Türkiye'de meyan bitkisinin Glycyrrhiza x iconica (endemik), G. asymmetrica (endemik), G. flavescens ssp. antalyensis (endemik), G. flavescens ssp. flavescens, G. echinata ssp. macedonica, G. echinata ssp. echinata, G. glabra olmak üzere 5 türü ve bu türlerden ikisine ait 2 varyete bulunmaktadır. Bu kapsamda; Türkiye'de yetişen meyan bitkisine ait 5 türün glisirizin, gliseritinik asit, karbenoksolon, likuiritin içeriklerinin belirlenmesi için en uygun ekstraksiyon yönteminin belirlenmesi amacıyla, ilk olarak G. glabra türüne ait meyan köklerinden bu 4 biyoaktif maddenin ekstraksiyonu için mikrodalga destekli ekstraksiyon, ultrasonik işlemci ile ultrason destekli ekstraksiyon, su banyosu ile ultrason destekli ekstraksiyon ve klasik ekstraksiyon yöntemlerinin ekstraksiyon koşulları Cevap Yüzey Yöntemi - Merkezi Kompozit Dizaynı kuadratik model ile optimize edilmiştir. Koşulları optimize edilen bu ekstraksiyon yöntemleri en yüksek verimde 4 biyoaktif madde içeriğini veren yöntemin belirlenmesi amacıyla tek yönlü ANOVA testine tabi tutulmuştur. Mikrodalga destekli ekstraksiyon yöntemi en etkin biyoaktif madde ekstraksiyonunu sağlayan yöntem olarak belirlenmiştir. Bu yöntemle, G. glabra türüne ait meyan kökünde glisirizin, glisiritinik asit, likuiritin ve karbenoksolon değerleri sırasıyla; 76,2 ± 0,2 mg g-1; 1156,2 ± 0,8 µg g-1; 8,3 ± 0,1 mg g-1 ve 2,6 ± 0,1 µg g-1 olarak tespit edilmiştir. Mikrodalga destekli ekstraksiyon yöntemiyle Türkiye'de bulunan meyan türlerinin kök, yaprak ve sap kısımları ekstrakte edilmiş, bu 4 biyoaktif madde içeriği en yüksek meyan türü, Gaziantep ve Kahraman Maraş illerinde yetişen G. glabra olarak belirlenmiştir. Gaziantep'te yetişen meyan köküne ait glisirizin, glisiritinik asit, karbenoksolon ve likuiritin değerleri sırasıyla; 14,7 ± 0,1 mg g-1; 0,08 ± 0,01 µg g-1; 0,012 ± 0,001 µg g-1 ve 9,74 ± 0,05 mg g-1 olarak tespit edilmiştir. Kahraman Maraş'ta yetişen meyan köküne ait glisirizin, glisiritinik asit, karbenoksolon ve likuiritin değerleri sırasıyla; 12,0 ± 0,1 mg g-1; 0,13 ± 0,01 µg g-1; 0,015 ± 0,001 µg g-1 ve 4,92 ± 0,01 mg g-1 olarak tespit edilmiştir. G. glabra türüne ait meyan köküyle elde edilen sulu ekstrakttan toz formda fonksiyonel içecek elde etmek için Cevap Yüzey Yöntemi- Mekrezi Kompozit Dizaynı kuadratik model kullanılarak püskürtmeli kurutma yönteminin kurutma koşulları optimize edilmiştir. Optimize edilen kurutma koşulları ile elde edilen toz içecekte 300 günlük süre boyunca raf ömrü çalışmaları gerçekleştirilmiştir. Raf ömrü çalışmaları kapsamında; L* değeri; 85,99- 89,05 aralığında, a* değeri 3,10-1,81 aralığında; b* değeri 34,30-37,39 aralığında; C* değeri 34,34-37,44 aralığında; h değeri 85,87-87,57 aralığında değişiklik göstermiştir. Su aktivitesi değeri 0,240- 0,440 aralığında tespit edilmiştir. HS GC-MS yöntemiyle analiz edilen fonksiyonel toz içeceğin etil alkol miktarı, 0,11-0,07 mg g-1 aralığında; toplam fenolik madde miktarı ise 79,21- 71,32 mg GA g-1 aralığında bulunmuştur. Raf ömrü süresince gerçekleştirilen duyusal değerlendirme sonuçlarına göre; renk, koku, lezzet ve genel beğeni kriterleri bakımından elde ettiğimiz toz içecek, duyusal olarak kabul edilebilir durumunu sürdürmüştür. Ayrıca bu üründe biyoyararlılık testleri gerçekleştirilerek meyandan elde edilen toz içecekte bulunan fenolik bileşiklerin ve biyoaktif maddelerin mide-bağırsak koşulları altındaki stabiliteleri ve bu bileşenlerin vücutta oluşturduğu fayda düzeyi belirlenmiştir. Bu amaçla biyoyararlılık prosedürü uygulanan toz içecek ve içmeye hazır hale getirilen toz içecekte; toplam fenolik madde analizi, DPPH yöntemi ile antioksidan kapasitesi analizi, bakır (II) indirgeyici antioksidan kapasite analizi, toplam flavonoid analizi ve LC-MS/MS ile biyoaktif maddelerin analizi gerçekleştirilmiştir. In this context, for determination of glycyrrhizin, glycyrrhetinic acid, carbenoxolone and liquiritin content of 5 species of licorice to determine the most suitable extraction method, firstly, extraction parameters of four different extraction methods namely, microwave assisted extraction, ultrasound processor extraction, ultrasound assisted extraction with water bath and classical extraction method were optimized by using Response Surface Methodology – Central Composite Design quadratic model for the roots of G. glabra. After optimization, these methods were subjected to a one-way ANOVA test to determine the method yielding the highest yield of 4 bioactive substances. Microwave assisted extraction method was determined as the most efficient bioactive material extraction method. By this method, glycyrrhizin, glycyrrhetinic acid, liquiritin and carbenoxolone values of licorice root of G. glabra species were calculated as 76,2 ± 0,2 mg g-1; 1156,2 ± 0,8 µg g-1; 8,3 ± 0,1 mg g-1 and 2,6 ± 0,1 µg g-1 respectively. In order to produce a functional beverage in powder form from aqueous extract obtained with licorice root of G. glabra, the drying conditions of the spray drying method were optimized by using response surface methodology central composite design quadratic model. Shelf life studies were carried out for 300 days in powdered beverage obtained with optimized drying conditions.After microwave assisted extraction of roots, leaves and stems of species of licorice which grow in Turkey, the highest bioactive compounds content in terms of these four bioactive compounds were determined in G. glabra species which grows in Gaziantep and Kahraman Maraş province. Glycyrrhizin, glycyrrhitinic acid, carbenoxolone and liquiritin values of licorice grown in Gaziantep were determined as; 14,7 ± 0,1 mg g-1; 0,08 ± 0,01 µg g-1; 0,012 ± 0,001 µg g-1 and 9,74 ± 0,05 mg g-1 respectively. Glycyrrhizin, glycyrrhitinic acid, carbenoxolone and liquitin values of licorice grown in Kahraman Maraş were determined as; 12,0 ± 0,1 mg g-1; 0,13 ± 0,01 µg g-1; 0,015 ± 0,001 µg g-1 and 4,92 ± 0,01 mg g-1.Within the scope of shelf life studies; L * value; a * value; b * value; C * value and h value varied between 85,99 - 89,05, 3,10 - 1,81; 34,30 - 37,39; 34,34 – 37,44; 85,87-87,57 respectively. Water activity value was determined in the range of 0,240 - 0,440. The amount of ethyl alcohol of functional powder beverage analyzed by HS-GC-MS method was in the range of 0,11 - 0,07 mg kg-1; the total amount of phenolic matter was found in the range of 79,21 – 71,32 mg GA g-1. According to the results of sensory evaluation during shelf life; color, smell, flavor and general taste criteria of powdered beverage has maintained its sensory status. In addition, bioavailability tests were performed to determine the stability of phenolic compounds and bioactive substances in the powdered beverage obtained from licorice under gastrointestinal conditions and the level of benefit of these compounds in the body. For this purpose, the bioavailability procedure applied for both powdered beverage and ready to drink form of powdered beverage and total phenolic content analysis, antioxidant capacity analysis by DPPH method, copper (II) reducing antioxidant capacity analysis, total flavonoid analysis and analysis of bioactive substances by LC-MS / MS were carried out.
