This research explores the possibilities and limitations of WD-XRF, applied as a method for quantification of 20 elements in plant material, using spiked cellulose standards for calibration. Three different analytical methods were investigated: 1) standards created from pure spiked cellulose; 2) spiked cellulose mixed with 20% of binder and 3) spiked cellulose applied as a thin layer on an inert carrier. Sensitivity, linearity, limit of detection, limit of quantification, repeatability, intralaboratory reproducibility, and accuracy were determined and compared. The accuracy of the investigated methods was tested by analysis of standard reference materials and comparison with other routinely used analytical techniques (ICP-OES and ICP-MS). The comparison included real plant samples which were collected from the environment characterized by different pollution levels. The accuracy of the semiquantitative standardless method was also considered and compared with other investigated methods. Tested methods can be very precise, with good intralaboratory reproducibility over wide linear range.

Mycotoxins are widely studied by many research groups in all aspects, but the stability of these compounds needs further research for clarification. The objective of this study is to evaluate deoxynivalenol and zearalenone stability during all steps of the malting and brewing processes. The levels of these compounds decreased significantly during the production process (barley to beer). During the malting process, the DON levels decreased significantly in the steeping, germination, and malting steps (62%, 51.5%, and 68%, respectively). Considering ZEN, when the levels were compared between barley and the last step of the process, a significant decrease was observed. Most of the mycotoxins produced were transferred to the rootlets and spent grains, which is advantageous considering the final product. Furthermore, the mycotoxin dietary intake estimation was included in this study. The results proved that if the concentrations of target mycotoxins in raw material are under the limits established by the regulations, the levels decrease during the malting and brewing processes and make the beer secure for consumers. The quality of the five commodities involved in the beer process plays a decisive role in the creation of a safe final product.

Předkládaná disertační práce se zabývá přípravou nových substitučních derivátů kobalt bis(dikarbolidu) a ikosaedrálních karboranů, navržených primárně s cílem vyvinout nové léčivo proti AIDS založené na inhibici enzymu HIV-1 proteasy boranovými klastry, které by dokázalo překonat nedostatky stávajících organických inhibitorů. Většina syntetizovaných látek byla poskytnuta biochemikům k provedení testů kinetiky inhibice těchto látek na isolovaném enzymu (IC50), HIV-1 proteasy nebo dále testů ve tkáňových kulturách buněk infikovaných virem HIV (EC50). Hodnoty koncetrací testované látky, při kterých dochází k poklesu aktivity enzymu (štepení substrátu) právě o 50 % jsou definovány jako koncentrace IC50 (in vitro) a EC50 (in vivo). Tato data byla zařazena do knihovny látek spolu s druhem mechanismu, kterým jsou dané látky schopny enzym selektivně inhibovat, za předpokladu, že byl stanoven. Tyto poznatky jsou klíčem k pochopení substruktury možného budoucího léčiva. Pro tyto účely byla autorem disertační práce syntetizována a charakterizována série celkem 14-ti nových sloučenin, obsahujících motiv dvou vzájmně propojených kobalt bis(dikarbolidových) jednotek, spojených flexibilním organickým řetězcem se zabudovanou funkční skupinou v centru molekuly. Dále bylo připraveno 14 nových B-N derivátů kobalt bis(dikarbolidu), které nesou nitriliovou, ammoniovou, amidovou a amidinovou funkční skupinu. Tyto látky mají použití jako nové stavební bloky pro organickou syntézu, použitelné pro inkorporaci tohoto typu látek do materiálů s řadou unikátních vlastností. Všechny nově připravené látky byly charakterizovány s využitím pokročilých instrumentálních metod strukturního výzkumu, tedy NMR, HPLC MS-ESI a RTG analýzy., The research in this Ph.D. thesis deals with the preparation of new substitutional derivatives of cobaltabisdicarbollide and icosahedral carboranes designed primarily to search for new drugs to inhibit the HIV-1 protease enzyme, which causes AIDS. The use of borane clusters overcomes the shortcomings of existing organic inhibitors. After synthesis of the compounds reported here, they were passed to biochemists who measured the inhibition kinetics of these substances on an isolated enzyme HIV-1 protease (IC50), and performed similar tests on HIV-1 infected cell cultures (EC50). For each compound, the concentration which caused an inhibition of enzyme activity (substrate cleavage) of 50 % of the concentrations of, IC50 (in vitro) and EC50 (in vivo) were included into boron compounds library. Additionally included was the specific mechanism by which the substances were able to selectively inhibit the enzyme. These findings are a key to possible future drug substructure determination. The author of this thesis synthesized and characterized a series of fourteen new compounds containing a motif of two units of cobalt bisdicarbollide connected with a flexible organic chain via a functional group embedded in the center of the molecule. Twelve new B-N derivatives offering cobalta bisdicarbollide nitrile, amino, amido and amidine functional groups were also prepared. These can serve as new building blocks in organic synthesis, suitable for the incorporation of these types of substances in materials and thereby introducing many unique features. Tato práce se zabývá strukturní charakterizací organokovových sloučenin se zaměřením na sloučeniny s polyizotopickým kovem. Pro většinu popisovaných experimentů v této práci byl použit hmotnostní spektrometr s elektrosprejovou ionizací a analyzátorem na principu iontové pasti, pomocí kterého byla získána hmotnostní spektra prvního řádu a tandemová hmotnostní spektra při snímání kladných i záporných iontů více než 250 organokovových sloučenin a následně studovány i fragmentární mechanismy. Pro srovnání a hlavně pro získání dalších informací (v případě experimentů tykajících se organokovových sloučenin s atomem niklu a germania a jejich komplexů s aminokyselinami) byl použit hmotnostní spektrometr s hybridním analyzátorem QqTOF. Vysoké rozlišení tohoto analyzátoru sloužilo pro potvrzení elementárního složení jednotlivých iontů., In this work, two independent electrospray mass spectrometers with different analyzers were used for performance of described experiments.
