Background: Tick-borne diseases cause economically significant losses to animal production globally, and anaplasmosis and theileriosis are associated with the greatest losses. However, the spread of the relevant pathogens in flocks of domesticated animals in southern Egypt is little understood. Accordingly, in this study, we aimed to determine the prevalences of Anaplasma ovis, Theileria ovis, and Theileria lestoquardi in southern Egyptian sheep and goats through blood tests, and to make a molecular characterization of the A. ovis detected in sheep targeting a specific gene. Results: We collected blood samples collected from 300 sheep and goats (n=150 /species) in Luxor Province in southern Egypt, and analyzed them for the presence of A. ovis, T. ovis and T. lestoquardi with screening by conventional and nested PCR targeting the msp4 and msp5, 18S rRNA, and merozoite surface protein genes. For A. ovis 140/300 samples (46.66%) were positive overall, with 90/150 (60%) and 50/150 (33.33%) positive samples in sheep and goats, respectively. Two major surface protein genes of A. ovis, msp4 and msp5, were sequenced using DNA extracted from sheep and goat blood samples, for phylogenetic analysis and genotyping. The msp4 gene sequence revealed no significant genetic diversity, to contrast to data on A. ovis strains from other countries. For T. lestoquardi, 8/150 (5.33%) samples were positive in sheep, but no samples were positive in goats (0%). For T. ovis, 32/150 (21.33%) samples were positive in sheep, but no samples were positive in goats (0%). Sequencing targeting the merozoite surface protein gene for T. lestoquardi and the small subunit ribosomal RNA gene for T. ovis revealed no significant genetic diversity in the study, another contrast to data on A. ovis strains from other countries. Conclusion: This study provides valuable data on phylogenetic and molecular classifications of A. ovis, T. ovis and T. lestoquardi found in southern Egyptian sheep and goats. It also represents the first report on detection and molecular characterization of T. lestoquardi in southern Egyptian sheep based on the specific merozoite surface protein gene, thus providing valuable data for molecular characterization of this pathogen in southern Egypt. Background: Gangba sheep as a famous breed of Tibetan sheep, its wool color is mainly white and black. Gangba wool is economically important as a high-quality raw material for Tibetan blankets and Tibetan serge. However, relatively few studies have been conducted on the wool color of Tibetan sheep. Results: To fill this research gap, this study conducted an in-depth analysis of two populations of Gangba sheep (black and white wool color) using whole genome resequencing to identify genetic variation associated with wool color. Utilizing PCA, Genetic Admixture, and N-J Tree analyses, the present study revealed a consistent genetic relationship and structure between black and white wool colored Gangba sheep populations, which is consistent with their breed history. Analysis of selection signatures using multiple methods (FST, π ratio, Tajima's D), 370 candidate genes were screened in the black wool group (GBB vs GBW); among them, MC1R, MLPH, SPIRE2, RAB17, SMARCA4, IRF4, CAV1, USP7, TP53, MYO6, MITF, MC2R, TET2, NF1, JAK1, GABRR1 genes are mainly associated with melanin synthesis, melanin delivery, and distribution. The enrichment results of the candidate genes identified 35 GO entries and 19 KEGG pathways associated with the formation of the black phenotype. 311 candidate genes were screened in the white wool group (GBW vs GBB); among them, REST, POU2F1, ADCY10, CCNB1, EP300, BRD4, GLI3, and SDHA genes were mainly associated with interfering with the differentiation of neural crest cells into melanocytes, affecting the proliferation of melanocytes, and inhibiting melanin synthesis. 31 GO entries and 22 KEGG pathways were associated with the formation of the white phenotype. Conclusions: This study provides important information for understanding the genetic mechanism of wool color in Gangba, and provides genetic knowledge for improving and optimizing the wool color of Tibetan sheep. Background: Bisphenol S (BPS) is a substitute for bisphenol A in plastic manufacturing and, as a potential endocrine disruptor, may alter the physiology of the oviduct, in which fertilization and early embryo development take place in mammals. The objective of this study was to assess the effect of a daily dietary exposure to BPS combined with a contrasted diet on the oviduct fluid proteome using an ovine model. Results: Eighty adult cyclic ewes were allotted to four groups (20/group): overfed (OF) consuming 50 µg/kg/day of BPS in their diet, underfed (UF) consuming 50 µg/kg/day of BPS, and non-exposed controls in each diet group. After three months, the mean body condition score, plasma levels of glucose and non-esterified fatty acids were significantly higher in OF than in UF females. The proteins in collected OF samples (50 µg) were analyzed by nanoliquid chromatography coupled with tandem mass spectrometry (nanoLC-MS/MS). Overall, 1563 proteins were identified, among which 848 were quantified. Principal component analysis of the data revealed a clear discrimination of samples according to the diet and a segregation between BPS-exposed and non-exposed females in overfed ewes. Hierarchical clustering of differentially abundant proteins (DAPs) identified two clusters of 101 and 78 DAPs according to the diet. Pairwise comparisons between groups revealed a stronger effect of BPS in OF than in UF females (70 vs. 24 DAPs) and a stronger effect of the diet in BPS-exposed than non-exposed females (56 vs. 36 DAPs). Functional analysis of DAPs showed an enrichment in metabolic processes, immune system, cell response to stress, and reproductive processes. Conclusions: This work highlights for the first time the important impact of BPS on the oviduct proteome, with larger effects seen in OF than UF females. Abomasal ulcers are recognized in sheep of all ages, but research regarding therapeutic interventions is limited. Proton Pump Inhibitors (PPIs) such as pantoprazole, are clinically used with a paucity of evidence regarding efficacy in mature sheep. Intravenous and subcutaneously administered pantoprazole dosed at 1.0 mg/kg in adult sheep will increase the pH of abomasal fluid compared to pre-administration baseline. The objectives were to assess the effect of pantoprazole, after single and multiple administration, on abomasal fluid pH in adult sheep. A third objective was to describe the pharmacokinetic parameters of IV and SC pantoprazole. Four clinically healthy adult Southdown ewes previously fitted with a gastrostomy tube in the abomasum were utilized in this randomized, 2-way cross-over trial. Ewes received pantoprazole (1.0 mg/kg) as a single and 3-dose regimen (every 24 hours). After a 10 day washout period the reverse treatment was applied. Blood for analysis of pantoprazole concentration was collected intermittently for 24 hours, and abomasal fluid pH was measured at intervals for a 96-hour period. The pH of the abomasal fluid was higher in pantoprazole treatments for up to 24 hours after dosing. Following intravenous administration of pantoprazole to study ewes, elimination half-life, volume of distribution, and clearance of pantoprazole was estimated as 3.29 hours, 0.35 L/kg, and 65.26 mL/hr/kg respectively. After subcutaneous dosing, maximum concentration, time to maximum concentration, half-life of elimination, and volume of distribution, were estimated as 2604 ng/mL, 0.55 hours, 2.48 hours, and 0.37 L/kg. In Sudan, resistance to benzimidazoles has been reported recently in cattle and goats from South Darfur. Herein, ivermectin efficacy against gastrointestinal nematodes (GINs) was evaluated in sheep and goats in three study areas in South Darfur. The faecal egg count reduction test (FECRT) was used to evaluate the efficacy of ivermectin in sheep and goats naturally infected with GINs in the region of Bulbul (goats: n = 106), Kass (goats: n = 40) and Nyala (Domaia (sheep: n = 47, goats: n = 77) and the University farm (goats: n = 52)), using different treatment plans, and the efficacy was evaluated 12 days after treatment. Ivermectin efficacy was also evaluated in goats experimentally infected using local Haemonchus contortus isolates from Kass and Nyala. Nematodes surviving ivermectin treatment in goats in Bulbul and Nyala were harvested and larvae used to infect worm-free male sheep (n = 6, ≤6 months old). Infected sheep were dosed subcutaneously with ivermectin every eight days with increasing doses from 0.2 mg/kg to 1.6 mg/kg bodyweight (bw). Reduced ivermectin efficacy was identified in sheep and goats in the four study locations. Using a paired statistic, the efficacy of a therapeutic dose in sheep was 75.6% (90% upper credible limit (UCrL): 77.5%), while twice the recommended dose led to a reduction of 92.6% (90% UCrL: 93.3%). In goats, the FECRs of a therapeutic dose were 72.9–95.3% (90% UCrL range: 73.6–95.7%) in Bulbul, Nyala Domaia, Nyala University farm and Kass. Twice the dose recommended for goats in Bulbul revealed a 90% UCrL of 87.6%. All post-treatment faecal cultures contained only Haemonchus spp. larvae. The experimental infection trials in sheep and goats supported our findings from field trials and calculated upper 90% CrL of below 98.9%. Genetic variations in the ovine ovulation rate, which are associated with the FecB mutation, provide useful models by which to explore the mechanisms regulating the development of mammalian antral follicles. In order to study the effects of the FecB mutation on cumulus cell differentiation, preovulatory follicles were aspirated and cumulus cells were isolated from three FecB genotypes (homozygous, heterozygous and wild type) of Small Tail Han (STH) sheep superstimulated with FSH. Transcriptome information from tens of thousands of cumulus cells was determined with the 10 × Genomics single-cell RNA-seq technology. Under the superovulation treatment, the observed number of preovulatory follicles in the ovaries of FecB carriers was still significantly higher than that in the wild-type (P < 0.05). The expression patterns of cumulus cells differed between FecB carriers and wild-type ewes. The screened cumulus cells could also be further divided into different cell clusters, and the differentiation states and fates of each group of cumulus cells also remained different, which supports the notion that heterogeneity in gene expression is prevalent in single cells. The oxidative phosphorylation pathway was significantly enriched in differentially expressed genes among the cell differentiation branch nodes of cumulus cells and among the differentially expressed genes of cumulus cells from the three genotypes. Combined with the important role of oxidative phosphorylation in the maturation of COCs, we suggest that the oxidative phosphorylation pathway of cumulus cells plays a crucial role in the differentiation process of cumulus cells and the mutation effect of the FecB gene. Body weight is an important economic trait for sheep meat production, and its genetic improvement is considered one of the main goals in the sheep breeding program. Identifying genomic regions that are associated with growth-related traits accelerates the process of animal breeding through marker-assisted selection, which leads to increased response to selection. In this study, we conducted a weighted single-step genome-wide association study (WssGWAS) to identify potential candidate genes for direct and maternal genetic effects associated with birth weight (BW) and weaning weight (WW) in Baluchi sheep. The data used in this research included 13,408 birth and 13,170 weaning records collected at Abbas-Abad Baluchi Sheep Breeding Station, Mashhad-Iran. Genotypic data of 94 lambs genotyped by Illumina 50K SNP BeadChip for 54,241 markers were used. The proportion of variance explained by genomic windows was calculated by summing the variance of SNPs within 1 megabase (Mb). The top 10 window genomic regions explaining the highest percentages of additive and maternal genetic variances were selected as candidate window genomic regions associated with body weights. Our findings showed that for BW, the top-ranked genomic regions (1 Mb windows) explained 4.30 and 4.92% of the direct additive and maternal genetic variances, respectively. The direct additive genetic variance explained by the genomic window regions varied from 0.31 on chromosome 1 to 0.59 on chromosome 8. The highest (0.84%) and lowest (0.32%) maternal genetic variances were explained by genomic windows on chromosome 10 and 17, respectively. For WW, the top 10 genomic regions explained 6.38 and 5.76% of the direct additive and maternal genetic variances, respectively. The highest and lowest contribution of direct additive genetic variances were 1.37% and 0.42%, respectively, both explained by genomic regions on chromosome 2. For maternal effects on WW, the highest (1.38%) and lowest (0.41%) genetic variances were explained by genomic windows on chromosome 2. Further investigation of these regions identified several possible candidate genes associated with body weight. Gene ontology analysis using the DAVID database identified several functional terms, such as translation repressor activity, nucleic acid binding, dehydroascorbic acid transporter activity, growth factor activity and SH2 domain binding. Background: The analysis of differentially expressed genes in muscle tissues of sheep at different ages is helpful to analyze the gene expression trends during muscle development. In this study, the longissimus dorsi muscle of pure breeding Hu sheep (H), Suffolk sheep and Hu sheep hybrid F1 generation (SH) and East Friesian and Hu sheep hybrid sheep (EHH) three strains of sheep born 2 days (B2) and 8 months (M8) was used as the research object, and transcriptome sequencing technology was used to identify the differentially expressed genes of sheep longissimus dorsi muscle in these two stages. Subsequently, GO and KEGG enrichment analysis were performed on the differential genes. Nine differentially expressed genes were randomly selected and their expression levels were verified by qRT-PCR. Results: The results showed that 842, 1301 and 1137 differentially expressed genes were identified in H group, SH group and EHH group, respectively. Among them, 191 differential genes were enriched in these three strains, including pre-folding protein subunit 6 (PFDN6), DnaJ heat shock protein family member A4 (DNAJA4), myosin heavy chain 8 (MYH8) and so on. GO and KEGG enrichment analysis was performed on 191 differentially expressed genes shared by the three strains to determine common biological pathways. The results showed that the differentially expressed genes were significantly enriched in ribosomes, unfolded protein binding, FoxO signaling pathway, glycolysis / glycogen generation and glutathione signaling pathway that regulate muscle protein synthesis and energy metabolism. The results of qRT-PCR were consistent with transcriptome sequencing, which proved that the sequencing results were reliable. Conclusions: Overall, this study revealed the important genes and signaling pathways related to sheep skeletal muscle development, and the result laid a foundation for further understanding the mechanism of sheep skeletal muscle development. Background: The Qinghai Tibetan sheep, a local breed renowned for its long hair, has experienced significant deterioration in wool characteristics due to the absence of systematic breeding practices. Therefore, it is imperative to investigate the molecular mechanisms underlying follicle development in order to genetically enhance wool-related traits and safeguard the sustainable utilization of valuable germplasm resources. However, our understanding of the regulatory roles played by coding and non-coding RNAs in hair follicle development remains largely elusive. Results: A total of 20,874 mRNAs, 25,831 circRNAs, 4087 lncRNAs, and 794 miRNAs were annotated. Among them, we identified 58 DE lncRNAs, 325 DE circRNAs, 924 DE mRNAs, and 228 DE miRNAs during the development of medullary primary hair follicle development. GO and KEGG functional enrichment analyses revealed that the JAK-STAT, TGF-β, Hedgehog, PPAR, cGMP-PKG signaling pathway play crucial roles in regulating fibroblast and epithelial development during skin and hair follicle induction. Furthermore, the interactive network analysis additionally identified several crucial mRNA, circRNA, and lncRNA molecules associated with the process of primary hair follicle development. Ultimately, by investigating DEmir's role in the ceRNA regulatory network mechanism, we identified 113 circRNA–miRNA pairs and 14 miRNA–mRNA pairs, including IGF2BP1-miR-23-x-novel-circ-01998-MSTRG.7111.3, DPT-miR-370-y-novel-circ-005802-MSTRG.14857.1 and TSPEAR-oar-miR-370-3p-novel-circ-005802- MSTRG.10527.1. Conclusions: Our study offers novel insights into the distinct expression patterns of various transcription types during hair follicle morphogenesis, establishing a solid foundation for unraveling the molecular mechanisms that drive hair development and providing a scientific basis for selectively breeding desirable wool-related traits in this specific breed. Background: ATPase activity and the antioxidant function of intestinal tissue can reflect intestinal cell metabolic activity and oxidative damage, which might be related to intestinal function. However, the specific influence of intestinal ATPase activity and antioxidant function on growth performance, feed conversion efficiency, and the intestinal microbiota in sheep remains unclear. Results: This study analyzed the correlation between ATPase activity and antioxidant function in the jejunum of 92 Hu sheep and their growth performance and feed conversion efficiency. Additionally, individuals with the highest (H group) and lowest (L group) jejunum MDA content and Na+ K+-ATPase activity were further screened, and the effects of jejunum ATPase activity and MDA content on the morphology and microbial community of sheep intestines were analyzed. There was a significant correlation between jejunum ATPase and SOD activity and the initial weight of Hu sheep (P < 0.01). The H-MDA group exhibited significantly higher average daily gain (ADG) from 0 to 80 days old and higher body weight (BW) after 80 days. ATPase and SOD activities, and MDA levels correlated significantly and positively with heart weight. The jejunum crypt depth and circular muscle thickness in the H-ATP group were significantly higher than in the L-ATP group, and the villus length, crypt depth, and longitudinal muscle thickness in the H-MDA group were significantly higher than in the L-MDA group (P < 0.01). High ATPase activity and MDA content significantly reduced the jejunum microbial diversity, as indicated by the Chao1 index and observed species, and affected the relative abundance of specific taxa. Among species, the relative abundance of Olsenella umbonata was significantly higher in the H-MDA group than in the L-MDA group (P < 0.05), while Methanobrevibacter ruminantium abundance was significantly lower than in the L-MDA group (P < 0.05). In vitro culture experiments confirmed that MDA promoted the proliferation of Olsenella umbonata. Thus, ATPase and SOD activities in the jejunum tissues of Hu sheep are predominantly influenced by congenital factors, and lambs with higher birth weights exhibit lower Na+ K+-ATPase, Ca2+ Mg2+-ATPase, and SOD activities. Conclusions: The ATPase activity and antioxidant performance of intestinal tissue are closely related to growth performance, heart development, and intestinal tissue morphology. High ATPase activity and MDA content reduced the microbial diversity of intestinal tissue and affect the relative abundance of specific taxa, representing a potential interaction between the host and its intestinal microbiota. The fatty acid-binding protein 4 (FABP4) plays a crucial role in the transportation and metabolism of fatty acids. It binds to long-chain fatty acids and facilitates their transport within cells. FABP4 is involved in lipid metabolism, insulin sensitivity, inflammation, and energy homeostasis. This study was conducted to assess the association between the FABP4 gene and growth traits in Karakul and Awassi sheep. A PCR-single strand conformation polymorphism (SSCP) protocol was utilized to assess the polymorphism of FABP4 PCR products with growth traits measured at birth, three, six, nine, and twelve-month intervals. One non-synonymous SNP was identified in the second exon, in which threonine was converted to aspartate in the 61st position in FABP4 (p.61Thr > Asp). This novel SNP showed significant associations with all growth traits measured at all age intervals. The results showed that lambs with the TT genotype exhibited higher growth traits than those with the GT and GG genotypes, respectively. The conducted prediction showed a clearly deleterious effect of p.61Thr > Asp on FABP4 structure, which was accompanied by reduced fatty acid binding efficiency. Owing to the predicted damaging effects caused by p.61Thr > Asp on FABP, lower levels of lipid transport and its consequent increased weight gain and other growth trait indices are expected. Therefore, a putative mechanism through which lambs with these genotypes exhibit higher growth traits is proposed. Meta-analysis is used to analyze the effects of sesame meal replacing soybean meal diet on growth performance and meat quality of mutton sheep, so as to provide basis for the feasibility of sesame meal as feed protein source for mutton sheep. Relevant literatures were collected in Chinese and English databases by keywords combination. According to the inclusion and exclusion criteria, eight literatures were included, including 138 lambs fed with sesame meal instead of soybean meal in the experimental group and 66 lambs fed with soybean meal in the control group. Average daily gain and feed conversion ratio were used as growth performance parameters. A total of 204 lambs were collected with pH, shear force and water holding capacity at 24 h after slaughter as the main parameters of mutton quality. Meta-analysis was performed by RevMan software. According to the forest map, the heterogeneity of the included literature was checked. The source of heterogeneity was determined by subgroup analysis. Finally, sensitivity analysis was per‐ formed to evaluate the stability and the publication bias of the literature was evaluated according to the funnel plot. The results showed that the replacement of soybean meal with sesame meal increased the average daily gain of mutton sheep significantly (P≤0.05) and reduced the feed-meat ratio significantly (P≤ 0.05), there was no effect on the pH，shear force (P>0.05), and water holding capacity (P≤0.05). Sub‐group analysis showed that when the replacement ratio of sesame meal was less than 20%, the average daily gain of mutton sheep was significantly increased (P<0.01) and the feed weight ratio was significantly reduced (P≤0.05). Feeding period more than 70 days could significantly increase the average daily gain (P≤0.05) and significantly reduced the feed conversion ratio (P≤0.05). Sensitivity analysis showed that the results of Meta-analysis were stable and reliable, and funnel plot showed that there was no publication bias for the collected literature. In summary, replacing soybean meal with sesame meal can significantly improve the growth performance of mutton sheep. When the proportion of sesame meal replacing soybean meal in the diet is less than 20% and the feeding-period is more than 70 days, the growth performance is the most significant, and there is no negative impact on meat quality. The objective of this study was to investigate the impact of probiotics on the growth performance of sheep. A meta-analysis model was established to assess the effects of probiotics on sheep's growth performance. Computer searches were conducted in CNKI, Wan Fang Date, Science Direct, and Web of Science databases from January 2008 to January 2023 for experimental studies examining the effects of probiotics on sheep's growth performance. Meta-analysis was performed using StataSE 17 (64- bit) and Review Manager 5.4.1 to analyze each effect index. A total of 40 Chinese and English studies on probiotics were collected, involving a sample size of 1 004 subjects. The addition of probiotics to feed has a significant improvement effect on average daily weight gain (P<0.05); Adding probiotics to feed can significantly reduce the feed to weight ratio of sheep (P<0.05). Further meta-analysis revealed that Clostridium and Lactobacillus exhibited greater advantages over Bacillus and yeast in enhancing sheep's growth performance. [ABSTRACT FROM AUTHOR]

This experiment was aimed to investigate the effects of feeding α-ketoglutarate on sperm quality and plasma biochemical indices of Hu sheep in autumn in Northeastern China. A total of 32 Hu sheep with well body condition and similar body weight were randomly divided into four groups, each Hu sheep was fed with 0, 5, 10 mg/d and 20 mg/d α-ketoglutarate separately. The experiment was con‐ ducted in Yichun, Heilongjiang from August to October, with an average temperature of 10.3 ℃ . The transition period was seven days, and the experimental period was 56 days. The jugular vein blood of Hu sheep was collected on the 28th, 42th, and 56th experiment days respectively to assessed plasma testosterone, melatonin, interleukin (2, 4, 6 and 10), adrenocorticotropic hormone, tumor necrosis factor-α, malondialdehyde content, total antioxidant capacity, and antioxidant enzymes activity. Hu sheep semen was collected to test sperm viability, sperm plasma membrane integrity, and sperm abnormality rate. The results showed that α-ketoglutarate had no significant effects on sperm abnormality rate, plasma testosterone, interleukin (2, 6 and 10) content in Hu sheep (P>0.05). Supplementary feeding for each Hu sheep 20 g/d of α-ketoglutarate significantly enhanced sperm viability, sperm plasma mem‐ brane integrity, plasma total antioxidant capacity, superoxide dismutase, glutathione peroxidase, and catalase activity in Hu sheep (P<0.05), and also significantly reduced plasma malondialdehyde, adrenocorticotropic hormone, interleukin-4, and tumor necrosis factor- α content in Hu sheep (P<0.05). Description of each supplementing with 20 g/d α-ketoglutarate can ameliorate the plasma reproductive hormone contents, antioxidant capacity, immune index contents and sperm quality of Hu sheep in autumn. Cost-effective methods are crucial in small-scale operations to assess the physicochemical properties of milk, identify potential clinical or subclinical mastitis, and test β-lactams in small ruminants. The aim was to evaluate the importance of physicochemical characteristics, somatic cells, and antibacterial substances as factors for monitoring the quality and safety of bulk tank milk from sheep and goats. Furthermore, we aimed to elucidate the influence of grazing capacity on the composition of sheep milk. The fat content of sheep milk (n=119) was found to be 7.7%, with a range of 5.3% to 9.9%. The average fat content of dairy goats (n=33) was found to be 4.9%, with a range of 2.8-6.7%. The results indicate that 2.8% of sheep milk samples and 7.8% of goat milk samples exhibited clear positive reactions when assessed using the California mastitis test. β-lactam antibiotics were present in 0.7% of sheep milk samples, but not in any of the goat milk samples using lateral flow strips. Toxoplasmosis is a cosmopolitan zoonotic infection that has significant effects on public health and causes economic losses in the livestock industry. The current study was designed to detect the Toxoplasma parasite in sheep blood samples and tissue samples of slaughtered sheep at the Sulaimani abattoir using molecular technique. A total of 300 peripheral sheep blood samples were randomly collected from 20 small ruminant flocks at 4 locations in the Sulaymaniyah province, northern Iraq. Also, 150 meat samples from thigh muscle, heart, and diaphragm were collected from slaughtered sheep. All collected blood samples were subjected to polymerase chain reaction (PCR) amplification to confirm Toxoplasma infection; in addition, meat samples were also analyzed for Toxoplasma by PCR following the digestion process. Of the 300 amplified blood samples, 94 were considered positive for Toxoplasma gondii, with a prevalence rate of 31.3%. The overall prevalence of Toxoplasma among meat samples was 34%. The diaphragm reported a higher infection rate (46%) than the heart (32%), while the femoral muscle reported an infection rate of 24%. Aged animals (older than 24 months) presented a higher infection rate (32.8%) than younger animals (28.9%). Sports-related muscle injuries account for 10–55% of all injuries, which is a growing concern, especially given the aging world population. To evaluate the process of skeletal muscle injury and compare it with muscle lesions observed in humans, we developed a novel in vivo model in sheep. In this model, muscle injury was induced by an ultrasound-guided transverse biopsy at the myotendinous junction of the medial gastrocnemius muscle. Twelve male sheep were examined at 3, 7, 14, and 28 days post-injury. Histological, immunofluorescence, and MRI analyses indicate that our sheep model could resemble key human clinicopathological features. Statistically significant differences (p < 0.05) were observed in collagen I, dMHC, α-SMA, and CD68 immunohistochemical detection when comparing injured and healthy muscles. The injured gastrocnemius muscle exhibited elevated levels of type I collagen, infiltration of CD68(+) macrophages, angiogenesis, and the emergence of newly regenerated dMHC(+) myofibers, which persisted for up to 4 weeks post-injury. Similarly, the progression of muscle injury in the sheep model was assessed using advanced clinical 3 T MRI and compared with MRI scans from human patients. The data indicate that the sheep muscle injury model presents features similar to those observed in human skeletal muscle injuries. The stems of Cynomorium songaricum are used in traditional Chinese medicine as a tonic and also used locally as a food material and livestock feed. It is known that some of the falvan‐3‐ol monomers and dimers that entered the milk of dairy sheep fed with C. songaricum stems are biotransformation products of the original flavan‐3‐ol polymers in C. songaricum stems. This study was performed to investigate the biotransformation process of the flavan‐3‐ols in dairy sheep and to evaluate the bioactivities. The results showed that procyanidin A2 and epicatechin could be released from the polymeric flavan‐3‐ols of C. songaricum through rumen microbial metabolism. On traumatic and lipopolysaccharide (LPS)‐induced inflammation models of Tg (mpx: EGFP) zebrafish larvae and LPS‐induced liver injury models of Tg (fabp10a: DsRed) zebrafish larvae, the milk from sheep fed with C. songaricum stems showed stronger anti‐inflammatory and hepatoprotective activities compared to blank milk. The absorbed chemical constituents of C. songaricum stems and the metabolites also exhibited anti‐inflammatory and hepatoprotective activities, with the dimeric flavan‐3‐ols being more effective than the monomers. The milk, the absorbed chemical constituents of C. songaricum stems, and the metabolites alleviated the increased level of reactive oxygen species induced by LPS in zebrafish larvae. Previous studies found that the BMP7 gene is differentially expressed in Hu sheep lamb skin of different pattern types and plays a key role in the proliferation of Hu sheep DPCs. However, the molecular mechanism of the differential expression of the BMP7 gene remains unclear. Here, we aim to investigate the cause of the BMP7 gene differentially expressed in Hu sheep lamb skin of different pattern types. Our findings demonstrate that DNA methylation modification can affect the expression of the BMP7 gene and can regulate the proliferation and cell cycle of Hu sheep DPCs. Previous studies have shown that the BMP7 gene is differentially expressed in Hu sheep lamb skin of different pattern types, and its expression level is significantly correlated with hair follicle indices of different pattern types, but the molecular mechanism of the differential expression of the BMP7 gene remains unclear. This study investigated the effect of DNA methylation on the transcriptional expression of BMP7. Firstly, we found that the mRNA expression of the BMP7 gene and the activity of the core promoter of the BMP7 gene were upregulated after 5-Aza-Deoxycytidine-induced demethylation treatment using qRT-PCR and double luciferase reporter assay. Then, we found that the proliferation of Hu sheep DPCs in vitro was promoted after 5-Aza-Deoxycytidine-induced demethylation treatment through qRT-PCR, CCK-8, and EdU assay, and that the overexpression of DNMT1 in DPCs induced the opposite effect. In addition, the results of the cell cycle assay reveal that the percentage of cells in the S phase was increased after 5-Aza-Deoxycytidine-induced demethylation treatment, and that the percentage of cells in the S phase was decreased after overexpression of DNMT1 in DPCs. However, the nutrient requirements of this breed are scarcely studied, especially regarding minerals. This study aims to evaluate the effects of dietary calcium levels on the growth performance, calcium utilization, and serum biochemical indexes of Yunnan semi-fine wool rams as well as to estimate their calcium requirement. Diets with calcium levels ranging from 0.73% to 0.89% can improve the growth performance and calcium utilization efficiency of rams, which could serve as baseline when formulating diets for Yunnan semi-fine wool rams. Calcium (Ca) is required for the growth and development of sheep, but the requirement of Yunnan semi-fine wool (YSW) rams remains uncovered. The current study aims to estimate the Ca requirement of growing YSW rams based on their growth performance, Ca utilization, and serum biochemical indexes. Forty-five YSW rams (10-month-olds) were randomly allocated to five dietary treatments with varying Ca levels of 0.50% (D1), 0.68% (D2), 0.73% (D3), 0.89% (D4), and 0.98% (D5). A higher value for average daily gain and a lower value for the feed conversion ratio were observed in the D3 group compared to the D5 group (p < 0.05). The dry matter intake amount changed quadratically with the increased Ca levels (p < 0.05). The levels of Ca intake, fecal Ca, and excreted Ca were significantly higher in the D5 group than those in the D1 group (p < 0.05). The apparent Ca digestibility rate and the Ca retention rate were significantly higher in the D4 group than in the D1 group (p < 0.05). The serum Ca concentration increased linearly with the incremental levels of dietary Ca (p < 0.05). The activity of alkaline phosphatase was significantly higher in the D1 group than in the D2 group (p < 0.05). The serum levels of hydroxyproline, osteocalcin, and calcitonin decreased from the D1 group to the D2 group, and then significantly ascended (p < 0.05) with the dietary Ca levels from the D3 group to the D5 group. The serum parathyroid hormone content was elevated from the D1 group to the D3 group and then decreased from the D4 group to the D5 group. The Lycium barbarum branches and leaves (LBL) are known to contain a range of active substances that have positive effects on animal immunity and antioxidation. This study aimed to examine how LBL impacts the growth and slaughter performance as well as rumen fermentation and microbiota in Hu sheep. A total of 50 male Hu sheep of indigenous origin, aged 3 months, were randomly divided into 5 groups of 10 sheep each. The groups were given different levels of LBL supplementation (0%, 3%, 6%, 9%, and 12%) to evaluate growth performance and nutrient apparent digestibility. Rumen fluid samples were collected for analysis of the fermentation parameters and rumen chyme was examined to study the rumen microbiota. The slaughter performance, meat quality, and organ index were evaluated at the conclusion of the experiment. The results showed that the final body weight and average daily gain of the LBL1 group were significantly higher than those of the CON group, LBL3 group, and LBL4 group (p < 0.05). The average dry matter intake of the LBL4 group was significantly lower than that of other experimental groups (p < 0.05). The apparent digestibility of CP in the LBL1 and LBL2 groups was higher than that in other experimental groups (p < 0.05). At the same time, the eye muscle area and grade-rule (GR) value of Hu sheep in the LBL1 group significantly increased and the quality of Hu sheep meat improved (p < 0.05). There was no significant difference in organ weight and organ index between the experimental groups (p > 0.05). The pH of the rumen fluid in the LBL1 group was significantly lower than that in the CON group (p < 0.05). There was no significant difference in the NH3-N content between the experimental groups (p > 0.05). Growing and mature Blackhead Ogaden sheep and Somali goats were used to evaluate the effect of salinity levels (up to 17 g/L of TDSs) of drinking water on water intake and loss, nutrient utilization, thermoregulation, and blood characteristics. Drinking water, total water intake, urine excretion, and total water loss were higher in both young and mature sheep and goats, while apparent dry matter digestibility decreased for high levels of TDSs compared with fresh water. There were no adverse effects of high salinity levels tested on feed intake, body weight change, different blood variables, and thermoregulatory mechanisms, reflecting considerable tolerance of relatively high levels of salinity in drinking water by these goat and sheep breeds. This experiment was conducted to evaluate the effects of drinking water salinity levels on water intake and loss, feed intake and digestion, body weight (BW), thermoregulation, and blood characteristics on growing and mature (18.8 ± 0.39 and 21.8 ± 0.40 kg BW, and 0.6–1 and 1.5–2 years of age, respectively) Blackhead Ogaden sheep and Somali goats. The animals were assigned to a 4 (water salinity) × 2 (sheep and goat species) × 2 (growing and mature animals) factorial arrangement for the 60-day experimental period and 10-day digestibility determination. Water treatments were fresh water (FRW) and low (SW-L), moderate (SW-M), and high (SW-H) levels of salinity (i.e., the addition of NaCl to obtain 10, 13.5, and 17 g of total dissolved salts (TDSs)/L, respectively). The salinity of drinking water did not affect feed intake, BW, thermoregulatory traits (respiration rate, rectal temperature, and heart rate), or blood parameters (p > 0.05); however, drinking water, total water intake, urine excretion, and total water loss increased (p < 0.01) while apparent dry matter digestibility decreased quadratically (p < 0.01) with increasing water salinity. Analysis of the interaction between water treatment and species showed that PCV (p = 0.059) and hemoglobin (p = 0.070) levels tended to be higher in sheep than in goats drinking FRW, and AST activities were greater (p = 0.036) in goats consuming SW-M than in sheep consuming water with the same salinity level. In conclusion, increasing the salinity level of drinking water by adding NaCl to up to 17 g/L of TDSs had no adverse effect on the water intake, feed intake, BW, and health status of growing and mature Blackhead Ogaden sheep and Somali goats. Fibroblast growth factor 5 (FGF5) plays key roles in promoting the transition from the anagen to catagen during the hair follicle cycle. The sheep serves as an excellent model for studying hair growth and is frequently utilized in various research processes related to human skin diseases. We used the CRISPR/Cas9 system to generate four FGF5-edited Dorper sheep and only low levels of FGF5 were detected in the edited sheep. The density of fine wool in GE sheep was markedly increased, and the proportion of fine wool with a diameter of 14.4–20.0 μm was significantly higher. The proliferation signal in the skin of gene-edited (GE) sheep was stronger than in wild-type (WT) sheep. FGF5 editing decreased cortisol concentration in the skin, further activated the activity of antioxidant enzymes such as Glutathione peroxidase (GSH-Px), and regulated the expression of Wnt signaling pathways containing Wnt agonists (Rspondins, Rspos) and antagonists (Notum) in hair regeneration. We suggest that FGF5 not only mediates the activation of antioxidant pathways by cortisol, which constitutes a highly coordinated microenvironment in hair follicle cells, but also influences key signals of the Wnt pathway to regulate secondary hair follicle (SHF) development. Overall, our findings here demonstrate that FGF5 plays a significant role in regulating SHF growth in sheep and potentially serves as a molecular marker of fine wool growth in sheep breeding. The study's aim was to determine the effect of using sheep tail fat (STF) on carboxymethyl‐lysine (CML) content and other properties of heat‐treated sucuk (HTS), a type of semi‐dry fermented sausage. Three mixtures were prepared: 100% beef fat (BF), 50% BF + 50% STF, and 100% STF. After production (fermentation, heat treatment, and drying), the samples were cooked at 180°C for 0, 1, 3, and 5 min to determine the effect of cooking time on CML, thiobarbituric acid reactive substance (TBARS), total sulfhydryl, and carbonyl contents. The lowest pH value (5.50) was observed in the presence of STF. The most oleic acid (46.02%) was observed in the 100% STF group. The score of taste and general acceptability decreased with increasing STF. Using STF had no significant effect on TBARS, total sulfhydryl, carbonyl, or CML content. These parameters were affected by cooking time. The mean CML content increased from 55.77 to 72.90 μg/g after 5 min of cooking. CML correlated more strongly with TBARS than sulfhydryl or carbonyl. Elevated cardiac troponin I (cTnI), a myocardial damage biomarker, has been reported in cord blood of neonates delivered vaginally or by cesarean section. Although the neonatal peak likely reflects the physiological adjustment to extrauterine life, a better understanding of serial prepartum changes is required to determine physiological causes of fetal cTnI release. We longitudinally sampled eight healthy lambs (20 days before spontaneous birth to 5 days postnatal), and from three fetuses receiving intravenous IGF-1. Samples were collected into heparin, and the plasma was stored at −80°C for later determination of high-sensitivity (hs) cTnI levels (BeckmanCoulter UniCel DxI Access IA; log transformed detection limit = 0.30, quantification limit = 0.78, 99th percentile = 1.78). Positive and negative control samples were drawn from an adult ewe during a terminal experiment (myocardial ischemia) and similarly assessed. hs-cTnI data were log transformed from ng/L. Log(hs-cTnI) was 1.47 ± 0.30 (means ± SD) at 20 days before birth and declined to 1.02 ± 0.65 in fetuses 12 ± 4 h before birth (P < 0.0001, R2 = 0.7869). Birth stimulated a delayed, transient peak in hs-cTnI (P = 0.0058). Newborn (43 ± 19 min postnatal) levels were 1.39 ± 0.40 (P = 0.0650 vs. fetus on day of birth) and 2.14 ± 0.63 the day after birth (P = 0.0331 vs. newborn). The second day after birth, levels declined to 1.65 ± 0.48 (P = 0.0238 vs. day 1). IGF-1 infusion increased hs-cTnI levels 25–50% over baseline (P = 0.0252, R2 = 0.9938). Baseline adult ewe log(hs-cTnI) was below the limit of detection; 3 h following coronary artery ligation, levels were 3.21. In conclusion, we newly report that fetal hs-cTnI levels decline concomitantly with reduced proliferation of cardiomyocytes toward term. The resistance of the Haemonchus contortus, a parasite causing severe anemia in ruminants, to commercial anthelmintics emphasizes the need for alternative bio-anthelmintics. This study aimed to identify the molecular, chemical composition, and in vitro anthelmintic activity of Sargassum duplicatum against H. contortus. Molecular identification employed the Chelex method, with DNA diversity and phylogeny assessed using maximum likelihood in IQ-tree. The analyzed chemical composition included proximate, total flavonoid, and total phenols. Adult worm motility test (AWMT) and egg hatch inhibiting test (EHIT) were conducted at concentrations of 2, 4, and 6 mg/mL of S. duplicatum ethanolic extract. AWMT followed a completely randomized factorial design (5 replications, each with 5 worms), while EHIT used a completely randomized one-way design (5 replications, each with H. contortus egg batches from 3 adult female worms). The cox1 gene sequence revealed the Sargassum sample as S. duplicatum (KP101270.1) with 99.83% similarity. The results indicated that the identified concentrations of S. duplicatum ethanolic extract, and the observation time significantly influenced motility and egg hatchability (p<0.05). Both factors exhibited a significant interaction (p<0.05). Concentrations of 4-6 mg/mL reduced worm motility by up to 50% (LD50) within 6-8 hours, while concentrations of 2-6 mg/mL inhibited egg hatchability by more than 87% during 24 hours of incubation. OBJECTIVE Foot-and-mouth disease (FMD) is a highly contagious disease in ruminants that causes significant economic losses worldwide. However, the prevalence of FMD virus (FMDV) in small ruminants has been overlooked in Pakistan. This study aimed to determine the prevalence of FMD in sheep and goats in the border area between Pakistan and Afghanistan. ANIMALS 800 sheep and goats belongs to age groups of 6 month to > 2 years. RESULTS For NSP, 340/800 (42.5%) of samples were positive, while SP analysis revealed that serotype O (44.5%) was the most common in sheep and goats, followed by Asia-1 (42%) and A (32%) serotypes. Sheep (39%; 95% CI, 34 to 44) had a higher (P < .05) prevalence of FMD than goats (46%; 95% CI, 41 to 51). Statistically significant (P < .05) differences in the seroprevalence of FMD-SP and FMD-NSPs were observed between various agencies (areas) of the study area. Risk factors such as age, sex, breed, season, flock size, body condition, animal movement, and production system were significantly (P < .05) associated with FMDV prevalence. CONCLUSIONS This study showed that FMD is highly prevalent in sheep and goats in the border areas of Pakistan and Afghanistan. Therefore, outbreak investigation teams should be arranged at the border level to develop FMD risk-based surveillance and control plans for small ruminants in order to mitigate infection risks. Q fever has become one of the most common causes of abortion in sheep herds, resulting in significant financial losses for Egyptian farmers. The goal of this study was to establish Coxiella burnetii seroprevalence and molecular detection in three sheep farms in Egypt. A total of 184 sheep of various ages and sexes had their serum samples and vaginal swabs taken. All serum samples were checked for the presence of C. burnetii antibodies by using an ELISA, while 50 vaginal swabs were randomly chosen for molecular detection of the C. burnetii IS1111 gene. The overall seroprevalence of Q fever in sheep was 37.5%, and it was more common in females (39.5%) than in males (8.3%). Antibodies to C. burnetii were found in more than half of pregnant ewes (47.7%). Antibodies to C. burnetii were found in 47.7% of pregnant ewes, compared to 31.4% in non-pregnant and abortive ewes (43.8%). Seroprevalence was observed to be significantly higher in sheep older than 3 years (71.2%). The presence of the C. burnetii IS1111 gene was found in 20% of the molecularly analysed vaginal swabs. Based on the abortion history and pregnancy state of the studied sheep, no statistical significance was identified, since the C. burnetii gene was present in equal percentages in both aborted and nonaborted ewes. A comparison of ELISA and PCR results for vaginal swab samples revealed a statistically non-significant link between the two procedures' results. Shiga toxin-producing Escherichia coli (STEC) strains have emerged as important foodborne pathogens of global public health concern, causing life-threatening diseases. Animals and their products have been documented as important reservoirs for STECs, especially E. coli O157. The aim of this study was to investigate STECs from healthy and diarrhoeic sheep in Sanandaj, Iran. In the current study, a total of 81 sheep faecal samples were taken (22 from diarrhoeic sheep and 59 from healthy sheep). E. coli and subsequently STEC strains was detected according to standard protocol (cultural characterisation and PCR assays). Finally, the frequency of Shiga-toxin producing gene(s) (stx1, stx2), intimin (eaeA) and enterohaemolysin (hlyA) was detected among STEC isolates using duplex PCR. Totally, 42 E. coli were isolated from 81 faecal samples (51.85% contamination). Of these, 34 isolates (80.9%) were identified as STEC patotypes based on Sorbitol-MacConkey (SMAC) medium culturing and also the presence of stx1 and/or stx2. Of these, only 3 isolates (7.1%) were identified as serotype O157:H7 based on PCR assay. In addition, the results showed that STEC bacteria were significantly more prevalent in diarrhoeic samples than in healthy samples (50% vs. 22.1%). Overall, the PCR results showed that 33 (97%), 12 (35.3%) and 8 (23.5%) isolates carried stx1, stx2 and hlyA, respectively. The eaeA gene was not found in any isolate. The number of isolated STEC bacteria in spring (10 isolates) and winter (14 isolates) were significantly higher than those in summer (4 isolates) and autumn (6 isolates) (P=0.039). Also, the number of STEC in diarrhoea samples was significantly higher compared to non-diarrhoea samples (P=0.032). In conclusion, the present study revealed high prevalence rate of STEC including serotype O157:H7 and non-O157:H7 in sheep faeces which highlights the importance of sheep as a reservoir of STEC pathogen in Sanandaj region. One of the most important challenges related to fresh meat products (especially sheep liver) is the short shelf-life of the product. For this purpose, synthetic preservative compounds are used to extend their shelf life, which have many side effects on human health. In this research, the effect of using an edible coating containing phenolic compounds of pomegranate peel cross-linked in the structure of aloe vera hydrogel was studied to extend the shelf life of sheep liver. Physicochemical (pH, peroxide index, total volatile nitrogen, thiobarbituric acid index and color parameters L* a *b * ) and sensory (aroma, color, texture and overall acceptability) properties of the product in storage days of 1, 3 and 7 days and in the storage temperature of +4oC was evaluated. The results indicated that, the lowest value of monohydroperoxide formation (i.e. peroxide index) was observed in test samples coated with aleo vera gel containing phenolic compound of pomegranate peel extract that corresponding to the lowest total volatile nitrogen. Generally, in accordance to the results of sensory evaluation and quality tests, using of aleo vera gel containing bioactive compounds of pomegranate peel extract is recommended for shelf life extending of sheep liver. Agrivoltaic systems combine solar energy and agriculture, promoting dual land use. Although grazing sheep in these systems is common, research on intentionally designed pastures to increase production is lacking. This study seeks to compare the herbage growth and lamb production in simple, diverse, and legume pastures in an agrivoltaic system over 2 years in Oregon. Annual herbage production in diverse pastures (4583 kg DM ha−1) was comparable to simple pastures (4457 kg DM ha−1), but higher than legume pastures (p <.05; 4023 kg DM ha−1) for the 2020/2021 growing season. In the 2021/2022 growing season, diverse pastures had a higher yield (4660 kg DM ha−1, p <.05) than simple pastures (4052 kg DM ha−1), which were similar to legume pastures (3955 kg DM ha−1). Liveweight gain (LWG; g head−1 d−1) tended (p =.06) to be higher from lambs in diverse and legume pastures compared to simple pastures in spring 2021. Liveweight production (LWP; kg ha−1 d−1) of legume pasture were greater (P < 0.05) than other pasture types, while simple pastures had the lowest LWP during this period. In summer 2021, LWG tended to be greater in lambs grazing diverse versus simple pastures. No difference was detected (all p >.05) in LWG or LWP at any grazing period in 2022. Overall, efficiency of pasture production was greatly reduced by lower light availability in all pasture types. Ladakh, one of the highest inhabited regions globally, hosts the unique Changthangi goat, renowned for producing Pashmina, the world's most luxurious natural fiber. In comparison, the fiber derived from Changthangi sheep is considered next only to Pashmina. This research endeavors to compare the skin transcriptome profiles of Changthangi goats and Changthangi sheep, aiming to discern the molecular determinants behind the recognition of Changthangi goats as the source of Pashmina. Drawing upon previously conducted studies, a collective of 225 genes correlated with fiber characteristics were extracted from the differentially expressed genes noticed between the two species (p-value of ≤ 0.05 and a log2 fold change of ≥ 1.5). These genes were analyzed using DAVID software to understand their biological functions and to identify enriched KEGG and Reactome pathways. The protein-protein interaction networks were constructed using Cytoscape, cytoHubba, and STRING to focus on key genes and infer their biological significance. Comparative transcriptome analysis revealed significantly higher expression of genes involved in signaling pathways like Wnt, MAPK, PI3K-Akt, Hedgehog, associated with fiber development and quality in Changthangi goats. These pathways play crucial roles in hair follicle (HF) formation, maintenance of epidermal stem cells, and fiber characteristics. Findings also highlight the enrichment of cell adhesion molecules and ECM-receptor interaction, emphasizing their roles in HF structure, growth, and signaling. The process of organizing and interpreting sensory information, known as perceptual processing, is a key feature of adaptation allowing animals to integrate, assess and cope with challenges in their environments. Here, we examined the durations of vigilance bouts and stares in bighorn sheep, Ovis canadensis , as an indicator of time spent vigilant and perceptual processing, respectively. We tested the extent to which vigilance was shaped by relevant ecological factors, focusing on predation risk. Specifically, we tested whether stare duration increased when sheep were younger, part of smaller groups, further from others, further from escape terrain or closer to obstructive cover, all of which are expected to affect individual predation risk. Perceptual processing in bighorn sheep conformed to the 1–4 s perceptual window found in other mammals, consistent with the idea of a phylogenetically conserved mechanism across mammalian species but was variable within individuals, indicating that perceptual processing is plastic to relevant ecological contexts. As expected, stare duration was much shorter than vigilance bout duration. Stare duration increased with decreasing distance to obstructive cover, which could indicate perceptual processing costs of searching for cryptic predators. We also found a significant difference in vigilance bout duration between the sexes: age was positively correlated with vigilance bout duration in males but not in females. Vigilance bout durations were longer when animals were further from escape terrain. The present study evaluated the effect of increasing levels of wet brewery residue (WBR) as a source of roughage on nutrient intake, performance and ingestive behaviour in lambs finished in confinement. Thirty-two uncastrated male lambs were used, weaned at 50 days of age. The treatments comprised increasing levels of WBR as roughage in the diet: 31, 44, 57 and 70%, on a dry-matter. The concentrate consisted of disintegrated corn, soybean meal, calcitic limestone and mineral salt. There was a quadratic effect (P ≤ 0.05) from the increasing levels of WBR on the intake of DM, OM, CP and TCH. The intake of EE, NDF and ADF increased linearly (P ≤ 0.01), while TDN intake showed a linear reduction (P < .0001) for increases in the level of WBR. There was a quadratic effect (P ≤ 0.010) on the average daily weight gain (ADG) from the addition of WBR to the diet. The time spent on feeding and rumination (P ≤ 0.05), expressed in minutes, showed a linear increase. Feeding rate for DM showed decreasing linear behaviour (P ≤ 0.006), while NDF rumination rate showed increasing linear behaviour (P ≤ 0.013) due to the increased levels of WBR in the diet. There was a linear increase in the time spent on each feeding activity (P ≤ 0.007) with the increase in WBR in the diet. Objective: This study aimed to investigate the effect of Codonopsis pilosula polysaccharide (CPP) on the motility, mitochondrial integrity, acrosome integrity rate, and antioxidant ability of sheep sperm after preservation at 4°C. Methods: Semen from healthy adult rams were collected and divided into four groups with separate addition of 0, 200, 400, and 1,000 mg/L CPP. Sperm motility was analyzed using the Computer-Assisted Semen Analysis software after preservation at 4°C for 24, 72, 120, and 168 h. Sperm acrosome integrity rate was analyzed by Giemsa staining at 24, 72, and 120 h, and mitochondrial membrane integrity was analyzed by Mito-Tracker Red CMXRos. The total antioxidant capacity (T-AOC) and malondialdehyde (MDA) content of spermatozoa were measured after 120 h of preservation. Results: The sperm viability and forward-moving sperm under 200 mg/L CPP were significantly higher than that in the control group at 72 h (61.28%±3.89% vs 52.83%± 0.70%, 51.53%±4.06% vs 42.84%±1.14%), and 168 h (47.21%±0.85% vs 41.43%±0.37%, 38.68%±0.87% vs 31.68%±0.89%). The percentage of fast-moving sperm (15.03%±1.10% vs 11.39%±1.03%) and slow-moving sperm (23.63%±0.76% vs 20.29%±1.11%) in the 200 mg/L group was significantly higher than control group at 168 h. The mitochondrial membrane integrity of the sperm in the group with 200 mg/L CPP was significantly higher than those in the control group after storage at 4°C for 120 h (74.76%±2.54% vs 65.67% ±4.51%, p<0.05). The acrosome integrity rate in the group with 200 mg/L (87.66%±1.26%) and 400 mg/L (84.00%±2.95%) was significantly higher than those in the control group (80.65%±0.16%) after storage for 24 h (p<0.05). CPP also increased T-AOC and decreased the MDA concentration after preservation at 4°C (p<0.05). Background: The skin is inhabited by a variety of micro‐organisms, with bacteria representing the predominant taxon of the skin microbiome. In sheep, the skin bacterial community of healthy animals has been addressed in few studies, only with culture‐based methods or sequencing of cloned amplicons. Objectives: The objectives of this study were to determine the sheep skin bacterial community composition by using metabarcoding for a detailed characterisation and to determine the effect of body part, breed and environment. Materials and Methods: Overall, 267 samples were taken from 89 adult female sheep, belonging to three different breeds and kept on nine different farms in Switzerland. From every individual, one sample each was taken from belly, left ear and left leg and metabarcoding of the 16S rRNA V3–V4 hypervariable region was performed. Results: The main phyla identified were Actinobacteriota, Firmicutes, Proteobacteria and Bacteriodota. The alpha diversity as determined by Shannon's diversity index was significantly different between sheep from different farms. Beta diversity analysis by principal coordinate analysis (PCoA) showed clustering of the samples by farm and body site, while breed had only a marginal influence. A sparse partial least squares discriminant analysis (sPLS‐DA) revealed seven main groups of operational taxonomic units (OTUs) of which groups of OTUs were specific for some farms. Conclusions and Clinical Relevance: These findings indicate that environment has a larger influence on skin microbial variability than breed, although the sampled breeds, the most abundant ones in Switzerland, are phenotypically similar. Changes in wool prices, labour costs and animal welfare have sparked interest in several new sheep traits. Shedding, the ability of moulting or seasonal wool loss in sheep, is one of them. Here, we aimed to develop breeding values for shedding using existing data from New Zealand flocks. We used breeder-collected shedding scores, 0 = 'no shedding on any part of the body' to 10 = 'wool shed fully from all the body', for Wiltshire and Wiltshire-composite lambs born between 2003 and 2022. After QC, 20,653 phenotypic records from 25,066 animals with pedigree information (393 sires and 7439 dams from 12 flocks) were included in the analyses. Genetic parameters and breeding values were obtained using a genetic additive model with pedigree data (BLUP model), assuming shedding scores were quantitative. The genetic model incorporated the following variables: date of birth, sex, birth and rearing rank, age of dam and contemporary group. We found a high heritability for lamb shedding, 0.54 (0.02), and estimated a low-to-moderate prediction accuracy of breeding values (ranging from 0.09 to 0.36) using forward-validation. Our results confirmed that shedding can be subject to selection and that pedigree-based breeding values for this trait can be a useful tool for breeders. [ABSTRACT FROM AUTHOR]

Removal of ewes from a flock before the end of their productive life, sometimes described as ewe wastage, can potentially lead to reduced flock productivity. While the main reasons for culling ewes are largely known, their relative importance to New Zealand farmers and farmers' rational behind their culling decisions are not. Therefore, this study involved semi-structured interviews with 38 sheep farmers from throughout New Zealand to explore their ewe culling decisions. Farmers consistently culled mixed-age and two-tooth ewes who failed to become pregnant or who had a vaginal prolapse. For other culling reasons, farmers' use of them and their rationale for doing so was diverse and varied between farmers based on ewe age-group, severity, season, perceived economic consequences and farmers' personal preference. With decreasing wool values, interest is increasing regarding shedding sheep. To investigate this, two long-term studies introducing Wiltshire genes into Romney flocks were initiated. Data from these two studies provide phenotypic relationships between a range of shedding scores at different ages. The data included shedding scores (on a 0–5 scale) repeated on lambs (∼5 months), hoggets (∼14–18 months) and two-tooths (∼27 months), and lamb fleece weights. Positive relationships between shedding scores on the same animals were observed. Lamb fleece weight was negatively correlated with all shedding scores. Lamb shedding score in February had a correlation of 0.54 (P < 0.001) with the February score as a hogget at Riverside farm. Scoring wool shedding is a laborious activity requiring individual animals to be scored in the shearing position. Therefore, lamb fleece weight was investigated for its relationship with shedding scores, as a potentially easier alternative. Lamb fleece weight had a greater correlation with February hogget shedding score than with the lamb shedding score (−0.76 vs −0.52, P < 0.001). This study indicated that February scores are an accurate predictor of future shedding phenotypes, and when used in conjunction with fleece weight, are a good predictor of phenotypes expressed at later ages. Measuring the methane to carbon dioxide ratio (CH4/CO2) from animals could be useful to predict CH4 yield when dry matter intake (DMI) cannot be measured. The objectives were to (1) evaluate the relationship of CH4/CO2 with CH4 yield; (2) compare the CH4/CO2 of 10–60 simulated spot-samples with the CH4/CO2 calculated with data from 48-h of respiration chamber measurements. The DMI and CH4 and CO2 emissions measured every 5–6 min in respiration chambers from a previous experiment with 70 lambs fed ryegrass substituted with 0%, 25%, 50%, 75% and 100% of forage rape were retrieved. Emission data were used to perform simulations of 10, 20, 30, 40, 50 and 60 randomly selected spot-samples per lamb. The CH4/CO2 of 20 or more spot-samples was useful to predict CH4 yield and detect differences between dietary treatments, while the precision of the prediction increased when increasing the number of spot-samples up to 50 samples per lamb. Twenty spot-samples were sufficient to obtain accurate CH4/CO2 estimates; however, variance decreased (precision improved) with an increasing number of spot-samples per lamb up to 50 spot-samples. The objective of this study was to estimate the phenotypic correlations between udder and teat morphology traits, milk production traits, and somatic cell score in dairy sheep from a flock in New Zealand. A total of 162 lactating ewes were scored for morphology traits during the milk production season of 2021–2022. The 130-d lactation yields of milk, fat, protein, and lactose were obtained with 2–4 test-days from each ewe and modelled using random regression with orthogonal polynomials. Age had a significant effect on all udder and teat traits. Coat-colour (genetic variety within the breed; white or black) was a significant effect for teat angle and udder separation. Udders that were above the hook were associated with lower milk, fat, protein, and lactose yields. Udders with well-defined separation between halves were associated with higher milk, protein, and lactose yields, and with lower somatic cell count. Well-attached udders were associated with lower fat yield and lower somatic cell score. Teats with a backwards angle were associated with lower milk and lactose yields. Further studies are needed to estimate heritability and genetic correlations between these traits to determine whether these traits should be implemented in breeding programs for dairy sheep in New Zealand. Background: Maedi-visna virus (MVV) is a lentivirus that infects monocyte/macrophage lineage cells in sheep, goats, and wild ruminants and causes pneumonia, mastitis, arthritis, and encephalitis. The immune response to MVV infection is complex, and a complete understanding of its infection and pathogenesis is lacking. This study investigated the in vivo transcriptomic patterns of lung tissues in sheep exposed to MVV using the RNA sequencing technology. Result: The results indicated that 2,739 genes were significantly differentially expressed, with 1,643 downregulated genes and 1,096 upregulated genes. Many variables that could be unique to MVV infections were discovered. Gene Ontology analysis revealed that a significant proportion of genes was enriched in terms directly related to the immune system and biological responses to viral infections. Kyoto Encyclopedia of Genes and Genomes analysis revealed that the most enriched pathways were related to virus-host cell interactions and inflammatory responses. Numerous immune-related genes, including those encoding several cytokines and interferon regulatory factors, were identified in the protein-protein interaction network of differentially expressed genes (DEGs). The expression of DEGs was evaluated using real-time polymerase chain reaction and western blot analysis. CXCL13, CXCL6, CXCL11, CCR1, CXCL8, CXCL9, CXCL10, TNFSF8, TNFRSF8, IL7R, IFN-γ, CCL2, and MMP9 were upregulated. Immunohistochemical analysis was performed to identify the types of immune cells that infiltrated MVV-infected tissues. B cells, CD4+ and CD8+ T cells, and macrophages were the most prevalent immune cells correlated with MVV infection in the lungs. Conclusion: Overall, the findings of this study provide a comprehensive understanding of the in vivo host response to MVV infection and offer new perspectives on the gene regulatory networks that underlie pathogenesis in natural hosts. Piroplasmosis and anaplasmosis are important zoonotic diseases of animal origin, which can be transmitted by ticks to infect animals. However, there is limited information on the infection of piroplasmosis and anaplasmosis in Tan sheep in Ningxia, China. In order to understand the prevalence of piroplasmosis and anaplasmosis in Tan sheep in Ningxia, 150 blood samples of Tan sheep from farms in five urban areas of Ningxia were detected by PCR, and some positive samples were sequenced to establish a phylogenetic tree. PCR revealed that the prevalence of Anaplasma spp. in Tan sheep in Ningxia was 28.0%. The overall prevalence of Piroplasmids was 33.3%, of which Theileria spp. and Babesia spp. were 20.7% and 12.7%, respectively. Among the samples of different ages, the highest detection rates of Piroplasmids and Anaplasma spp. were found in Tan sheep aged 20–30 months, and the detection rate of Theileria spp., Babesia spp., and Anaplasma spp. were 25.4%, 23.6%, and 36.3%, respectively. In this study, one Theileria species was identified as Theileria uilenbergi, two Babesia species were identified as Babesia molasi and Babesia ovis, and two Anaplasma species were identified as Anaplasma ovis and Anaplasma phagocytophilum, and the dominant species were A. ovis and T. uilenbergi. To the best of our knowledge, this is the first report detailing the infection rate and genotype of Piroplasmids and Anaplasma spp. in Tan sheep in Ningxia, China. The results of this study provide valuable data for understanding the epidemiology of tick-borne disease in Tan sheep in Ningxia, China, and lay a theoretical foundation for the prevention and control of piroplasmosis and anaplasmosis in Tan sheep in Ningxia, northwest China. Background: Gastrointestinal nematodes (GINs) have a major impact on sheep production, health, and welfare worldwide. Norway is no exception, but there are only a few studies on the prevalence of GINs in Norwegian sheep. The aim of this study was to investigate the current occurrence of the most important nematodes in sheep flocks in Norway. Faecal samples were collected from flocks in 2021/2022, mainly from three geographical regions in Norway, i.e., northern, eastern, and western. In each of 134 flocks included, individual samples from 10 lambs (autumn) were pooled. Third stage larvae (L3) were cultivated and harvested (Baermann method) from the pooled samples. The DNA was then extracted and further analysed using droplet digital PCR (ddPCR). This enables assessment of the proportions of the three most important nematode species/genera, i.e., H. contortus, T. circumcincta, and Trichostrongylus. The fractional abundance/relative proportion of each species/genus was assessed by performing duplex assays with universal strongyle and species/genus-specific primers and probe sets. In addition, the occurrence of Nematodirus eggs was assessed by standard faecal egg counts (i.e., McMaster method). Results: Of the 134 flocks sampled, 24 were from the northern region, 31 from eastern, and 71 from western Norway. In addition, some flocks from central (n = 7), and southern (n = 1) Norway were included. Among the sampled flocks, T. circumcincta occurred most commonly (94%), followed by H. contortus (60%) and Trichostrongylus (55%), and Nematodirus (51%). In general, mixed infections were observed, with 38% and 18% of flocks infected with three or all four genera, respectively. Conclusions: The results of this study indicate that GINs are widespread in Norway. Teladorsagia circumcincta seems to be present in most flocks based on this screening. Moreover, the results show that Nematodirus spp. infect lambs throughout the country, predominantly N. battus, and indicate that this nematode has become more abundant, which could lead to an increase in nematodirosis. Nutritive value of five Cenchrus ciliaris (buffel grass) genotypes (IG96-50, IG96-96, IG96-358, IG96-401 and IG96-403) weredetermined. Their sugar contents (>70 mg/g of dry matter) and ensiling potential were evaluated using in vitro batch culture and in vivo studies. Research indicated significant differences (P < 0.05) in the dry matter, organic matter, ether extract, neutral detergent fiber, acid detergent fiber, cellulose and lignin contents of the C. ciliaris genotypes tested. Genotypes also differed (P < 0.05) in total carbohydrates, structural carbohydrates, non-structural carbohydrates and protein fractions. Genotype IG96-96 had the lowest total digestible nutrients, digestible energy and metabolizable energy contents (377.2 g/kg, 6.95 and 5.71 MJ/kg of dry matter, respectively), and net energy values for lactation, maintenance and growth. After 45 days of ensiling, C. ciliaris silages differed (P < 0.05) in dry matter, pH, and lactic acid contents, and their values ranged between 255–339, 4.06–5.17 g/kg of dry matter and 10.8–28.0 g/kg of dry matter, respectively. Maize silage had higher (P < 0.05) Organic Matter (919.5g/kg of dry matter), ether extract (20.4g/kg of dry matter) and hemi-cellulose (272.3 g/kg of dry matter) than IG96-401 and IG96-96 silages. The total carbohydrates and non-structural carbohydrates of maize silage were higher (P < 0.05), while structural carbohydrates were comparable (P < 0.05) with C. ciliaris silages. Sheep on maize silage had (P < 0.05) higher metabolizable energy, lower crude protein, and digestible crude protein intake (g/kg of dry matter) than those on C. ciliaris silage diets. Nitrogen intake and urinary-N excretion were higher (P < 0.05) on genotype IG96-96 silage diet. Overall, this study suggested that certain C. ciliaris genotypes, notably IG96-401 and IG96-96, exhibited nutritive values comparable to maize silage in sheep studies, offering a promising avenue for future exploration as potential alternatives in diversified and sustainable livestock nutrition programs. Preclinical in vivo evaluation is an essential step in the progression of new cardiac devices into patient use, with studies predominantly performed in the domestic sheep model. A growing area of interest in cardiac device development is transcatheter mitral valve replacement (TMVR). Clinically, multimodal imaging, or computed tomography (CT) and echocardiography (echo) are used extensively to preoperatively determine mitral valve morphology prior to an intervention, but there is no description on how these modalities can be implemented to support preclinical studies. The purpose of this study is to apply clinically relevant CT and echo acquisition and assessment techniques to a large group of naive research sheep in order to analyze and report modality-related effects on mitral valve dimensional reference intervals in the sheep model. To this end, fifty-five adult domestic sheep underwent preoperative CT and echo exams and resultant images were analyzed using a landmark-based multiplanar measurement protocol and compiled into a master dataset for statistical analysis. We found moderate agreement between CT and echo-derived measurements of the mitral valve in sheep and propose the first clinically-relevant dimensional indices for the sheep's naive mitral valve which can be used to guide future studies evaluating novel TMVR devices. This study is the first of its kind in proposing a reproducible method for detailed examination of the mitral valve in the sheep model using clinically-relevant multimodal imaging. Mycoplasmal pneumonia in sheep and goats usually result covert but huge economic losses in the sheep and goat industry. The disease is prevalent in various countries in Africa and Asia. Clinical manifestations in affected animals include anorexia, fever, and respiratory symptoms such as dyspnea, polypnea, cough, and nasal discharge. Due to similarities with other respiratory infections, accurate diagnosis can be challenging, and isolating the causative organism is often problematic. However, the utilization of molecular techniques, such as PCR, allows for rapid and specific identification of pathogens. Thus, a goat infection model with Mycoplasma was established and the pathogen was tested using PCR. The results indicated that this approach could be effectively utilized for the rapid detection of mycoplasma in clinical settings. Additionally, the prevalence of contagious pleuropneumonia of sheep in Qinghai Province was further investigated through PCR analysis. A total of 340 nasal swabs were collected from 17 sheep farms in Qinghai province. Among these samples, 84 tested positive for Mycoplasma mycoides subsp. capri (Mmc) and 148 tested positive for Mycoplasma ovipneumoniae (Movi), resulting in positive rates of 24.71% and 43.53% respectively. Furthermore, our investigation revealed positive PCR results for nasal swabs, trachea, and lung samples obtained from sheep exhibiting symptoms suggestive of mycoplasma infection. Moreover, three distinct strains were isolated from these positive samples. Additionally, the inflammatory cytokines of peripheral blood mononuclear cells (PBMCs) were assessed using RT-PCR. The findings demonstrated a high susceptibility of sheep to Movi in Qinghai province, with infected sheep displaying an inflammatory response. Consequently, the outcomes of this study will furnish valuable epidemiological insights for the effective prevention and control of this disease within Qinghai Province. [ABSTRACT FROM AUTHOR]
