Abstract

International audience; In the sub-Antarctic region, climate change is particularly rapid, while their freshwater ecosystems, such as ponds, host plant species with limited spatial distributions. These particular systems and their plant communities remain however poorly known and the context of their changing habitat calls for deeper insights into these systems. We performed an extensive survey of 45 ponds over three locations of the Iles Kerguelen during the winter and summer seasons of two years, which included the measurement of 12 abiotic parameters and the assessment of the plant community composition. Overall, our results showed that Iles Kerguelen ponds are shallow freshwater ecosystems harbouring species-poor plant communities with high even- ness, of which structure did not vary among the sampled locations. Three different habitats were identified among ponds in relation with nutrient supply by marine animals, which ultimately influenced plant community structure and cover. We highlight that these habitats are mostly determined by water depth, nutrients, and temperature (mean and variance). Present plant community composition was more strongly correlated with plant community composition in the previous year than with abiotic conditions. Overall, this study provides new knowledge on the aquatic plant communities and the functioning of Iles Kerguelen freshwater ecosystems, which can serve as a basis for future studies dealing with the impact of climate change in the sub-Antarctic region.

Abstract

International audience; Plant genomes are characterized by large and complex gene families that often result in similar and partially overlapping functions 1. This genetic redundancy severely hampers current efforts to uncover novel phenotypes, delaying basic genetic research and breeding programs 2. Here, we describe the development and validation of Multi-Knock, a genome-scale CRISPR toolbox that overcomes functional redundancy in Arabidopsis by simultaneously targeting multiple gene-family members, thus identifying genetically hidden components. We computationally designed 59,129 optimal single guide RNAs (sgRNAs) that each target 2 to 10 genes within a family at once. Furthermore, partitioning the library into ten sub-libraries directed towards a different functional group allows flexible and targeted genetic screens. From the 5,635 sgRNAs targeting the plant transportome, we generated over 3,500 independent Arabidopsis lines that allowed us to identify and characterize the first known cytokinin tonoplast-localized transporters in plants. With the ability to overcome functional redundancy in plants at the genome-scale level, the developed strategy can be readily deployed by scientists and breeders for basic research and to expedite breeding efforts.

Abstract

Variation in species richness across the tree of life, accompanied by the incredible variety of ecological and morphological characteristics found in nature, has inspired many studies to link traits with species diversification. Angiosperms are a highly diverse group that has fundamentally shaped life on earth since the Cretaceous, and illustrate how species diversification affects ecosystem functioning. Numerous traits and processes have been linked to differences in species richness within this group, but we know little about how these interact and their relative importance. Here, we synthesized data from 152 studies that used state-dependent speciation and extinction (SSE) models on angiosperm clades. Intrinsic traits related to reproduction and morphology were often linked to diversification but a set of universal drivers did not emerge as traits did not have consistent effects across clades. Importantly, dataset properties were correlated to SSE model results - trees that were larger, older, or less well-sampled tended to yield trait-dependent outcomes. We compared these properties to recommendations for SSE model use and provide a set of best practices to follow when designing studies and reporting results. Finally, we argue that SSE model inferences should be considered in a larger context incorporating species’ ecology, demography and genetics.

Abstract

International audience; In Arabidopsis, the receptor-like kinase (RLK) FERONIA (FER) senses peptide ligands in the plasma membrane (PM), modulates plant growth and development, and integrates biotic and abiotic stress signaling for downstream adaptive responses. However, the molecular interplay of these diverse processes is largely unknown.Here, we show that FER, the receptor of Rapid Alkalinization Factor 1 (RALF1), physically interacts with C2 domain ABA-related (CAR) proteins to control the nano-organization of the PM. During this process, the RALF1-FER pathway upregulates CAR protein translation, and then more CAR proteins are recruited to the PM. This acts as a rapid feedforward loop that stabilizes the PM liquid-ordered phase. FER interacts with and phosphorylates CARs, thereby reducing their lipid-binding ability and breaking the feedback regulation at later time points.The formation of the flg22-induced FLS2-BAK1 immune complex, which depends on the integrity of FER-containing nanodomains, is impaired in fer and pentuple car14569 mutant.Together, we propose that the FER-CAR module controls the formation of PM nano-organization during RALF signaling through a self-contained amplifying loop including both positive and negative feedback.

Abstract

International audience; Climate change is mostly driven by the increase of atmospheric CO2 concentration. It leads to an increase in ambient temperature and temperature extremes, as well as to a reduction in water availability (Box 1). Despite the apparent benefit that an increase of CO2 and ambient temperature might have on the production of plant biomass, climate change severely challenges the plant life cycle, and thus increasingly leads to food insecurity. Investigating the molecular mechanisms of plant adaptation to climate change is thus a pressing issue. This was the theme of the EMBO Workshop ‘Molecular responses of plants facing climate change’ in June 2022 in Montpellier, France. Here, we summarize some key insights presented during the conference on the investigation of molecular mechanisms of plant adaptation to environmental constraints in a changing climate.

Abstract

International audience; In the context of global warming, a clear understanding of microrefugia—microsites enabling the survival of species populations outside their main range limits—is crucial. Several studies have identified forcing factors that are thought to favor the existence of microrefugia. However, there is a lack of evidence to conclude whether, and to what extent, the climate encountered within existing microrefugia differs from the surrounding climate. To investigate this, we adopt a “bottom-up” approach, linking marginal disconnected populations to microclimate. We used the southernmost disconnected and abyssal populations of the circumboreal herbaceous plant Oxalis acetosella in Southern France to study whether populations in sites matching the definition of “microrefugia” occur in particularly favorable climatic conditions compared to neighboring control plots located at distances of between 50 to 100 m. Temperatures were recorded in putative microrefugia and in neighboring plots for approximately 2 years to quantify their thermal offsets. Vascular plant inventories were carried out to test whether plant communities also reflect microclimatic offsets. We found that current microclimatic dynamics are genuinely at stake in microrefugia. Microrefugia climates are systematically colder compared to those found in neighboring control plots. This pattern was more noticeable during the summer months. Abyssal populations showed stronger offsets compared to neighboring plots than the putative microrefugia occurring at higher altitudes. Plant communities demonstrate this strong spatial climatic variability, even at such a microscale approach, as species compositions systematically differed between the two plots, with species more adapted to colder and moister conditions in microrefugia compared to the surrounding area

The loss of immunity repression or early senescence activation compromises symbiont survival and leads to the formation of nonfunctional nodules (fix−). Despite many studies exploring an overlap between immunity and senescence responses outside the nodule context, the relationship between these processes in the nodule remains poorly understood. To investigate this phenomenon, we selected and characterized three Medicago mutants developing fix− nodules and showing senescence responses. Analysis of specific defense (PATHOGENESIS-RELATED PROTEIN) or senescence (CYSTEINE PROTEASE) marker expression demonstrated that senescence and immunity seem to be antagonistic in fix− nodules. The growth of senescence mutants on non-sterile (sand/perlite) substrate instead of sterile in vitro conditions decreased nodule senescence and enhanced defense, indicating that environment can affect the immunity/senescence balance. The application of wounding stress on wild-type (WT) fix+ nodules led to the death of intracellular rhizobia and associated with co-stimulation of defense and senescence markers, indicating that in fix+ nodules the relationship between the two processes switches from opposite to synergistic to control symbiont survival during response to the stress. Our data show that the immune response in stressed WT nodules is linked to the repression of DEFECTIVE IN NITROGEN FIXATION 2 (DNF2), Symbiotic CYSTEINE-RICH RECEPTOR-LIKE KINASE (SymCRK), and REGULATOR OF SYMBIOSOME DIFFERENTIATION (RSD), key genes involved in symbiotic immunity suppression. This study provides insight to understand the links between senescence and immunity in Medicago nodules. Analyses of Medicago mutants with non-functional nodules highlight the relationship and mechanisms controlling the establishment of the immune and senescence programs during nodule organogenesis.

Abstract

International audience; The plant cytoskeleton regulates fundamental biological processes, including cell division. How to experimentally perturb the cytoskeleton is a key question if one wants to understand the role of both actin filaments (AFs) and microtubules (MTs) in a given biological process. While a myriad of mutants are available, knock-out in cytoskeleton regulators, when nonlethal, often produce little or no phenotypic perturbation because such regulators are often part of a large family, leading to functional redundancy. In this review, alternative techniques to modify the plant cytoskeleton during plant cell division are outlined. The different pharmacological and genetic approaches already developed in cell culture, transient assays, or in whole organisms are presented. Perspectives on the use of optogenetics to perturb the plant cytoskeleton are also discussed.

Abstract

The nucleotides guanosine tetraphosphate and pentaphosphate (or (p)ppGpp) are implicated in the regulation of chloroplast function in plants. (p)ppGpp signalling is best understood in the model vascular plant Arabidopsis thaliana in which it acts to regulate plastid gene expression to influence photosynthesis, plant development and immunity. However, little information is known about the conservation or diversity of (p)ppGpp signalling in other land plants. We studied the function of ppGpp in the moss Physcomitrium (previously Physcomitrella) patens using an inducible system for triggering ppGpp accumulation. We used this approach to investigate the effects of ppGpp on chloroplast function, photosynthesis and growth. We demonstrate that ppGpp accumulation causes a dramatic drop in photosynthetic capacity by inhibiting chloroplast gene expression. This was accompanied by the unexpected reorganisation of the thylakoid system into super grana. Surprisingly, these changes did not affect gametophore growth, suggesting that bryophytes and vascular plants may have different tolerances to defects in photosynthesis. Our findings point to the existence of both highly conserved and more specific targets of (p)ppGpp signalling in the land plants that may reflect different growth strategies.

Abstract

Seedling root traits impact plant establishment under challenging environments. Pearl millet is one of the most heat and drought tolerant cereal crops that provides a vital food source across the sub-Saharan Sahel region. Pearl millet’s early root system features a single fast-growing primary root which we hypothesize is an adaptation to the Sahelian climate. Using crop modelling, we demonstrate that early drought stress is an important constraint in agrosystems in the Sahel where pearl millet was domesticated. Furthermore, we show that increased pearl millet primary root growth is correlated with increased early water stress tolerance in field conditions. Genetics including GWAS and QTL approaches identify genomic regions controlling this key root trait. Combining gene expression data, re-sequencing and re- annotation of one of these genomic regions identified a glutaredoxin-encoding genePgGRXC9as the candidate stress resilience root growth regulator. Functional characterization of its closest Arabidopsis homologAtROXY19revealed a novel role for this glutaredoxin (GRX) gene clade in regulating cell elongation. In summary, our study suggests a conserved function for GRX genes in conferring root cell elongation and enhancing resilience of pearl millet to its Sahelian environment.

Abstract

International audience; Over the past decades, the atmospheric CO2 concentration and global average temperature have been increasing, and this trend is projected to soon become more severe. This scenario of climate change intensifies abiotic stress factors (such as drought, flooding, salinity, and ultraviolet radiation) that threaten forest and associated ecosystems as well as crop production. These factors can negatively affect plant growth and development with a consequent reduction in plant biomass accumulation and yield, in addition to increasing plant susceptibility to biotic stresses. Recently, biostimulants have become a hotspot as an effective and sustainable alternative to alleviate the negative effects of stresses on plants. However, the majority of biostimulants have poor stability under environmental conditions, which leads to premature degradation, shortening their biological activity. To solve these bottlenecks, micro- and nano-based formulations containing biostimulant molecules and/or microorganisms are gaining attention, as they demonstrate several advantages over their conventional formulations. In this review, we focus on the encapsulation of plant growth regulators and plant associative microorganisms as a strategy to boost their application for plant protection against abiotic stresses. We also address the potential limitations and challenges faced for the implementation of this technology, as well as possibilities regarding future research.

Abstract

International audience; Centromeres are critical for cell division, loading CENH3 or CENPA histone variant nucleosomes, directing kinetochore formation and allowing chromosome segregation(1,2). Despite their conserved function, centromere size and structure are diverse across species. To understand this centromere paradox(3,4), it is necessary to know how centromeric diversity is generated and whether it reflects ancient trans-species variation or, instead, rapid post-speciation divergence. To address these questions, we assembled 346 centromeres from 66 Arabidopsis thaliana and 2 Arabidopsis lyrata accessions, which exhibited a remarkable degree of intra- and inter-species diversity. A. thaliana centromere repeat arrays are embedded in linkage blocks, despite ongoing internal satellite turnover, consistent with roles for unidirectional gene conversion or unequal crossover between sister chromatids in sequence diversification. Additionally, centrophilic ATHILA transposons have recently invaded the satellite arrays. To counter ATHILA invasion, chromosome-specific bursts of satellite homogenization generate higher-order repeats and purge transposons, in line with cycles of repeat evolution. Centromeric sequence changes are even more extreme in comparison between A. thaliana and A. lyrata. Together, our findings identify rapid cycles of transposon invasion and purging through satellite homogenization, which drive centromere evolution and ultimately contribute to speciation.

Since the beginning of research on the genetics of speciation, several questions have emerged such as: What are the genetic bases of incompatibilities? How many loci are necessary to prevent hybridization and how are they distributed along genomes? Can speciation occur despite gene flow and how common is ecological speciation? Early stages of divergence are key to understand the ecology and genetics of speciation, and semi-isolated species where hybrids can still be produced are particularly relevant.

Abstract

International audience; Analysis of plant metabolite 13C-enrichments with gas-chromatography mass spectrometry (GC/MS) has gained interest recently. By combining multiple fragments of a trimethylsilyl (TMS) derivative, 13C-positional enrichments can be calculated. However, this new approach may suffer from analytical biases depending on the fragments selected for calculation leading to significant errors in the final results. The goal of this study was to provide a framework for the validation of 13C-positional approaches and their application to plants based on some key metabolites (glycine, serine, glutamate, proline, α-alanine and malate). For this purpose, we used tailor-made 13C-PT standards, harboring known carbon isotopologue distributions and 13C-positional enrichments, to evaluate the reliability of GC-MS measurements and positional calculations. Overall, we showed that some mass fragments of proline_2TMS, glutamate_3TMS, malate_3TMS and α-alanine_2TMS had important biases for 13C measurements resulting in significant errors in the computational estimation of 13C-positional enrichments. Nevertheless, we validated a GC/MS-based 13C-positional approach for the following atomic positions: (i) C1 and C2 of glycine_3TMS, (ii) C1, C2 and C3 of serine_3TMS, and (iii) C1 of malate_3TMS and glutamate_3TMS. We successfully applied this approach to plant 13C-labeled experiments for investigating key metabolic fluxes of plant primary metabolism (photorespiration, tricarboxylic acid cycle and phosphoenolpyruvate carboxylase activity).

Abstract

Cystathionine-β-synthase (CBS) domains are found in proteins of all living organisms and have been proposed to play a role as energy sensors regulating protein activities through their adenosyl ligand binding capacity. In plants, members of the CBSX protein family carry a stand-alone pair of CBS domains. In Arabidopsis (Arabidopsis thaliana), CBSX1 and CBSX2 are targeted to plastids where they have been proposed to regulate thioredoxins (TRXs). TRXs are ubiquitous cysteine thiol oxido-reductases involved in the redox-based regulation of numerous enzymatic activities as well as in the regeneration of thiol-dependent peroxidases. In Arabidopsis, 10 TRX isoforms have been identified in plastids and divided into five sub-types. Here, we show that CBSX2 specifically inhibits the activities of m-type TRXs toward two chloroplast TRX-related targets. By testing activation of NADP-malate dehydrogenase and reduction of 2-Cys peroxiredoxin, we found that TRXm1/2 inhibition by CBSX2 was alleviated in the presence of AMP or ATP. We also determined, by pull-down assays, a direct interaction of CBSX2 with reduced TRXm1 and m2 that was abolished in the presence of adenosyl ligands. In addition, we report that, compared with wild-type plants, the Arabidopsis T-DNA double mutant cbsx1 cbsx2 exhibits growth and chlorophyll accumulation defects in cold conditions, suggesting a function of plastidial CBSX proteins in plant stress adaptation. Together, our results show an energy-sensing regulation of plastid TRX m activities by CBSX, possibly allowing a feedback regulation of ATP homeostasis via activation of cyclic electron flow in the chloroplast, to maintain a high energy level for optimal growth.

Abstract

We analyzed changes in the long-term vegetation cover and in fire activity over the past 5000 years in the Ecuadorian páramo using a sediment core from Papallacta (Ecuador). The chronology is constrained by three tephra layers and 32 AMS 14C ages, and 168 samples yielded a high-resolution record of environmental changes. We estimated the upslope wind convectivity as the ratio between pollen transported from the Andean cloud forest and Poaceae pollen to distinguish changes in atmospheric moisture from changes in soil moisture. The record showed that the two sources of moisture, either from year-round adiabatic cloud dripping linked to SASM activity or to ENSO variability at decadal-scale, influenced vegetation-cover changes. Between 5000 and 2450 cal yr BP, both soil moisture and biomass burning were higher than after 2450 cal yr BP. The shift between the two states matches the zonal increase in summer insolation that drove the ITCZ to its southernmost position. Our results underline resilience to volcanic activity, the importance of the upslope convective dripping with the lowest convective index observed at ~4500 cal yr BP, the anomalous last century with the highest convective activity and the driest soil conditions recorded in the last 5000 years, the recent increase in fire activity and the link between soil moisture and the position of the ITCZ.

Abstract

International audience; It has been argued that the mechanisms structuring ecological communities may be more generalizable when based on traits than on species identities. If so, patterns in the assembly of community-level traits along environmental gradients should be similar in different places in the world. Alternatively, geographical change in the species pool and regional variation in climate might result in site-specific relationships between community traits and local environments. These competing hypotheses are particularly untested for animal communities. Here we test the geographical constancy of trait-based assembly patterns using a widespread multi-trophic community: aquatic macroinvertebrates within bromeliads. We used data on 615 invertebrate taxa from 1,656 bromeliads in 26 field sites from Mexico to Argentina. We summarized invertebrate traits with four orthogonal axes, and used these trait axes to examine trait convergence and divergence assembly patterns along three environmental gradients: detrital biomass and water volume in bromeliads, and canopy cover over bromeliads. We found no overall signal of trait-based assembly patterns along any of the environmental gradients. However, individual sites did show trait convergence along detrital and water gradients, and we built predictive models to explore these site differences. Sites that showed trait convergence along detrital gradients were all north of the Northern Andes. This geographical pattern may be related to phylogeographical differences in bromeliad morphology. Bromeliads with low detritus were dominated by detritivorous collectors and filter feeders, where those with high detritus had more sclerotized and predatory invertebrates. Sites that showed the strongest trait convergence along gradients in bromeliad water were in regions with seasonal precipitation. In such sites, bromeliads with low water were dominated by soft-bodied, benthic invertebrates with simple life cycles. In less seasonal sites, traits associated with short-term desiccation resistance, such as hard exoskeletons, were more important. In summary, we show that there are strong geographical effects on the trait-based assembly patterns of this invertebrate community, driven by the biogeography of their foundational plant species as well as by regional climate. We suggest that inclusion of biogeography and climate in trait-based community ecology could help make it a truly general theory.

Abstract

International audience; Improving the global monitoring of above-ground biomass (AGB) is crucial for forest management to be effective in climate mitigation. In the last decade, methods have been developed for estimating AGB from terrestrial laser scanning (TLS) data. TLS-derived AGB estimates can address current uncertainties in allometric and Earth observation (EO) methods that quantify AGB. We assembled a global dataset of TLS scanned and consecutively destructively measured trees from a variety of forest conditions and reconstruction pipelines. The dataset comprised 391 trees from 111 species with stem diameter ranging 8.5 to 180.3 cm and AGB ranging 13.5–43,950 kg. TLS-derived AGB closely agreed with destructive values (bias 1,000 kg). More effort should go to further understanding and constraining several TLS error sources. We currently lack an objective method of evaluating point cloud quality for tree volume reconstruction, hindering the development of reconstruction algorithms and presenting a bottleneck for tracking down the error sources identified in our synthesis. Since quantifying AGB with TLS requires only a fraction of the efforts as compared to destructive harvesting, TLS-calibrated ASMs can become a powerful tool in AGB upscaling. TLS will be critical for calibrating/validating scheduled and launched remote sensing initiatives aiming at global AGB mapping.

Abstract

International audience; Woody encroachment and forest progression are widespread in forest-savanna transitional areas in Central Africa. Quantifying these dynamics and understanding their drivers at relevant spatial scales has long been a challenge. Recent progress in open access imagery sources with improved spatial, spectral and temporal resolution combined with cloud computing resources, and the advent of relatively cheap solutions to deploy laser sensors in the field, have transformed this domain of study. We present a study case in the Mpem & Djim National Park (MDNP), a 1,000 km2 protected area in the Centre region of Cameroon. Using open source algorithms in Google Earth Engine (GEE), we characterized vegetation dynamics and the fire regime based on Landsat multispectral imagery archive (1975–2020). Current species assemblages were estimated from Sentinel 2 imagery and the open source biodivMapR package, using spectral dissimilarity. Abstract

Many climbing plants have microspines on their stems, which facilitate attachment and prevent slipping and falling from host plant supports. Extending via growth through complex environments and anchoring stems to substrates with minimal contact forces are key benefits for climbing plants. Microspines are also highly desirable features for new technologies and applications in soft robotics. Using a novel sled-like device, we investigated static and sliding attachment forces generated by stems in 10 species of tropical climber from French Guiana differing in size and climbing habit. Eight species showed higher static and sliding forces when their stems were pulled in the basal direction against a standard surface than in the apical direction. This anisotropic behaviour suggests that tropical climbers have evolved different ratchet-like mechanisms that allow easy sliding forwards but are resistant to slipping downwards. The presence of a downwards ‘stick-and-slip’ phenomenon, where static attachment is not significantly stronger than maximal sliding attachment, was present in most species apart from three showing relatively weak attachment by microspines. This indicates that diverse microspine attachment strategies exist in climbing plants. This diversity of functional properties offers a range of potential design specifications for climbing strategies on different substrates for artificial climbing artefacts.

Abstract

International audience; Early plant primary succession in recently deglacierized terrains is a good indicator of the species losses and gains that affect novel alpine plant communities migrating under the pressure of climate warming. In the tropical alpine -páramo- region of the northern Andes, home to the world's greatest alpine phyto-diversity, forced primary succession will condition the conservation of many species. Using a post-glacial chronosequence between the little ice age and present below the Conejeras glacier (Colombia) as a space-for-time substitution approach, we sought to determine how time since deglacierization affects the composition, the biogeographic origin and the growth form distribution of novel tropical alpine plant communities. Using an array of multivariate techniques and the Dirichlet model, we assessed relationships among plant communities and with environmental factors. Communities established in less than 169 years lacked a number of characteristic and endemic species usually found in the national park Los Nevados, such as Calamagrostis effusa, Senecio isabelis and Espeletia hartwegiana. Moreover, these communities have been colonized by non-native species, e.g., Rumex acetosella. Upright shrubs and large tussock grasses, characteristic of the alpine tropics, established slowly because they required highly organic, slow-developing soils. Taxa of tropical biogeographic origin were under-represented early after deglacierization in comparison with temperate taxa. These results suggest the existence of a strong climatic debt for some native species, tropical growth forms and taxa of tropical origin, which may translate into significant taxonomic and functional losses whereas the few observed gains concern the establishment of non-native species.

Abstract

International audience; Clustered organization of biosynthetic non-homologous genes is emerging as a characteristic feature of plant genomes. The co-regulation of clustered genes seems to largely depend on epigenetic reprogramming and three-dimensional chromatin conformation. In this study, we identified the long non-coding RNA (lncRNA) MARneral Silencing (MARS), localized inside the Arabidopsis marneral cluster, which controls the local epigenetic activation of its surrounding region in response to abscisic acid (ABA). MARS modulates the POLYCOMB REPRESSIVE COMPLEX 1 (PRC1) component LIKE HETEROCHROMATIN PROTEIN 1 (LHP1) binding throughout the cluster in a dose-dependent manner, determining H3K27me3 deposition and chromatin condensation. In response to ABA, MARS decoys LHP1 away from the cluster and promotes the formation of a chromatin loop bringing together the MARNERAL SYNTHASE 1 (MRN1) locus and a distal ABA-responsive enhancer. The enrichment of co-regulated lncRNAs in clustered metabolic genes in Arabidopsis suggests that the acquisition of novel non-coding transcriptional units may constitute an additional regulatory layer driving the evolution of biosynthetic pathways

Abstract

International audience; 1. Neutral landscape models have many applications in ecology, such as supporting spatially-explicit simulations, developing, and evaluating landscape indices. However, current approaches provide few options to produce large landscapes with controlled composition and fragmentation indices. 2. We introduce flsgen (Fragmented Landscape Generator), a new neutral landscape generator that address this limitation by providing a high level of control over 14 landscape indices. The main novelty of flsgen is the decomposition of landscape generation into two steps: the solving of a constraint satisfaction problem and the generation of a landscape raster with a stochastic algorithm. The latter relies on a continuous environmental gradient that influences the landscape’s spatial configuration. 3. flsgen can generate fine-grained artificial landscapes in small amounts of time, which makes it suited to produce large landscape series systematically. We demonstrate the features of flsgen through three illustrative use cases. 4. flsgen is a practical and efficient tool that expand the current possibilities of neutral landscape models and widen their potential applications. To facilitate its uptake, flsgen is available as free and open-source software through a Java API, a command-line interface, or an R package.

International audience; The justification of the 4 genera that currently compose the class Pavlovophyceae is based on a low number of species and a relative paucity of available, traceable and referenced cultures. Previous integrative phylogeny work revealed strains that can refine and strengthen our knowledge of the genera in the class. The application of multiple light and electron microscopy techniques allowed us to prioritize the cytomorphological characters (pyrenoid, thylakoid, stigma, knob-scales, life stage / life cycle) used for the taxonomy of these algae and to describe two new species: Exanthemachrysis fresneliae Véron sp. nov. and Rebecca billardiae Véron sp. nov. Consequently, revisions of the two genera Exanthemachrysis Lepailleur emend. Véron and Rebecca Green emend. Véron were made. In addition, the genus Pavlova Butcher emend Véron is revised in the light of these characters. Particular emphasis is placed on the life stages and habitat of the species.

Abstract

International audience; Native plants of the Sahara are remarkably resistant to prolonged droughts and are important sources of forage, food, and medicine. Their regeneration from seeds after strong anthropogenic disturbances is a challenging step toward their conservation and to mitigate desertification. We studied the germination responses of 43 dominant native species of the Sahara desert that could be propagated from seeds for future ecological restoration projects, exploring the effects of cold stratification and temperatures on seed germination. Seeds were stratified at 4°C for 3 days and then tested for germination at 5, 10, 15, 20, 25, 30, 35, and 40°C. While cold stratification negatively affected four species, it improved the germination of 36 species. Five of these species required cold stratification to germinate. Temperatures during the germination trial significantly influenced seed germinability. Twenty-three species (54%) germinated to higher percentages at intermediate temperatures (20 to 30°C) similar to the maximum temperatures of the rainiest months, when germination in the field is high. Thirteen species (30%) also germinated at a higher temperature (40°C). Seven species (16%) germinated best at low temperatures (5 to 10°C). Our results suggest that cold stratification is an efficient and easy treatment to apply prior to sowing seeds in the field for restoration.

Abstract

International audience; Clarifying generic circumscriptions within Oleaceae improves its complicated infrafamilial classification. Focusing on the small and disjunctly-distrubted subtribe Schreberinae, we use phylogenomic data from plastid and nuclear DNA from an extensive sampling to assess its phylogenetic patterns and biogeographic history. Results show paraphyly among its members, with Comoranthus nested within Schrebera, leading us to synonymize these genera. Schrebera s.l. occurs in four major regions of the world, but its center of origin remains uncertain with three possible ancestral ranges identified. However, the diversification of this genus is estimated to have started in the Early Oligocene (ca. 34 Mya), and its current distribution pattern is best explained by dispersal between landmasses rather than continental vicariance. Madagascar is the center of diversity of Schrebera s.l., where ten endemic species, of which five are new, were recovered from analyses of morphological, molecular, and ecoclimatic data. Therefore, a taxonomic treatment of the species from Madagascar and the Comoro Islands is presented. As currently circumscribed, Schrebera s.l., and thus the monotypic subtribe, includes 16 species (one in South America, two in Southeast Asia, three in Africa, of which one is shared with Madagascar, and 11 in Madagascar and the Comoro Islands).

Abstract

International audience; Assessment methods have been developed to estimate a preliminary conservation status for species and subsequently to facilitate the building of Red Lists. Such pre-assessment methods could be particularly useful in the French Overseas Territories (FOTs) where Red Lists tend to be outdated or absent and where a high number of endemic species face detrimental anthropogenic pressures. We first aimed to conduct a preliminary assessment (hereafter, pre-assessment) of the conservation status of endemic plants from Guadeloupe, Martinique, Réunion, Mayotte, French sub-Antarctic islands, New Caledonia, and Scattered Islands. We then compared the various methods used in conducting the pre-assessment and discussed ways to adapt these methods to small territories. We compiled occurrence data of endemic species identified thanks to a previous taxonomic work and pre-assessed their conservation status under Red List criteria A and B and the use of a Random Forest algorithm. We then measured the accuracy, specificity, and sensitivity of each method based on existing Red Lists. The Random Forest algorithm and a method based on range-size performed best at correctly attributing conservation status. Using these pre-assessment methods, we estimated that up to 60% of the endemic flora of the FOTs is potentially threatened. Range restriction but also anthropogenic pressures were key factors that explained these risks. Pre-assessment methods are useful tools to get a first measure of species conservation status. These methods should be adapted to the territories considered and their conservation issues in order to reach a good performance.

Abstract

The brassinosteroid (BR) hormone and its plasma membrane (PM) receptor BR INSENSITIVE1 (BRI1) are one of the best-studied receptor–ligand pairs for understanding the interplay between receptor endocytosis and signaling in plants. BR signaling is mainly determined by the PM pool of BRI1, whereas BRI1 endocytosis ensures signal attenuation. As BRs are ubiquitously distributed in the plant, the tools available to study the BRI1 function without interference from endogenous BRs are limited. Here, we designed a BR binding-deficient Arabidopsis (Arabidopsis thaliana) mutant based on protein sequence-structure analysis and homology modeling of members of the BRI1 family. This tool allowed us to re-examine the BRI1 endocytosis and signal attenuation model. We showed that despite impaired phosphorylation and ubiquitination, BR binding-deficient BRI1 internalizes similarly to the wild type form. Our data indicate that BRI1 internalization relies on different endocytic machineries. In addition, the BR binding-deficient mutant provides opportunities to study non-canonical ligand-independent BRI1 functions.

International audience; The plant hormone auxin is certainly the most studied developmental regulator in plants. The many functions of auxin during development, from the embryo to the root and shoot construction, are mediated by an ever-growing collection of molecular regulators, with an overwhelming degree of both ubiquity and complexity that we are still far from fully understanding and that biological experiments alone cannot grasp. In this review, we discuss how bioinformatics and computational modeling approaches have helped in recent years to explore this complexity and to push the frontiers of our understanding of auxin biology. We focus on how analysis of massive amounts of genomic data and construction of computational models to simulate auxin-regulated processes at different scales have complemented wet experiments to increase the understanding of how auxin acts in the nucleus to regulate transcription and how auxin movement between cells regulates development at the tissular scale. Abstract

International audience; Within a landscape, multiple habitats exist for soil microbial communities. But how these habitats shape community composition requires an understanding of the way in which microbial diversity is impacted across a broad range of spatial scales. Mountain ecosystems are excellent systems to study microbial communities, because a multitude of climate and soil variables change within a relatively small distance. We investigated microbial community structure in bulk and rhizosphere soils beneath three plant species, Vaccinium myrtillus, Juniperus communis and Picea abies, that structure local plant communities along an elevation gradient in the French Alps. We examined the impact that climate, soil properties, plant diversity and plant root chemical and morphological traits had on microbial αand β-diversities. The most abundant bacterial phyla detected in both bulk and rhizosphere soils were Proteobacteria, Actinobacteria, Acidobacteria and Verrucomicrobia. Along the elevation gradient, bacterial phyla did not display a clear distribution pattern between bulk and rhizosphere soils. For fungi, dominant phyla were Ascomycota and Basidiomycota, and contrasting distribution patterns were found between bulk and rhizosphere soils. Overall, bacterial and fungal α-diversity responded differently to elevation as well to soil compartments (bulk versus rhizosphere soil), revealing no significant patterns in bulk soil beneath any of the structuring plant species, but increasing in the rhizosphere compartment of P. abies just below the treeline. Changes in bacterial β-diversity with elevation were related mostly to soil physical and chemical properties. Bacterial and fungal α-diversity in rhizosphere communities were more related to plant species identity, vegetation diversity and belowground plant traits compared to soil properties, whilst the opposite was found for bulk soil. Our results highlight that environmental changes at the landscape scale (e.g. associated to elevation, soil properties or climate), impact significantly soil microbial communities, but vegetation refines communities at a local scale via the rhizosphere niche.

Abstract

International audience; Understanding of the mechanisms underlying species coexistence within plant communities is crucial to predict their fate given the current context of biodiversity loss. Freshwater ecosystems are among the most abiotically constrained habitats because they harbour clonal macrophyte communities. Strong habitat filtering in these ecosystems influences the functional composition and diversity of macrophyte communities, determining the outcome of interactions between neighbours and ultimately affecting local spatial arrangement between neighbours (LSA). This influence may be modulated by environmental conditions in abiotically constrained habitats, such as freshwater ecosystems.We focused on macrophyte communities living in ponds in the Iles Kerguelen, in the sub-Antarctic. These freshwater ecosystems are especially abiotically constrained (cold climate), and their plant communities are remarkably species-poor, simplifying the study of interactions between neighbours. We measured several abiotic variables in the ponds, species LSA and interspecific interactions (using the log response ratio metric), and the functional composition of the community using aerial, root and clonal traits. We also determined the biomass of the whole macrophyte community.Our results showed that LSA does not effectively assess interactions between neighbours at very small scales, neither at the community (one species vs. all neighbouring species) nor the species level (between pairs of species). Secondly, aerial (leaf and stem) and root traits related to resource acquisition played a more important role in interactions between neighbours than clonal traits (i.e. internode length and specific internode mass related to space acquisition and resource storage, respectively).Depending on the target species, (1) interactions responded positively or negatively to mean trait and functional diversity of the community; and (2) different traits of neighbours (aerial or root traits) triggered an interaction response. Lastly, abiotic variables, in particular water temperature and light intensity, influenced macrophyte community biomass and plant community structure (i.e. species richness, functional diversity and LSA), either directly or through the modulation of other abiotic variables.Our results lead to the following conclusions: (1) interactions between neighbours do not reflect their local spatial arrangements but are strongly associated with abiotic variables and neighbourhood functional traits (depending on species); (2) joint integration of biotic and abiotic variables in multivariate analyses enables better inference of biodiversity responses to environment, and the subsequent consequences for ecosystem function, especially in the context of climate change.

Abstract

Phylogenetically closely related plant species often share similar trait states (phylogenetic signal), but local assembly may favor dissimilar relatives and thereby decouple the diversity of a trait from the diversity of phylogenetic lineages. Associated fauna might either benefit from plant trait diversity, because it provides them complementary resources, or suffer from it due to dilution of preferred resources. We hence hypothesize that decoupling of trait and phylogenetic diversity weakens the relationship between the plant-trait diversity and the abundance and diversity of associated fauna. Studying permanent meadows, we tested for combined effects of plant phylogenetic diversity and diversity of two functional traits (specific leaf area, leaf dry matter content) on major groups of soil fauna (earthworms, mites, springtails, nematodes). We found that only in phylogenetically uniform plant communities, was uniformity in the functional traits associated with (i) high abundance in springtails, and (ii) high abundance of the sub-group that feeds more directly on plant material (in springtails and mites) or those that are more prone to disturbance (in nematodes), and (iii) high diversity in all three groups tested (springtails, earthworms, nematodes). Our results suggest that soil fauna profits from the resource concentration in local plant communities that are uniform in both functional traits and phylogenetic lineages. Soil fauna would hence benefit from co-occurrence of closely related plants that have conserved the same trait values, rather than of distantly related plants that have converged in traits. This might result in faster decomposition and a positive feedback between trait conservatism and ecosystem functioning.

Abstract

International audience; MAPKs are universal eukaryotic signaling factors whose functioning is assumed to depend on the recognition of a common docking motif (CD) by its activators, substrates, and inactiva- tors. We studied the role of the CD domain of Arabidopsis MPK4 by performing interaction stu- dies and determining the ligand-bound MPK4 crystal structure. We revealed that the CD domain of MPK4 is essential for interaction and activation by its upstream MAPKKs MKK1, MKK2, and MKK6. Cys181 in the CD site of MPK4 was shown to become sulfenylated in response to reactive oxygen species in vitro. To test the function of C181 in vivo, we generated wild-type (WT) MPK4-C181, nonsulfenylatable MPK4-C181S, and potentially sulfenylation mimicking MPK4-C181D lines in the mpk4 knockout back- ground. We analyzed the phenotypes in growth, development, and stress responses, reveal- ing that MPK4-C181S has WT activity and complements the mpk4 phenotype. By contrast, MPK4-C181D cannot be activated by upstream MAPKK and cannot complement the pheno- types of mpk4. Our findings show that the CD motif is essential and is required for activation by upstream MAPKK for MPK4 function. Furthermore, growth, development, or immunity functions require upstream activation of the MPK4 protein kinase.

Abstract

As auxin is a major regulator of plant development, studying the signaling mechanisms by which auxin influences cellular activities is of primary importance. In this review, we describe current knowledge on the different modalities of signaling, from the well-characterized canonical nuclear auxin pathway, to the more recently discovered or re-discovered non-canonical modes of auxin signaling. In particular, we discuss how both the modularity of the nuclear auxin pathway and the dynamic regulation of its core components allow specific transcriptomic responses to be triggered. We highlight the fact that the diversity of modes of auxin signaling allows for a wide range of time scales of auxin responses, from second-scale cytoplasmic responses to minute-/hour-scale modifications of gene expression. Finally, we question the extent to which the temporality of auxin signaling and responses contributes to development in both the shoot and the root meristems. We conclude by stressing the fact that future investigations should allow an integrative view to be built not only of the spatial control, but also of the temporality of auxin-mediated regulation of plant development, from the cell to the whole organism.

Abstract

In plants, as in animals, organ growth depends on mechanical interactions between cells and tissues, and is controlled by both biochemical and mechanical cues. Here, we investigate the control of seed size, a key agronomic trait, by mechanical interactions between two compartments: the endosperm and the testa. By combining experiments with computational modelling, we present evidence that endosperm pressure plays two antagonistic roles: directly driving seed growth, but also indirectly inhibiting it through tension it generates in the surrounding testa, which promotes wall stiffening. We show that our model can recapitulate wild type growth patterns, and is consistent with the small seed phenotype of the haiku2 mutant, and the results of osmotic treatments. Our work suggests that a developmental regulation of endosperm pressure is required to prevent a precocious reduction of seed growth rate induced by force-dependent seed coat stiffening.

Abstract

The estimation of metabolic fluxes in photosynthetic organisms represents an important challenge that has gained interest over the last decade with the development of 13C-Metabolic Flux Analysis at isotopically non-stationary steady-state. This approach requires a high level of accuracy for the measurement of Carbon Isotopologue Distribution in plant metabolites. But this accuracy has still not been evaluated at the isotopologue level for GC-MS, leading to uncertainties for the metabolic fluxes calculated based on these fragments. Here, we developed a workflow to validate the measurements of CIDs from plant metabolites with GC-MS by producing tailor-made E. coli standard extracts harboring a predictable binomial CID for some organic and amino acids. Overall, most of our TMS-derivatives mass fragments were validated with these standards and at natural isotope abundance in plant matrices. Then, we applied this validated MS method to investigate the light/dark regulation of plant TCA cycle by incorporating U-13C-pyruvate to Brassica napus leaf discs. We took advantage of pathway-specific isotopologues/isotopomers observed between two and six hours of labeling to show that the TCA cycle can operate in a cyclic manner under both light and dark conditions. Interestingly, this forward cyclic flux mode has a nearly four-fold higher contribution for pyruvate-to-citrate and pyruvate-to-malate fluxes than the phosphoenolpyruvate carboxylase (PEPc) flux reassimilating carbon derived from some mitochondrial enzymes. The contribution of stored citrate to the mitochondrial TCA cycle activity was also questioned based on dynamics of 13C-enrichment in citrate, glutamate and succinate and variations of citrate total amounts under light and dark conditions. Interestingly, there was a light-dependent 13C-incorporation into glycine and serine showing that decarboxylations from pyruvate dehydrogenase complex and TCA cycle enzymes were actively reassimilated and could represent up to 5% to net photosynthesis.

Abstract

International audience; Plants have developed fine-tuned cellular mechanisms to respond to a variety of intracellular and extracellular signals. These responses often necessitate the rearrangement of the plant cytoskeleton to modulate cell shape and/or to guide vesicle trafficking. At the cell periphery, both actin filaments and microtubules associate with the plasma membrane that acts as an integrator of the intrinsic and extrinsic environments. At this membrane, acidic phospholipids such as phosphatidic acid, and phosphoinositides contribute to the selection of peripheral proteins and thereby regulate the organization and dynamic of the actin and microtubules. After recognition of the importance of phosphatidic acid on cytoskeleton dynamics and rearrangement, it became apparent that the other lipids might play a specific role in shaping the cytoskeleton. This review focuses on the emerging role of the phosphatidylinositol 4,5bisphosphate for the regulation of the peripherical cytoskeleton during cellular processes such as cytokinesis, polar growth, biotic and abiotic responses.

Abstract

Aim : To investigate the geographic patterns and ecological correlates in the geographic distribution of the most common tree dispersal modes in Amazonia (endozoochory, synzoochory, anemochory and hydrochory). We examined if the proportional abundance of these dispersal modes could be explained by the availability of dispersal agents (disperser‐availability hypothesis) and/or the availability of resources for constructing zoochorous fruits (resource‐availability hypothesis). Time period : Tree‐inventory plots established between 1934 and 2019. Major taxa studied : Trees with a diameter at breast height (DBH) ≥ 9.55 cm. Location : Amazonia, here defined as the lowland rain forests of the Amazon River basin and the Guiana Shield. Methods : We assigned dispersal modes to a total of 5433 species and morphospecies within 1877 tree‐inventory plots across terra‐firme, seasonally flooded, and permanently flooded forests. We investigated geographic patterns in the proportional abundance of dispersal modes. We performed an abundance‐weighted mean pairwise distance (MPD) test and fit generalized linear models (GLMs) to explain the geographic distribution of dispersal modes. Results : Anemochory was significantly, positively associated with mean annual wind speed, and hydrochory was significantly higher in flooded forests. Dispersal modes did not consistently show significant associations with the availability of resources for constructing zoochorous fruits. A lower dissimilarity in dispersal modes, resulting from a higher dominance of endozoochory, occurred in terra‐firme forests (excluding podzols) compared to flooded forests. Main conclusions : The disperser‐availability hypothesis was well supported for abiotic dispersal modes (anemochory and hydrochory). The availability of resources for constructing zoochorous fruits seems an unlikely explanation for the distribution of dispersal modes in Amazonia. The association between frugivores and the proportional abundance of zoochory requires further research, as tree recruitment not only depends on dispersal vectors but also on conditions that favour or limit seedling recruitment across forest types. Abstract

Excess and limited trace metal contents in soils and plants can limit crop yields and pose a risk for the environment and human health. This mini-review reports on the emerging approach of combining X-ray absorption spectroscopy (XAS) with isotope analyses to improve the understanding of metal speciation and dynamics in soil-plant systems. In soils and their components, shifts in isotope compositions could be in some cases linked to changing metal speciation and thereby provide information on processes that control the phytoavailability of metals. In plants, the XAS-isotope approach has potential to improve the understanding of how complex interactions of metal speciation, redox processes, and membrane transport control metal uptake and translocation to edible plant parts. Yet, the XAS-isotope approach proves to be in a rather exploratory phase, and many research gaps remain. International audience; In general, cell growth is a mechanical process that balances internal and externalstresses allowing or limiting expansion. Plant cells are often compared to“hydraulicmachines”due to the similar concept of balanced counterforces between the primarywall stresses and the turgor pressure. Knowledge of the mechanics of plant root cellsis essential to understand how plant wall works and, therefore, how plants grow. Onemethod for qualitative and quantitative analysis of mechanical properties is atomicforce microscopy (AFM) that measures mechanical properties of living cells or tissuesunder conditions close to relevant physiological environments (Arnould et al., 2017,Kozlova et al., 2019, Milani et al., 2011, 2014, Peaucelle et al., 2012, 2011, Torode et al.,2018, Yakubov et al., 2016, Zdunek and Kurenda, 2013, Zhao et al., 2005). In our recentstudy, AFM has been used to investigate properties of root epidermal cells (Balzergueet al., 2017), which are the cells that form the outermost layer of the root. These epi-dermal cells were studied in situ on living seedlings and were therefore still influ-enced by the inherent multicellular properties of the root, which is an improvementover studies conducted on isolated living cells.This chapter explains and illustrates the different steps required to prepare anddeposit the seedling, and in particular the root tip, of the plant on a glass slideready for nanomechanical measurements by AFM

Therefore, we applied combined nanoscale secondary ion mass spectrometry (NanoSIMS) and synchrotron-based techniques, micro-X-ray fluorescence (µ-XRF) and micro-X-ray absorption near-edge structure (µ-XANES) to examine the speciation and localisation of Cr and the distribution of (micro-) nutrients in rice root tip and mature region. µ-XRF mapping revealed that the distribution of Cr and (micro-) nutrients varied between root regions. Cr K-edge XANES analysis at Cr hotspots attributed the dominant speciation of Cr in outer (epidermal and sub-epidermal) cell layers of the root tips and mature root to Cr(III)-FA (fulvic acid-like anions) (58–64%) and Cr(III)-Fh (amorphous ferrihydrite) (83–87%) complexes, respectively. The co-occurrence of a high proportion of Cr(III)-FA species and strong co-location signals of 52Cr16O and 13C14N in the mature root epidermis relative to the sub-epidermis indicated an association of Cr with active root surfaces, where the dissolution of IP and release of their associated Cr are likely subject to the mediation of organic anions. The results of NanoSIMS (poor 52Cr16O and 13C14N signals), dissolution (no IP dissolution) and µ-XANES (64% in sub-epidermis >58% in the epidermis for Cr(III)-FA species) analyses of root tips may be indicative of the possible re-uptake of Cr by this region. The results of this research work highlight the significance of IP and organic anions in rice root systems on the bioavailability and dynamics of heavy metals (e.g. Cr).

International audience; Chromatin remodeling is the dynamic modification of chromatin architecture by ATPdependent chromatin remodeling proteins complexes. These complexes recognize chromatin modifications such histone acetylation, control chromatin states between relaxation and condensation, and thus regulate gene activity. Histone lysine acylations mirror acetylation, but entail histone acylation of histone lysine residues with either short-chain fatty acids, such as malonylation (Kmal), propionylation (Kpr), butyrylation (Kbu), crotonylation (Kcr), β-hydroxybutyrylation (Kbhb), 2-hydroxyisobutyrylation (Khib), or dicarboxylic acids such as succinylation (Ksu). HAT and HDAC are involved in lysine acylation and deacylation. Post-translational modification (PTM) is the covalent modification of proteins following protein biosynthesis by ribosomes. As an example, phosphorylation is adding a phosphate to a hydroxyl group of an amino acid residue. Among other types of PTMs are ubiquitination, acetylation or Snitrosylation. PTMs usually respond to cell signaling and regulate the modified protein function. Stress granules are dense aggregations composed of proteins and RNAs that can appear in different cell compartments when the cell is under stress.

Abstract

International audience; Cross-kingdom interactions with plants were frequently related to microbial pathogens and herbivores. Yet, mutualistic interactions that involve multiple partners can confer cross-kingdom functional benefits, which have been understudied. Ant gardens (AGs) are recognized as one of the most sophisticated of all symbioses between ants and flowering plants, forming good models to study cross-kingdom interactions. The aim of this study was to examine whether ant-plant interactions can influence the community composition of root-associated fungi. We assessed whether two AG ant species, Camponotus femoratus and Neoponera goeldii, confer different physico-chemical properties to their nests, and affect root fungal community composition and fungal functional guilds in the bromeliad Aechmea mertensii. The diversity and community composition of root-associated fungi depended on ant species identity. The two ants had a contrasting influence on the structure and chemistry of the nest, and on the floristic diversity of the AGs. Multiple drivers may therefore determine the root-associated fungal communities. As the outcome of the ant-bromeliad interaction depends on the ant species, and because the plants are also involved in interactions with root-associated symbionts, this study provided evidence that ecologically relevant symbioses can be mediated by cross-kingdom interactions.

Abstract

International audience; Tree architecture shows large genotypic variability, but how this affects water-deficit responses is poorly understood. To assess the possibility of reaching ideotypes with adequate combinations of architectural and functional traits in the face of climate change, we combined high-throughput field phenotyping and genome-wide association studies (GWAS) on an apple tree (Malus domestica) core-collection. We used terrestrial light detection and ranging (T-LiDAR) scanning and airborne multispectral and thermal imagery to monitor tree architecture, canopy shape, light interception, vegetation indices and transpiration on 241 apple cultivars submitted to progressive field soil drying. GWAS was performed with single nucleotide polymorphism (SNP)-by-SNP and multi-SNP methods. Large phenotypic and genetic variability was observed for all traits examined within the collection, especially canopy surface temperature in both well-watered and water deficit conditions, suggesting control of water loss was largely genotype-dependent. Robust genomic associations revealed independent genetic control for the architectural and functional traits. Screening associated genomic regions revealed candidate genes involved in relevant pathways for each trait. We show that multiple allelic combinations exist for all studied traits within this collection. This opens promising avenues to jointly optimize tree architecture, light interception and water use in breeding strategies. Genotypes carrying favourable alleles depending on environmental scenarios and production objectives could thus be targeted.

Abstract

KAI2 are plant α/β hydrolase receptors, which perceive smoke-derived butenolide signals (karrikins) and putative endogenous, yet unidentified phytohormones (KAI2-ligands, KLs). The number of functional KAI2 receptors varies among plant species. It has been suggested that KAI2 gene duplication and sub-functionalization plays an adaptative role for diverse environments or ligand diversification by altering the receptor responsiveness to specific KLs. Legumes represent one of the largest families of flowering plants and contain many essential agronomic crops. Prior to legume diversification, KAI2 underwent duplication, resulting in KAI2A and KAI2B. Integrating plant genetics, ligand perception and enzymatic assays, and protein crystallography, we demonstrate that Pisum sativum KAI2A and KAI2B act as receptors and enzymes with divergent ligand stereoselectivity. KAI2B has a stronger affinity than KAI2A towards the KAI2-ligand (-)-GR24 and remarkably hydrolyses a broader range of substrates including the strigolactone-like isomer (+)-GR24. We determine the crystal structures of PsKAI2B in apo and butenolide-bound states. The biochemical and structural analyses as well as recorded mass spectra of KAI2s reveal a transient intermediate on the catalytic serine and a stable adduct on the catalytic histidine, further illuminating the role of KAI2 not only as receptors but also as bona fide enzymes. Our work uncovers the stereoselectivity of ligand perception and catalysis by evolutionarily diverged KAI2 receptors in KAR/KL signaling pathways and proposes adaptive sensitivity to KAR/KL and strigolactone phytohormones by KAI2B.

Abstract

International audience; As sessile organisms, plants have evolved sophisticated mechanisms of gene regulation to cope with changing environments. Among them, long non-coding RNAs (lncRNAs) are a class of RNAs regulating gene expression at both transcriptional and post-transcriptional levels. They are highly responsive to environmental cues or developmental processes and are generally involved in fine-tuning plant responses to these signals. Roots, in addition to anchoring the plant to the soil, allow it to absorb the major part of its mineral nutrients and water. Furthermore, roots directly sense environmental constraints such as mineral nutrient availability and abiotic or biotic stresses and dynamically adapt their growth and architecture. Here, we review the role of lncRNAs in the control of root growth and development. In particular, we highlight their action in fine-tuning primary root growth and the development of root lateral organs, such as lateral roots and symbiotic nodules. Lastly, we report their involvement in plant response to stresses and the regulation of nutrient assimilation and homeostasis, two processes leading to the modification of root architecture. LncRNAs could become interesting targets in plant breeding programs to subtly acclimate crops to coming environmental changes.

International audience; Publié en 1701, la version du Dictionnaire universel de Furetière révisée par l’émigré protestant Henri Basnage de Beauval contient de nombreuses mentions de la flore et de la faune brésiliennes, ce qui peut parait surprenant dans un dictionnaire de français. L’explication de cette singularité lexicographique se trouve dans le fait que Basnage a fait appel à un expert en histoire naturelle, le Dr. Régis, médecin huguenot réfugié à Amsterdam. Régis a créé des entrées très détaillées à partir de nombreuses sources en botanique et de récits des voyageurs de l’époque. L’approche qu’il propose dans ces entrées est véritablement encyclopédique. Dans ce texte, nous analysons les entrées concernant des arbres du Brésil afin de retrouver l’ensemble des textes sources employés par Régis. Nous comparons les entrées du Dictionnaire universel avec l’autre grand dictionnaire encyclopédique de l’époque, le Vocabulario portugais compilé par Bluteau (1712-1728). Les sources utilisées par Bluteau nous permettent quant à elles de mieux comprendre les descriptions de la flore brésilienne à une époque où la classification des plantes restait à faire et où la terminologie botanique était encore balbutiante.

Abstract

The shortage of reliable primary taxonomic data limits the description of biological taxa and the understanding of biodiver sity patterns and processes, complicating biogeographical, ecological, and evolutionary studies. This deficit creates a significant taxo nomic impediment to biodiversity research and conservation planning. The taxonomic impediment and the biodiversity crisis are widely recognized, highlighting the urgent need for reliable taxonomic data. Over the past decade, numerous countries worldwide have devoted considerable effort to Target 1 of the Global Strategy for Plant Conservation (GSPC), which called for the preparation of a working list of all known plant species by 2010 and an online world Flora by 2020. Brazil is a megadiverse country, home to more of the world’s known plant species than any other country. Despite that, Flora Brasiliensis, concluded in 1906, was the last comprehensive treatment of the Brazilian flora. The lack of accurate estimates of the number of species of algae, fungi, and plants occurring in Brazil contributes to the prevailing taxonomic impediment and delays progress towards the GSPC targets. Over the past 12 years, a legion of taxonomists motivated to meet Target 1 of the GSPC, worked together to gather and integrate knowledge on the algal, plant, and fungal diversity of Brazil. Overall, a team of about 980 taxonomists joined efforts in a highly collaborative project that used cybertaxonomy to prepare an updated Flora of Brazil, showing the power of scientific collaboration to reach ambitious goals. This paper presents an overview of the Brazilian Flora 2020 and provides taxonomic and spatial updates on the algae, fungi, and plants found in one of the world’s most biodiverse countries. We further identify collection gaps and summarize future goals that extend be yond 2020. Our results show that Brazil is home to 46,975 native species of algae, fungi, and plants, of which 19,669 are endemic to the country. The data compiled to date suggests that the Atlantic Rainforest might be the most diverse Brazilian domain for all plant groups except gymnosperms, which are most diverse in the Amazon. However, scientific knowledge of Brazilian diversity is still un equally distributed, with the Atlantic Rainforest and the Cerrado being the most intensively sampled and studied biomes in the coun try. In times of “scientific reductionism”, with botanical and mycological sciences suffering pervasive depreciation in recent decades, the first online Flora of Brazil 2020 significantly enhanced the quality and quantity of taxonomic data available for algae, fungi, and plants from Brazil. This project also made all the information freely available online, providing a firm foundation for future research and for the management, conservation, and sustainable use of the Brazilian funga and flora.
