Bu çalışmada, artrit ve akut yaralanmalarda enflamasyonu önlemek ve ağrıyı dindirmek için kullanılandiklofenak sodyumun, koyun dalak dokusundan saflaştırılan glukoz-6-fosfat dehidrogenaz enzimi (G6PD; E.C. 1.1.1.49) ve glutatyon redüktaz enzimi (GR; E.C. 1.8.1.7) üzerine in vitro etkileri araştırıldı. Çalışmanın ilkaşamasında G6PD ve GR enzimleri koyun dalak dokusundan, amonyum sülfat (NH4)2SO4 çöktürmesi ve 2', 5' ADP-Sepharose-4B jel afinite kromatografisi ile saflaştırıldı. Enzimlerin saflık dereceleri sodyum dodesilsülfat poliakrilamid jel elektroforezi (SDS-PAGE) metodu ile kontrol edildi. Çalışmanın ikinci aşamasında saf olarak elde edilen enzim aktiviteleri üzerine diklofenak sodyumun etkileri araştırıldı. Enzimler üzerinde inhibisyon etkisi gösteren diklofenak sodyumun için % Aktivite-[I] grafiği çizilerek IC50 değerleri hesaplandı. Ayrıca beş farklı substrat konsantrasyonu ve üç farklı inhibitör konsantrasyonlarıyla ölçümler yapılarak Lineweaver-Burk grafikleri çizildi. Elde edilen bu grafiklerden diklofenak sodyumun G6PD enzimini 0.51 mM, IC50 değeri ve 0.48 ± 0.01 mM Ki değeri ile yarışmasız olarak inhibe ettiği belirlendi. GR enzimini ise 6.55 mM IC50 değeri ve 15.69 ± 0.84 mM Ki değeri ile yarışmasız olarak inhibe ettiği tespit edildi., In this study, in vitro effects of diclofenac sodium, which is used to prevent inflammation and alleviate pain in arthritis and acute injuries, on glucose-6-phosphate dehydrogenase enzyme (G6PD; EC 1.1.1.49) and glutathione reductase enzyme (GR; EC 1.8.1.7) purified from sheep spleen tissue were investigated. In the first stage of the study, G6PD and GR enzymes were purified from sheep spleen tissue by ammonium sulfate (NH4)2SO4 precipitation and 2', 5' ADP-Sepharose-4B gel affinity chromatography. The purity levels of the enzymes were checked by the sodyum dodesilsülfat poliakrilamid jel elektroforezi (SDS-PAGE) method. In the second stage of the study, the effects of diclofenac sodium on the pure enzyme activities were investigated. IC50 values were calculated by plotting the % Activity-[I] graph for diclofenac sodium, which has an inhibitory effect on enzymes. In addition, Lineweaver-Burk graphs were drawn by making measurements with five different substrate concentrations and three different inhibitor concentrations. From these graphs, it was determined that G6PD enzyme inhibited with 0.51 mM IC50 value and 0.48 ± 0.01 mM Ki value non competitively, by diclofenac sodium. It was determined that the GR enzyme inhibited with an 6.55 mM IC50 value of and 15.69 ± 0.84 mM Ki value non competitively.

Lung cancer is one of the most common malignancies and aggressive tumour, which causes the highest cancer-related death in the world. Ifosfamide is an alkylating antineoplastic agent used in the treatment of cancer including, lung cancer. Curcumin, a flavonoid from the rhizome of Curcuma longa has been shown to chemotherapeutic activity through inhibition of cell growth, cell cycle arrest, and pro-apoptotic activity in human cancer. However, the effects of Ifosfamide and Curcumin combination on non-small cell lung cancer (NSCLC) remain unclear. In this study, the influences of Ifosfamide, cell proliferation, ROS accumulation, apoptosis, migration, and its combined effects with curcumin were investigated in NSCLC cell line A549 by RT-Cell analyse, DCFDA assay, western blot and wound healing-migration assays. A549 cells were treated with the Ifosfamide-Curcumin combination and the results showed that the combination was effective in ROS accumulation, which contributed to inducing apoptosis via increasing pro-apoptotic Bax and decreasing anti-apoptotic TIGAR, and depression of cell migration. The treatment of combination significantly induced apoptosis, together with the downregulation of TIGAR and upregulation of pro-apoptotic Bax protein. Moreover, Curcumin and Ifosfamide combination suppressed cell migration into the scratched area as compared to untreated cells. Our findings suggest that Curcumin enhanced the efficacy of Ifosfamide in human non-small cell lung cancer cell line., Akciğer kanseri, dünyada kansere bağlı en yüksek ölüme neden olan kötü huylu ve saldırgan tümörlerden biridir. İfosfamid, akciğer kanserinin de dâhil olduğu birçok kanser tedavisinde kullanılan alkilleyici bir antineoplastik ajandır. Curcuma longa rizomundan elde edilen bir flavonoid olan kurkumin'in, insan kanserlerinde hücre büyümesinin inhibisyonu, hücre döngüsü dudurulması ve pro-apoptotik etki yoluyla kemoterapötik aktiviteye sahip olduğu gösterilmiştir. Bununla birlikte, İfosfamid ve Kurkumin kombinasyonunun küçük hücreli olmayan akciğer kanseri (NSCLC) üzerindeki etkileri henüz doğrudan ele alınmamıştır. Bu çalışmada akciğer kanseri hücre hattı A549'da, İfosfamid ve Kurkumin kombinasyonunun, hücre proliferasyonu, hücre içi ROS birikimi, apoptozis ve hücre göçü üzerine olan etkileri gerçek-zamanlı hücre analizi, DCFDA floresan işaretleme, western blot ve yara iyileşme-hücre göçü testleri ile araştırılmıştır. A549 hücrelerinde, İfosfamid-kurkumin kombinasyonu ile tedavi sonrasında, hücre içi ROS birikiminde ve pro-apoptotik Bax proteinin düzeyinde bir artış olduğu belirlendi. Ayrıca, kombinasyonun, tedavi edilmeyen ve tek ajan ile tedavi edilen hücrelere kıyasla, anti-apoptotik TIGAR protein düzeyinin azalmasında ve çizilen alana hücre göçünün bastırılmasında etkili olduğu belirlendi. Bulgularımız, Kurkuminin, küçük hücreli olmayan akciğer kanseri hücre hattın A549’da İfosfamidin etkinliğini artırdığını göstermektedir.

The Muslims who entered Africa in the middle of seventh century conquered Spain at the beginning of eighth century and in this context within a time span less than a century the southern shores of Mediterranean Sea encountered a new political formation. The Emirate of Al-Andalus did not adopt a naval policy in its first decades but as from IX century, marine groups independent of the state emerged in the eastern shores of the country. In this century the emirate had to strenghten its navy because of several motives and those aforementioned marine groups advanced to eastern destinations like Sicily and Crete. Those groups made jihad-oriented incursions against Christian shores or conducted piracy and sea trade. The groups that remained in Andalus constituted human source of the sea army of the emirate in the context of Andalusian emirs’ effort to found an official fleet after the Viking attacks in 844. Especially the groups called bahriyyun living on the southern shores of the country became the workers of the first dockyard in Sevilla and performed as warriors and carried out other services. At the end of IX century, the groups of bayriyyun began to settle down in the site of Pechina which was the predecessor of Almería that would be the main naval base of Caliphate of Andalus. Not all the groups were enrolled in the state service and some of them engaged in sea incursions and trade. At the end of the century, one of this group had to land in southern shores of France because of a storm and, in an unplanned way, created coastal microstate of Fraxinetum that would live about eighty years. Backed by new warriors and equipment flowing from Andalus, these warriors increased in numbers gradually in Provance which was in a political turmoil and they conducted raids spreading towards Italy and Switzerland by using mountain passes in the Alps. They both took prisoners by targeting passenger groups in the passes and they got booty by attacking cities. Monarchs of the region demanded Byzantine naval assistance to rid themselves of the warriors of Fraxinetum but political discords among them enabled this small base to survive until 973. Fraxinetum had an indirect relation with Andalus and functioned as a different type of sugr (buffer zone) against Latin and German states. This study was prepared to give information about the naval base founded by these Muslim marines and their activities in Western Mediterranean and the region of the Alp Mountains. Although the proximity of this region to Andalus geography, in our country studies were not realized about the place of Fraxinetum in Islamic history.Our article aims to a be an introduction to further studies on the topic., VII. asır ortalarında Afrika kıtasına giren Müslümanlar VIII. asrın başlarında İspanya’yı fethetmişler ve böylece bir asırdan daha kısa bir zamanda Akdeniz’in güneyi yeni bir siyasî yapıyla karşılaşmıştır. İspanya’da kurulan Endülüs Emevi Devleti ise ilk yıllarında bir donanma siyaseti geliştirmemiş ancak IX. asırdan itibaren ülkenin doğu sahillerinde devletten bağımsız denizci gruplar ortaya çıkmıştır. Bu asırda hem devlet çeşitli sebeplerden dolayı donanmasını güçlendirmek zorunda kalmış hem de bu denizci gruplar Sicilya ve Girit gibi daha doğudaki hedeflere gitmişlerdir. Bu Müslüman denizciler cihad amacıyla Hristiyan sahillerine akınlarda bulunmuşlar ya da korsanlık ve deniz ticareti gibi faaliyetleri yürütmüşlerdir. Endülüs’te kalan denizci gruplar 844 yılındaki Viking saldırısından sonra Endülüs emirlerinin resmî bir donanma teşkilatı oluşturma çabaları çerçevesinde devletin deniz ordusunun insan kaynağını teşkil etmiştir. Bilhassa ülkenin güney ve güneydoğu sahillerinde yaşayan ve Bahriyyun olarak tabir edilen denizcilerin bir kısmı Sevilla’daki ilk tersanenin çalışanları oldukları gibi gemilerde askerlik ve diğer hizmetlerde bulunmuşlardır. Bahriyyun taifesi IX. asrın sonlarına doğru ileride Endülüs halifelik donanmasının ana üssü olacak olan Almería şehrinin öncülü olan Pechina mevkiine yerleşmeye başlamışlardır. Denizci grupların tamamı devlet hizmetine hemen girmemiş ve bazıları deniz akınları ve ticaretle meşgul olmuşlardır. IX. asrın sonlarında bu gruplardan biri Fransa’nın güney sahillerine bir fırtına sebebiyle çıkmak zorunda kalmış ve planlanmayan bir şekilde burada seksen yıl kadar devam edecek olan Fraxinetum (Cebelu’l-Kılal) kıyı devletçiğini teşkil etmişlerdir. Endülüs’le insan ve malzeme takviyesi amacıyla denizden bağlantı kuran bu savaşçılar siyasî kargaşa içinde bulunan Provans bölgesinde zaman içinde çoğalmışlar ve Alp dağlarındaki geçitleri kullanmak suretiyle İtalya ve İsviçre’ye kadar yayılarak saldırılarda bulunmuşlardır. Dağ geçitlerindeki yolcu gruplarını hedef alarak esirler elde ettikleri gibi şehirlere saldırarak buralardan ganimet elde etmişlerdir. Bölgenin siyasî egemenleri Fraxinetum’daki bu savaşçılardan kurtulmak için Bizans’tan deniz yardımı talep etmişler ancak kendi aralarındaki siyasî anlaşmazlıklar bu küçük üssün 973 tarihine kadar yaşamasına imkân vermiştir. Endülüs Emevileri ile dolaylı bir ilişki içinde olan Fraxinetum bölgedeki Latin ve Cermen devletleri karşısında farklı bir tür sugr (tampon bölge) işlevi görmüştür. Bu çalışma Fraxinetum’da Müslüman denizciler tarafından oluşturulan deniz üssü ve Batı Akdeniz’deki ve Alp Dağları bölgesindeki faaliyetleri hakkında bilgi vermek amacıyla hazırlanmıştır. Endülüs coğrafyasına yakınlığına rağmen Fraxinetum’un İslam tarihindeki yeri hakkında ülkemizde çalışmalar yapılmamıştır. Makalemiz bu konuda başka çalışmalar için bir giriş yapmak amacını taşımaktadır.

Bu çalışmada Japon bıldırcın eritrosit dokularından 6-fosfoglukonat dehidrogenaz enzimi (6PGD) 52,84 EU.mg-1 spesifik aktiviteyle ve % 69 verimle 2ˈ, 5ˈ-ADP Sepharose 4B afinite kromatografisi kullanılarak saflaştırıldı. Japon bıldırcın eritrositlerinden saflaştırılan 6PGD enzimi için saflaştırma katsayısı 4984 olarak bulundu. Enzim saflığının kontrolü için SDS-poliakrilamid jel elektroforezi (SDS-PAGE) yapıldı ve tek bant gözlendi. SDS-PAGE yöntemi kullanılarak enzimin alt birimlerinin molekül kutlesi 81 kDa olarak hesaplandı. Optimum iyonik şiddet 0,5 M Tris-HCl, optimum pH ve stabil pH 0,5 M Tris-HCl tamponu pH 8,0 olarak bulundu. Optimum sıcaklık 60 ºC olarak bulundu. Ayrica Japon bıldırcın 6PGD enziminin KM ve Vmax değerleri Lineweaver-Burk grafiklerinden sırasıyla 6PGA substratı için KM değeri 0,120 mM, Vmax değeri 0,191 EU/mL, NADP+ substratı için KM değeri 0,017 mM, Vmax değerise 0,228 EU/mL olarak hesaplandı., In this study, the 6PGD enzyme from Japanese quail erythrocytes was purified with specific activity of 52.84 EU/mg and 69% yield of purification by 2ˈ, 5ˈ- ADP Sepharose 4B affinity gel in a single chromatographic method. The purification folds of the enzyme were 4984 folds. The purified enzyme was checked using SDS polyacrylamide gel electrophoresis (SDS-PAGE) method; the result of gel showed a single band. The subunit molecular weight of the enzyme was calculated as 81 kDa by the SDS-PAGE method. The characterization studies of the 6PGD enzyme from erythrocytes of Japanese quail showed: the optimum ionic strength to be at 0.5 M Tris-HCl, optimum and stable pH values to be at 0.5 M Tris-HCl buffers pH 8.0. The optimal temperature for the enzyme activity was found at 60 ˚C. Finally, the KM and Vmax values for the 6PGD enzyme from Japanese quail’s erythrocytes were calculated respectively for the 6PGA the KM value found as 0.120 mM, Vmax value as 0.191 EU/mL and for NADP+ the KM value as 0.017 mM and Vmax value as 0.228 EU/mL.
