Uyku yoksunluğu öğrenme-belleği bozmakta; strese, inflamasyona ve davranış değişikliklerine neden olmaktadır. Bu çalışmada, üridinin REM uyku yoksunluğu oluşturulan sıçanlarda öğrenme-bellek, sitokinler ve anksiyete üzerindeki etkileri incelenmiştir. Yetişkin erkek Wistar albino sıçanlar (n=150) A, B ve C gruplarına ayrılmıştır. REM uyku yoksunluğu oluşturmak için modifiye edilmiş çoklu platform yönteminden yararlanılmıştır. Tüm sıçanlar takip edildikleri kafeslere ve üridin (1mmol/kg, i.p.)- salin (1ml/kg, i.p.) enjeksiyonuna göre 6 alt gruba ayrılmıştır. A ve B grubundaki sıçanlar 96 saat, C grubundaki sıçanlar 24 saat boyunca uygun kafeslerde takip edilmiştir. A ve B gruplarına 4 gün günde iki, 5. gün bir defa; C grubuna birinci gün günde 2, 2. gün bir defa enjeksiyon yapılmıştır. A grubunda öğrenme-bellek performansı Morris su tankı testi ile, B ve C gruplarında anksiyete durumu ve lokomotor aktivite açık alan ve yükseltilmiş artı labirent testleri ile analiz edilmiştir. Testlerin ardından A grubundaki sıçanlardan elde edilen serum örneklerinde kortikosteron; hipokampus homojenatlarında sitokin seviyeleri ELISA ve RT-PCR yöntemleri ile ölçülmüştür. Çalışmamızda REM uyku yoksunluğunun öğrenmeyi bozduğu ve üridin uygulamasının bozulan öğrenme parametreleri üzerine olumlu etki ettiği gösterilmiştir. REM uyku yoksunluğu oluşturulan sıçanlarda kortikosteron seviyelerinin arttığı ancak üridinin bu artış üzerine etkisi olmadığı gözlenmiştir. REM uyku yoksunluğu oluşturulan sıçanların hipokampus homojenatlarında IL-1β, IL-4 ve IL-6 sitokinlerinin mRNA ekspresyonları ile birlikte IL-1β, IL-4, IL-6, IL-10 ve TNF-α sitokin seviyelerinin arttığı saptanmıştır. Üridinin REM uyku yoksunluğu oluşturulan sıçanlarda IL-1β mRNA ekspresyonlarını azalttığı gözlenmiştir. Yirmi dört ve 96 saatlik REM uyku yoksunluğu sonucunda lokomotor ve keşfetme davranışlarının arttığı tespit edilmiştir. Üridinin bu davranışlar üzerine olumlu etki ettiği gözlenmiştir. Sleep deprivation impairs learning-memory and causes stress, inflammation and behavioral changes. In this study, effects of uridine on learning-memory, cytokines and anxiety in REM sleep deprived (REMSD) rats were investigated. Adult male Wistar albino rats (n=150) were randomized to A, B and C groups. REMSD was induced by modified multiple platform method. Rats were divided into 6 subgroups according to cages they were placed and uridine (1 mmol/kg, i.p.)-saline (1ml/kg, i.p.) injections. Groups A and B were tracked for 96h and treated with uridine or saline twice for 4 days and once on the 5thday whereas C was tracked for 24h and treated twice on 1st and once on next day. Group A was tested on Morris water maze for learning-memory whereas B and C were tested on open field and elevated-plus maze for anxiety. Afterwards, corticosterone levels from serum and cytokine levels from hippocampi of group A were analyzed by ELISA and RT-PCR. We conclude that REMSD in impairs learning and uridine treatment ameliorates this impairment. It has been observed that corticosterone levels increased in REMSD rats, whereas uridine had no effect. REMSD increased IL-1β, IL-4, IL-6, IL-10 and TNF-α cytokine levels as well as mRNA expressions of IL-1β, IL-4 and IL-6. mRNA expressions of IL-1β were decreased in REMSD rats by uridine treatment. It has been determined that locomotor and exploratory behaviors increase as a result of 24 and 96 hours of REMSD. It has been observed that uridine has a positive effect on these behaviors.

Domates (Solanum lycopersicum L.) bitkisi anavatanı Güney Amerika’nın batı kıyıları olan Solanaceae familyası türlerinden biridir. Sebze üretimi ve tüketimi içerisinde hem ülkemizde hem de dünya genelinde en çok üretilen ve tüketilen bitkiler arasında yer almaktadır. Domatesin pas akarına karşı toleransını arttıracak (zararın azalması ve hızlıca iyileşmesi ile bitki gelişimini teşvik etme gibi) alternatif çözümlere ihtiyaç duyulmaktadır. Bu çalışmayla birlikte, kontrollü koşullarda domates pas akarı bulaştırılan domates fidelerinde sadece üridin veya sadece akarisitlerin (abamectin, acequinocyl, pyridaben, spiromesifen) yarı dozlarının uygulaması ila bunların kombinasyonlarının akar zararına olan etkilerinin belirlenmesi amaçlanmıştır. Literatür taraması yapıldığında bu konuyla ilgili herhangi bir araştırmanın mevcut olmadığı gözlemlenmiştir. Denemede akarisit uygulanan kombinasyonlarda abamectin 0,25 mg/L, acequinocyl 62,5 mg/L, pyridaben 15 mg/L ve spiromesifen 25 mg/L konsantrasyonları kullanılmıştır. Bitkilerde büyüme oranı, klorofil miktarı, yaprak oransal su kapsamı ve turgor kaybı gibi fizyolojik parametreler ölçülmüştür. Bununla birlikte MDA, glutatyon ve askorbik asit miktarı ile antioksidatif savunma sistemine ait bazı enzimlerin CAT, APOX ve SOD aktivitesindeki değişimler de incelenmiştir. Travmatik beyin hasarı dünya çapında morbidite ve mortalitesi yüksek bir acil durumdur. Hastaneye başvuran hastaların çoğuna hafif travmatik beyin hasarı tanısı konmakla birlikte hastalarda uzun dönemde kalıcı hasarlar bırakabilmektedir. Travmatik beyin hasarı fizyopatolojik süreçlerinde birbirini takip eden; primer hasar ve sekonder hasar dönemi yer almaktadır. Primer beyin hasarı travmadan hemen sonra oluşmakta iken sekonder hasar primer hasardan sonra saatler ve günler sonra ortaya çıkmkatadır. Çalışmamızın amacı; deneysel olarak travmatik beyin hasarı modeli uygulaması ile sekonder hasar dönemi ile ilişkili oksidatif, inflamatuar ve apoptotik parametrelerde meydana gelen değişiklikleri incelemek ve Üridin ve/veya hipotermi kombine tedavisinin nöroprotektif etkisini araştırmaktır. Çalışmamızda, Marmarou ve arkadaşlarının tanımladığı serbest ağırlık düşürme modeli kullanılarak travmatik beyin hasarı oluşturulmuş ve hafif hipotermi (32-34.0±1°C) 4 saat boyunca uygulanmıştır. Üridin travmatik beyin hasarını takiben 7 gün boyunca 500mg/kg intraperitoneal uygulanmıştır. Takiben beyin dokusu analizleri, TTC, Western Blot ve ELISA kitleri kullanılarak yapılmıştır. Oksidatif parametreler içerisinden Glutatyon Peroksidaz kombine tedavi uygulaması ile artarken, Katalaz ve Myeloperoksidaz azalmıştır. İnflamatuar belirteçlerden olan İnterlökin-1β ve İnterlökin-6 ise kombine tedavi uygulaması ile azalma göstermiştir. Nekrotik süreçte görev alan Poli ADP-riboz polimeraz-1 ise TBH sonrası artarken, hipotermi, üridin ve kombine tedavi uygulaması ile azalmıştır. Traumatic brain injury is accepted as an emergency situation with high morbidity and mortality rates worldwide. Although most of the patients admitted to the hospital are diagnosed with mild traumatic brain injury, permanent damage could be seen in the long term period. The physiopathological processes of traumatic brain injury are classified as; primary injury and secondary injury period. While primary brain injury occurs immediately after trauma, secondary injury occurs hours and days after primary injury. The aim of this study is to examine the changes in oxidative, inflammatory and apoptotic parameters within the secondary injury period in an experimental model of traumatic brain injury and to investigate the neuroprotective effect of Uridine and/or hypothermia combination therapy. In our study, the free weight drop model defined by Marmarou et al. was used and mild hypothermia (32-34.0±1°C) was applied for 4 hours. Uridine was administered intraperitoneally in 500mg/kg dose for 7 days following traumatic brain injury. Subsequently, brain tissue analyzes were performed using TTC, Western Blot and ELISA kits. Among the oxidative parameters, Glutathione Peroxidase was increased with the combination therapy, while Catalase and Myeloperoxidase were decreased. Interleukin-1β and Interleukin-6, which are accepted as inflammatory markers, decreased with combination therapy. Poly ADP-ribose polymerase-1, which is involved in the necrotic process, was increased after TBI and decreased with hypothermia, uridine and combination therapy. The main goal in the approach to traumatic brain injury is to protect the patient against the effects that occur during the secondary injury period. In our study, the efficacy of Uridine-hypothermia combination therapy was demonstrated after seven days of treatment, which is the first in the literature in this respect. With other studies to be conducted in this area, it is aimed to be a treatment approach that can be used in routine practice in mild traumatic brain injury and to be included in trauma patient management guidelines.

Bu tez çalışmasının amacı, farklı konsantrasyonlarda üridin eklenerek polikaprolakton (PCL) polimerinden elektro çekim yöntemi ile nanolifli yüzeylerin üretilmesi ve yara örtüsü olarak kullanımlarının araştırılmasıdır. Çalışma, nanolifli yüzeylerin üretimi, karakterizasyonu ve in vitro-in vivo araştırmalarının yapılması olmak üzere iki kısımdan oluşmaktadır. İlk kısımda, farklı konsantrasyonlarda PCL polimer çözeltilerinden nanolifli yüzeyler üretilmiş ve üridin katkısı yapılacak olan en uygun polimer çözelti konsantrasyonu belirlenmiştir. Optimum polimer konsantrasyonunun belirlenmesinden sonra, polimer çözeltilerine farklı oranlarda (ağırlıkça %0,1, %0,5 ve %1) üridin eklenmiş ve nanolif üretimine imkân veren en uygun üretim parametreleri kullanılarak elektro çekim yöntemi ile nanolifli yüzeyler elde edilmiştir. Üretilen numunelerin yüzey karakterizasyonları taramalı elektron mikroskobu (SEM) ve temas açısı ölçümleri ile, içyapı analizleri ise Fourier Dönüşümlü Kızılötesi Spektroskopi (FTIR) ile tespit edilmiştir. Çalışmanın ikinci kısmında üridin katkılı nanolifli yüzeylerin yara örtüsü olarak performansı in vitro ve in vivo olarak incelenmiştir. In vitro çalışmalarda üridinin salıverilmesi ultraviyole görünür ışık (UV-VIS) spektrofotometresi ile takip edilmiştir. Eklenen üridin konsantrasyonu arttıkça yüzeyden patlama salıverilmesinin arttığı tespit edilmiştir. In vivo deneylerde, üridin katkılı ve katkısız nanolifli yüzeylerin yanısıra, yara örtüsü performansını karşılaştırabilmek amacı ile ticari bir yara örtüsü kullanılmıştır. Deneylerde Sprague Dawley cinsi dişi sıçanların sırt bölgelerinde yara modeli oluşturulmuş ve yara kapanma oranı belirli günlerde takip edilmiştir. Bu oranlar incelendiğinde, üridin içeren yara örtülerinin, katkısız nanolifli yara örtüsünden daha iyi performans sergilediği, ticari yara örtüsüne göre ilk günlerde yara iyileşmesini daha çok hızlandırdığı görülmüştü. The aim of this thesis is to produce nanofibrous surfaces from polycaprolactone (PCL) polymer by adding uridine at different concentrations by electrospinning method and to investigate their use as a wound dressing. The study consists of two parts. In the first part, nanofiber surfaces were IX, 132produced from PCL polymer solutions at different concentrations. After determining the optimum polymer concentration, uridine was added at different rates (0.1%, 0.5% and 1%(w/w)) to the polymer solutions and nanofiber surfaces were obtained by electrospinning method using the most suitable production parameters that allow nanofiber production. The surface characterizations of the samples were determined by scanning electron microscopy (SEM), contact angle measurements, internal structure analyzes were determined by Fourier Transform Infrared Spectroscopy (FTIR). In the second part of the study, the performance of uridine-added nanofiber surfaces as a wound dressing was investigated in vitro and in vivo. The release of uridine was followed by Ultraviolet Visible light (UV-VIS) spectrophotometer. It was determined that as the added uridine concentration increased, the burst release from the surface increased. In the in vivo experiments, a commercial dressing was used to compare the dressing performance with and without uridine added nanofibrous surfaces. A wound model was created in the back regions of Sprague Dawley females and the rate of wound closure was followed on certain days. When these ratios were examined, it was observed that uridine-containing dressings performed better than the pure nanofiber dressings and accelerated wound healing in the first days compared to commercial dressings.

Bir pirimidin nükleozidi olan üridin, vücutta serbest halde bulunabildiği gibi nükleotidlerin fosfatlı hallerinde de bulunabilir. Nükleotid şekerlerin ve nükleik asitlerin yapısına girer, glikojen biyosentezi, protein ve lipid glikolizasyonu gibi birçok fizyolojik fonksiyonda yer alır. Üridin nükleotidleri pürinerjik reseptörler üzerinden hücresel etkilerini gösterirler. Hemostaz, dolaşım sistemi içinde bulunması gereken kanın, herhangi bir hasar durumunda bu sistem dışına çıkmasını engellemek, kanamayı durdurmak amacıyla işlev gören fizyolojik bir sistemdir. Kan kaybını önleyerek hayat kurtarıcı olabildiği gibi, patofizyolojik şartlarda normal akışın olması gereken damar yatağında anormal hemostazın oluşması ile hayatı tehdit eden sonuçlara da yol açabilmektedir. Hemostazın hücresel bileşeni olan trombositler üzerinde pürinerjik reseptörlerin bazı alt tipleri tanımlanmıştır. Özellikle adenin ve nükletidlerinin trombositler üzerindeki etkilerine dair aryrıntılı çalışmalar bulunsa da üridin ve nükleotidlerine dair bu çalışmalar sınırlıdır. Bu çalışmada üridin ve nükleotidlerinin hemostaz parametreleri üzerine etkileri optik agregometre ile hücresel düzeyde, tromboelastogram ile sistem düzeyinde araştırılmıştır.Agregometre deneylerinde üridinin 10, 100 ve 1000 M son konsantrasyonları, UMP, UDP ve UTP'nin 1, 10, 50, 100, 500 ve 1000 M son konsantrasyonları çalışıldı. Trombositten zengin plazma üzerine üridin veya nükleotidi eklenerek 3 dakika inkübe edildi. Ardından ADP, kollajen veya epinefrin ile agregayon uyarılarak elde edilen agregasyon eğrileri kontrol grubuna göre karşılaştırıldı. Tromboelastogram deneylerinde üridin ve nükleotidlerinin 1000 M son konsantrasyonları çalışıldı. Tromboelastogram parametreleri kontrol grubuna göre değerlendirildi.Agregometrede üridinin düşük konsantrasyonda kollajen ile uyarılan agregasyonu aktive ettiği, UDP ve özellikle UTP'nin konsantrasyona bağlı olarak ADP ve kollajen ile uyarılan agregasyonu inhibe ettiği bulunmuştur. UDP ve UTP, epinefrin ile uyarılan agregasyonlarda düşük konsantrasyonlarda aktivasyon, yüksek konsantrasyonlarda inhibisyon yönünde sonuçlar vermiştir. Tromboelastogramda üridin ve UMP'nin etkisi gözlenmezken, UDP'nin hiperkoagülan, UTP'nin hipokoagülan etkisi gözlenmiştir.UMP'nin etkisiz olması, UDP ve özellikle UTP'nin konsantrasyon artışla birlikte etkilerinin gözlenmesi, trombositler üzerindeki inhibitör etkilerinin fosfat gruplarıyla ilişkili olduğunu düşündürmektedir. UTP'nin inhibitör etkisi tromboelastogram ile de gözlenmiştir. Tromboelastogramdaki UDP'nin aktivasyon yönündeki etkileri, UTP'ye göre zıt ve ilk defa ortaya konan bulgulardır. Uridine, a pyrimidine nucleoside, may be present in the body in free form or in phosphated forms of nucleotides. Uridine enters the structure of nucleotide sugars and the structure of nucleic acids. It is involved in many physiological functions such as glycogen biosynthesis, protein and lipid glycolization. The uridine nucleotides exert their cellular effects through purinergic receptors.Hemostasis is a physiological system that functions to prevent the blood that should be present in the circulatory system from going out of the system and to stop the bleeding in case of any damage. It can be life-saving by preventing blood loss, and can lead to life-threatening consequences with abnormal hemostasis in the vascular bed. Several subtypes of purinergic receptors have been identified on platelets. Although there are detailed studies on the effects of adenine and nucletides on platelets, these studies on uridine and nucleotides are limited. In this study, the effects of uridine and nucleotides on hemostasis parameters were investigated at cellular level by optical aggregometer, and at system level by thromboelastogram.In the aggregometer experiments, the final concentrations of uridine 10, 100 and 1000 M and final concentrations of 1, 10, 50, 100, 500 and 1000 M of UMP, UDP and UTP were studied. Uridine or nucleotide was added to the platelet-rich plasma and incubated for 3 minutes. Then, aggregation curves obtained by stimulating aggregation with ADP, collagen or epinephrine were compared according to the control group. In thromboelastogram experiments, final concentrations of 1000 M of uridine and nucleotides were studied. Thromboelastogram parameters were evaluated according to the control group.In the aggregometer, uridine was found to activate low concentration collagen-induced aggregation, UDP and especially UTP inhibited ADP and collagen-induced aggregation, depending on the concentration. UDP and UTP showed low concentrations of activation and high concentrations of inhibition in epinephrine-induced aggregations. While thromboelastogram showed no effect of uridine and UMP, hypercoagulant effect of UDP and hypocoagulant effect of UTP were observed.The ineffectiveness of UMP, the observation of the effects of UDP and especially UTP with increasing concentration suggest that inhibitory effects on platelets are related to phosphate groups. The inhibitory effect of UTP was also observed by thromboelastogram. The effects of UDP on activation in thromboelastogram are opposite and first-time findings compared to UTP. 64

Preterm yenidoğanlara neonatal yoğun bakım ünitelerinde, resüsitasyon, pulmoner hipertansiyon ve solunum sıkıntısı gibi nedenlerden dolayı oksijen tedavisi uygulanmaktadır. Term ve preterm yenidoğanlar, endojen radikal süpürücü sistemlerinin tam olarak olgunlaşmamış olması sebebiyle reaktif oksijen türlerinin neden olduğu bozulmalara duyarlıdırlar. Reaktif oksijen türleri, fizyolojik olarak antioksidan sistemler aracılığı ile düzenlenmektedir. Reaktif oksijen türleri ve antioksidan arasındaki dengenin bozulması DNA ve mitokondriyal membran, apoptoz ve hücresel fonksiyon bozukluğuna bağlı olarak oksidatif strese neden olabilir. Oksidatif stresin hiperoksiyi takiben akciğer, retina ve muhtemelen genel doku hasarından sorumlu olduğu kabul edilmiştir. Bu çalışmanın amacı, yenidoğan sıçan beyninde hiperoksi uygulaması sonrasında oluşan oksidatif stresin neden olduğu bozulmalara karşı üridinin, antioksidan sistem aracılığıyla olası tedavi etkinliğini araştırmaktır. Çalışmamızda 53 adet neonatal Spraque-Dawley cinsi sıçan doğumdan 6 gün sonra randomize olarak 5 gruba ayrılmıştır. Oda havasındaki sıçanlara 6. gün tek doz intraperitoneal serum fizyolojik (%0,9 NaCl) veya diğer gruplara hiperoksi maruziyetinden 15 dakika önce tek doz 100 mg/kg, 300 mg/kg, 500 mg/kg dozunda üridin intraperitoneal (i.p.) enjeksiyonu yapılmıştır. 48 saat boyunca oda havasında (%21 O2) tutulmuş veya hiperoksiye (%80 O2) maruz bırakılan sıçanlar takiben dekapite edilerek tüm beyin homojenatlarından Western Blot yöntemiyle DJ-1 proteininin analizi ve Malondialdehit (MDA), Myeloperoksidaz (MPO), Süperoksit Dismutaz (SOD), Glutatyon Peroksidaz (GSH-Px) enzimlerinin biyokimyasal analizleri yapılmıştır. Oxygen therapy is applied to preterm neonates in neonatal intensive care units for reasons such as resuscitation, pulmonary hypertension and respiratory distress. Term and preterm neonates are susceptible to deterioration caused by reactive oxygen species because their endogenous radical scavenging systems are not fully mature. Reactive oxygen species are regulated physiologically by antioxidant systems. Disruption of the balance between reactive oxygen species and antioxidants may cause oxidative stress due to DNA and mitochondrial membrane, apoptosis and cellular dysfunction. The aim of this study was to investigate the possible treatment efficiency of uridine through antioxidant system against the deterioration caused by oxidative stress after hyperoxia application in pups brain. Rats in room air were injected intraperitoneally (i.p.) a single dose of either saline (0.9% NaCl) or uridine100 mg/kg, 300 mg/kg, 500 mg/kg 15 minutes prior to hyperoxia exposure to on day 6. Rat pups which were kept either at room air (21% O2) or exposed to hyperoxia (80% O2) for 48 hours, were subsequent to decapitated. The homogenates obtained from the whole brain were analyzed by Western blotting DJ-1 protein, and the biochemical analysis of Malondialdehyde (MDA), Myeloperoxidase (MPO), Superoxide Dismutase (SOD), Glutathione Peroxidase (GSH-Px) enzymes were conducted.

Bir pirimidin nükleozidi olan üridin vücutta serbest halde bulunabildiği gibi nükleotidlerin (mono-, di- ve tri-fosfatlı halleri), nükleotid şekerlerin (UDP-glukoz ve UDP-galaktoz) ve nükleik asitlerin (RNA) yapisina girerek glikojen biyosentezi, protein ve lipid glikolizasyonu gibi fizyolojik fonksiyonlarda yer alır. Nükleotid reseptörlerinin tanımlanmasıyla, pirimidin nükleozidlerinin sinirsel iletide de (pirimidinerjik transmisyon) rolü olduğu öne sürülmektedir. Üridin özellikle RNA'dan serbestleşmek suretiyle gıdalarla vücuda alınabildiği halde hücrelere nasıl alındığı tam olarak bilinmemektedir. İn vitro çalışmalarda pirimidin nükleozid taşınmasının ve dokulara alınmasının hormonlarla (insülin, glukagon) kontrol edilebileceği ve pirimidinlerin dokularda nükleotid ve nükleik asitlerin yapısına katılabileceği belirtilmiştir.Çalışmamızda; insülin'in sıçanlarda üridin düzeylerine etkilerinin araştırılması planlanmıştır. Bu amaçla ilk olarak; açlık ve tokluk durumlarında sıçanların kan glukoz ve serum üridin düzeyleri incelendi. Arteryel kanül yerleştirilmiş olan sıçanlardan öncelikle 24 saat açlık sonrası, daha sonra da 1 saatlik beslenmeyi takiben çeşitli zaman aralıklarında kan alınarak kan glukoz ve serum üridin düzeylerinin zamana bağlı değişimleri incelendi. Benzer bir kurguda kanül takılmadan 24 saat açlık ve sonrasında 2 saatlik tokluk noktalarında sakrifiye edilen sıçanlardan kan örnekleri toplanarak serum insülin düzeyleri analiz edildi. İkinci olarak insülin'in periton içi yolla farklı dozlarda enjeksiyonu sonrası serum üridin düzeylerinin değişimleri incelendi. İnsülin enjeksiyonunu takiben farklı zaman aralıklarında femoral arterine yerleştirilen kateterden kan toplanarak üridin, glukoz ve insülin düzeyi ölçümleri yapıldı. Son çalışmada ise serum üridin düzeylerindeki değişimlerin glukozun hücre içine girip girmemesine bağlı olup olmadığı hiperinsülinemik öglisemik-klemp yöntemi uygulanarak araştırılmıştır. Bulgularımız, serum insülin düzeylerinin artmış olduğu tokluk anında üridin düzeylerinin azaldığını, subkutan (s.c.) insülin uygulamasının serum üridin konsantrasyonunu doza ve zamana bağlı olarak azalttığını ve üridin düzeylerinde insülin tarafından uyarılan azalmaların hücre içine Glukoz girişinden bağımsız olduğunu göstermiştir. Çalışmamızdan elde edilen bulgular literatürde ilk kez dolaşımdaki üridin'in hücrelere alınmasının insülin tarafından teşvik edildiğini ve bu işlemin hücrelere glukoz girişinden bağımsız olduğunu düşündürmektedir. İnsülin uyarısıyla üridin'in hücre içine alınmasının moleküler mekanizmalarının aydınlatılması için daha fazla çalışmaya ihtiyaç bulunmaktadır.Anahtar Sözcükler: Üridin, İnsülin, Hiperinsülinemik-Öglisemik klemp, hücre içine alım Uridine, a pyrimidine nucleoside, is an endogenous compound detected both in free form and in the form of mono-, di- and trinucleotides and nucleotide sugars (UDP-glucose and UDP-galactose). It is also an important part of ribonucleic acid (RNA) and plays critical roles in many physiological functions like biosynthesis of glycogen and glycosylation of proteins and lipids. With the identification of nucleotide receptors, role of pyrimidine nucleosides (pyrimidinergic transmission) in the communication of nerve cells has also been proposed. The mechansim for the uptake of uridine by the cells of body, after being released from the RNA as a food ingredient, is yet unknown. In in-vitro experiments, the transport and uptake of uridine by the tissues has been proposed to be associated with hormones (insulin, glucagon) so that uridine can be incorporated in nucleotides and nucleic acids. The aim of our experiment was to determine the effects of insulin on levels of uridine in rats. Therefore, firstly, levels of glucose and uridine were investigated during fasting and in fully-fed animals. Blood samples from rats inserted with arterial cannulas were obtained in fasting state after 24 hour food restriction, and then at certain time points 1 hour following feeding in order to investigate time-dependent changes in glucose and uridine levels. In a separate group of animals, serum insulin levels were analyzed once during fasting state and 2 hours after feeding has started. In a second protocol, serum uridine levels were determined after intraperitoneal injection of different doses of insulin. Following insulin injection, a catheter was placed in the femoral artery and blood samples were collected at different time intervals and were then processed for measurement of levels of uridine, glucose and insulin. Finally, the association of cellular glucose uptake in the decreased serum uridine concentrations was investigated using hyperinsulinemic-euglycemic clamp method. Our findings showed that serum uridine concentrations were decreased at the time of insulin increase in fully-fed animals, subcutaneous insulin administration decreased serum uridine in a dose- and time-dependent manner and that such changes in uridine concentrations were not associated with cellular uptake of glucose. Data obtained in our study suggest, for the first time in the literature, that insulin stimulates cellular uptake of uridine and this process is independent from that of glucose. (thesis): Palmar-plantar erythrodysesthesia (PPE) frequently accompanies the therapy with a continuous 5-FU infusion or peroral capecitabine (5-FU prodrug). In the most severe cases this adverse effect leads to discontinuation of a needful therapy. Local 10 % uridine ointment is used to prevent and treat the said adverse event. Nevertheless, this method is not generally accepted as an effective one because it has never been proved in a randomized controlled clinical trial. Most probably, a direct effect of a cytostatic compound on the skin of hands and foots causes PPE. The toxicity of 5-FU is mediated primarily by its incorporation into RNA and by thymidylate synthase (TS) inhibition and subsequent DNA synthesis disruption. The importance of particular 5-FU toxicity mechanisms varies in different cell types. For choosing the best PPE local antidote it is necessary to find out which molecular mechanism applies in keratinocytes. We have chosen pyrimidine nucleosides as the most suitable compounds for the local PPE therapy because the uridine ointment is already being used in several oncology centers in the Central Europe. Neonatal hipoksik-iskemik beyin hasarı serebral palsi, epilepsi, öğrenme güçlüğü ve neonatal mortalite gibi nörolojik defisitlere sebep olur. Bu çalışmada, sağ kommon karotid arter oklüzyonu ile iskemi oluşturularak, hipoksiye maruz bırakılan sıçanlarda üridin tedavisi ile gözlemlenen nöroproteksiyonda histon asetilazların katılımı araştırıldı.Anestezi altında yedi günlük sıçanlara sağ kommon karotid arter koterizasyonunu takiben 150 dakika süre ile hipoksik karışım solutuldu. Deney hayvanları; Sham grubu (n:8); intraperitoneal (i.p.) olarak serum fizyolojik uygulanan Kontrol grubu (n:14) ve i.p. 500 mg/kg doz üridin uygulanan Üridin grubu (n:14) olmak üzere 3 gruba ayrıldılar. Serum fizyolojik ve üridinin ilk dozu iskemi-hipoksi uygulamasından hemen sonra, 2. ve 3. dozları ise yavru sıçanlar sırasıyla 8 ve 9 günlük iken uygulandılar.Bu çalışmada hipoksi-iskemi sürecinde 7 günlük yenidoğan sıçanlarda üridin uygulamasının histon deasetilaz aktivitesini azaltarak nöroprotektif etki gösterdiği gözlemlenmiştir.Anahtar Kelimeler: Neonatal hipoksi-iskemi, histon deasetilaz, üridin. In this study, in rats exposed to hypoxia forming ischemia via the right common carotid artery occlusion in the neuroprotection observed with uridine treatment whether the contribution of histone acetylases were investigated.Seven-day old pups under anesthesia, mixture of hypoxic was inhalated for 150 minutes following the right common carotid artery cauterization. Subjects were divided into 3 groups including; Sham group (n:8); Control group (n:14) intraperitoneally (i.p.) administered with saline and Uridine group (n:14) injected i.p. with 500 mg/kg administered uridine.The first dose of saline and uridine were performed immediately after the ischemia-hypoxia, 2. and 3. doses were respectively administered when the newborn pups were 8 and 9-day-old.In this study, it is observed that uridine administration has influence on neuroprotective effect with reducing histone deacetylase activity in process of hypoxia-ischemia in 7-day-old newborn rats.Key words: Neonatal hypoxia-ischemia, histone deacetylase, uridine. 42

Pirimidin nükleozidleri üridin ve sitidin, vücutta serbest halde bulunabildiği gibi nükleotidlerin, nükleotid şekerlerin ve nükleik asitlerin yapısına girerek çeşitli fizyolojik fonksiyonlarda da yer alırlar. Farklı çalışmalarda insan serum üridin konsantrasyonları erişkinlerde 3-6 mikromolar arasında bildirilirken, sitidin düzeyleri ise 0.1 mikromolar kadar düşük, hatta bazen tespit edilemeyecek düzeylerdedir.Çalışmamızın amacı literatürde ilk kez gebeliğin çeşitli dönemlerinde ve yenidoğanlarda serum üridin ve sitidin konsantrasyonlarını analiz etmektir.Uludağ Üniversitesi Tıp Fakültesi Kadın Hastalıkları ve Doğum Polikliniği'ne başvuran ve herhangi bir hastalığı olmayan 20-40 yaş arası 1., 2., ve 3. trimester gebeler ile aynı polikliniğe gebelik dışı bir nedenle başvuran 20-40 yaş arası kadınlar, sırasıyla gebe ve kontrol grubu olarak çalışmamıza dahil edildi. Ayrıca Uludağ Üniversitesi Tıp Fakültesi Kadın Hastalıkları ve Doğum Kliniği'nde doğum yapan gebeler (anne grubu) ve bu gebelerin normal yolla doğan term (38-42 hafta) bebekleri (yenidoğan grubu) çalışmamıza dahil edildi. Kontrol, gebe ve anne gruplarından rutin tahlil için alınan kanlardan serum örneği elde edilirken, yenidoğan serum örnekleri doğum esnasında umblikal kord kanı alınarak toplandı. Alınan örneklerden pirimidin ölçümleri Tıbbi Farmakoloji laboratuvarında, glukoz ve insülin analizleri ise Biokimya Merkez Laboratuarında yapıldı. Gönüllülerin demografik bilgileri Avicenna HBYS üzerinden dosya taraması yapılarak elde edildi.Bulgularımız kontrol grubundaki gönüllüler ile gebeliğin herhangi bir dönemindeki ve doğum yapan annelerdeki serum üridin konsantrasyonları arasında anlamlı bir fark bulunmadığını göstermiştir. Ancak yenidoğan serum üridin konsantrasyonlarının annelerininkine ve dolayısıyla sağlıklı erişkin kadın ve gebelerinkine göre iki kat yüksek olduğu bulunmuştur. Anne ve yenidoğan serum üridin konsantrasyonları arasında anlamlı bir korelasyon mevcuttur.Anahtar Sözcükler: Üridin, Gebe, Yenidoğan The pyrimidine nucleosides uridine and cytidine could either present freely or play roles in several physiological functions by incorporating into nucleotides, nucleotide sugars, and nucleic acids. Various studies reported human serum uridine concentrations in 3-6 micromolar range, while cytidine levels are generally lower than 0.1 micromolar, and below detection limits in most conditions.The aim of the present study was to analyze serum uridine and cytidine concentrations during pregnancy and in newborns, for the first time in the literature.Pregnant subjects at the age of 20-40 in their 1st, 2nd and 3rd trimesters as well as age-matched women without pregnancy who admitted to Uludag University Medical School Department of Obstetrics and Gynecology were included in the study as pregnant and control groups, respectively. In addition, mothers who gave birth in the maternity ward of the same department and their newborns born at term (38-42 weeks) via normal vaginal route were included in the study as mother and newborn groups, respectively. Serum samples were extracted from control, pregnant and mother blood they donated for routine analyses and from newborns by obtaining blood from the umbilical cord. Pyrimidine analyses were performed in Pharmacology Department while glucose and insulin analyses were performed in Biochemistry Core Laboratory. Demographic data of the subjects were derived from Avicenna HBYS system.Our data show that serum uridine concentrations are comparable between subjects in control, pregnant and mother groups. However, uridine concentrations in newborns were twice as much as those in mothers, as well as control and pregnant subjects. U okviru istraživanja protutumorskih spojeva iz skupine „malih molekula” s ciljem dobivanja boljih bioloških učinaka u tretiranju tumorskih stanica sintetizirani su 1,4-disupstituirani 1,2,3-triazolni derivati; gvanozina i aciklovira (5, 10–12 i 14), uracila (19, 20 i 25–33), N-1-sulfoniluracila (34–41) i uridina (46–55). Pronađena je nova metoda sinteze nestabilnog 8-hidrazingvanozina 3 u mikrovalnom reaktoru (MR) koji je zatim uz NaN3 preveden u 8-azidgvanozin 4 i na njemu su ispitani uvjeti za Cu(I) kataliziranu 1,3-dipolarnu cikloadiciju (CuAAC) s terminalnim alkinskim alkoholima. Metoda je primijenjena na sintezu 1,2,3-triazolnih derivata aciklovira (10–12 i 14). U svrhu priprave triazolnih C5 modificiranih pirimidinskih derivata, Sonogashira reakcijom sintetizirani su 5-etiniluracil 18 i 5-etiniluridin 45 (MR), azidi su generirani in situ iz alkil-halogenida i NaN3 ili je upotrijebljen 5-azido šećer 24, te su CuAAC reakcijama u odličnim iskorištenjima dobiveni derivati uracila (19, 25–28, 30–33), a u mikrovalnom reaktoru derivati uridina (46–48, 50–53, 55). Kondenzacijom 1,4-disupstituiranih C5-triazolnih derivata uracila (19, 28–31) uz DBU i sulfonil kloride pripravljeni su odgovarajući N-1-sulfoniluracilni derivati (34, 37–39 i 41) u dobrim iskorištenjima, dok kondenzacijom N1′ nesupstituiranog triazolnog derivata 20 nastaju 35 i 36 u tragovima. Antiproliferativno djelovanje 1,4-disupstituiranih 1,2,3-triazolnih derivata ispitano je na osam staničnih linija (MDCK, HeLa, CaCo-2, NCI-H358, K562, Jurkat, HuT 78 i Raji) te je detaljno analizirana korelacija strukture i biološke aktivnosti. As a part of investigation aimed to identify new small molecules with improved biological properties in the in vitro screening for antitumor activity the 1,4-disubstituted 1,2,3-triazole derivatives of guanosine, acyclovir (5, 10–12, and 14), uracil (19, 20, and 25–33), N-1-sulfonyl uracil (34–41) and uridine (46–55) were prepared. New synthetic method for preparation of unstable 8-hydrazine guanosine 3 by using microwave reactor (MR) was developed. Product 3 was converted into 8-azido guanosine 4 in the reaction with NaN3. The latter compound was used in numerous experiments directed to find optimal reaction conditions for Cu(I) catalyzed 1,3-dipolar cycloaddition reaction (CuAAC) with terminal alkynols. By the latter method 1,2,3-triazole acyclovir derivatives (10–12, and 14) were prepared. In order to prepare C5-triazolyl pyrimidine derivatives starting 5-etynyl uracil 18 and 5-etynyl uridine 45 (MR) derivatives were synthesized using Sonogashira coupling; the azides were generated in situ from alkyl halogenides and NaN3 or 5-azido sugar 24 was used. By subsequent CuAAC reaction the C5-triazolyl uracil (19, 25–28, 30–33) and uridine (46–48, 50–53, 55) (MR) derivatives were prepared in excellent yields. Condensation reaction of 1,4-disubstituted C5 triazole uracil derivatives (19, 28–31) with DBU and sulfonyl chlorides gave the respective N-1-sulfonyl uracil derivatives (34, 37–39, and 41) in very good yields, while condensation reaction with N1' unsubstituted triazole derivative 20 resulted with only trace amounts of 35 and 36. All 1,4-disubstituted 1,2,3-triazole derivatives were tested for the antiproliferative activity on the panel of eight cell lines (MDCK, HeLa, CaCo-2, NCI-H358, K562, Jurkat, HuT 78, and Raji), and detailed analysis of structure/activity relationship (SAR) was presented.

Väitekirja elektrooniline versioon ei sisalda publikatsioone., Iga raku elutegevuse üks olulisemaid protsesse on valgusüntees, mille käigus mRNA nukleotiidse järjestuse järgi sünteesitakse vastavalt geneetilisele koodile valgud. Valgusünteesi eest vastutab valkudest ja RNA-st (rRNA) koosnev kompleks, ribosoom, mis koosneb kahest ebavõrdse suurusega osast (suurest ja väiksest subühikust). Väiksem subühik vastutab geneetilise koodi järgimise ja suurem valkude valmimise eest. Ribosoom koosneb RNA-st (rRNA) ja valkudest (r-valk). rRNA on ribosoomi funktsioneerimise jaoks kõige olulisem komponent. Lisaks konserveerunud struktuurile sisaldab rRNA ka mitmeid mittestandardseid nukleotiide, mille paiknemine on olnud teada juba mõnda aega, kuid nende rolli kohta ribosoomides on endiselt väga vähe andmeid. Pseudouridiinid on enim levinud RNA modifikatsioonid ja nende tekkimise eest vastutavad valgud – pseudouridiini süntaasid. RluD on pseuouridiini süntaas, mis koosneb kahest osast: S4-sarnasest ja katalüütilisest osast. RluD põhjustab RNA-s pseudouridiinide tekkimise kolme kohta, millest kõik on väga kõrgelt konserveerunud ja asuvad suure subühiku rRNA heeliksis 69 (H69). Vaatamata tähtsusele, ei ole senini teada, kuidas tunneb RluD ära just need uridiinid, mida ta muudab. Käesolevas töös kasutasime geneetilisi meetodeid, et tekitada mutatsioone rRNA ja RluD geenide kindlatesse kohtadesse, tuvastamaks kuidas need mutatsioonid muudavad RluD poolt sünteesitavate pseudouridiinide tekkimist. Uurimise käigus leidsime, et RluD on spetsiifiline vaid positsioonide 1911, 1915 ja 1917 suhtes kui rRNA on ribosoomide koosseisus, kuid kaotab olulisel määral oma spetsiifilisusest, kui rRNA ei ole r-valkudega seotud. RluD aktiivsuse jaoks on oluline kontakteerumine nukleotiidiga positsioonis 1916, mis on ainuke leitud spetsiifilisuse määraja rRNA-s. Saadud andmed annavad ka alust väita, et RluD seondumine rRNA-ga on kaheetapiline. Antud töös saadud tulemused aitavad paremini mõista protsesse, mis on vajalikud funtsionaalsete ribosoomide tekkimiseks rakkudes., One of the most important processes in every cell lifecycle is protein synthesis performed by ribosomes. Ribosomes are composed of two unequal subunits (large and small subunit). Small subunit is responsible for following the genetic code and large subunit is responsible for synthesis of proteins. Ribosome consists of ribosomal RNA (rRNA) and r-proteins. Structure of rRNA is very conserved and it carries the catalytic role in protein synthesis. rRNA contains many modified nucleotides which are also highly conserved. Although modified nucleotides are known for a long time, there is little information about their function in rRNA. The most common modifications in rRNA are pseudouridines which are synthesized by proteins called pseudouridine synthases. E.coli RluD is pseudouridine synthase that synthesizes three highly conserved pseudouridines located in a very important structural element of the ribosome, helix 69 (H69). Little is known about how RluD recognizes its substrate nucleotides, or why these modifications are needed. We used genetic manipulations to introduce site specific point mutations into rRNA and RluD genes in order to determine the effect of the mutations on the occurrence of RluD directed pseudouridines. Our results show that RluD is specific to positions 1911, 1915, and 1917 in fully assembled ribosomes. However, RluD exhibits loosened specificity on protein free rRNA. Our results suggest that RluD exhibits a two-step binding to its substrate. The S4-like domain of RluD is responsible for initial binding that guides the catalytic domain to the substrate. We found that the catalytic domain of the RluD probably contacts nucleotide at position 1916 of the 23S rRNA to achieve its specificity. We also found that all RluD directed pseudouridines occur concurrently and independent of each other to the rRNA. Our results help to better understand the mechanism of ribosomal biogenesis in cells.

Bu çalışmada Parkinson hastalık tedavisinde kullanılan Levodopa ve Karbidopa ilaçlarının genotoksik, bir pirimidin nükleozidi olan Üridin'in antigenotoksik etkileri belirlenmeye çalışıldı. Bu amaçla in vivo fare kemik iliği Mikronükleus ve Komet Testleri gerçekleştirildi.Çalışmada Levodopa ilacı 10, 50 ve 100 mg/kg dozlarda intraperitoneal (i.p.) yolla farelere enjekte edildi. Bu ilaç ile 24 saat muamele sonunda gerçekleştirilen fare kemik iliği Mikronükleus Testinde her üç doz grubunda meydana gelen mikronükleuslu polikromatik eritrositlerin (MNPCE) oranının kontrol grubuna göre anlamlı olarak arttığı gözlendi. Komet Testi sonuçlarına göre, Levodopa uygulama gruplarında komet kuyruk uzunluğu, kuyruk % DNA, genetik hasar indeksi (GHİ) ve hasarlı hücre yüzdesinde doza bağlı olarak bir artış olduğu aynı zamanda kafa % DNA'sında istatiksel olarak bir azalmanın meydana geldiği belirlendi.Çalışmada aynı yolla uygulanan Karbidopa ilacının 1, 5 ve 10 mg/kg dozlarının genotoksik etkileri gözlenmedi.Levodopa ve Karbidopa ilaçları birlikte 10/1, 50/5 ve 100/10 mg/kg dozlarda farelere i.p. olarak uygulandı. İn vivo fare kemik iliği Mikronükleus ve Komet Testi verilerine göre 50/5 ve 100/10 mg/kg doz gruplarındaki PCE oranındaki artışın istatistiksel olarak anlamlı olduğu belirlendi (p < 0,0001). Komet Testi verilerine göre, kombine ilaç uygulamalarının Komet kuyruk uzunlukları negatif kontrol grubu ile karşılaştırıldığında 50/5 ve 100/10 mg/kg'lık doz gruplarında istatistiksel olarak anlamlı bir artış bulundu. Kuyruk % DNA oranlarında her üç doz grubunda meydana gelen artış, kafa % DNA verilerinde doz artışına bağlı olarak meydana gelen azalış ve genetik hasar indeks değerlerinde her üç doz grubunda meydana gelen artış negatif kontrolle kıyaslandığında istatistiksel olarak anlamlı olduğu belirlendi. Hasarlı hücre yüzde oranları kontrol grubu ile karşılaştırıldığında ise, 50 ve 100 mg/kg doz gruplarındaki artışın istatistiksel olarak anlamlı olduğu sonuçlarına ulaşıldı.Yapılan in vivo Mikronükleus ve Komet Testi verileri değerlendirildiğinde, kullanılan 100 mg/kg Üridin dozunun genotoksik etki göstermediği gözlendi. Üridin'in Levodopa'nın 10, 50 ve 100 mg/kg dozları ile kombine bir şekilde uygulanması sonucunda, Levodopa dozlarının meydana getirdiği MNPCE oranlarını sırasıyla 1,8, 2,15 ve 2,85 kat azalttığı belirlendi. Komet kuyruk uzunluğu oranlarını sırasıyla 2,09, 2,69 ve 4,94 kat azalttığı ve GHİ değerlerini ise, sırasıyla 2,59, 6,64 ve 6,18 kat azalttığı belirlendi.Üridin'in 100 mg/kg dozunun Levodopa/Karbidopa kombinasyonunun 10/1, 50/5 ve 100/10 mg/kg doz grupları ile birlikte uygulaması sonucunda gerçekleştirilen fare kemik iliği Mikronükleus ve Komet Testi verilerine göre, uygulanan Üridin dozunun Levodopa/Karbidopa kombine dozlarının meydana getirdiği MNPCE oranlarını sırasıyla 1,03, 2,37 ve 3,83 kat, komet kuyruk uzunluğu oranlarını sırasıyla 2,35, 3,43 ve 4,29 kat, kuyruk % DNA oranlarını sırasıyla 5,73, 15,74 ve 17,13 kat GHİ oranlarını ise, sırasıyla 3,23, 5,73 ve 5,93 kat azalttığı ve hesaplanan hasarlı hücre yüzdesi oranlarını düşürdüğü sonucuna ulaşıldı.Sonuç olarak, çalışmada genotoksik etkileri belirlenmeye çalışılan Levodopa ve Karbidopa ilaçlarının in vivo fare kemik iliği Mikronükleus ve Komet Testlerinde, Karbidopa uygulamasının genotoksik etki göstermediği sonucuna ulaşıldı. Levodopanın tek başına ve Karbidopa ile birlikte uygulanması sonucunda mikronükleus frekansının istatistiksel anlamlı bir şekilde arttığı belirlendi (p < 0,0001). Çalışmada kullanılan Üridin'in genotoksik etkisinin bulunmadığı, bunun yanı sıra Levodopa ve Levodopa/Karbidopa kombinasyonu ile birlikte kullanıldığında Levodopa ve Levodopa/Karbidopa kombinasyonunun meydana getirdiği genotoksik hasarları geri çevirdiği sonucuna ulaşıldı. In this study, genotoxic effects of Levodopa and Carbidopa, drugs that are used in treatment Parkinson's disease, and antigenotoxic effects of uridine, a pyrimidine nucleoside, are investigated. In vivo mouse bone marrow micronucleus test and Comet assay are used for this purpose.Levodopa was administered to the mice by intraperitoneal injection at 10, 50 and 100 mg/kg doses. As a result of the mouse bone marrow micronucleus assay after 24 hours of treatment, the ratio of polychromatic erythrocyte with micronucleus increased significantly in all of the three dose groups compared to the control group. According to Comet assay results, an increase in Comet tail length, tail DNA percentage, DNA damage index and percentage of damaged cells in a dose dependent manner and a statistical decrease in head DNA percentage were determined in Levodopa treatment groups.Carbidopa administered to the mice by intraperitoneal injection at 1, 5 and 10 mg/kg doses had no genotoxic effects.Levodopa and Carbidopa were administered in combination to the mice by intraperitoneal injection at 10/1, 50/5 and 100/10 mg/kg doses. As a result of in vivo mouse bone marrow micronucleus assay and Comet assay, increase in the ratio of polychromatic erythrocyte with micronucleus in 50/5 and 100/10 mg/kg dose groups was statistically significant compared to the control group (p < 0,0001). According to Comet assay results, there was a statistically significant increase in Comet tail lengths in 50/5 and 100/10 mg/kg dose groups in combination treatment compared to negative control group. Increase in tail DNA percentage in all three dose groups, decrease in head DNA percentage with increasing doses and increase in DNA damage index values in all three dose groups were statistically significant compared to negative control. It was also found that increase in the percentage of damaged cells compared to control group in 50/5 and 100/10 mg/kg dose groups was statistically significant.According to Comet assay and in vivo micronucleus assay, Uridine treatment at 100 mg/kg dose had no genotoxic effect. Combination of Uridine with 10, 50 and 100 mg/kg doses of Levodopa decreased the ratio of polychromatic erythrocyte with micronucleus 1,8-, 2,15- and 2,85-fold respectively. The same combination decreased Comet tail lengths 2,09-, 2,69- and 4,94-fold respectively and decreased genetic damage index values 2,59-, 6,64- and 6,18-fold respectively.According to Comet assay and in vivo mouse bone marrow micronucleus assay performed with Uridine at 100 mg/kg dose and Levodopa/Carbidopa combination at 10/1, 50/5 and 100/10 mg/kg doses, administered Uridine dose decreased the ratio of polychromatic erythrocyte with micronucleus 1,03-, 2,37- and 3,83 -fold, the ratio of Comet tail lengths 2,35-, 3,43- and 4,29-fold, the ratio of tail DNA percentage 5,73-, 15,74- and 17,13-fold, the ratio of genetic damage index 3,23-, 5,73- and 5,93-fold respectively and the ratio of percentage of damaged cells, which would in turn arise from Levodopa/Carbidopa combination doses.In conclusion, genotoxic effects of the drugs, Levodopa and Carbidopa are studied by using in vivo mouse bone marrow micronucleus test and Comet assay and it was found that Carbidopa treatment alone has no genotoxic effect. A statistically significant increase in micronucleus frequency was determined as a result of the treatment with Levodopa alone and its combination with Carbidopa (p < 0,0001). It was concluded that Uridine had no genotoxic effect; furthermore it reverts the genotoxic damage caused by Levodopa/Carbidopa combination when it is used in combination with these agents.

HINTERGRUND: Die Leber nimmt bei der Regulierung und Aufrechterhaltung des Uridinplasmaspiegels eine zentrale Rolle ein. Die Synthese von Uridin hängt dabei wesentlich von der intakten Funktion hepatozytärer Mitochondrien und des mitochondrialen Enzymes Dihydroorotatdehydrogenase (DHODH) ab. Bei Patienten unter HIV-Therapie zeigten sich in Studien verminderte Uridinplasmaspiegel, wobei die dafür verantwortlich gemachte therapieassoziierte mitochondriale Toxizität durch Uridinsupplementation reduziert werden konnte. Schädigungen von Leber und Mitochondrien im Rahmen chronischer Lebererkrankungen wie Hepatitis C (CHC), B (CHB), alkoholischer und nichtalkoholischer Fettleber (AFLD und NAFLD) könnten ebenfalls zu einem Abfall des Uridinplasmaspiegels führen. METHODIK: Nach Etablierung einer HPLC-Methodik zur Evaluation von Uridin im menschlichen Serum wurden Uridinplasmaspiegel von Patienten mit oben genannten Erkrankungen mit denjenigen einer gesunden Kontrollgruppe verglichen. Ferner wurden die Spiegel mit Demografie, Genotypen, Viruslasten, antiviraler Therapie und Therapiedauer sowie mit sonographischen wie histologischen Leberbefunden und Laborparametern korreliert. ERGEBNISSE: Sämtliche 130 Patienten zeigten einen signifikant niedrigeren Uridinplasmaspiegel als die aus 14 Probanden bestehende gesunde Kontrollgruppe, lagen jedoch noch im physiologischen Normbereich gesunder Erwachsener. In absteigender Reihenfolge zeigten die grössten Unterschiede die Gruppen mit chronischer Hepatits C (n = 69), Hepatitis B (n = 37) sowie AFLD/AFLD (n = 24). Demographische Faktoren, chronischer Alkoholkonsum, histologischer Grad der Leberentzündung und -fibrose, sowie Diabetes mellitus zeigten keinen Einfluss. Spezifika der Virushepatitiden zeigten, abgesehen von der Viruslast bei CHC mit Tendenz zu eher niedrigen Uridinplasmasppiegeln bei hohen Lasten ebenfalls keine Signifikanzen. Eine Leberverfettung zeigte hinsichtlich des Uridinplasmaspiegels lediglich im Vergleich der Gruppe AFLD/NAFLD zur Kontrollgruppe signifikant reduzierte Werte. In Bezug auf das absolute wie relative Vorliegen einer Leberzirrhose zeigten sich ebenfalls signifikant erniedrigte Spiegel, und auch bei Korrelation des Child-Pugh-Index sowie laborchemischen Lebersynthesemarkern zeigten sich mit Zunahme der Leberschädigung reduzierte Uridinspiegel. SCHLUSSFOLGERUNG: Der Uridinplasmaspiegel scheint bei chronischen Lebererkrankungen wie Hepatitis C und B sowie alkoholischer und nichtalkoholischer Fettleber relativ erniedrigt zu sein, ebenso bei der Leberzirrhose. Um den Einfluss einer Mitochondrienschädigung genauer einzuordnen wären zukünftige Untersuchungen unter Einbezug oxidativer Marker ebenso interessant wie Überlegungen, Uridin bei fortgeschrittenen chronischen Lebererkrankungen zu substituieren., BACKGROUND: The liver plays a pivotal role in regulating and maintaining uridine levels in plasma. Synthesis of uridine is mainly depending on the intact function of mitochondria and the mitochondrial enzyme dihydroorotate dehydrogenase in liver cells. HIV therapy associated mitochondrial toxicity, affecting reduced uridine plasma levels could be diminished by uridine supplementation. Moreover, in cases of liver or mitochondrial damage induced by chronic liver diseases such as hepatitis C (CHC) and B (CHB), alcoholic and non-alcoholic induced fatty liver (AFLD and NAFLD) can result in reduced uridine levels in plasma. METHODS: An HPLC-based method for evaluating uridine levels in human serum was established and utilized to compare uridine levels in patients suffering from the aforementioned diseases with controls. Moreover, uridine levels were evaluated with regard to (a) demographic data, (b) genotypes, (c) viral load, (d) antiviral treatment, (e) treatment duration, (f) sonographic and histological findings, and (g) laboratory values. RESULTS: Uridine levels in plasma of all 130 patients were significantly reduced compared to those of the 14 healthy individuals, however, were still within the physiological range observed in healthy adults. Maximum deviations decreased in the following order: Hepatitis C (n=69), hepatitis B (n=37) and AFLD/NAFLD (n=24). Demographic factors, chronic alcoholic diseases, histological grade of liver inflammation and fibrosis, as well as diabetes mellitus; had no impact on uridine plasma levels. Apart from CHC, which tends to reduce levels of uridine in plasma with a high viral load; specifics of the infectious hepatitis indicated no significant change. In cases of fatty liver, only when comparing the AFLD/NAFLD group to controls, significantly reduced levels of uridine in plasma could be confirmed. Additionally, in the case of existent relative or absolute cirrhosis, as well as with an increasing Child-Pugh-Index and decreasing liver synthesis products, significant reduced uridine plasma levels could be detected. CONCLUSION: The present data indicates reduced levels of uridine in plasma in cases of chronic liver diseases (such as hepatitis C and B), alcoholic and non-alcoholic steatohepatitis, as well as cirrhosis. To assess the impact of mitochondrial damage, future investigations should include oxidative markers and evaluate the potential of substituting uridine in cases of advanced chronic liver diseases.

The effects of extracellular adenosine on Drosophila melanogaster cell lines Cl.8+, Mbn-2, S2 were examined. The cells were grown in the complete medium and tested for growth rate with in presence or absence of adenosine, dipyridamole, uridine and deoxycoformycin. The S2 cells were tested for cAMP production in presence or absence of adenosine.

HINTERGRUND: Langzeitnebenwirkungen der antiretroviralen Therapie werden u.a. auf mitochondriale Toxizität von NRTIs zurückgeführt, wobei es zu einer Dysfunktion der Atmungskette und nachgeschalteter Stoffwechselwege wie der de-novo-Pyrimidinsynthese kommen kann. Experimentell konnte gezeigt werden, dass das Pyrimidinnukleosid Uridin einige dieser Toxizitätseffekte reduzieren konnte. METHODIK: Nach Etablierung einer HPLC-Methodik zur Evaluation von Uridin im menschlichen Serum wurden Uridinspiegel von HIV-positiven Patienten mit Uridinspiegeln einer HIV-negativen Kontrollgruppe verglichen sowie mit Demografie, Nebenwirkungen bzw. Koerkrankungen, antiretroviraler Therapie und Therapiedauer korreliert. ERGEBNISSE: Die 182 HIV-Patienten wiesen signifikant niedrigere Uridinspiegel als die 30 Kontrollpersonen auf, blieben aber noch im physiologischen Normbereich gesunder Erwachsener. Demografische Faktoren sowie Marker der HIV-Infektion zeigten keine signifikante Korrelation mit Uridin. HIV-Patienten mit Begleiterkrankungen boten im Vergleich zu HIV-Patienten ohne Begleiterkrankungen signifikant verminderte Uridinspiegel. Bei Differenzierung nach den einzelnen Befunden zeigte sich, dass nur Patienten mit Diabetes mellitus Typ 2 und chronischen Lebererkrankungen wie Steatosis Hepatis signifikant erniedrigte Spiegel aufwiesen. Im Vergleich von 4 HIV-Patientengruppen mit unterschiedlicher antiretroviraler Therapie verhielten sich die Uridinspiegel analog zur potenziellen mitochondrialen Toxizität des jeweiligen Therapieregimes. Die Patientengruppe mit dideoxy-NRTI-haltiger Therapie wies die niedrigsten Uridinplasmaspiegel auf und zeigte im Vergleich zur Patientenguppe ohne antiretrovirale Therapie signifikant erniedrigte Uridinplasmaspiegel. Mit zunehmender Therapiedauer zeigte sich bei der Therapiegruppe mit dideoxy-haltiger HAART abfallende, bei den anderen 3 Therapiegruppen konstante bis leicht ansteigende Spiegel. SCHLUSSFOLGERUNG: Bei HIV-positiven Patienten scheint die Einnahme von (di)deoxy-NRTIs sowie Koerkrankungen wie Fettleber und Diabetes mellitus Typ 2 mit signifikant niedrigeren Uridinspiegeln assoziiert zu sein. Ob eine Uridinsubstitution zur Vermeidung von NRTI-assoziierten Langzeitnebenwirkungen sinnvoll ist, müssen weitere Untersuchungen zeigen., BACKGROUND: Long-term side effects of antiretroviral therapy are mainly attributed to mitochondrial toxicity of nucleoside analogue reverse transcriptase inhibitors (NRTI) by causing respiratory chain dysfunction leading to inhibition of downstream metabolic pathways such as a de novo-pyrimidine synthesis. In vitro and in vivo studies indicated that uridine, a pyrimidine precursor, may prevent and reduce long term toxicity of NRTI treatment. METHODS: Uridine plasma levels of HIV-positive patients and HIV-negative controls were assessed by using high-performance liquid chromatography, compared to each other and correlated with demography, markers of HIV, comorbidities respectively long term side effects, antiretroviral therapy and duration of therapy. RESULTS: Uridine plasma levels of 182 patients were significantly decreased compared to HIV-negative controls, although still remaining in lower physiologic range of healthy adults. Demographic factors and markers of HIV-infection both evidenced no correlation with uridine plasma levels. HIV-patients with comorbidities showed significantly decreased uridine plasma levels compared to HIV-positives without comorbidities. This effect was mainly due to chronic liver disease such as fatty liver disease and diabetes mellitus type two. When comparing HIV-patients, divided into four different antiretroviral therapy containing groups, uridine plasma levels correlated with the potential mitochondrial toxicity of each treatment regimen. Furthermore, HIV-positive patients with dideoxy-NRTI containing therapy had statistically significant lower uridine plasma levels compared to HIV-positives without antiretroviral therapy. Regarding the observation period, patients with dideoxy-NRTI containing therapy showed decreasing uridine plasma levels whereas patients without such therapy had constant or slowly increasing levels. CONCLUSION: Dideoxy-NRTI containing HAART or comorbidities such as fatty liver disease and diabetes mellitus may be associated with statistically significant decreased uridine plasma levels in HIV-positive patients. Further investigation is necessary to evaluate whether supplementation of uridine may be beneficial to prevent or reduce NRTI side affects.
