Ergenlik döneminin endokrin düzenleyicileri olan Kisspeptin, Leptin, GnRH ve LH arasındaki ilişkide çinko önemli rol oynar. Beslenmenin hormonal aktörleri olan Leptin ve NPY'nin fonksiyonlarında çinkonun düzenleyici rolüne dikkat çekilir. Öğrenme ve bellek fonksiyonlarında çinkonun hipokampal rolü vazgeçilmezdir. Bellek, ergenlik ve beslenme davranışları arasında da karşılıklı bir ilişki vardır. Bu çalışmanın amacı ergenlik, beslenmenin düzenlenmesi ve bellekte etkili olan moleküler yolaklarda çinkonun olası etkilerini araştırmaktır. Toplam 40 adet sıçan eşit sayıda 4 gruba ayrıldı: Grup 1; çinko eksik diyet, Grup 2; normal diyet, Grup 3; çinko takviyeli diyet, Grup 4; Kontrol. Çalışmada ilk 3 grubu oluşturan hayvanlar gebelik başlangıcından doğum sonrası yavrularından ayrılacakları 21. güne kadar çinko eksik diyetle beslenen annelerin yavrularından elde edildi. Kontrol grubunu oluşturan hayvanlar ise standart sıçan yemiyle beslenen annelerden doğan yavrular arasından rastgele olarak seçildi. Çalışmada bellek performansı Morris su labirenti ile değerlendirildikten 24 saat sonra hayvanlar sakrifiye edilerek kan ve hipotalamus ile hipokampus doku örnekleri alındı. Maternal çinko eksikliği oluşturulduktan sonra çinko eksik diyetle beslenen sıçanlarda ortaya çıkan bellek fonksiyonundaki bozukluklar çinko desteğiyle önlendi. Hipokampusta çinko eksikliğinde baskılanan ZnT3 ve BDNF gen ekspresyonu, standart sıçan yemiyle beslenen hayvanlarda düzelmezken, çinko desteği hipokampal ZnT3 ve BDNF gen ekspresyonundaki baskılanmayı tersine çevirdi. Çinko eksikliğinde baskılanan hipokampal NPY, Kisspeptin ve GnRH gen ekspresyon düzeyleri çinko desteğiyle normale döndü. Hipotalamus GATAD 1 gen ekspresyonundaki artış çinko eksikliğiyle şiddetlenirken, hipotalamus NPY ve GnRH gen ekspresyonunda görülen çinko eksikliğindeki baskılanma çinko desteğiyle önlendi. Hipotalamusta çinko eksikliğinde baskılanan Kisspeptin gen ekspresyonu, standart sıçan yemiyle beslenen hayvanlarda düzelmezken, çinko desteği bu değişiklikleri ortadan kaldırdı. Çinko eksikliğinde kan düzeylerinde baskılanma görülen Kisspeptin, Leptin, GnRH, LH, FSH seviyeleri çinko desteğiyle normale değerlerine ulaştı. Yine çinko eksikliğinde baskılanan serum NPY seviyeleri, maternal çinko eksikliği oluşturulduktan sonra standart sıçan yemiyle beslenen hayvanlarda düzelmezken, çinko desteği bu baskılanmayı tersine çevirdi. Mevcut çalışmada maternal çinko eksikliği oluşturulduktan sonra çinko eksik diyetle beslenen erkek sıçanlarda bozulmuş bellek performansı, ergenliği ve beslenmeyi düzenleyen hormonal parametrelerdeki bozulmanın çinko desteğiyle düzeltilebileceği gösterilmiştir., Zinc plays an important role in the relationship between Kisspeptin, Leptin, GnRH, and LH, which are endocrine regulators of the puberty term. Attention is drawn to the regulatory role of zinc in the functions of leptin and NPY, which are hormonal actors of feeding. In learning and memory, the hippocampal role of zinc is indispensable. There is also an interrelationship between memory, puberty, and feeding behavior. This study aims to investigate the possible effects of zinc on the molecular pathways that affect puberty, the regulation of feeding, and memory. A total of 40 rats were divided equally into four groups as follows: Group 1, zinc-deficient diet, Group 2: normal diet, Group 3: zinc-supplemented diet, Group 4, control. The animals constituting the first 3 groups were produced from the dams fed a zinc-deficient diet from the beginning of the pregnancy until the 21st day after birth when they were to be separated from pups. Animals in the control group were randomly selected from the pups born to dams fed the standard diet. The memory performance was evaluated by the Morris water maze. 24 h after the memory test, animals were sacrificed, and blood and tissue samples of the hypothalamus and hippocampus were collected. After the maternal zinc deficiency was established, the impairment in memory in rats fed a zinc-deficient diet was prevented by zinc supplementation. ZnT3 and BDNF gene expression suppressed by zinc deficiency in the hippocampus did not improve in animals fed the standard diet, whereas zinc supplementation reversed the suppression in hippocampal ZnT3 and BDNF gene expression. Hippocampal NPY, Kisspeptin, and GnRH gene expression levels were suppressed in zinc deficiency and reestablished with zinc supplementation. Hypothalamic GATAD1 gene expression was increased in zinc deficiency. The suppression of NPY and GnRH gene expression in the hypothalamus in zinc deficiency was prevented by zinc supplementation. Hypothalamic Kisspeptin gene expression, which was suppressed by zinc deficiency, did not improve in animals fed the standard diet, whereas zinc supplementation restored normal expression levels. Serum Kisspeptin, Leptin, GnRH, LH, and FSH levels were suppresed in zinc deficiency and recovered by zinc supplementation. Serum NPY levels suppressed by zinc deficiency did not improve in animals fed the standard diet, whereas zinc supplementation reversed this suppression.
