The Mekong and Chao Phraya rivers harbor a species-rich freshwater mussel assemblage containing a large radiation of the Pseudodontini species. Members of the genera Bineurus Simpson 1900 and Thaiconcha Bolotov et al., 2020 primarily inhabit small and medium-sized tributaries of these rivers. Here, we present an integrative taxonomic review of these genus-level clades. We show that Bineurus contains four species: B. mouhotii (Lea, 1863), B. exilis (Morelet, 1866) stat. rev., B. anodontinum (Rochebrune, 1882) stat. rev., and B. loeiensis sp. nov. In its turn, Thaiconcha comprises three species: T. callifera (Martens, 1860), T. munelliptica sp. nov., and T. thaiensis sp. nov. Two species, Pseudodon ovalis Morlet, 1889 and P. thomsoni Morlet, 1884, are considered here as questionable taxa. These findings further highlight that Southeast Asia represents a significant evolutionary hotspot of freshwater mussels, which requires further international collaborative research and conservation efforts. Following the disappearance of the seven Buckingham Member (Unit 10) Cypraeidae from the Myakka Lagoon System (Myakka), there was a hiatus represented by two strata at Sarasota (Pinecrest Members 9 & 8) in the early Piacenzian Pliocene, 3.6 million years ago (“mya"), from which no Cypraeidae have been recorded. This was followed by the warmest and most tropical conditions of the entire Pliocene. The sea grass beds and mud flats, which were the preferred habitats of the Pinecrest Cypraeidae, returned in force. The depositional strata which resulted, Pinecrest (Unit 7) and its Kissimmee River Valley Equivalent, were very thick and probably represent a long period of uninterrupted deposition. Eight new species in six genera emerged in Myakka. In the Kissimmee River Valley (Kissimmee), six new species in three genera (one new) emerged as the first and earliest known Cypraeidae species from that area. This represented geographically separate, but parallel, evolutionary tracks. Pinecrest Members 6 & 5 represented a repeat of the conditions experienced during Pinecrest Members 9 & 8, resulting in the disappearance of all Pinecrest (Unit 7) Cypraeidae from Myakka and Kissimmee. Evolutionary and ecological hypotheses of the freshwater mussel subfamily Ambleminae are intensely geographically biased—a consequence of the complete exclusion of Mesoamerican taxa in phylogenetic reconstructions of the clade. We set out to integrate a portion of the Mesoamerican freshwater mussel assemblage into existing hypotheses of amblemine classification and evolution by generating a molecular phylogeny that includes four previously unsampled Mesoamerican genera and nine species endemic to that region. Given the traditionally hypothesized affinity to Nearctic mussels and the understanding that classification should reflect common ancestry, we predicted that (a) Mesoamerican genera would be recovered as members of the recognized tribes of the Ambleminae, and (b) genera would be supported as monophyletic. The mutilocus phylogeny (COI + 28S + 16S) reported herein does not fully support either of those hypotheses. Neither Cyrtonaias nor Psorula were supported as monophyletic and we predict several other Mesoamerica genera are also non‐monophyletic. The reconstructed phylogeny recovered four independent lineages of Mesoamerican freshwater mussels and these clades are distributed across the phylogeny of the Ambleminae, including the tribe Quadrulini (Megalonaias), Lampsilini (two lineages: Cyrtonaias explicata/Sphenonaias microdon, and Pachynaias), and a previously unrecognized, exclusively Mesoamerican and Rio Grande clade consisting of the genera Psoronaias, Psorula and Popenaias. The latter clade possesses several morphological characteristics that distinguish it from its sister taxon, tribe Lampsilini, and we recognize this newly identified Mesoamerican clade as a fifth tribe of the Ambleminae attributable to the Popenaiadini Heard & Guckert, 1970. Mitochondrial genomes have proved to be a powerful tool in resolving phylogenetic relationship. In order to understand the mitogenome characteristics and phylogenetic position of the genus Dosinia, we sequenced the complete mitochondrial genomes of Dosinia altior and Dosinia troscheli (Bivalvia: Veneridae), compared them with that of Dosinia japonica and established a phylogenetic tree for Veneridae. The mitogenomes of D. altior (17,536 bp) and D. troscheli (17,229 bp) are the two smallest in Veneridae, which include 13 protein-coding genes, 2 ribosomal RNA genes, 22 tRNA genes, and non-coding regions. The mitogenomes of the Dosinia species are similar in size, gene content, AT content, AT- and GC- skews, and gene arrangement. The phylogenetic relationships of family Veneridae were established based on 12 concatenated protein-coding genes using maximum likelihood and Bayesian analyses, which supported that Dosininae and Meretricinae have a closer relationship, with Tapetinae being the sister taxon. The information obtained in this study will contribute to further understanding of the molecular features of bivalve mitogenomes and the evolutionary history of the genus Dosinia. Aglajidae is a family of predominantly colourful shallow water marine slugs widely distributed in coral, rocky and sandy habitats of tropical and temperate waters worldwide. The monophyly of the group is supported by morphological traits, but recent molecular phylogenetic studies questioned not only the monophyletic status of the family but also the definition of its traditional genera and relationships. Therefore, in order to test the monophyly of the family and of its genera and to elucidate internal sister relationships, we here present a new multilocus phylogenetic study based on Bayesian inference and maximum likelihood analyses of the largest ever assembled taxon and gene data sets of Aglajidae including 412 specimens from 63 species (74% of the global diversity) with representatives of type species of all genera and 835 DNA sequences of the gene markers COI, 16S rRNA, 28S rRNA and Histone-3. The genera Aglaja, Chelidonura, Odontoglaja and Philinopsis were confirmed as not monophyletic. Fifteen evolutionary lineages of generic status were recovered and seven are here described as new, namely Biuve, Camachoaglaja, Mannesia, Mariaglaja, Niparaya, Spinophallus and Tubulophilinopsis. The valid taxonomic status of Spinoaglaja is confirmed and Migaya is considered to be a junior synonym of Nakamigawaia. Our results support the hypothesis that events of radula loss or gain have happened independently at least twice in the evolutionary history of Aglajidae, but lack of node support in the deeper parts of our tree obscured inference on the sister relationships of genera. Recent research efforts have significantly advanced our knowledge on Asian freshwater mussel (Bivalvia: Unionida) diversity and distribution. Here we provide a modern consensus of the diversity, biogeography and conservation of Unionida in the region comprising East and Southeast Asia (excluding Wallacea) and Asian Russia. A data review confirmed the presence of 228 native and 3 non-native Unionida (98% Unionidae, 2% Margaritiferidae), rendering the region a global hotspot of freshwater mussel diversity. Species richness was highest in China (particularly Yangtze basin) in absolute numbers and Cambodia when correcting for country area, and decreased gradually towards the south and steeply towards the north and east. Six of the seven unionid subfamilies are native to the region, with species richness peaking in Southeast Asia for Rectidentinae, Gonideinae, Parreysiinae and Modellnaiinae, China for Anodontinae and Unioninae, and Asian Russia for Margaritiferidae. Conservation status and data collected after 1980 were not available for 61 and 24% of species, respectively. Dams, deforestation and pollution are likely the major threats to mussels in the region, though data in this respect are scarce. A group of limpets,Cellana karachiensis, exhibiting phenotypic plasticity were examined from Gujarat coastline India, using molecular phylogeny. Previous examination of the COI genes established the presence of three different haplotypes X, Y and Z, while present study showed three more haplotypes X1, X2and Z1. Thus, a total of six COI gene haplotypes, having 99.23% to 99.85% sequence similarity, were observed with variations at six sites. Bayesian phylogenetic analysis shows divergence of lineages X–Y, X1–X2and Z–Z1. Careful observation of nucleotide alterations showed a nonrandom mutation with more A↔G and C↔T transitions between closely related species of the genusCellana.A stretch of 17 base pair, within COI region, was marked as region with high degree of variability between species ofCellana. Results suggest that this could be the beginning of speciation, with partial or complete reproductive barrier or these are already distinct species in different stages of evolution. The large, burrowing bivalve Laternula elliptica is an abundant component of shallow-water soft-substrate communities around Antarctica but its congeners are temperate and tropical in distribution and their phylogenetic relationships are obscure. A new molecular analysis of Laternulidae species shows that there are two distinct clades, one of Exolaternula species, E. spengleri and E. liautaudi, possessing a ligamental lithodesma and a larger clade of species lacking the lithodesma. Of the latter, Laternula elliptica is a sister taxon to temperate and tropical species, including those that live around the coasts of Australia from Tasmania to Darwin. It is suggested that L. elliptica was left isolated around Antarctica following the opening of the Tasman Gateway and initiation of the Circum- Antarctic Current as Australia drifted northwards following the final breakup of Gondwana. A further scenario is that as Australia moved closer to Asia, species spread into tropical habitats and more widely to the Red Sea and Japan. Exolaternula species have a likely Tethyan origin and the present-day range is from the Arabian Gulf, around southern Asia and as far north as southern Russia. [ABSTRACT FROM AUTHOR]

The genus Candidula (Geomitridae), consisting of 28 species in Western Europe as currently described, has a disjunct distribution in the Iberian Peninsula, Italy, the Balkans, the Aegean Islands, and one species on the Canary Islands. Although the genus is seemingly well defined by characters of the reproductive system, the relationships within the genus are still unclear and some authors have indicated a possible subgeneric division based on the internal morphology of the dart sac. Despite substantial phylogenetic incongruence, we present a well-resolved molecular phylogeny of Candidula based on two mitochondrial genes ( COI and 16S rRNA), the nuclear rDNA region (5.8S rNRA + ITS2 + 28S rRNA) and seven additional nuclear DNA regions developed specifically for this genus (60SL13, 60SL17, 60SL7, RPL14, 40SS6, 60SL9, 60SL13a), in total 5595 bp. Six reciprocally monophyletic entities including Candidula species were recovered, grouping into two major clades. The incorporation of additional geomitrid genera allowed us to unequivocally demonstrate the polyphyly of the genus Candidula . One major clade grouped species from southern France and Italy with the widely distributed species C. unifasciata . The second major clade grouped all the species from the Iberian Peninsula, including C. intersecta and C. gigaxii . Candidula ultima from the Canary Islands was recovered as separated lineage within the latter clade and related to African taxa. The six monophyla were defined as six new genera belonging to different tribes within the Helicellinae. Thus, we could show that similar structures of the stimulatory apparatus of the genital system in different taxa do not necessarily indicate a close phylogenetic relationship in the Geomitridae. More genera of the family are needed to clarify their evolutionary relationships, and to fully understand the evolution of the stimulatory apparatus of the genital system within the Geomitridae. Many marine mollusks attain or increase their predatory efficiency using complex chemical secretions, which are often produced and delivered through specialized anatomical structures of the foregut. The secretions produced in venom glands of Conus snails and allies have been extensively studied, revealing an amazing chemical diversity of small, highly constrained neuropeptides, whose characterization led to significant pharmacological developments. Conversely, salivary glands, the other main secretory structures of molluscan foregut, have been neglected despite their shared occurrence in the two lineages including predatory members: Gastropoda and Cephalopoda. Over the last few years, the interest for the chemistry of salivary mixtures increased based on their potential biomedical applications. Recent investigation with -omics technologies are complementing the classical biochemical descriptions, that date back to the 1950s, highlighting the high level of diversification of salivary secretions in predatory mollusks, and suggesting they can be regarded as a pharmaceutical cornucopia. As with other animal venoms, some of the salivary toxins are reported to target, for example, sodium and/or potassium ion channels or receptors and transporters for neurotransmitters such as, glutamate, serotonin, neurotensin, and noradrenaline, thus manipulating the neuromuscular system of the preys. Other bioactive components possess anticoagulant, anesthetic and hypotensive activities. Here, we overview available knowledge on the salivary glands of key predatory molluscan taxa, gastropods, and cephalopods, summarizing their anatomical, physiological and biochemical complexity in order to facilitate future comparative studies on main evolutionary trends and functional convergence in the acquisition of successful predatory strategies. To achieve a better understanding of the formation and evolutionary history of molluscan shell microstructures, we analysed crystallographic textures of shell microstructures of selected species of Protobranchia, the most ancient group of Bivalvia. Our dataset covers four of the five protobranch superfamilies. Shell layers of five species of Nuculoidea, four species of Solemyida and seven species of Nuculanoidea were analysed and classified according to their textural crystallographic patterns into six different types. Our investigation revealed that some microstructures (e.g. the radially elongate simple prismatic structure) have structure-specific textural patterns, while other microstructures (e.g. the homogeneous structure) show taxon-specific differences despite morphological similarities. The former result means that each microstructure accurately reflects a unique crystallographic structure; the inconsistency in the latter, however, calls into question the homologies among morphologically defined microstructures. In comparison with the latest molecular phylogenetic trees, our data showed that crystallographic textures are phylogenetically constrained within Protobranchia. Each nuculoidean species had two to three distinct textural patterns, and one or two similar textural patterns were recognized within each species of Solemyida and Nuculanoidea. DNA methylation is a critical epigenetic regulator of development in mammals and social insects, but its significance in development outside these groups is not understood. Here we investigated the genome-wide dynamics of DNA methylation in a mollusc model, the oyster Crassostrea gigas, from the egg to the completion of organogenesis. Large-scale methylation maps reveal that the oyster genome displays a succession of methylated and non methylated regions, which persist throughout development. Differentially methylated regions (DMRs) are strongly regulated during cleavage and metamorphosis. The distribution and levels of methylated DNA within genomic features (exons, introns, promoters, repeats and transposons) show different developmental lansdscapes marked by a strong increase in the methylation of exons against introns after metamorphosis. Kinetics of methylation in gene-bodies correlate to their transcription regulation and to distinct functional gene clusters, and DMRs at cleavage and metamorphosis bear the genes functionally related to these steps, respectively. This study shows that DNA methylome dynamics underlie development through transcription regulation in the oyster, a lophotrochozoan species. To our knowledge, this is the first demonstration of such epigenetic regulation outside vertebrates and ecdysozoan models, bringing new insights into the evolution and the epigenetic regulation of developmental processes. The consequences of emerging marine diseases on the evolutionary trajectories of affected host populations in the marine realm are largely unexplored. Evolution in response to natural selection depends on the genetic variation of the traits under selection and the interaction of these traits with the environment (GxE). However, in the case of diseases, pathogen genotypes add another dimension to this interaction. Therefore, the study of disease resistance needs to be extended to the interaction of host genotype, pathogen genotype and environment (GxGxE). In this study, we used a full-sib breeding design crossing two genetically differentiated populations of the Pacific oyster Crassostrea gigas (Thunberg, 1793), to determine the influence of host genotype, pathogen genotype and temperature on disease resistance. Based on a controlled infection experiment on two early life stages, that is, D-larvae and Pediveliger larvae at elevated and ambient water temperatures, we estimated disease resistance to allopatric and sympatric Vibrio sp. by measuring survival and growth within and between genetically differentiated oyster populations. In both populations, survival was higher upon infection with sympatric Vibrio sp., indicating that disease resistance has a genetic basis and is dependent on host genotype. In addition, we observed a significant GxGxE effect in D-larvae, where contrary to expectations, disease resistance was higher at warm than at cold temperatures. Using thermal reaction norms, we could further show that disease resistance is an environment dependent trait with high plasticity, which indicates the potential for a fast acclimatization to changing environmental conditions. These population-specific reaction norms disappeared in hybrid crosses between both populations which demonstrates that admixture between genetically differentiated populations can influence GxGxE interactions on larger scales. Tergipedidae represents a diverse and successful group of aeolid nudibranchs, with approximately 200 species distributed throughout most marine ecosystems and spanning all biogeographical regions of the oceans. However, the systematics of this family remains poorly understood since no modern phylogenetic study has been undertaken to support any of the proposed classifications. The present study is the first molecular phylogeny of Tergipedidae based on partial sequences of two mitochondrial (COI and 16S) genes and one nuclear gene (H3). Maximum likelihood, maximum parsimony and Bayesian analysis were conducted in order to elucidate the systematics of this family. Our results do not recover the traditional Tergipedidae as monophyletic, since it belongs to a larger clade that includes the families Eubranchidae, Fionidae and Calmidae. This newly recovered clade is here referred to as Fionidae, the oldest name for this taxon. In addition, the present molecular phylogeny does not recover the traditional systematic relationships at a generic level, and therefore, systematic changes are required. We recognize the following clades within Fionidae: Calma, Cuthona, Cuthonella, Eubranchus, Fiona, Murmania, Tenellia, Tergipes, Tergiposacca gen. nov., Rubramoena gen. nov. and Abronica gen. nov. The type species of Tergiposacca, T. longicerata nov. sp. is described. The other two new genera have a previously described species as their type species. Most of these taxa, with the exceptions of Eubranchus, Tergipes and Fiona are composed of radically different constituent species from their traditional membership, but appear to be supported by morphological synapomorphies as well as molecular data. Aenigmastyletus, Catriona, Phestilla, Tenellia and Trinchesia are nested within other clades and, thus are here considered as synonyms of the larger clades. The phylogenetic position and validity of Myja, Guyvalvoria, Leostyletus and Subcuthona still need to be tested in future studies when material becomes available. [ABSTRACT FROM AUTHOR]

Recent research on molluscan nervous systems provides a unique perspective on the evolution of neural circuits. Molluscs evolved large, encephalized nervous systems independently from other phyla. Homologous body-patterning genes were re-specified in molluscs to create a plethora of body plans and nervous system organizations. Octopuses, having the largest brains of any invertebrate, independently evolved a learning circuit similar in organization and function to the mushroom body of insects and the hippocampus of mammals. In gastropods, homologous neurons have been re-specified for different functions. Even species exhibiting similar, possibly homologous behavior have fundamental differences in the connectivity of the neurons underlying that behavior. ABSTRACT The idea that physical processes involved in biological development underlie morphogenetic rules and channel morphological evolution has been central to the rise of evolutionary developmental biology. Here, we explore this idea in the context of seashell morphogenesis. We show that a morphomechanical model predicts the effects of variations in shell shape on the ornamental pattern in ammonites, a now extinct group of cephalopods with external chambered shell. Our model shows that several seemingly unrelated characteristics of synchronous, ontogenetic, intraspecific, and evolutionary variations in ornamental patterns among various ammonite species may all be understood from the fact that the mechanical forces underlying the oscillatory behavior of the shell secreting system scale with the cross-sectional curvature of the shell aperture. This simple morphogenetic rule, emerging from biophysical interactions during shell formation, introduced a non-random component in the production of phenotypic variation and channeled the morphological evolution of ammonites over millions of years. The related concepts of disguising an object or physically changing it to resemble something entirely different have long captivated the human imagination. Although such notions are seemingly derived from fables and science fiction, cephalopods have perfected analogous capabilities over millions of years of natural evolution. Consequently, these invertebrates have emerged as exciting sources of inspiration for futuristic adaptive camouflage and shapeshifter-like technologies. Herein, we provide an overview of selected literature examples that have used cephalopods as models for the development of novel adaptive materials, devices, and systems. We in turn highlight some significant remaining challenges and potential future directions for such studies. The Potomida genus (Bivalvia, Unionida) has a Circum-Mediterranean distribution and like other freshwater mussel species, its populations have suffered dramatic declines. Although this genus is currently considered as monotypic, it has a long history of taxonomic revisions and presently many aspects of its systematics and evolutionary history are unclear. We sampled a total of 323 individuals from 39 different sites across the Potomida genus distribution, and sequenced two mitochondrial (16S rDNA and Cytochrome c Oxidase Subunit I) and one nuclear (28S rDNA) genes to clarify its phylogeny and phylogeographic history. Our results show that the genus includes two well-supported clades, one comprising solely the western Mediterranean species Potomida littoralis , and the other including two eastern Mediterranean species, the Greek endemic P. acarnanica and the Anatolian and Middle Eastern P. semirugata . We suggest that Potomida started radiating during the upper Miocene, and that both vicariance and dispersal events shaped the diversification and distribution of the genus along the Mediterranean region. P. littoralis is further divided in two mitochondrial lineages, one restricted to Europe and the other occurring mostly in North Africa. Moreover, some European basins present both lineages in sympatry. The conservation status of the three recognized species should be reevaluated, particularly P. acarnanica , since it is restricted to two Greek river basins presenting a high risk of extinction. Overall, our results clarify some important gaps in knowledge concerning the phylogeny, phylogeography and evolution of the Potomida genus in the Mediterranean region with important taxonomical, ecological and conservational implications. The current phylogenetic framework for the South American Hyriidae is solely based on morphological data. However, freshwater bivalve morphology is highly variable due to both genetic and environmental factors. The present study used both mitochondrial (COI and 16S) and nuclear (18S-ITS1) sequences in molecular phylogenetic analyses of nine Neotropical species of Hyriidae, collected from 15 South American rivers, and sequences of hyriids from Australia and New Zealand obtained from GenBank. The present molecular findings support traditional taxonomic proposals, based on morphology, for the South American subfamily Hyriinae, currently divided in three tribes: Hyriini, Castaliini and Rhipidodontini. Phylogenetic trees based on COI nucleotide sequences revealed at least four geographical groups of Castalia ambigua : northeast Amazon (Piriá, Tocantins and Caeté rivers), central Amazon, including C. quadrata (Amazon and Aripuanã rivers), north (Trombetas river), and C. ambigua from Peru. Genetic distances suggest that some specimens may be cryptic species. Among the Hyriini, a total evidence data set generated phylogenetic trees indicating that Paxyodon syrmatophorus and Prisodon obliquus are more closely related, followed by Triplodon corrugatus . The molecular clock, based on COI, agreed with the fossil record of Neotropical hyriids. Little is known about the evolutionary history of Helix , despite the fact that it includes the largest land snails in the western Palaearctic, some of which (e.g. H. pomatia Linnaeus, 1758) are valuable human food. We compared two groups of Helix with apparently contrasting evolutionary histories: the widespread species H. pomatia and the group distributed along the Italian Apennine chain, a relatively unknown set of species with a restricted distribution over a range of altitudes. To reconstruct the evolutionary trajectories of these two groups, we analysed morphological (shell and genitalia) and molecular characters (mitochondrial and nuclear markers) in a total of 59 populations from northern and central Europe ( H. pomatia ) and along the Apennine chain (various species). We also reconstructed the phylogeny and the evolutionary history of the genus by combining our data with that currently available in the literature. We found that spatial changes did not merely imply fragmentation of populations, but also implied environmental changes (woodlands vs. grasslands) that may have triggered the observed phenotypic diversification. We also found that Anatolia is the ancestral range of Helix and is therefore an important area for the Palaearctic diversity. The results provide insights into the evolutionary history of species richness and more generally into the processes that may have shaped the distribution and diversification of these organisms across Europe and the peri-Mediterranean area. Noninvasive methods in shell shape variation may help to understand evolution, ecology, stress and role of molluscan in aquatic ecosystems. Imaging analysis is a suitable diagnostic tool in morphological studies to (1) evaluate the health status of investigated animals, and (2) monitor sea coastal habitats. We introduce the feasibility of the cone-beam computed tomography as an optimal technique for 3D surface scanning to obtain virtual valve surfaces of Mytilus galloprovincialis, and analyze them exploiting the geometric morphometric facilities. Statistical output revealed morphological difference between mussels coming from different extensive rearing systems highlighting how the entire valve surface contributed to discriminate between groups when we compared 2-and 3D analyses. Many factors drive the morphological differences observed in the valve shape variation between the two sites, such as geographical genetic differentiation, natural environmental effects and culture conditions. The simplicity of the proposed methodology avoids damage and handling of individuals, makes this approach useful for morphological data collection, and helps to detect detrimental agents for sea ecosystems by using molluscans. The Ammonoidea is a group of extinct cephalopods ideal to study evolution through deep time. The evolution of the planispiral shell and complexly folded septa in ammonoids has been thought to have increased the functional surface area of the chambers permitting enhanced metabolic functions such as: chamber emptying, rate of mineralization and increased growth rates throughout ontogeny. Using nano-computed tomography and synchrotron radiation based micro-computed tomography, we present the first study of ontogenetic changes in surface area to volume ratios in the phragmocone chambers of several phylogenetically distant ammonoids and extant cephalopods. Contrary to the initial hypothesis, ammonoids do not possess a persistently high relative chamber surface area. Instead, the functional surface area of the chambers is higher in earliest ontogeny when compared to Spirula spirula. The higher the functional surface area the quicker the potential emptying rate of the chamber; quicker chamber emptying rates would theoretically permit faster growth. This is supported by the persistently higher siphuncular surface area to chamber volume ratio we collected for the ammonite Amauroceras sp. compared to either S. spirula or nautilids. We demonstrate that the curvature of the surface of the chamber increases with greater septal complexity increasing the potential refilling rates. We further show a unique relationship between ammonoid chamber shape and size that does not exist in S. spirula or nautilids. This view of chamber function also has implications for the evolution of the internal shell of coleoids, relating this event to the decoupling of soft-body growth and shell growth. Life-history evolution often results in trade-offs between reproduction, growth and longevity. We investigated the relationships among these traits in a hermaphroditic freshwater snail, Helisoma trivolvis, by manipulating opportunities for reproduction and measuring body size and egg production throughout the lifespan. Snails were placed in one of four treatment groups: snails isolated for their entire lives, snails housed with a partner for 1 week at 18 weeks, snails housed with a partner for 1 week at 26 weeks and snails afforded 1 week of mating opportunities six times between 18 and 97 weeks. We monitored egg production and shell diameter regularly throughout the experiment until all snails had died. Isolated snails laid almost no eggs, confirming a low level of self-fertilization in this species. For both groups of snails with only one mating opportunity, the average duration of egg production was 16.8 weeks. Snails in the multiply-mated group continued to lay about 1 egg mass per day with an average of 19-25 eggs per mass for 48 weeks and then egg production decreased. Although all of the snails grew consistently throughout the experiment, when snails were actively laying eggs their growth slowed relative to those no longer producing eggs. There were no significant differences in longevity among snails that were isolated and never mated, those that mated once (either early or later in life) and those that had multiple mating opportunities and continued to lay eggs throughout their lifetimes. Understanding the extent of local adaptation is critical for conservation and restoration planning in an era of environmental change. Adaptive differentiation among populations may mediate a species' response to environmental stress, yet these evolutionary processes are seldom studied in marine systems. We used native Olympia oysters ( Ostrea lurida ) as a model organism for studying local adaptation with applications to marine restoration. We tested whether populations of oysters in California estuaries are locally adapted to their home sites and to stressful low salinity events, which are predicted to increase in frequency with climate change. We spawned oysters from three sites in San Francisco Bay and raised their offspring under common laboratory conditions. These oysters were then reciprocally transplanted among the three field sites. At two of the three sites, oysters of local origin tended to survive better than those from other populations, suggesting that local adaptation may occur even within a single estuary. In a subsequent experiment, we raised oysters from two sites in San Francisco Bay and one site in Tomales Bay through two generations under common conditions and then subjected these oysters to a low salinity challenge in the laboratory. Oysters with the highest survival originated from the site with the lowest field salinity, suggesting that natural selection may favor stress tolerant phenotypes in certain regions. As interest grows in restoring heavily impacted native species, our results indicate that considering local adaptation may be essential to deciding how and where to conserve and restore species faced with changing conditions. Background: Shell color polymorphisms of Mollusca have contributed to development of evolutionary biology and population genetics, while the genetic bases and molecular mechanisms underlying shell pigmentation are poorly understood. The Pacific oyster (Crassostrea gigas) is one of the most important farmed oysters worldwide. Through successive family selection, four shell color variants (white, golden, black and partially pigmented) of C. gigas have been developed. To elucidate the genetic mechanisms of shell coloration in C. gigas and facilitate the selection of elite oyster lines with desired coloration patterns, differentially expressed genes (DEGs) were identified among the four shell color variants by RNA-seq. Results: Digital gene expression generated over fifteen million reads per sample, producing expression data for 28,027 genes. A total number of 2,645 DEGs were identified from pair-wise comparisons, of which 432, 91, 43 and 39 genes specially were up-regulated in white, black, golden and partially pigmented shell of C. gigas, respectively. Three genes of Abca1, Abca3 and Abcb1 which belong to the ATP-binding cassette (ABC) transporters super-families were significantly associated with white shell formation. A tyrosinase transcript (CGI_10008737) represented consistent up-regulated pattern with golden coloration. We proposed that white shell variant of C. gigas could employ “endocytosis” to down-regulate notch level and to prevent shell pigmentation. Conclusion: This study discovered some potential shell coloration genes and related molecular mechanisms by the RNA-seq, which would provide foundational information to further study on shell coloration and assist in selective breeding in C. gigas. Both historical and contemporary processes influence the genetic structure of species, but the relative roles of such processes are still difficult to access. Population genetic studies of species with recent evolutionary histories such as the New Zealand endemic scallop Pecten novaezelandiae (<1 Ma) permit testing of the effects of recent processes affecting gene flow and shaping genetic structure. In addition, studies encompassing the entire distributional range of species can provide insight into colonization processes. Analyses of genetic variation in P. novaezelandiae (952 individuals from 14 locations, genotyped at 10 microsatellite loci) revealed a weak but significant regional structure across the distributional range of the species, as well as latitudinal gradients of genetic diversity and differentiation: estimates of migration rates supported these patterns. Our results suggest that the observed genetic structure and latitudinal gradients reflect a stepping-stone model of colonization (north to south) and emerging divergence of populations as a result of ongoing limitations to gene flow and insufficient time to reach migration-drift equilibrium. The low levels of interpopulation and interregional genetic differentiation detected over hundreds of kilometres reflect the recent evolutionary history of P. novaezelandiae and stand in contrast to patterns reported for other evolutionary older species at the same spatial scale. The outcomes of this study contribute to a better understanding of evolutionary processes influencing the genetic variation of species and provide vital information on the genetic structure of P. novaezelandiae. Evolutionarily conserved signaling intermediate in Toll pathways (ECSIT) is a multifunctional adaptor protein that plays a key role in the regulation of the oxidative phosphorylation (OXPHOS) system, bone morphogenetic protein (BMP) pathway and Toll-like receptor (TLR) signaling pathway in mammals. However, the function of ECSIT homologs in mollusks, the second most diverse group of animals, is not well understood. In this study, we identified an ECSIT homolog in the Hong Kong oyster Crassostrea hongkongensis ( Ch ECSIT) and investigated its biological functions. The full-length cDNA of Ch ECSIT is 1734 bp and includes an open reading frame (ORF) of 1074 bp that encodes a polypeptide of 451 amino acids. The predicted Ch ECSIT protein shares similar structural characteristics with other known ECSIT family proteins. Quantitative real-time PCR analysis revealed that Ch ECSIT mRNA is broadly expressed in all of the examined tissues and at different stages of embryonic development; its transcript level could be significantly up-regulated by challenge with microorganisms ( Vibrio alginolyticus , Staphylococcus haemolyticus and Saccharomyces cerevisiae ). In addition, Ch ECSIT was found to be located primarily in the cytoplasm, and its overexpression stimulated the transcriptional activity of an NF-κB reporter gene in HEK293T cells. The aim of this work is to investigate the sequence conservation and the evolution of repeated DNA in related species. Satellite DNA is a component of eukaryotic genomes and is made up of tandemly repeated sequences. These sequences are affected by high rates of mutation that lead to the occurrence of species-specific satellite DNAs, which are different in terms of both quantity and quality. In this work, a novel repetitive DNA family, named PjHhaI sat, is described in Pecten jacobaeus. The quantitative analyses revealed a different abundance of this element in the molluscan species investigated in agreement with the 'library hypothesis' even if, in this case, at a high taxonomic level. In addition, the qualitative analysis demonstrated an astonishing sequence conservation not only among scallops but also in six other molluscan species belonging to three classes. These findings suggest that the PjHhaI sat may be considered as the most ancients of DNA described so far, which remained 'frozen' during molluscan evolution. The widespread distribution of this sat DNA in molluscs as well as its long evolutionary preservation open up questions on the functional role of this element. Molluscs are economically and ecologically important components of aquatic ecosystems. In addition to supporting valuable aquaculture and wild-harvest industries, their populations determine the structure of benthic communities, cycling of nutrients, serve as prey resources for higher trophic levels and, in some instances, stabilize shorelines and maintain water quality. This paper reviews existing knowledge of the ecology of host–parasite interactions involving marine molluscs, with a focus on gastropods and bivalves. It considers the ecological and evolutionary impacts of molluscan parasites on their hosts and vice versa, and on the communities and ecosystems in which they are a part, as well as disease management and its ecological impacts. An increasing number of case studies show that disease can have important effects on marine molluscs, their ecological interactions and ecosystem services, at spatial scales from centimeters to thousands of kilometers and timescales ranging from hours to years. In some instances the cascading indirect effects arising from parasitic infection of molluscs extend well beyond the temporal and spatial scales at which molluscs are affected by disease. In addition to the direct effects of molluscan disease, there can be large indirect impacts on marine environments resulting from strategies, such as introduction of non-native species and selective breeding for disease resistance, put in place to manage disease. Much of our understanding of impacts of molluscan diseases on the marine environment has been derived from just a handful of intensively studied marine parasite–host systems, namely gastropod–trematode, cockle–trematode, and oyster–protistan interactions. Understanding molluscan host–parasite dynamics is of growing importance because: (1) expanding aquaculture; (2) current and future climate change; (3) movement of non-native species; and (4) coastal development are modifying molluscan disease dynamics, ultimately leading to complex relationships between diseases and cultivated and natural molluscan populations. Further, in some instances the enhancement or restoration of valued ecosystem services may be contingent on management of molluscan disease. The application of newly emerging molecular tools and remote sensing techniques to the study of molluscan disease will be important in identifying how changes at varying spatial and temporal scales with global change are modifying host–parasite systems. Bilaterally asymmetrical glochidia (i.e. bivalved parasitic larvae bearing a large marginal appendage on a single valve) have been reported from five Asian freshwater mussel genera belonging to two separate subfamilies, the Gonideinae (i.e. Pseudodon, Solenaia, and Physunio) and Rectidentinae (i.e. Contradens and Trapezoideus). This classification requires that the bilaterally asymmetrical glochidium-bearing mussels are not monophyletic, and suggests that this atypical larval morphology evolved twice in the same geographic region. Although homoplastic glochidium characters are known (e.g. marginal appendages and size), we hypothesized that bilaterally asymmetrical glochidia represent a novel morphological synapomorphy. We tested the monophyly of the mussels bearing bilaterally asymmetrical glochidia using a molecular matrix consisting of representatives from all six freshwater mussel families and three molecular markers ( 28S, 16S, and COI). Bayesian inference, maximum likelihood, and ancestral state reconstruction were employed to estimate the phylogeny and larval trait transformations. The reconstructed phylogeny rejects the monophyly of the asymmetrical glochidium-bearing mussels and resolves two putative origins of asymmetrical glochidia; however, ancestral state reconstruction supports asymmetrical glochidia as a synapomorphy of only one supraspecific taxon of the Rectidentinae. In the Gonideinae, asymmetrical glochidia were autapomorphic of Pseudodon cambodjensis ( Petit, 1865). That is, no other taxa resolved among the Gonideinae had bilaterally asymmetrical glochidia, including other Pseudodon species. We describe how the alleged intrageneric glochidial variation in Pseudodon, and in the other genera of the Gonideinae reported to have asymmetrical glochidia (i.e. Solenaia and Physunio), challenge the resolved convergence of asymmetrical glochidia. Soft-tissue attachment scars of two genera and four species of Late Jurassic craspeditid ammonites from the Russian Platform are described. A previously suggested relationship between lateral attachment scars and ammonoid hyponome is confirmed, however, a new interpretation is proposed for dorsal attachment scars: they could have been areas not only for attachment of the dorsal (nuchal) retractors, but also of the cephalic retractors. The new type of the soft-tissue attachment-anterior lateral sinuses, located between the lateral attachment scars and the aperture of the ammonite body chamber is described. Enclosed elliptical or subtriangular areas in apertural parts of the anterior lateral sinuses were found for the first time. Their presence and location suggest that this structure could have been used for attaching the funnel-locking apparatus, similar to those of coleoids. A transformation of shape and position of lateral attachment scars through the evolution of the Late Jurassic craspeditid lineage starting from platycones (Kachpurites fulgens) to keeled oxycones (Garniericeras catenulatum) is recognized. Molecular phylogenies of extant species are considered effective tools to infer mechanisms of speciation. Here, we benefit from this utility to investigate the evolutionary history of an organismal group linked to different aquatic ecosystems, the microgastropod genus Pseudamnicola (family Hydrobiidae). Previous studies have found around 45 species of the nominal subgenus P. (Pseudamnicola), most of them in coastal stream localities of several Mediterranean islands and mainland territories, whereas only 12 species of the other subgenus, P. (Corrosella), have been collected from springs and headwaters of mountainous regions of the Iberian Peninsula and south of France. As springs often act as isolated habitats affecting dispersion and constraining gene flow, we supposed that the temporal history and mode of diversification of species from both subgenera should differ and therefore be reflected in their phylogenetic patterns. To assess this hypothesis, we performed a molecular phylogeny based on mitochondrial and nuclear DNA sequences and later conducted an independent analysis to examine the potential effect of certain geographic and ecological variables in the genetic divergences of the subgenera. Additionally, we estimated the ancestral area of diversification of both groups. Published anatomical revisions and our molecular analyses suggest that the genus Pseudamnicola should be divided into three genera: the two previous subgenera plus a new one described here. As postulated, the evolution of the spring organisms was strongly related to habitat fragmentation and isolation, whereas dispersal followed by divergence seem to have been the most common speciation processes for euryhaline species inhabiting coastal streams and low river stages in which waters remain connected. On the contrary, rather than habitat fragmentation or dispersion, environmental conditions have played a larger role during the deep divergent split leading to the three genera. Background: Non-indigenous taxa currently represent a large fraction of the species and biomass of freshwater ecosystems. The accumulation of invasive taxa in combination with other stressors in these ecosystems may alter the habitats to which native taxa are adapted, which could elicit evolutionary changes in native populations and their ecological interactions. Assessing ecological and evolutionary consequences of invasions simultaneously may therefore be the most effective approach to study taxa with complex invasion histories. Here we apply such an integrated approach to the cerithioid gastropod Melanoides tuberculata, a model system in invasion biology. Results: Molecular phylogenetics and ancestral range reconstructions allowed us to identify several independent Asian invasions in Lakes Malawi and Tanganyika, the Congo River, Nigeria and Cameroon. Some invasive M. tuberculata populations display much variation in shell morphology, and overlap in morphospace with M. tuberculata populations native to Africa. Experiments confirmed great ecophenotyic plasticity in some invasive populations, which, in combination with the overlap in disparity with native populations, masks invaders and their dispersal through Africa. Finally, the results of geographic modeling indicate that cryptic M. tuberculata invasions occurred primarily in densely populated areas. Conclusions: We reveal the continental nature of invasions of Asian M. tuberculata to Africa. Several of the affected ecosystems have high endemicity in Cerithioidea: Lake Tanganyika has an unparalleled diversity in freshwater cerithioids (>10 endemic genera) and the Congo Basin and Lake Malawi are home to the two largest endemic species clusters of Melanoides in Africa (~12 and ~8 species, respectively). Cerithioids perform ecologically important functions in the benthic ecosystems of African freshwaters, but invaders and ecosystem change pose risks to their native diversity. It has long been known that species should not be distributed randomly in morphospace (a multidimensional trait space), even under simple models of evolution. However, recent studies suggest that position in morphospace can affect aspects of evolution such as the durations of clades and the species richness of their constituent taxa. Here we investigate the dynamics of morphospace occupancy in living and fossil marine bivalves using shell size and aspect ratio, two functionally important traits. Multiple lines of evidence indicate that the center of a family's morphospace today represents a location where taxonomic diversity is maximized, apparently owing to lower extinction rates. Within individual bivalve families, species with narrow geographic ranges are distributed throughout the morphospace but widespread species, which are generally expected to be extinction resistant, tend to be concentrated near the center. The morphospace centers of most species-rich families today (defined as the median value for all species in the family) tend to be close to the positions of the family founders, further suggesting an association between position in morphospace and net diversification rates. The interrelationships and evolutionary history of molluscs have seen great advances in the last decade. Recent phylogenetic studies have allowed alternative morphology-based evolutionary scenarios to be tested and, most significantly, shown that the aplacophorans are sister group to polyplacophorans (chitons), corroborating palaeontological and embryological evolutionary scenarios in which aplacophorans are secondarily simplified from a chiton-like ancestor. Aplacophoran morphology therefore does not represent the plesiomorphic condition for molluscs as a whole. The mollusc crown group radiated in the Early Cambrian, and rapidly thereafter, stem lineages to the major molluscan classes emerged: cephalopods, gastropods, bivalves (= pelecypods), monoplacophorans, rostroconchs (inferred stem scaphopods) and aculiferans. This attests to the fast, adaptive radiation of the crown group during the Cambrian explosion. Kimberella from the latest Ediacaran exhibits several molluscan traits, which justifies its position as a molluscan stem-group member, rather than as a more basal Lophotrochozoan. The genus Agriopleura (Radiolitidae) is restricted to the Lower Cretaceous of the Mediterranean region, including the Middle East, and this rudist is apparently absent from the New World. Agriopleura underwent a size increase from late Hauterivian to mid-late Barremian, matching Cope's rule, followed by a Lilliput phase in the early Aptian. The relative development of radial bands increased through time and represents a key evolutionary index. During its evolution, Agriopleura increased in species diversity and expanded geographically onto the European Mediterranean Tethyan margin from the late Hauterivian to Barremian, after which it disappeared from the region (pseudotermination). In the lower Aptian, after a Lazarus gap, the genus reappears in the southern Mediterranean Tethyan margin. Assuming that Archaeoradiolites is the direct descendant of Agriopleura, the disappearance of the genus in the mid-Aptian was a pseudoextinction and this coincided with a major crisis of shallow carbonate settings and their associated biota. The definition of Agriopleura species is based on a set of qualitative and quantitative external and internal characters. Five species are recognized: three Barremian species, Agriopleura blumenbachi, the type species , A. marticensis and A. carinata; one lower Aptian species A. libanica; and a new species A. sequana restricted to the upper Hauterivian. The morphology of gastropod shells has been a focus of analyses in ecology and evolution. It has recently emerged as an important issue in developmental biology, thanks to recent advancements in molecular biological techniques. The growing tube model is a theoretical morphological model for describing various coiling patterns of molluscan shells, and it is a useful theoretical tool to relate local tissue growth with global shell morphology. However, the growing tube model has rarely been adopted in empirical research owing to the difficulty in estimating the parameters of the model from morphological data. In this article, I solve this problem by developing methods of parameter estimation when (1) 3D Computed Tomography (CT) data are available and (2) only 2D image data (such as photographs) are available. When 3D CT data are available, the parameters can be estimated by fitting an analytical solution of the growing tube model to the data. When only 2D image data are available, we first fit Raup׳s model to the 2D image data and then convert the parameters of Raup׳s model to those of the growing tube model. To illustrate the use of these methods, I apply them to data generated by a computer simulation of the model. Both methods work well, except when shells grow without coiling. I also demonstrate the effectiveness of the methods by applying the model to actual 3D CT data and 2D image data of land snails. Molluscs are diverse and important enough to receive considerable scientific attention. We herein question recent opinions that the molluscan origin, deep inner relationships, and early evolution have been largely resolved. The traditional Testaria concept implied progressive evolution from worm-like aplacophoran to polyplacophoran and then conchiferan body plans; sole evidence for this came from morphocladistic analyses, which we conclude were confounded by homoplasy. The recently preferred Aculifera-Conchifera concept is supported by some but not all analyses using massive sequence data on rather small and uneven taxon sets. Recent results from mitogenomics indicate that gene-rearrangement events could cause sequence biases overriding potential phylogenetic signal. We discuss recent progress regarding multilocus marker analyses, particularly refining the neglected Serialia hypothesis with Monoplacophora sister to Polyplacophora. This third hypothesis for molluscan relationships is supported by an integrative interpretation and is roughly compatible with available fossil evidence if the first molluscs were small and had a true shell rather than a chiton-like body organization. Despite some claims of consensus, there are substantial discrepancies among recent molecular studies regarding class-level topologies. These may be symptomatic of a plethora of factors and evolutionary processes – obvious or more hypothetical – that might hinder successful reconstruction of early molluscan diversification. Certain species of chiton (Mollusca, Polyplacophora) have hundreds of small (< 100 µm) eyes embedded in their dorsal shell plates. These eyes each contain a retina, a layer of screening pigment, and a lens. Previously, we demonstrated that the eyes of chitons provide spatial vision. As in other camera-type eyes, the screening pigments in the eyes of chitons absorb off-axis light in order to preserve the contrast of images formed on the retina. Our results indicate that the red-brown, alkali-soluble screening pigment associated with the eyes of the chitonAcanthopleura granulata(Gmelin, 1791) is pheomelanin. Using high-performance liquid chromatography (HPLC) and MALDI-TOF mass spectroscopy, we find that degradingA. granulata’s screening pigment with alkaline hydrogen peroxide produces 6-(2-amino-2-carboxyethyl)-2-carboxy-4-hydroxybenzothiazole (BTCA), a diagnostic marker of pheomelanin. Chitons are the first molluscs demonstrated to use pheomelanin as a screening pigment in their eyes. Our results suggest that the image-forming eyes of chitons may have evolved separately from eyes that employ different types of screening pigment, such as those of most other invertebrates. Further, we hypothesize that change in the expression pattern of tyrosinase – an enzyme responsible for melanin synthesis in many other metazoans – may have contributed to the origin of screening pigments in chitons, a critical step in the evolution of their eyes. We present a critical evaluation of the Early Miocene terrestrial and aquatic mollusc fauna of the North Bohemian Lake in the Most Basin in the Czech Republic. In total, 90 species (8 aquatic and 81 terrestrial gastropods, one bivalve) are documented from that lake system that had formed within the North Bohemian Rift. Only three of these species are newly recorded for the Most Basin, suggesting that the fauna is well sampled. Based on historical collections of the Natural History Museum in Vienna, which were partly acquired by quantitative bulk samples from Tuchořice, a rough estimate of the composition of the terrestrial assemblage can be presented. More than 80% of the >30,400 shells are represented by carychiids, vertiginids and valloniids, suggesting the presence of densely forested wetlands fringing the North Bohemian Lake. About 57% of the terrestrial species are known so far exclusively from the Most Basin. This high degree of "endemism", however, is rather a result of the still very fragmentary knowledge of coeval European faunas. Discus rasseri Harzhauser, Neubauer & Georgopoulou sp. nov. and Discus zagorseki Harzhauser, Neubauer & Georgopoulou sp. nov. are described as new species and Esuinella Harzhauser, Neubauer & Georgopoulou gen. nov. Despite the importance of Nucella as a model system in numerous fields of biology, no phylogenetic analysis of the genus, including every widely recognized species, has been conducted. We have analysed about 4,500 bp of DNA from six different genes (three mitochondrial, three nuclear) from each taxon in the genus. Our results showed western Pacific N. heyseana and N. freycinetii as distinct and distantly related, but found no evidence that N. elongata is distinct from N. heyseana. We also resolved N. heyseana as the closest living relative of the North Atlantic N. lapillus and, using the fossil record for calibration, inferred a minimum separation time between Atlantic and Pacific lineages of at least 6.2 Ma, slightly pre-dating the opening of the Bering Strait. Comparative analyses showed egg size to be evolutionarily labile, but also revealed a highly significant negative relationship between egg size and the nurse-egg-to-embryo ratio. The negative correlation indicates that evolutionary changes in egg size among species are balanced by changes in the number of nurse eggs allocated to each offspring, indicating that interspecific variation in the nurse-egg-to-embryo ratio has not been driven by divergent selection on hatching size, but may instead be a response to variation in other factors, such as parent-offspring conflict. North Pacific specimens of Melanochlamys sea slugs were examined morphologically (including the male reproductive morphology, shell and external coloration) and were sequenced for three genes (mitochondrial COI and 16S and nuclear H3). Phylogenetic and species delimitation analyses were used to determine the evolutionary relationships and species diversity among the specimens examined. Both molecular and morphological data revealed an unexpected high level of cryptic diversity. At least four distinct species occur on the Northwestern Pacific. Melanochlamys ezoensis occurs in Russia and temperate and cold areas in Japan. Three additional undescribed species occur in Japan and/or South Korea. One of the undescribed species occurs both in South Korea and in Japan, but only in Tokyo Bay, suggesting that it could be non-native in Japan. Two distinct species occur on the Northeastern Pacific coast; Melanochlamys diomedea is widespread from Southern California to Alaska, whereas M. ezoensis was found only in San Francisco Bay, suggesting a human-mediated introduction. This is further supported by the absence of records of M. ezoensis in San Francisco prior to 2001. The species diversity of Melanochlamys in the Northwestern Pacific is much greater than in the Northeastern Pacific; it is hypothesized that differences in geographic and ocean current system complexity might account for different responses to glacial extinction and postglacial expansion. The distribution of additive vs. non-additive genetic variation in natural populations represents a central topic of research in evolutionary/organismal biology. For evolutionary physiologists, functional or whole-animal performance traits ('physiological traits') are frequently studied assuming they are heritable and variable in populations., Physiological traits of evolutionary relevance are those functional capacities measured at the whole-organism level, with a potential impact on fitness. They can be classified as capacities (or performances) or costs, the former being directly correlated with fitness and the latter being inversely correlated with fitness (usually assumed as constraints)., In spite of their obvious adaptive significance, the additive genetic variation in physiological traits, and its relative contribution to phenotypic variance (or narrow-sense heritability) in comparison with maternal, dominance or epistatic variance, is known only for a few groups such as insects and mammals., In this study, we assessed the additive and maternal/non-additive genetic variation in a suite of physiological and morphological traits in populations of the land snail Cornu aspersum., Except for dehydration rate ( h2 = 0·32 ± 0·15), egg mass ( h2 = 0·82 ± 0·30) and hatchling mass ( h2 = 1·01 ± 0·31; population = fixed effect), we found very low additive genetic variation. Large non-additive/maternal effects were found in all traits. Cage effects did not change the results, indicating low contribution of common environmental variance to our results. No differences were found between the phenotypic and non-additive genetic variance/covariance matrices., Even though we compared populations across 1300 km in a common garden set-up, our results suggest an absence of physiological as well as morphological differentiation in these populations., These results contrast with previous analyses in the original distributional range of this species, which found high additive genetic variation in morphological traits. Spiralian embryogenesis is deeply conserved and seems to have been in place in the last common ancestor of the large assemblage of protostome phyla known as the Lophotrochozoa. While the blastula fate maps of several spiralian embryos have been determined, little is known about the events that link the early embryo and the larva. For all cells in the Ilyanassa blastula, we determined the clonal morphology at four time points between the blastula and veliger stages. We found that ectomesoderm comes mostly from 3a and 3b, but also from 2c and 2b. We also observed the ingression and early proliferation of 3a- and 3b-derived ectomesoderm. We found cells in the 2b clone that marked the anterior edge of the blastopore and later the mouth and cells in the 3c/3d clones that marked the posterior edges of these structures. This demonstrates directly that the mouth forms in the same location as the blastopore. In the development of the shell field, we observed dramatic cell migration events that invert the positions of the 2b and 2d clones that contribute to the shell. Using time-lapse imaging, we followed and described the cleavage pattern of the conserved endomesodermal blast cell, 4d, up to 4d + 45 h, when there were 52 cells in the clone. Our results show the growth and movement of clones derived from cells of the spiralian blastula as they transform into the trochophore-like and veliger stages. Abstract: Larval modes of development affect evolutionary processes and influence the distribution of marine invertebrates in the ocean. The decrease in pelagic development toward higher latitudes is one of the patterns of distribution most frequently discussed in marine organisms (Thorson's rule), which has been related to increased larval mortality associated with long pelagic durations in colder waters. However, the type of substrate occupied by adults has been suggested to influence the generality of the latitudinal patterns in larval development. To help understand how the environment affects the evolution of larval types we evaluated the association between larval development and habitat using gastropods of the Muricidae family as a model group. To achieve this goal, we collected information on latitudinal distribution, sea water temperature, larval development and type of substrate occupied by adults. We constructed a molecular phylogeny for 45 species of muricids to estimate the ancestral character states and to assess the relationship between traits using comparative methods in a Bayesian framework. Our results showed high probability for a common ancestor of the muricids with nonpelagic (and nonfeeding) development, that lived in hard bottoms and cold temperatures. From this ancestor, a pelagic feeding larva evolved three times, and some species shifted to warmer temperatures or sand bottoms. The evolution of larval development was not independent of habitat; the most probable evolutionary route reconstructed in the analysis of correlated evolution showed that type of larval development may change in soft bottoms but in hard bottoms this change is highly unlikely. Lower sea water temperatures were associated with nonpelagic modes of development, supporting Thorson's rule. We show how environmental pressures can favor a particular mode of larval development or transitions between larval modes and discuss the reacquisition of feeding larva in muricids gastropods. Abstract: Jumonji (Jmj) proteins are histone demethylases, which control the identity of stem cells. Jmj genes were characterized from plants to mammals where they have been implicated in the epigenetic regulation of development. Despite the Pacific oyster Crassostrea gigas representing one of the most important aquaculture resources worldwide, the molecular mechanisms governing the embryogenesis and reproduction of this lophotrochozoan species remain poorly understood. However, annotations in the C. gigas EST library suggested the presence of putative Jumonji genes, raising the question of the conservation of this family of histone demethylases in the oyster. Using Primer walking, 5′-RACE PCR and in silico analyses, we characterized nine Jumonji orthologues in the oyster, called Cg-Jmj, bearing conserved domains critical for putative histone demethylase activity. Phylogenic analyses revealed that oyster Jumonji cluster into two distinct groups: ‘single-domain Jmj’ and ‘multi-domain Jmj’, and define 8 subgroups corresponding to each cognate orthologues in metazoans. RT-qPCR investigations showed specific regulations of Cg-Jmj mRNAs during the early development and along the reproduction cycle. Furthermore, in situ and in toto hybridizations indicate that oyster Jumonji genes are transcribed mostly within the gonad in adult oysters whereas they display a ubiquitous expression during embryonic and larval development. Our study demonstrates the presence of nine Jumonji orthologues in the oyster C. gigas. Their domain conservation and their expression profile suggest an implication during reproduction and development, questioning about the epigenetic regulation by histone methylation in lophotrochozoans. Octopuses of the family Octopodidae are singular among cephalopods in their reproductive behavior, showing two major reproductive strategies: the first is the production of few and large eggs resulting in well-developed benthic hatchlings (holobenthic life history); the second strategy is the production of numerous small eggs resulting in free-swimming planktonic hatchlings (pelago-benthic life history). Here, we utilize a Bayesian-based phylogenetic comparative method using a robust molecular phylogeny of 59 octopus species to reconstruct the ancestral states of development type in benthic octopuses, through the estimation of the most recent common ancestors and the rate of gain and loss in complexity (i.e., planktonic larvae) during the evolution. We found a high probability that a free-swimming hatchling was the ancestral state in benthic octopuses, and a similar rate of gain and loss of planktonic larvae through evolution. These results suggest that in benthic octopuses the holobenthic strategy has evolved from an ancestral pelago-benthic life history. During evolution, the paralarval stage was reduced to well-developed benthic hatchlings, which supports a 'larva-first' hypothesis. Difficulties in elucidating the evolutionary history of the octopods have arisen from problems in identifying informative morphological characters. Recent classifications have divided the largest group, the incirrate octopods, into five groups. These include the pelagic superfamily Argonautoidea and three gelatinous pelagic families (Vitreledonellidae, Bolitaenidae, Amphitretidae). All benthic incirrate octopods have been accommodated in the family Octopodidae, itself divided into four subfamilies, Octopodinae, Eledoninae, Bathypolypodinae and Graneledoninae, which are defined by the presence or absence of an ink sac, and uniserial or biserial sucker arrangements on the arms. We used relaxed clock models in a Bayesian framework and maximum likelihood methods to analyse three nuclear and four mitochondrial genes of representatives from each of the previous subfamilies. Strong evidence indicates that the family Octopodidae is paraphyletic and contains the gelatinous pelagic families. The subfamilies of Octopodidae recognised in earlier works do not reflect evolutionary history. The following clades were supported in all analyses: (1) Eledone/Aphrodoctopus, (2) Callistoctopus/ Grimpella/ Macroctopus/Scaeurgus, (3) Abdopus/ Ameloctopus/Amphioctopus/ Cistopus/Hapalochlaena/ Octopus, (4) Enteroctopus/Muusoctopus/ Vulcanoctopus, (5) Vitreledonella/ Japetella, (6) Southern Ocean endemic and deep-sea taxa with uniserial suckers. These clades form the basis for a suite of taxa assigned family taxonomic rank: Amphitretidae, Bathypolypodidae, Eledonidae, Enteroctopodidae, Megaleledonidae and Octopodidae sensu nov. The Evolution Megalab database of Cepaea nemoralis provides shell morph frequencies in sets of samples collected from throughout the geographical range during the mid-20th and early 21st centuries. Using data from Britain only, historic and recent samples from woodland, hedgerow and grassland habitats, compared over different geographical distances apart, have been examined for change in frequency with time. There is evidence of changes in mean morph frequency over the last half century in some but not all habitats. Yellow and mid-banded morphs have increased in woodland, unbanded and mid-banded have increased in hedgerow habitats, while for grassland there were too few samples to draw a conclusion. Early and late samples are rarely situated very close to each other. Pairings over longer distances produce larger datasets but their value declines as they are increasingly influenced by factors associated with metapopulation structure; the maximum usable distance is not more than a few km. Cave-dwelling animals can be classified based on their occurrence in and relationship to the subterranean environment. Subsurface distribution data and studies addressing the initial causes for animals to enter underground habitats are sparse. By retrieving occurrence data from two voluntary biospeleological collections in Central Germany, the degree of cave penetration in terrestrial gastropods was investigated, thus to infer potential evolutionary drivers. In total, 66 identified gastropod species entered the subterranean environment with 23 of the species also recorded from the dark zone. Gastropods possessed variable degrees of cave penetration and no obligate cave forms were observed. A decline of occurrence records from the entrance to the dark zone was the most prominent pattern. Nevertheless, several species were collected from all three light zones (i.e. entrance, transition and dark). A variety of potential reasons can be inferred to explain their subsurface appearance: exploitation of alternative food sources, aphotic above-ground movement, mating sites, temporal, seasonal and long-term refugia, and chance. Moreover, the results imply a frequent migration of species between the interconnected light zones and the surface. Consequently, terrestrial gastropods should be considered when investigating the origin and importation of allochthonous resources in caves. [ABSTRACT FROM AUTHOR]
