Objective: Aim of this study was to evaluate the effect of topical intranasal insulin on healing of nasal mucosa in a rat model. Methods: Forty-eight Wistar rats, weighing between 250 and 300 g and aged 10–12 weeks were used and randomized into two equal groups. 1.9 mm curette was introduced through the left nostril and 1.9 mm mucosa from the left nasal septum was curetted. Postoperatively, animals in the control group received 1 mL of physiologic saline, 3 times a day in a nasal irrigation fashion. Animals in the experimental group received 1 mL of 5 IU/mL regular insulin in saline solution. Subjects were sacrificed after 5, 10, and 15 days and macroscopic and histomorphometric evaluations were performed. Results: There were no mucosal synechiae and septal perforation macroscopically. Histological examination revealed that the defect size reduction was 21% in the saline group versus 56% in the insulin group on the fifth day (p = 0.006). There was 62% defect reduction in the saline group versus 79% in the insulin group on the 10th day (p = 0.034). On the 15th day, only 67% of saline group animals had complete defect closure, whereas 100% of animals treated with insulin had complete closure (92% vs 100% mucosal defect reduction, p = 0.036). Both edema and inflammation were less in the insulin group on 15th day (p = 0.006; p = 0.023, respectively). Conclusion: The results from this study support the safety and efficacy of topical insulin on wound healing in the literature. Fourty six percent of male patients display infertility due to germ cell damage induced by chemo-radiotherapy for treatment of childhood cancers. It is not possible to collect mature sperm from these patients since spermatogenesis is not present start until puberty. Monolayer long term culture of spermatogonial stem cells is challenging. Mesenchymal stem cells have a similar embryonic origin with the Sertoli cells that have a crucial role in regulation of spermatogenesis. Sertoli cells share gene expression profile and differentiation potential of mesenchymal stem cells. We hypothesized that mesenchymal stem cells can support the testicular niche and maintain the stem cell pool in an in vitro co-culture system by supporting Sertoli cells. The aim of this study is to protect stem cell viability of newborn C57BL/6 mice testes for 1, 2, 4 and 6 weeks by using an syngenic bone marrow derived mesenchymal stem cell supported indirect air liquid interphase co-culture system. The co-culture system has been set and maintained successfully using isolated mesenchymal stem cells of passage 3. 19. yüzyılda ortaya çıkan klasik polisiye romanında, Mandel (1996) in tartıştığı üzere aksayandüzeni onararak rasyonaliteyi yeniden sağlamaya çalışan bir dedektif karakteri bulunmaktadır.Dedektifin toplumsal konumu da tarihsel ve toplumsal değişikliklerle ilişkilidir. Nitekim klasikpolisiye romanında bilim ve akıl kavramlarının vurgulanması Aydınlanma ve pozitivizmleilişkilidir. Dönemsel olarak değerlendirildiğinde, iki dünya savaşı arasındaki dönem İngilizpolisiyesinin Altın Çağ dönemi olarak adlandırılmaktadır. Bu dönemde kadın yazarlar ve kadındedektifler ön planda olup şiddetin hakim olmadığı, huzurlu ve rahatlatıcı mekanlarda işlenenpolisiye kurgular dikkat çekicidir.Bu döneme 1920’li yıllarda Amerika’da yaygınlaşan, özellikle erkeksi dedektif karakterlerinağır bastığı hard-boiled geleneği eşlik eder. 1970’lerin sonunda, Amerika’da polisiye edebiyattakadın bakış açısı ön plana çıkmıştır. Dedektif karakterlerin toplumdaki ataerkil değerlere göretanımlanmaması, kadının kendi öznelliğinin farkında olması, toplumun farklı kesimlerindenkadınların kurguya dâhil edilmesi bu döneme ilişkin göze çarpan değişikliklerdir. Bu çalışmada,polisiye romanda modern kadın dedektif figürlerinin ortaya çıkışının kadın hareketi ve feministyaklaşımla ilişkili olduğu tartışılmaktadır. Yürümek,bir mobilite biçimi olarak sosyal teori alanında daha çok kentleşme vemodernite içerisinde tartışılmaktadır. Yürümek, Urry’nin (2000, 2007)tartıştığı üzere yönetimsellikle yakından ilgili olup kamusal alanda toplumsalcinsiyet ve sosyal sınıflara göre tarihsel olarak değişiklik göstermiştir.Nitekim bu farklılık edebiyata da yansımıştır. Makalede bu değişimin kentselmekânda nasıl gerçekleştiği ele alınmaktadır. Yürümek sosyolojik olarak dakentsel mekânı ilk çalışan teorisyenlerin kullandığı bir teknik olarak Simmel,Engels ve Benjamin’in çalışmalarında görülmektedir. Yürümek sosyal teori içerisindekenti demokratikleştirişinden; Sürrealizmde estetik ve eleştirel bir pratikolarak ele alınışına; Situasyonistlerde kapitalizme ve modernist şehirplancılığına karşı bir direniş pratiği olmasına kadar farklı şekillerde elealınmıştır. Böylelikle bu çalışmada yürümenin sadece sıradan bir hareketlilikbiçimi olmaktan öte kentsel mekânda önemli bir pratik eylemlilik ve sosyolojikolarak da kenti eleştirel olarak yeniden düşünmek ve keşfetmek için biraraştırma tekniği olduğu hatırlatılmaktadır., Walking as a form of mobility isdiscussed mostly in social theory with regard to urbanization and modernity. AsUrry (2000, 2007) argues, walking is closely related to governmentality and hasshown alterations historically in terms of gender and social classes in publicspace. Hence, this differentiation has also reflections in the literature.Therefore, this paper will evaluate how this differentiation occurs in urbanspace. Walking sociologically as a technique is used by first theorists(Simmel, Engels and Benjamin) studying urban space. Walking has been evaluatedin social theory in different urban realms: from as an element contributing tocity’s democratication to aesthetic and critical practice in Surrealism; and aresistance to capitalist and modernist urban planning of Situationists. Thus,it is reminded in this paper that walking is more than a mundane mobilityfeature but an important practical performance and research technique inexploring and reconsidering city critically.

Mevsimlik tarım işçiliği, hanehalkı olarak yapılan ve geçici bir süre göçe dayalı olan işgücüdür. Çingene topluluğu içinde mevsimlik tarım işçiliğini yoğun olarak yapılan işler arasında görmekteyiz. Çingene topluluğu da kendi içerisinde homojen değildir. Türkiye’de Çingeneler başlıca üç grup altında görülebilir: Romanlar, Domlar ve Lomlar. Türkiye’nin batısında yaşayan Romanlar ile Doğu ve Güneydoğu’da yaşayan Domların mevsimlik tarım işçiliğine yönelme nedenleri farklıdır. Bu farklılıklar, Türkiye’deki göç pratiklerinin sonucu olarak görülebilir. Romanlar 1950’lerdeki tarımsal dönüşümden etkilenmelerine karşın, Domlar 1990’lardaki zorunlu göçten etkilenmişlerdir. Bu çalışma, Edirne'deki Roman topluluğu ile Diyarbakır'daki Dom topluluğunun farklı çoğunluklar ile (sırasıyla Edirne'deki Türkler ve Diyarbakır'daki Kürtler) entegrasyon seviyeleri ve siyasi topluluğa aidiyetlerini, vatandaşlık haklarından (sivil, sosyal, siyasi ve kültürel) yararlanma seviyelerini ve ulus ötesi vatandaşlığın Roman ve Dom derneklerine etkilerini karşılaştırmayı amaçlamaktadır. Bu çalışma aynı zamanda iki topluluğun sosyal entegrasyonunu, modern vatandaşlığın eşitlik ilkesine dayanarak tam vatandaşlık haklarından ne ölçüde yararlanabildiklerini açığa çıkarmaya çalışmaktadır. Çalışmanın temel argümanı, Roman topluluğunun Dom topluluğuna göre vatandaşlık haklarına daha fazla erişebildiğini ileri sürmektedir. Bu durum, Roman topluluğunun Edirne ve Türkiye'de etnik çoğunluk olan Türklerle; öte yandan Dom topluluğunun Diyarbakır'da çoğunluk olup Türkiye'de azınlık olan Kürtlerle yaşamasıyla ilgilidir. En önemlisi, Roman topluluğunun devlet ve ulusötesi alanla Dom topluluğuna göre yakın ilişkileri bulunmaktadır.Bu çalışmada, etnisitenin iki topluluğun tam vatandaşlık haklarından yararlanmasında ortak bir engel olduğu; fakat iki topluluğun farklı tarihsel, sosyal ve ekonomik dönüşümler geçirdiği ileri sürülmektedir. Sosyal dışlanma iki toplulukta da farklı seviyelerde görülmektedir. Dolayısıyla bu çalışma, vatandaşlığın eşitlik ilkesinin iki topluluk için de neden bozulduğunu ortaya çıkarmaya çalışmaktadır. 1990'larda uygulanan zorunlu göç pratiği ve müzisyenlik mesleğinin kaybolmaya yüz tutması Dom topluluğu için emek pazarındaki sosyal dışlanmanın temel etkenlerini oluştururken, Roman topluluğu tarımsal modernizasyonun etkileri ile birlikte temel olarak kentleşme ve modernleşme ekseninde son 40?50 yıl içerisinde etnisite ve sınıf oluşumu açısından kendilerini yeniden konumlandırmışlardır. Bu çalışma, Dom topluluğunun Roman topluluğuna göre daha sınırlı vatandaşlık haklarına sahip olduklarını öne sürmektedir. Bu farklılık yoksulluğun etkileri ve çoğunluğa entegrasyon seviyeleri ile bağlantılı olarak açıkça görülebilir. This study aims to compare Roma community in Edirne and Dom community in Diyarbakır with regard to their integration levels to different majorities (respectively Turks in Edirne and Kurds in Diyarbakır) and belonging to the political body (state), access to citizenship rights (civil, social, political and cultural) and the affect of transnational citizenship on Roma and Dom communities. The main argument of this study asserts that Roma community can have more access to citizenship rights than Dom community. This is related with the fact that Roma community lives with Turks, who are the ethnic majority in Edirne and in Turkey, whereas Dom community lives mostly with Kurds, who are the majority in Diyarbakır but minority in Turkey. Foremost, Roma community has closer connections with state and transnational space than Dom community.The study has found that ethnicity appears as a common barrier for both communities in benefiting from full citizenship. However, it is noted that they experienced different historical, social and economic transformations. Social exclusion is observed at different levels for the two communities. Hence, the study tries to explain why the equality principle of citizenship is ruptured for both communities. While forced migration in 1990s and the gradual loss of musician craft were key factors for the exclusion of Dom community in the labor market, Roma community with affect of agricultural modernization, has repositioned themselves in terms of ethnicity and class formation in last 40-50 years owing mainly to urbanization and modernization. The study has found that Dom community has very limited citizenship rights compared to Roma community. The differences can be obviously seen with regard to impact of poverty and their integration levels to the majority. 344

The emphasis on the individual and rational mind appeared in the age of Enlightenment. However, today in our world there are fundamental social challenges suppressing the individual which can be observed along with the growing restlessness, the damaging of spiritual coherence of the individuals through forcing them to take risk in their working life and loss of universal values and meanings. Modernity and new forms of capitalism create such corrosive effects on the individual, while remedies are inherent in the same dynamics. Hence, these dynamics can be seen through a life coaching profession, which is an effort of neoliberalism in building up homo economicus. Life coaching as a new profession appeared in the United States and Europe within the last 10-15 years and also has been expanded in Turkey with the effect of the globalization. Intended for the training of this profession, a new market has come into existence which is supported by international life coaching certificate programs which exceed nation-state. Aynı zamanda içerisinde ikinci el kıyafet, antika değeri olan eşyalar, tablolar, ayakkabılar, el tamir aletleri ağırlıklı olmak üzere birçok ikinci el ürünü barındırmaktadır. Bu çalışma ile birlikte bir tüketim mekânı olarak bitpazarının yoksulluk, tüketim modası ile olan ilişkisi ve mekânsal yönleri ortaya çıkarılmak istenmektedir. Bunun için 2017-2018 yılları arasında, İzmir ve Eskişehir kentlerinde saha çalışması yürütülerek bitpazarına gelip ikinci el eşya alan 15 müşteri ve tezgâh açan 11 satıcı ile yarı-yapılandırılmış görüşme, yapılandırılmamış gözlem ve fotoğraf çekimi teknikleri ile veri toplanarak yürütülmüştür. Bitpazarı insanların sosyalleştiği bir alışveriş mekânı olması bakımından ekonomik ve kültürel öğelerin temsil edildiği bir alandır. Bunun için çalışma boyunca bitpazarının tüketim yönlü özellikleri ve piyasası, bitpazarında bireylerin `yoksulluk halleri` ile bitpazarının mekânsal boyutunun ekonomik ve kültürel yönlerine odaklanılmıştır. İzmir ve Eskişehir kentlerinin bitpazarlarının ekonomik ve kültürel yönlerinin incelendiği bu çalışma her iki bitpazarının sınıfsal olarak kentlerin orta sınıf altında bireylerinden satıcı ve müşterilerinin kullandığı alternatif bir alışveriş mekânı olduğunu göstermektedir. Bunun yanında bitpazarı ikinci el giyim modası, koleksiyonculuk ve antikacılığın süreklilik sağlamasında önemli bir rol oynamaktadır. The Flea Market, unlike the other district bazaars, is a commerce space where second hand commodities or in other words previously used daily necessaries are sold cheaper then brand new products by the individuals who cannot afford to buy it. This place at the same time contains a great many of second hand commodities mostly consist of second hand clothes, antique objects, paintings, books, shoes and hand repair tools. The flea markets historically shows a continuity since the beginning of marketplaces emergence to nowadays. This thesis have importance for revealing the interaction between internet and the flea market in terms of lower income or the poor groups consumption patterns who predominantly make use of the flea market and examining the consumption oriented features of the desire to move up the middle class by utilising flea market with an aim of vintage fashion or collecting purposes. This study aims to conceive the spatial dimension of flea market as a commerce space and its relation with poverty and consumption trends. For that purpose a field work with 15 buyers and 11 sellers was conducted between 2017-2018 in İzmir and Eskişehir cities by using semi-structured interview, unstructured observation and photographing techniques. Flea market, a commerce space in which people socialize,is a place where economic and cultural elements are represented. Throughout the study, the aim was to focus on the consumption based characteristics and the market economy of the flea market, the poverty states of individuals in the flea market and the economic and cultural aspects of the spatial dimension of it. This study in which the economic and cultural aspects of flea markets of İzmir and Eskişehir cities are investigated, shows that lower middle class individuals of both sellers and buyers utilizes from flea market as an alternative shopping place. Besides, flea market has a role in providing a continuity to second hand fashion, collecting and antiquities. 172

27. Cannabinoid receptor ligands modulate fibrosis and inflammation in idiopathic pulmonary fibrosis: a preliminary study.

- Abstract
Background/aim: No specific pharmacological treatment regimen for idiopathic pulmonary fibrosis (IPF) exists. Therefore, new antiinflammatory therapeutic strategies are needed. Cannabinoids (CBs), known for their inflammation-modulating and antifibrotic effects, may be potential medication candidates for treating IPF. We aim to evaluate the inflammation-modulating and antifibrotic effects of CB receptor (CBR) agonists and antagonists in lipopolysaccharide-stimulated normal human lung fibroblast, epithelial cells, IPF fibroblast cells, and monocytes., Materials and Methods: We detected CBRs in normal human lung fibroblasts (LL24) and IPF fibroblast cells (LL29), epithelial cells (A549) and monocytes (THP-1) by flow cytometry. We determined TGF-β1, IL-8, and TNF-α inflammatory cytokines in the LL24, LL29, A549, and THP-1 cell culture supernatants on days 1 and 5 by ELISA. We evaluated the cell viability in LL24, LL29, and A549 cells on days 1, 3, and 5 spectrophotometrically and detected collagen Type I (ColI) production in the LL24 and LL29 cell culture supernatants on days 1, 3, and 5 by ELISA., Results: LL24, LL29, A549, and THP-1 cells exhibited CB1 (CB1R) and CB2 (CB2R) receptors. CB1R and CB2R agonists WIN55,212-2 and JWH015 inhibited fibroblastic and epithelial cell proliferation on day 5. TGF-β1 and TNF-α release increased, while IL-8 release decreased in LL24, LL29, A549, and THP-1 cells in response to the administration of WIN55,212-2 and JWH015 at a 10
-2 mM concentration. CB1R and CB2R antagonists AM251 and AM630 did not block agonistic responses, suggesting a nonclassical CBR-mediated pathway. CB2R agonist JWH015 decreased ColI expression in IPF lung fibroblasts LL29 on day 3., Conclusion: These results suggest that CB signaling regulates the progression of pulmonary inflammation and fibrosis via CBR activation. - Abstract
Flow cytometry and immunohistochemistry techniques both determine the target protein by immunolabeling. Flow cytometric analysis quantifies total number of fluorescent labeled cells and qualify sup-populations according to size and granularity. Immunohistochemistry is able to map immune-labeled cells and extracellular matrix components under light and electron microscope by enzyme or fluorescent molecules. Real-time identification, in-time classification, and final plotting of spermatogonial lineage are of crucial importance for monitoring the fertility potential of spermatogonial stem cell microenvironment and predicting progress of spermatogenesis. - Abstract
Magnetic-activated cell sorting (MACS) is the technology that is recently used as a magnetic-based cell isolation/purification technique. This technique enables the isolation and selection of germ, hematopoietic, and somatic stem cells including skin stem cells (SkSCs). Here, we have tried to describe the isolation of stem cells by MACS using CD34 antigen for SkSCs, again CD34 for hematopoietic stem cells (HSCs) and Thy-1 for spermatogonial stem cells (SpSCs). MACS allowed the isolation of CD34+, CD34+, and Thy-1+ human SkSCs, HSCs, and SpSCs with minimum 98% purity.
