Planococcus Ferris (Hemiptera: Coccomorpha, Pseudococcidae) is a genus of mealybug containing 49 species worldwide. Many species of Planococcus are important pests of agricultural crops. Surveys have been set up in Bengkulu Province, Sumatra-Indonesia, on coffee plantations, in 2023. Planococcus bengkuluensis Zarkani & Kaydan sp. nov. was found in association with Coffea robusta Lindl. ex De Will. (Rubiaceae), described and illustrated. The new species has multilocular disc pores and oral collar tubular ducts on the posterior dorsum in limited numbers. It is another species of Planococcus possessing dorsal multilocular disc pores. This review discusses mealybug vectors of plant viruses, the economic losses they cause, mealybug species and their hosts. Among the numerous mealybug species, Planococcus species are the most effective vector of plant viruses, transmitting many Ampeloviruses. Diverse methods for the control and regulation of mealybugs are also discussed. Physical, cultural and biological control methods are labor-intensive but environmentally friendly compared to chemical methods. However, chlorpyrifos are one the active ingredients of insecticides effective against several mealybug species. Using plant products such as neem oil as a biocontrol method has been effective, similar to other insecticides. Notwithstanding, the biological method of controlling mealybugs is effectively slow but safe and highly recommended. The Anagyrus species have the highest success rate amongst other natural parasites of mealybugs. Also, farm sanitation and pruning as cultural methods help reduce mealybug populations.

Planococcus is a genus of Gram-positive bacteria known for potential industrial and agricultural applications. Here, we report the first draft genome sequence and phylogenomic analysis of a CRISPR-carrying, multidrug-resistant, novel candidate Planococcus sp. NCCP-2050T isolated from agricultural soil in Pakistan. The strain NCCP-2050T exhibited significant resistance to various classes of antibiotics, including fluoroquinolones (i.e., ciprofloxacin, levofloxacin, ofloxacin, moxifloxacin, and bacitracin), cephalosporins (cefotaxime, ceftazidime, cefoperazone), rifamycins (rifampicin), macrolides (erythromycin), and glycopeptides (vancomycin). Planococcus sp. NCCP-2050T consists of genome size of 3,463,905 bp, comprised of 3639 annotated genes, including 82 carbohydrate-active enzyme genes and 39 secondary metabolite genes. The genome also contained 80 antibiotic resistance, 162 virulence, and 305 pathogen–host interaction genes along with two CRISPR arrays. Based on phylogenomic analysis, digital DNA–DNA hybridization, and average nucleotide identity values (i.e., 35.4 and 88.5%, respectively) it was suggested that strain NCCP-2050T might represent a potential new species within the genus Planococcus. Astaxanthin and β-carotene are the most prominent carotenoids extensively used in pharmaceutics. Here, we present a halotolerant bacterium from Lake Wadi El-Natrun capable of producing astaxanthin and β-carotene analyzed by HPLC, ESI–MS, and infrared spectroscopy. The phenotypic and phylogenetic analyses classified the isolate as a novel strain of the genus Planococcus, for which the name Planococcus sp. Eg-Natrun is proposed. Carotenoid biosynthesis can exceptionally occur in a light-inducible or constitutive manner. The maximum carotenoid yields were 610 ± 13 µg/g (~ 38% β-carotene and ~ 21% astaxanthin) in a minimal medium with acetate and 1024 ± 53 µg/g dry cells in a rich marine medium. The carotenogenesis incentives (e.g., acetate) and disincentives (e.g., methomyl) were discussed. Moreover, we successfully isolated the CrtE gene, one of the astaxanthin biosynthesis genes, from the unknown genome using a consensus-based degenerate PCR approach. Abstract Background Extremophiles have attracted much attention in the last few decades, as they possess different properties by producing certain useful metabolites. However, the secondary metabolism of the extremophiles of Antarctic krill has received little attention. Results In this study, a new bacterial strain MSAK28401T from Antarctic krill was isolated and identified. The results of analysis on phenotypic, chemotaxonomic, and genomic characteristics showed that the strain MSAK28401T belongs to the genus Planococcus. Cells of this strain were coccoid (0.89–1.05 μm) and aerobic. The majority of the fatty acid content was C15:0 anteiso (37.67 ± 0.90%) followed by C16:1 ω7c alcohol (10.37 ± 1.22%) and C16:0 iso (9.36 ± 0.71%). The calculated average nucleotide identity and DNA–DNA hybridization values between the strain MSAK28401T and type strains P. citreus DSM 20549T and P. rifietoensis M8T were lower than 91 and 70%, respectively. The strain MSAK28401T (=KCTC 43283T and MCCC 1k05448T) represented a new member of the genus Planococcus and was named P. alpniumensis sp. nov. Moreover, genes involved in the degradation of aromatic compounds (e.g., salicylate, gentisate, and quinate) were found in the genome, implying that strain MSAK28401T has an aromatic compound as its potential metabolite. This work will help us understand the genomic characteristics and potential metabolic pathway of Planococcus from Antarctic krill. Conclusions This study reported the genomic information and phenotypic characteristics of the new strain P. alpniumensis MSAK28401T isolated from Antarctic krill, and provided the genome information of Planococcus strains for further studying the function roles in aromatic compound metabolism.

The biology of Horniolus fortunatus (Lewis, 1896) has been fragmentarily reported, but what the larvae and adults feed on and the immature stages are still unknown. Pseudo-nitzschia blooms often occur in coastal and open ocean environments, sometimes leading to the production of the neurotoxin domoic acid that can cause severe negative impacts to higher trophic levels. Increasing evidence suggests a close relationship between phytoplankton bloom and bacterial assemblages, however, the microbial composition and succession during a bloom process is unknown. Here, we investigate the bacterial assemblages before, during and after toxic and non-toxic Pseudo-nitzschia blooms to determine the patterns of bacterial succession in a natural bloom setting. Opportunistic sampling of bacterial community profiles were determined weekly at Santa Cruz Municipal Wharf by 454 pyrosequencing and analyzed together with domoic acid levels, phytoplankton community and biomass, nutrients and temperature. We asked if the bacterial communities are similar between bloom and non-bloom events and if domoic acid or the presence of toxic algal species acts as a driving force that can significantly structure phytoplankton-associated bacterial communities. We found that bacterial diversity generally increases when Pseudo-nitzschia numbers decline. Furthermore, bacterial diversity is higher when the low-DA producing P. fraudulenta dominates the algal bloom while bacterial diversity is lower when high-DA producing P. australis dominates the algal bloom, suggesting that the presence of algal toxin can structure bacterial community. We also found bloom-related succession patterns among associated bacterial groups; Gamma-proteobacteria, were dominant during low toxic P. fraudulenta blooms comprising mostly of Vibrio spp., which increased in relative abundance (6-65%) as the bloom progresses. On the other hand, Firmicutes bacteria comprising mostly of Planococcus spp. (12-86%) dominate during high toxic P. australis blooms, with the bacterial assemblage showing the same bloom-related successional patterns in three independent bloom events. Other environmental variables such as nitrate and phosphate and temperature appear to influence some low abundant bacterial groups as well. Our results suggest that phytoplankton-associated bacterial communities are strongly affected not just by phytoplankton bloom in general, but also by the type of algal species that dominates in the natural bloom.

Six novel bacterial strains, designated N016 T , N017, N022 T , N028, N056 T , and N064, were isolated from soil sampled on the Qinghai-Tibet Plateau. Cells were aerobic, orange or yellow, globular or rod-shaped, non-motile, non-spore-forming, Gram-stain-positive, catalase-positive and oxidase-negative. All the isolates were salt-tolerant and could grow in the range of 4-42 °C. Results of phylogenomic analyses based on 16S rRNA gene sequences and core genomic genes showed that the three pairs of strains (N016 T /N017, N022 T /N028, and N056 T /N064) were closely related to the members of the genus Planococcus , and clustered with Planococcus ruber , Planococcus glaciei , and Planococcus chinensis . The digital DNA-DNA hybridization and average nucleotide identity values of the six novel strains with other members of the genus Planococcus were within the ranges of 18.7-53 % and 70.58-93.49 %, respectively, all below the respective recommended thresholds of 70.0 % and 95-96 %. The genomic DNA G+C content of the six strains ranged from 43.5 to 46.0 mol%. The major fatty acids of the six strains were anteiso -C 15 : 0 , iso -C 14 : 0 , and C 16 : 1  ω 7 c alcohol. The predominant polar lipids of strains N016 T , N022 T , and N056 T were diphosphatidylglycerol, phosphatidylglycerol, and phosphatidylethanolamine. Menaquinones 7 and 8 were the respiratory quinones. The results of the above analyses indicated that the six strains represent three novel species of the genus Planococcus , for which the names Planococcus shenhongbingii sp. nov. (type strain N016 T =GDMCC 1.4062 T =JCM 36224 T ), Planococcus shixiaomingii sp. nov. (type strain N022 T =GDMCC 1.4063 T =JCM 36225 T ), and Planococcus liqunii sp. nov. (type strain N056 T =GDMCC 1.4064 T =JCM 36226 T ) are proposed.

A novel bacterial strain, APC 4016 T , was previously isolated from the skin of a snub-nosed spiny eel, Notacanthus chemnitzii , from a depth of 1000 m in the northern Atlantic Ocean. Cells were aerobic, cocci, motile, Gram-positive to Gram-variable staining, and gave rise to orange-pigmented colonies. Growth occurred at 4-40 °C (optimum, 25-28 °C), pH 5.5-12 (optimum, pH 7-7.5), and 0-12 % (w/v) NaCl (optimum, 1 %). 16S rRNA gene phylogenetic analysis confirmed that strain APC 4016 T belonged to the genus Planococcus and was most closely related to Planococcus okeanokoites IFO 12536 T (98.98 % 16S similarity). However, digital DNA-DNA hybridization and average nucleotide identity values between these two strains were low, at 20.1 and 83.8 %, respectively. Major (>10 %) cellular fatty acids of strain APC 4016 T were iso-C 14 : 0 , anteiso-C 15 : 0 and C 16 : 1 -ω-Alc. The predominant respiratory quinones were menaquinones 5, 6, 7 and 8. The major cellular polar lipids were phosphatidylglycerol, diphosphatidylglycerol and phosphatidylethanolamine, and three unknown lipids were also present. The draft genome sequence is 3.6 Mb with a G+C content of 45.25 mol%. This strain was previously shown to have antimicrobial activity and to encode bacteriocin and secondary metabolite biosynthetic gene clusters. Based on the phylogenetic analysis and its distinct phenotypic characteristics, strain APC 4016 T is deemed to represent a novel species of the genus Planococcus , and for which the name Planococcus notacanthi sp. nov. is proposed. The type strain of this species is APC 4016 T (=DSM 115753 T =NCIMB 15463 T ).

This study aims at exploiting salinity stress as an innovative, simple, and cheap method to enhance the production of antioxidant metabolites and enzymes from bacteria for potential application as functional additives to foods and pharmaceuticals. We investigated the physiological and biochemical responses of four bacterial isolates, which exhibited high tolerance to 20% NaCl (wt/vol), out of 27 bacterial strains isolated from Aushazia Lake, Qassim region, Saudi Arabia. The phylogenetic analysis of the 16S rRNA genes of these four isolates indicated that strains ST1 and ST2 belong to genus Bacillus, whereas strains ST3 and ST4 belong to genus Planococcus. Salinity stress differentially induced oxidative damage, where strains ST3 and ST4 showed increased lipid peroxidation, lipoxygenase, and xanthine oxidase levels. Consequently, high antioxidant contents were produced to control oxidative stress, particularly in ST3 and ST4. These two Planococcus strains showed increased glutathione cycle, phenols, flavonoids, antioxidant capacity, catalase, and/or superoxide dismutase (SOD). Interestingly, the production of glutathione by Planococcus strains was some thousand folds greater than by higher plants. On the other hand, the induction of antioxidants in ST1 and ST2 was restricted to phenols, flavonoids, peroxidase, glutaredoxin, and/or SOD. The hierarchical analysis also supported strain-specific responses. This is the first report that exploited salinity stress for promoting the production of antioxidants from bacterial isolates, which can be utilized as postbiotics for promising applications in foods and pharmaceuticals.

The marine environment represents a well-off and diverse group of microbes, which offers an enormous natural bioactive compounds of commercial importance. These natural products have expanded rigorous awareness due to their widespread stability and functionality under harsh environmental conditions. The genus Planococcus is a halophilic bacterium known for the production of diverse secondary metabolites such as 2-acetamido-2-deoxy-α-d-glucopyranosyl-(1, 2)-β-d-fructofuranose exhibiting stabilizing effect and methyl glucosyl-3,4-dehydro-apo-8-lycopenoate displaying antioxidant activity. The genus Planococcus is reported generally for hydrocarbon degradation in comparison with biosurfactant/bioemulsifier secretion. Although Planococcus was proposed in 1894, it seized long stretch (till 1970) to get accommodated under the genus Planococcus authentically. Large-scale biosurfactant production from Planococcus was reported in 2014 with partial characterization. For the first time in 2019, we documented genomic and functional analysis of Planococcus sp. along with the physico-chemical properties of its biosurfactant. In 2020, again we screened biosurfactant for pharmacological applications. The present review discusses the comprehensive genomic insights and physical properties of Planococcus-derived biosurfactant. Moreover, we also highlight the prospects and challenges in biosurfactant production from Planococcus sp. Among ∼102 reports on biosurfactant produced by marine bacteria, 43 were of glycolipid and 59 were non-glycolipid type. Under other biosurfactant type, they were identified as lipopeptide (20) like surfactin (5), glycolipoprotein/lipoprotein (12), and other non-glycolipid (22). Planococcus sp. generally produces glycolipid-type biosurfactant (4) and exopolysaccharides (2). The single report documented in the literature is on biosurfactant production (glycolipid +non glycolipid) by diverse marine microbes (39) suggesting their novelty and diversity for biosurfactant secretion.

Abstract The Planococcus sp. PAMC21323 is a yellow pigment-producing bacterium isolated from King George Island in Antarctica; it has a broad growth temperature range of 5–40 °C. Herein, we describe the complete genome sequence information of the genus Planococcus with its annotated sequence, genetic features for bioremediation, and oxidative stress capacity. The Planococcus sp. PAMC21323 possesses chromosomal DNA (3,196,500-bp) with plasmid DNA (3364-bp). The complete 3,199,864-bp of the genome consists of 3171 genes including 60 transfer RNAs and 24 ribosomal RNAs. Strain PAMC21323 encodes various genes associated with detoxification of heavy metal ions and aromatic hydrocarbons. Moreover, it is equipped with diverse stress response systems, which can be used to sense the internal and oxidative stresses caused by detoxification. This is the first report highlighting the genetic potential of Planococcus sp. PAMC21323 in bioremediation, suggesting application of this psychrotrophic strain in bioremediation in harsh environments.

The presence and spreading of Mexican black scale, Saissetia miranda (Cockerell and Parrott) on fig trees (Ficus carica) in the orchards of the middle of Iraq (south of Baghdad) during 2017 and 2018 was detected. The insect was identified in international scientific centers by specialists in scales insects. Also, it has several generations a year, including one generation during the springand then entered aestivation during the summer when the temperature exceeded 45o C and two generations during the autumn and then entered the winter when the temperature ranged between 12 -18 oC and entered the insect hibernation in the stage of eggs under complete insect cover. The population density of the insect in the autumn more than in the spring and the insect was found on the modern branches and leaves and fruits, but the numerical density on the modern branches more than on the rest of the plant parts. Female body shape S. miranda was convex with color of the outer shell gradually changes from white to dark brown then black, body width rate was 3.69 mm and length was 2.01 mm, average number of eggs per female 920 eggs under field conditions, pale yellow egg color gradually turns to reddish orange hatching crawling nymphs with the same color of eggs and four pairs of legs and then turn into static nymphs characterized by the shape of her body with the emergence of a plan similar to the letter H on the dorsal side. The appearance of the Mexican black scale with the Planococcus ficus was reported in Fig. Parasitic cases of parasites belonging to the families of Encyrtidae and Cecidomyidae were recorded. Planococcus sp. S5 belongs to strains that degrade naproxen, one of the most popular non-steroidal anti-inflammatory drugs. In the presented work, the immobilization of the S5 strain was carried out on the Loofah plant sponge, which improved the degradation efficiency, and kinetic studies indicated the abolition of the inhibition by the substrate observed in the free cell system. At the same time, after immobilization, evident changes were observed in the metabolic profile of the strain, which was related to the specific microenvironment of the carrier. The study also presents the naproxen degradation pathway in a system with the immobilized S5 strain for the first time. The analysis of intermediates formed during the decomposition of naproxen indicated that this decomposition occurs through naphthalene and salicylic acid. Furthermore, the degradation of naproxen via 3-hydroxybenzoic acid to gentisic acid is also possible. Moderate halophilic bacteria thrive in saline conditions and produce biosurfactant (BS) which facilitates the oil scavenging activity in the oil polluted surroundings. Production of such unusual bioactive molecules plays a vital role for their survival in an extreme and adverse environment. Current research deals with isolation of Planococcus maritimus strain SAMP MCC 3013 from Indian Arabian coastline sea water for BS production. The bacterium tolerated up to 2.7 M NaCl demonstrating osmotic stress bearable physiological systems. We used integrated approach to explore the genomic insight of the strain SAMP and displayed the presence of gene for BS biosynthesis. The genome analysis revealed this potential to be intrinsic to the strain. Preliminary screening techniques viz., surface tension (SFT), drop collapse (DC) and oil displacement (OD) showed SAMP MCC 3013 as a potent BS producer. BS reduced SFT of phosphate buffer saline (PBS) pH: 7.0 from 72 to 30 mN/m with a critical micelle concentration (CMC) value of 1.3 mg/mL. Subsequent investigation on chemical characterization, using thin layer chromatography (TLC), Fourier transform infrared spectroscopy (FT-IR), nuclear magnetic resonance (1H NMR and 13C NMR) and liquid chromatography mass spectrometry (LC-MS) revealed terpene containing BS having sugar, lipid moieties. The genomic sequence analysis of P. maritimus SAMP showed complete genes in the pathway for the synthesis of terpenoid. Probably terpenoid is the accountable backbone molecule for the BS production, but the later stages of terpenoid conversion to the BS could not be found. Moreover, it is important to highlight that till today; no single report documents the in-detailed physico-chemical characterization of BS from Planococcus sp. Based on genomic and functional properties, the term terpene containing BS is denoted for the surfactant produced by P. maritimus.

The vine mealybug (VMB), Planococcus ficus (Hemiptera: Pseudococcidae), is a key insect pest of vineyards. While pheromone-based mating disruption (MD) has been successfully tested against a wide range of insect pests, knowledge about its efficacy against key mealybug species, such as P. ficus, is scarce. In this study, a novel MD product, Isonet® PF, was evaluated by testing 300, 400, and 500 dispensers/ha at four study sites located in Northern (Veneto) and Southern (Sicily) Italy. Experiments were carried out over 2 years by monitoring the mealybug populations in wine grape and table grape vineyards managed with and without the application of MD. Pheromone dispensers were periodically collected during the grapevine-growing season, extracted, and analyzed by GC-MS, to determine their pheromone content and the release in mg/ha/day. The results showed that use of the MD dispenser Isonet® PF reduced the percentage of VMB-infested bunches and the number of VMB specimens per bunch compared with the untreated controls. This was recorded over 2 years at all experimental sites. Differences in the incidence of infested bunches among the three tested rates of Isonet® PF were not detected. Overall, the results presented here contribute to optimizing the sex pheromone dosage used in MD control programs against VMB allowing a reduction of broad-spectrum insecticides currently employed to manage this important pest. Moderate halophilic bacteria thrive in saline conditions and produce biosurfactant (BS) which facilitates the oil scavenging activity in the oil polluted surroundings. Production of such unusual bioactive molecules plays a vital role for their survival in an extreme and adverse environment. Current research deals with isolation of Planococcus maritimus strain SAMP MCC 3013 from Indian Arabian coastline sea water for BS production. The bacterium tolerated up to 2.7 M NaCl demonstrating osmotic stress bearable physiological systems. We used integrated approach to explore the genomic insight of the strain SAMP and displayed the presence of gene for BS biosynthesis. The genome analysis revealed this potential to be intrinsic to the strain. Preliminary screening techniques viz., surface tension (SFT), drop collapse (DC) and oil displacement (OD) showed SAMP MCC 3013 as a potent BS producer. BS reduced SFT of phosphate buffer saline (PBS) pH: 7.0 from 72 to 30 mN/m with a critical micelle concentration (CMC) value of 1.3 mg/mL. Subsequent investigation on chemical characterization, using thin layer chromatography (TLC), Fourier transform infrared spectroscopy (FT-IR), nuclear magnetic resonance (1H NMR and 13C NMR) and liquid chromatography mass spectrometry (LC-MS) revealed terpene containing BS having sugar, lipid moieties. The genomic sequence analysis of P. maritimus SAMP showed complete genes in the pathway for the synthesis of terpenoid. Probably terpenoid is the accountable backbone molecule for the BS production, but the later stages of terpenoid conversion to the BS could not be found. Moreover, it is important to highlight that till today; no single report documents the in-detailed physico-chemical characterization of BS from Planococcus sp. Abstract Pulp and paper industry is one of the high priority sectors that generates large amount of solid (sludge) and liquid (paper mill effluent) wastes. Making use of this huge quantity of paper mill sludge (PMS) in an environment friendly manner is challenging and cost involving. Similarly the present wastewater treatment strategies based on physical and chemical methods encounter certain serious setbacks including secondary pollution generation with expensive mode of abatement processes. The present study aims at a "waste treats waste" strategy for the bioremediation of paper mill effluent in fluidized bed reactor (FBR) using another waste of the same industry, PMS as immobilizing matrix for Planococcus sp. TRC1, a wastewater bacterial isolate. This study simultaneously explores this isolate for the yellowish orange pigment it produces (2.3 ± 0.2 mg/gm of dry bacterial biomass) and characterizes it as a member of the pharmacologically important carotenoid pigment family via UV-Vis spectrophotometry, TLC, FT-IR and 13C NMR. The antioxidant potential of this pigment was studied by DPPH assay (IC 50 = 33 ± 0.4 μg/ml) and H 2 O 2 assay (IC 50 = 147.4 ± 2.2 μg/ml). In FBR, the PMS immobilized bacteria showed removal of phenol, lignin, colour and COD from the effluent by 96%, 74%, 81% and 85% respectively after 60hr of treatment. The experimental data on immobilization fitted well with pseudo second-order (R2 = 0.955) and Freundlich adsorption isotherm (R2 = 0.996) models. The alterations in PMS before and after bacterial immobilization, as revealed by SEM and FT-IR, depicted the success of PMS as immobilization matrix. Phytotoxicity (90% seed germination) and mutagenicity studies confirmed that the treated effluent was substantially less toxic than its raw state. This study highlights a novel utilization possibility of PMS in an eco-friendly and economic way as immobilization matrix for Planococcus sp.TRC1 for paper pulp mill effluent treatment along with production of carotenoid pigment from this potential bacterial isolate as value added product. The vine mealybug (Planococcus ficus) is regarded as a key mealybug pest of grapevines in South Africa, with entomopathogenic nematodes (EPNs) being touted as a potential alternative to chemical control, although their vulnerability to above-ground environmental conditions has limited their use. In this study, tests were conducted to assess the ability of adjuvants to increase the deposition of S. yirgalemense on grapevine leaves. The combination of Nu-Film-P® and Zeba® resulted in significantly more infective juveniles (30) being deposited per 4 cm2 leaf disc than with either the control (14.8), or with Nu-Film-P® (23.3), although not significantly more than with Zeba® alone (29.2). The ability of S. yirgalemense, in conjunction with the two adjuvants, to control P. ficus on grapevine foliage was then assessed under controlled conditions. The application of S. yirgalemense with both Zeba® and Nu-Film-P® to P. ficus on leaf discs in a growth chamber resulted in 84% mortality, significantly greater than that attained by the application of S. yirgalemense with either Zeba® (47%), or water alone (26%). Similar results were observed in a glasshouse trial, in which the combination of S. yirgalemense, Zeba® and Nu-Film-P® offered 88% control of P. ficus on leaf discs hung on potted vines, compared with the control that was achieved with S. yirgalemense with either Zeba® (56%) or water alone (30%). This study demonstrates the potential of a combination of S. yirgalemense with adjuvants to give significant control of P. ficus on grapevine foliage, compared with using EPNs alone. One factor potentially affecting growth of wine spoilage microbes (e.g., Pediococcus spp.) is the presence of nutrients not consumed during alcoholic fermentation by Saccharomyces cerevisiae. To assess the impact of must nutrient supplementation on Pediococcus spp., synthetic grape musts containing low (55.2 mg N/L), medium (250 mg N/L), or high (530 mg N/L) concentrations of yeast assimilable nitrogen (YAN) were fermented by S. cerevisiae. Upon cessation of fermentative activity P. damnosus OW-2, P. inopinatus OW-8, P. parvulus WS-7C, WS-29A, OW-1, or P. pentosaceus ATCC 33316 were inoculated at 104 to 105 cfu/mL. With the exceptions of OW-1 and OW-2, none of the other species or strains grew in the synthetic wines unless yeast extract or peptone was added, suggesting the absence of an essential nutrient. Experiments were replicated using Cabernet Sauvignon musts containing low (66.9 mg N/L), medium (219 mg N/L), and high (438 mg N/L) YAN. In general, wines containing the greatest residual amino acid concentrations (high YAN) supported better growth of the aforementioned Pediococcus spp. However, low YAN wines containing negligible residual nitrogen achieved similar populations after a short period of initial inhibition, suggesting that 'excessive' nitrogen supplementation to musts does not have a large impact on growth of pediococci post alcoholic fermentation. The vine mealybug, Planococcus ficus Signoret (Hemiptera: Pseudococcidae), is a key insect pest of South African grapevine. The ability of mealybugs to avoid or resist the action of chemical pesticides has led to the investigation of alternative control methods, such as the application of entomopathogenic nematodes (EPNs). However, EPN application faces challenges, due to the maladaptation of EPN species to aboveground conditions. In this study, the ability of adjuvants to improve the control of P. ficus in grapevine using an indigenous nematode species, Steinernema yirgalemense, was investigated. A trial was performed to assess EPN survival on grapevine foliage, when applied in the morning (high humidity/ low temperature) compared with in the afternoon (high temperature / low humidity). In a semi-field trial, the combination of adjuvants Zeba® and Nu-Film-P® resulted in 66% control of P. ficus after 48 h, compared to the use of Zeba® alone (43%), and EPNs alone (28%). Additionally, lower concentrations of EPNs showed predictably lower mortality rates of P. ficus. Significantly, higher EPN survival was recorded at each time interval in the morning, compared with the corresponding interval in the afternoon. This study demonstrates the ability of S. yirgalemense, when applied with adjuvants and at an appropriate time of day, to control P. ficus on grapevine, under semi-field conditions. Planococcus ficus (Signoret) (Hemiptera: Pseudococcidae) constitutes a high impact pest in vineyards. Synthetic insecticides are inefficient to control the mealybug population and they affect human health and the environment. The insecticidal properties of eight volatile compounds against vine mealybugs and their grapevine leaf phytotoxicity in laboratory conditions were evaluated. 2-decanone, 3-decanone, α-methyl cinnamaldehyde and cinnamaldehyde produced a higher percentage of mealybug mortality in relation to control at a fumigant dose of 300 μL·L-1 air. The 3-octanone, cinnamyl chloride, 1-octen-3-ol and 3-octanol were not effective against P. ficus. Cinnamaldehyde and α-methyl cinnamaldehyde produced a low acetylcholinesterase inhibition (IC50= 2.67 μL·L-1 and 9.10 μL·L-1, respectively), whereas 2-decanone and 3-decanone did not cause enzyme inhibition. Cinnamaldehyde was not phytotoxic for grapevine leaves; therefore, this compound was selected for a contact application to improve its effectiveness, resulting in a LC50 of 394.36 μL·L-1 solution. BACKGROUND: Although mating disruption is increasingly being used to control the worldwide grapevine pest vine mealybug, Planococcus ficus (Signoret) (Hemiptera: Pseudococcidae), its mode of action remains unclear. A 3‐year field experiment was carried out to investigate the effects of mating disruption on the development and reproduction of the vine mealybug. The influence of mating disruption applied over consecutive years on the pest population density was also evaluated. RESULTS: The percentage of ovipositing females was significantly reduced in disrupted plots by 18.8–66.2%, depending on the year. The absence of ovipositing females in disrupted plots in the autumn of the second and third years indicates the effectiveness of mating disruption throughout the whole growing season. Mating disruption consistently prolonged the pre‐oviposition period in all years by up to 12.5 days. CONCLUSION: Our findings provide new insights into the mechanisms underlying the pheromone‐based control of the vine mealybug and indicate that the reduction of the pest population density is attributable to both a decrease and a delay in female mating. In addition, the population density of vine mealybugs under mating disruption decreased over the years, indicating that consecutive applications of this control strategy would significantly increase the effectiveness of control of the vine mealybug by mating disruption. © 2018 Society of Chemical Industry This study demonstrates the efficacy of mating disruption in reducing the population density of Planococcus ficus by both decreasing and delaying mating. Antarctic regions are characterized by low temperatures and strong UV radiation. This harsh environment is inhabited by psychrophilic and psychrotolerant organisms, which have developed several adaptive features. In this study, we analyzed two Antarctic bacterial strains, Planococcus sp. ANT_H30 and Rhodococcus sp. ANT_H53B. The physiological analysis of these strains revealed their potential to produce various biotechnologically valuable secondary metabolites, including surfactants, siderophores, and orange pigments. The genomic characterization of ANT_H30 and ANT_H53B allowed the identification of genes responsible for the production of carotenoids and the in silico reconstruction of the pigment biosynthesis pathways. The complex manual annotation of the bacterial genomes revealed the metabolic potential to degrade a wide variety of compounds, including xenobiotics and waste materials. Carotenoids produced by these bacteria were analyzed chromatographically, and we proved their activity as scavengers of free radicals. The quantity of crude carotenoid extracts produced at two temperatures using various media was also determined. This was a step toward the optimization of carotenoid production by Antarctic bacteria on a larger scale.

ABSTRACTMealybugs (Hemiptera: Pseudococcidae) are important pests of the grapevine Vitis spp. and are responsible for direct and indirect damage to production. The main mealybug species present in wine grapevine (Vitis vinifera L.) in Southern Brazil were identified and their incidence evaluated. Bunch-samples (n = 50) from 131 vineyards located in the Serra Gaúcha Region (RS) of Brazil were analyzed at harvest, and the occurrence of mealybugs in the roots was evaluated at the time of eradication of plants for replanting. Mealybugs were reared in laboratory until adulthood for species determination. The species Dysmicoccus brevipes (Cockerell, 1983), Dysmicoccus sp., Planococcus citri (Risso, 1813), Pl. minor(Maskell, 1897), Pseudococcus viburni (Signoret, 1875) and Pseudococcus sp. were identified in bunches. Dysmicoccus sp., D. umbambae Granara de Willink, 2009, Pl. citri and Pseudococcus sp. were found in the roots. Pl. citri (31.4%) and Dysmicoccus sp. (22.7%) were the most common species found in wine grape bunches in the Serra Gaúcha Region.

It is well known that dissolved salts can significantly lower the freezing point of water and thus extend habitability to subzero conditions. However, most investigations thus far have focused on sodium chloride as a solute. In this study, we report on the survivability of the bacterial strain Planococcus halocryophilus in sodium, magnesium, and calcium chloride or perchlorate solutions at temperatures ranging from +25°C to −30°C. In addition, we determined the survival rates of P. halocryophilus when subjected to multiple freeze/thaw cycles. We found that cells suspended in chloride-containing samples have markedly increased survival rates compared with those in perchlorate-containing samples. In both cases, the survival rates increase with lower temperatures; however, this effect is more pronounced in chloride-containing samples. Furthermore, we found that higher salt concentrations increase survival rates when cells are subjected to freeze/thaw cycles. Our findings have important implications not only for the habitability of cold environments on Earth but also for extraterrestrial environments such as that of Mars, where cold brines might exist in the subsurface and perhaps even appear temporarily at the surface such as at recurring slope lineae. In this study a survey for determining the parasitoid complex of Planococcus ficus (Signoret) (Hemiptera: Coccomorpha: Pseudococcidae) was conducted in Adana and Mersin Provinces, Turkey. Five primary, two primary/secondary, two secondary parasitoid species associated with P. ficus, were found. Intracellular subtilisin proteases (ISPs) have important roles in protein processing during the stationary phase in bacteria. Their unregulated protein degrading activity may have adverse effects inside a cell, but little is known about their regulatory mechanism. Until now, ISPs have mostly been described from Bacillus species, with structural data from a single homolog. Here, we study a marine ISP originating from a phylogenetically distinct genus, Planococcus sp. The enzyme was successfully overexpressed in E. coli, and is active in presence of calcium, which is thought to have a role in minor, but essential, structural rearrangements needed for catalytic activity. The ISP operates at alkaline pH and at moderate temperatures, and has a corresponding melting temperature around 60 °C. The high‐resolution 3‐dimensional structure reported here, represents an ISP with an intact catalytic triad albeit in a configuration with an inhibitory pro‐peptide bound. The pro‐peptide is removed in other homologs, but the removal of the pro‐peptide from the Planococcus sp. AW02J18 ISP appears to be different, and possibly involves several steps. A first processing step is described here as the removal of 2 immediate N‐terminal residues. Furthermore, the pro‐peptide contains a conserved LIPY/F‐motif, which was found to be involved in inhibition of the catalytic activity. [ABSTRACT FROM AUTHOR]

Abstract: BACKGROUND: Mating disruption (MD) employs high doses of a pest's synthetic sex pheromone in agricultural plots, to interfere with its reproduction. MD is assumed to have few behavioral effects on non‐target arthropods, because sex pheromones are highly species‐specific and non‐toxic. Nevertheless, some natural enemies use their host's sex pheromones as foraging cues, and thus may be attracted to MD plots. To investigate this hypothesis, we compared parasitoid and spider assemblages in paired plots in five Israeli vineyards during 2015. One plot was MD‐treated against two key pests, Lobesia botrana (Denis & Schiffermüller) and Planococcus ficus (Signoret). Both plots were insecticide‐treated as needed. Natural enemies were suction‐sampled and collected from pheromone‐baited monitoring traps. RESULTS: The total abundance, species diversity and species composition of most natural enemies were unaffected by MD. An important exception involved P. ficus' main parasitoid, Anagyrus sp. nr. pseudococci (Girault). Anagyrus sp. nr. pseudococci females were mainly captured in control plots, while male captures were low and not influenced by MD. Parasitized P. ficus occurred only in MD plots. CONCLUSION: Non‐target effects of MD involved mostly A. sp. nr. pseudococci females and hardly affected other natural enemies. Planococcus ficus, the vine mealybug, is the dominant mealybug pest of grapes in South Africa. To provide an alternative for chemical control, entomopathogenic nematodes (EPNs) were investigated as a biological control agent to be used in an integrated pest management system. Four local EPN species were screened for efficacy against female P. ficus, the most potent of which were Heterorhabditis noenieputensis, with 90% mortality, and Steinernema yirgalemense, with 63%. Since S. yirgalemense was previously shown to be highly effective against a range of pests, the effects of temperature and humidity on the infectivity of S. yirgalemense to female P. ficus were also assessed. The application of S. yirgalemense at 25°C yielded the highest mortality, of 72%, followed by 45% mortality at 30°C, and only 9% mortality when applied at 15°C. Steinernema yirgalemense performed best at 100% relative humidity (RH), resulting in 70% mortality. Decreasing RH levels resulted in decreased mortality (61% mortality at 80% RH, 40% mortality at 60% RH). As a soil-based organism, S. yirgalemense is most effective as a biocontrol agent of P. ficus under conditions of moderate temperature and high humidity. Here, extracellular synthesis of silver nanoparticles (AgNPs) was carried out by Planococcus plakortidis strain BGCC-51 isolated from dye industry effluent soil. The microbes were isolated, screened, and characterised by molecular analysis (accession number KX776160). The optimisation of synthesis of AgNPs to determine the optimum substrate level (1-5 mM), pH (5-9), and temperature (25-55°C) were further carried out. P. plakortidis strain BGCC-51 gave best yield of AgNPs at substrate concentration 5 mM, pH 8, and at 35°C. Synthesised AgNPs were characterised by scanning electron microscope and high-resolution transmission electron microscope. The size of synthesised AgNPs was in the range of 20-40 nm having spherical morphology. The AgNPs were found to show antimicrobial activity against bacteria such as Escherichia coli (ATCC 25922), Pseudomonas aeruginosa (ATCC 27853), and Staphylococcus aureus (ATCC 29213). A psychrophilic extracellular protease was isolated from the marine bacterium Planococcus sp. M7 found in the deep-sea mud of the Southern Indian Ocean. The mature protease is about 43 kDa and contains 389 amino acids. Sequence alignment revealed that the protease whose catalytic triad was comprised of Ser224, Lys249, and Gln253 contains a catalytic module belonging to the serralysin-type protease family 41, and displays 46.55% identity with the experimentally verified serine protease from Bacillus subtilis str. 168. The enzyme displayed an alkaline mesophilic preference with an optimum pH of 10.0 and an optimum temperature of 35 °C. The enzyme retained its activity from 5 to 35 °C and was resistant to repeated freezing and thawing, but was completely inactivated at 55 °C. Calcium ions had a protective effect against thermal denaturation. More than 60% of the maximum activity was retained at pH values in the range of 5.0-11.0. Almost no activity loss was detected after 1 h of incubation at pH 8.0-10.0 and 20 °C, or with 1.0% SDS. Most important, this protease also showed good stability and compatibility with the standard enzyme-free detergent, which indicates its special interest for applications in detergent industry. The effect of increasing mating delay on the reproductive performance and population growth rates of the vine mealybug, Planococcus ficus (Signoret) (Hemiptera: Pseudococcidae), was investigated under laboratory conditions. Virgin females were mated at 1, 3, 5, 7, 14, 21 and 28 days after emergence and reproductive and life table parameters were estimated. The pre-oviposition period (number of days between mating and the onset of oviposition) significantly decreased in females mated within 7 days, whereas females mated at older ages showed equivalent pre-oviposition periods (<4 days). The length of the oviposition period did not vary with increasing age at mating. Female longevity significantly increased in females mated at 21 and 28 days, as a consequence of a longer pre-reproductive period. Fecundity and sex ratio were not affected by the female age at mating, whereas fertility was higher in mealybugs mated at older ages. Additional field observations highlighted that young and old virgin females were equally able to attract males, as both mated on the same day as the field release. Mating delay also affected the life table parameters of P. ficus, as the intrinsic and finite rates of increase did not differ in mealybugs mated within 7 days and significantly decreased in females mated at older ages. The mean generation time and the population doubling time were overall similar in females mated at 1–7 days, and increased significantly in females that experienced longer mating delays. In terms of the mating disruption control of P. ficus, our findings indicate that this method would be effective if mating is delayed >7 days, as shorter delays in mating did not reduce the population growth rates. Determining the most likely source of an invasive pest species might help to improve their management by establishing efficient quarantine measures and heading the search of efficient biological control agents. Planococcus ficus is an invasive mealybug pest of vineyards in Argentina, California, Mexico, Peru and South Africa. This mealybug pest had a previously known geographic distribution spanning southern Europe, the Middle East, and parts of northern Africa. In North America, Pl. ficus was first discovered in the early 1990s and soon thereafter in Mexico. To determine the origin of invasive populations in North America, Pl. ficus from California and Mexico were compared with material throughout its presumptive native range in the Mediterranean region, as well as material collected from an older invasion in South Africa and recently invaded Argentina. From each sample location, genomic DNA was sequenced for the nuclear internal transcribed spacer one (ITS1) and the mitochondrial cytochrome c. oxidase one (CO1). Phylogenetic analyses of CO1, ITS1 and concatenated CO1 and ITS1 data-sets using Bayesian and neighbor-joining analysis support two major divisions: a European grouping (Europe, Tunisia, Turkey) and a Middle Eastern grouping (Israel and Egypt). The invasive populations in Argentina and South Africa align with the European group and the invasive populations in North America align with the Middle Eastern group, with one Israel sample aligning closely with the North American clade, suggesting that Israel was the origin of those populations. A novel type strain, Planococcus faecalis AJ003 T , isolated from the feces of Antarctic penguins, synthesizes a rare C30 carotenoid, glycosyl-4,4ʹ-diaponeurosporen-4ʹ-ol-4-oic acid. The complete genome of P. faecalis AJ003 T comprises a single circular chromosome (3,495,892 bp; 40.9% G + C content). Annotation analysis has revealed 3511 coding DNA sequences and 99 RNAs; seven genes associated with the MEP pathway and five genes involved in the carotenoid pathway have been identified. Plants have evolved a variety of defense traits against foliar herbivory, including the production of primary and secondary metabolites, the allocation of chemical compounds, and morphological plasticity. Using two vegetative generations of the invasive clonal species Alternanthera philoxeroides , we investigated the effects of maternal and offspring herbivory by Planococcus minor on the integrative defense strategy of plants. Herbivory severely inhibited leaf, stolon and root growth, as well as the production of primary metabolites (soluble sugars, starch, and total non-structural carbohydrates in stolons), and decreased average leaf area and specific leaf area of the second-generation A. philoxeroides . The changes in growth measures of the first-generation A. philoxeroides with herbivory were consistent with that of the second generation. By contrast, herbivory basically did not affect the concentration of non-structural carbohydrate compounds in the roots, and even increased the concentrations of N and total phenols in taproots. Furthermore, herbivory-induced maternal effects also reduced the growth of the second-generation plants. The results suggest that A. philoxeroides is capable of adapting to herbivory by P. minor , mainly via the allocation of available resources in belowground organs, and that the herbivory effect can persist across vegetative generations.
