1.ÖZETSisplatin’e bağlı ürogenital organ toksisitesine karşı kuversetinin olası koruyucu etkilerinin incelenmesiAmaç: Çalışmanın amacı, sisplatin’e bağlı ürogenital organ toksisitesine karşı kuversetinin olası koruyucu etkilerinin incelemektir.Gereç ve Yöntem: Çalışmamızda 4 aylık erkek sıçanlar kullanılmıştır. Hayvanlar 4 gruba (kontrol+ SF, kontrol+kuversetin, sisplatin+SF, sisplatin+kuversetin) ayrılmıştır. Hayvanlara eter anestezisi uygulanarak oral gavaj aracılığı ile 21 gün boyunca SF ve kuversetin uygulanmıştır. Dekapitasyon sonrası kan ve doku (böbrek, mesane, testis ve korpus kavernozum) örnekleri alınmıştır. Kanda DNA hasarı, dokularda apoptotik belirteçler, antioksidan ve oksidatif stres parametreleri incelenmiştir. Mesane ve korpus kavernozum dokularında izole organ banyosunda izometrik kasılma çalışmaları yapılmıştır. Bulgular Graphpad Prism v5 programı ile değerlendirilmiştir.Bulgular: Yapılan izometrik kasılma çalışmaları sonucu korpus kavernozum ve mesane dokularının kasılmalarında iyileşme gözlemlenirken, kavernoz dokunun gevşeme yanıtları incelendiğinde kuversetinin herhangi iyileştirici etkisi gözlemlenmemiştir. Alınan dokularda, apoptotik belirteçlerden kaspaz-3 ve oksidatif stres parametrelerinden 8-OHdG, malondialdehit, glutatyon, süperoksit dismutaz düzeylerinde olumlu etkileri olduğu belirlenmiştir. Sonuç: Sisplatin’e bağlı ürogenital organ toksisitesine karşı kuversetinin koruyucu etkileri gözlemlenmiştir.Anahtar sözcükler: Sisplatin, kuversetin, antioksidan sistem, apoptoz, ürogenital organlar.2.SUMMARYInvestigation of the possible protective effects of quersetin against cisplatin-induced urogenital organ toxicity.Aim: The aim of this study is investigate of the possible protective effects of quersetin against cisplatin-induced urogenital organ toxicity.Material and Method: In the study, animals were divided into 4 groups (control+SF, control+quercetin, cisplatin+ SF, cisplatin+quercetin). For 21 days, quercetin and SF was administered to the animals via oral gavage under ether anesthesia. After decapitation, blood and tissue (kidney, bladder, testis, corpus cavernosum) samples were collected. In blood DNA damage and in tissue samples apoptotic, antioxidant and oxidative stress parameters were examined. In isolated tissue bath, isometric contraction study was perfomed with corpus cavernosum and bladder tissues. Data was assessed with Graphpad Prism v5 program.Results: As a result of performed isometric contraction of the corpus cavernosum and bladder tissues improvements was observed but when we examined cavernosum tissue’s relaxation responses, there was not any healing effects of quercetin. Apoptotic parameter caspase-3 and oxidative stress parameters, 8-OHdG, malondialdehyde, glutathione, superoxide dismutase showed significant improvements.Conclusion: Quercetin improved cisplatin dependent reproductive organ toxicity and provided benefits in apoptosis, oxidative stress parameters.Key Words: Cisplatin, quercetin,antioxidant system,apoptosis, reproductive organ
