In this study the antiproliferative effects of Paclitaxel (PAC), Epirubicin (EPI) and Tamoxifen (TAM) on growth kinetics of Ehrlich Ascites Tumor (EAT) cells were examined in culture. An estrogen-receptor-positive ER (+) hyperdiploid EAT cell line growing in vitro was also analysed in the present study. IC50 doses of PAC, EPI and TAM (12 mu g/ml, 12 mu g/ml and 2 mu g/ml, respectively) were used. Cells were treated with the above doses for 0, 4, 8, 16, 24 and 32 hrs. At the end of these periods, living cell numbers were determined by collecting EAT cells in every group for growth study rate and for MTT assay. Therefore, the mitotic index was determined in the same experimental groups. The proliferation of EAT cells, inhibited by PAC, EPI and TAM concentrations was compared to control with increasing treatment time (4-32 hrs). Treatment of PAC, EPI and TAM alone for 24 hrs decreased the proliferation rate of EAT cells by 50% with respect to control. The inhibition of proliferation rate was higher in double drug treatment than that in single drug treatment with increased treatment time. In the treatment of three drugs applied for 32 hrs, this effect reached a maximum and proliferation rate decreased by 12% as compared to the (100%) control. In our studies, when the mitotic index parameter data were evaluated to determine which phase of the cell cycle was affected by PAC to cause the repression of cell reproduction, it was found that PAC exerted of its cytotoxic effect by causing cell accumulation at mitosis. The accumulation of the cells resulted in an increase in mitotic index values, which was an expected consequence of PAC treatment. It was observed that depending on the drug treatments, inhibition of proliferation rate and mitotic index in EAT cells were increased with respect to control, being with statistically significant occurrence (p < 0.01-p < 0.001). As a result, concomitant treatment combined with hormonal therapy has given improved results compared with single treatment and PAC+ EPI + TAM treatments had a maximum synergistic effect for 32 hrs (p < 0.001).

5.0ZET Asellus aquaticus (Crustacea : Isopoda) Ovaryumunun İnce Yapısı Bu çalışmada A.aquaticus (Crustacea : Isopoda) ovaryumunun genel yapısı, stereomikroskop ile, ovaryum duvarı, ovidukt, oosit ve folikül hücreleri de, faz kontrast mikroskobu, ışık mikroskobu ve elektron mikroskobu ile incelenmiştir. A. aquaticus ' un ovaryumlan bağırsağın dorsolateralinde, birbirinden bağımsız uzanan iki uzun tüpsü yapı şeklindedir. Germaryum bölgesi, ovaryumun ventralinde ince bir bant şeklindedir. Ovaryum duvarı, birkaç katlı telsi yapıdan oluşmuştur. Ovidukt ovaryumun tam ortasında değil, ovaryumun ilk üçte birlik kısmının sonunda bulunmaktadır. Ovidukt kılıfında kas hücrelerinin varlığı tespit edilmiştir. Oosit ve folikül hücreleri farklı gelişim evrelerinde incelenmiştir. Oosit gelişiminin ilk evrelerinde, oositler küçük, nukleuslan oldukça homojen dağılımlı kromatine sahip ve nukleolus belirgin ve büyüktür. Bu evrede oositler sitoplazmik yapılar bakımından fakir ve homojen görünüşlüdür. Büyük ve belirgin nukleolusun yanında ayırıcı bir özellik olarak, mitokondirilerin nukleusun hemen etrafında yığıldıkları görülmüştür. Ayrıca bu evrede oositlerin etrafinı folikül hücreleri çevrelemez. Oosit gelişimi ilerledikçe, nukleusun hacmindeki artışın yanısıra sitoplazma ve nukleus yapısmda farklılaşmalar görülür. Nukleusun etrafındaki mitokondriler yavaş yavaş sitoplazmamn her tarafına yayılır. Folikül hücreleri oositleri kuşatmaya başlar. 52Oosit gelişiminin daha ileri evrelerinde folikül hücreleri, oositi tamamen kuşatırlar. Oosit sitoplazmasında gittikçe artan irili ufaklı granüller görülür. Bu granüller oositin depolamaya başladığı vitellus maddesidir. Daha sonra oosit yüzeyinde görülen mikrovilluslar, folikül hücrelerine uzanırlar. Bu sırada mitokondriler oosit çevresine doğru yoğunlaşırlar. Oosit zarında mikropinositoz çöküntüleri görülür. Vitellogenez evresinin ilerlemesiyle, oositte vitellin zar belirginleşir ve vitellus maddesinden oluşan granüllerin sayısı artar. Folikül hücrelerinin sitoplazmasında granüllü endoplazmik retikulum ve Golgi kompleksi görülür. Vitellogenezin sonuna doğru artık olgunlaşmış oositler kendileri genişlerken, folikül hücrelerinin de yer yer inceldiği ve kaybolduğu görülür. 53 SUMMARY infrastructure of the Ovary of Asellus aquaticus (Crustacea: Isopoda) In this study, the overall structure of the ovary of A.aquaticus was studied at stereomicroscope, while ovarian wall, oviduct, oocytes and follicle cells were studied at phase microscope, light microscope and electron microscope. Separate ovaries of A.aquaticus, long tube-shaped, are located dorsolateral to the gut. Germinal zone is confined to ventral region of the ovary as a thin band. The ovarian wall is composed of a few of fibrous layers. The position of the oviduct is at the end of the first one-third part of the ovary, not centrally. The sheat of the oviduct was found to possess some muscle cells. Oocytes and follicle cells in different developmental stages were examined in the present study. In the first stages of development oocytes are small, and possess nuclei with homogenously distrubuted chromatine and a big and prominent nucleolus. The oocytes at this stage are homogeneous in appearance and poor in cytoplasmic structures. In addition to distinctive big nucleoli at this stage, mitochondria are seen to be located around the nucleus. Moreover follicle cells do not cover the oocytes at this stage. As the oocyte developes there seems a differantiation in the structure of cytoplasm and nucleus, in addition to an increase in the volume of nucleus. Mitochondria surrounding the nucleus are gradually scattered throughout the cytoplasm, and follicle cells begin to surround the oocytes. 54At the further stages of oocyte development, follicle cells fully surround the oocytes. Increased granules in varying sizes are formed in oocyte cytoplasm, thus vitellogenic material starting to be accumulated by oocytes. After that, microvilli on the oocyte surface are extended towards the follicle cells, and mitochondria accumulate at the surrounding of the oocyte. There also appear micropinocytotic invaginations on the oocyte membrane. Along with the progression of vitellogenic stages, vitellin membrane becomes conspicuous, and the number of granules comprising vitellogenic material increases. Endoplasmic reticulum and Golgi complexes are abundantly formed in the cytoplasm of the follicle cells. As mature oocytes become anlarged towards the end of vitellogenesis the folicle cells seem to become thinner and even disappeared in some places. 55 53

IV OZ Normal, Tumoral ve Rejeneratif Büyümeler Arasındaki Kinetik İlişkiler Bu çalışmada; normal büyüme, fare kemik iliğinde mitotik indeks ve S fazındaki hücre yüzdesi; rejeneratif büyüme, parsiyal hepatek- tomi yapılan karaciğerde rejenerasyon yüzdesi, DNA dağılımı ve S fazında ki hücre yüzdesi; tumoral büyüme ise, EAT hücrelerinde çoğalma hızı, DNA dağılımı ve S fazındaki hücre yüzdesi parametreleri incelenerek araş tırılmış ve hücre kinetiğinde meydana gelen değişikliklere göre bu üç tip büyümenin birbirlerini ne şekilde etkiledikleri değerlendirilmiştir. Elde edilen sonuçlara göre, normal büyümeyi, rejeneratif büyüme tek başına ya da tumoral büyüme ile birlikte stimule etmekte, tumoral büyüme ise, yalnız başına inhibisyona yol açmaktadır. Dolayısıyla, rejeneratif büyümenin tumoral büyümeye göre dominant bir etkiye sahip olduğu düşünülmektedir. Diğer taraftan tumoral büyüme rejeneratif büyüme üze rinde stimule edici bir etki, rejeneratif büyüme tumoral büyüme üzerinde inhibe edici bir etki oluşturmaktadır. So, regenerative growth is thought to have a dominant effect according to the tumoral growth. On the other hand, tumoral growth has a stimulative effect on regenerative growth and regenerative growth has an inhibitory effect on tumoral growth. 59

42 - ÖZET X ışınlarının ve iyonizasyon meydana getiren ışınlar yayan radyoaktif maddelerin keşfedilmelerinden hemen sonra, bu ışınların canlılarda bazı zararlı etkiler meydana getir dikleri tesbit edilmiş ve bundan sonra radyasyonla çalışanla rı bu etkilerden korumak üzere bazı sınırlamalar konulmaya başlanmıştır. 1953 yılında kurulan ICRP kuruluşu ile de bu sınırlamalar uluslararası bir düzeye getirilmiştir. Bu neden le radyoaktif madde veya radyasyon kaynakları ile çalışılma dan önce bu kaynakların etrafı, canlılara zararsız hale geti rilmek amacı ile zırhlanmakta ve sonra çalışılmaya başlanmak tadır. Bu çalışmada ICRP kuruluşunun sınırlamalarına bağlı kalınarak, noktasal bir gama kaynağı olan Co terapi cihazı nın konulacağı mekanın kaynaktan yayınlanacak radyasyonu za rarsız bir düzeye indirmek üzere, zırhlama hesapları yapıl- 6 f) mıştır.
