldehyde. After wavelengths optimization ( EX = 335 nm, EM = 420 nm), pH value (pH = 6) and detection temperature (T = 4 °C), the analytical parameters were determined: calibration, R2 = 0.9999 with range of linearity 0 100 mol/l; intraassay (n = 10), CV(%) = 3.43 %; interassay (n = 10), CV(%) = 8.17 %; REC(%) = 100.95 % and the detection limit was 0.30 mol/l. The coefficient of correlation, which was acquired after comparision to HPLC/FL, was r = 0.993. We can assume, we have optimized the reliable, sensitive and accurate assay for determination of GSSG.
