The genealogical divergence index (gdi) was developed to aid in molecular species delimitation under the multispecies coalescent model, which has been shown to delimit genetic structures but not necessarily species. Although previous studies have used meta-analyses to show that gdi could be informative for distinguishing taxonomically good species, the biological and evolutionary implications of divergences showing different gdi values have yet to be studied. I showed that an increase in gdi value was correlated with later stages of divergence further along a speciation continuum in an Amazonian Hercules beetle system. Specifically, a gdi value of 0.7 or higher was associated with diverge between biological species that can coexist in geographic proximity while maintaining their evolutionary independence. Divergences between allopatric species that were conventionally given subspecific status, such as geographic taxa that may or may not be morphologically divergent, had gdi values that fell within the species delimitation ambiguous zone (0.2 < gdi < 0.7). However, the results could be drastically affected by the sampling design, i.e. the choice of different geographic populations and the lumping of distinct genetic groups when running the analyses. Different gdi values may prove to be biologically and evolutionarily informative should additional speciation continua from different empirical systems be investigated, and the results obtained may help with objectively delimiting species in the era of integrative taxonomy. The New World genus Dynastes reaches its northernmost extent in the United States with two species, Dynastes grantii and D. tityus (Linnaeus), the latter of which is widespread in eastern United States, including Virginia. Notes on the biology and distribution of D. tityus in Virginia are presented, along with observations on their feeding and mating behaviors on green ash trees (Fraxinus pennsylvanica Marshall) in Southampton County during the summer of 2019. The apparent dependence on ash trees of both species of American Dynastes that occur north of Mexico for attracting and locating mates is briefly discussed in light of their possible endangerment due to the ever-expanding range of the invasive Emerald Ash Borer, Agrilus planipennis Fairmaire. Beetle elytra are thickened forewings, they are lightweight and tough to protect the hindwings without hindering flight capacities. Dynastes hercules elytra are known for their hygrochromic properties. However, the whole structure of the elytron remains to be characterized. Scanning Electron Microscopy (SEM), Transmission Electron Microscopy (TEM) and to our knowledge for the first time X-Ray tomography were undertaken on adult male Dynastes hercules to characterize their multi-scale structure. Trabeculae present a periodic arrangement over a short distance. Two inferred models describe the heights of plies in endocuticles of dorsal and ventral cuticles. The prevalence and consequences of genetic introgression between species have been intensively debated. I used Hercules beetles as examples to test for conditions that may be associated with the occurrence of introgression. RADseq data were used to reconstruct the species tree and history of introgression between Hercules beetles. Image data from museum specimens were used to investigate the phenotypic similarity of two adaptive traits between species from two distinct climatic realms (Nearctic vs. Neotropical). Genetic introgression was identified between Hercules beetles living in geographic proximity (parapatric). Phylogenetic relatedness and phenotypic similarity did not predict nor preclude genetic introgression between species. Phenotypic assimilation in body coloration was evident between distantly related Hercules beetles codistributed in Central America, where directional introgression was also statistically supported from the putative donor to receiver lineages. The number of introgressed loci was significantly higher between species with than without phenotypic similarity. I discuss the implications of recent studies on adaptive genetic introgression by providing supporting evidence from the Hercules beetle system. [ABSTRACT FROM AUTHOR]

With the recent attention and focus on quantitative methods for species delimitation, an overlooked but equally important issue regards what has actually been delimited. This study investigates the apparent arbitrariness of some taxonomic distinctions, and in particular how species and subspecies are assigned. Specifically, we use a recently developed Bayesian model-based approach to show that in the Hercules beetles (genus Dynastes) there is no statistical difference in the probability that putative taxa represent different species, irrespective of whether they were given species or subspecies designations. By considering multiple data types, as opposed to relying exclusively on genetic data alone, we also show that both previously recognized species and subspecies represent a variety of points along the speciation spectrum (i.e., previously recognized species are not systematically further along the continuum than subspecies). For example, based on evolutionary models of divergence, some taxa are statistically distinguishable on more than one axis of differentiation (e.g., along both phenotypic and genetic dimensions), whereas other taxa can only be delimited statistically from a single data type. Because both phenotypic and genetic data are analyzed in a common Bayesian framework, our study provides a framework for investigating whether disagreements in species boundaries among data types reflect (i) actual discordance with the actual history of lineage splitting, or instead (ii) differences among data types in the amount of time required for differentiation to become apparent among the delimited taxa. We discuss what the answers to these questions imply about what characters are used to delimit species, as well as the diverse processes involved in the origin and maintenance of species boundaries. This letter presents a biomimetic humidity sensor inspired by the humidity-dependent color change observed in the cuticle of the Hercules beetle. A thin-film-type humidity sensor with nanoporous structures (three-dimensional photonic crystals) mimicking the spongy multilayer in the beetles was designed and fabricated using the colloidal templating method and a hydrophilic surface treatment. The visible color of the fabricated humidity sensor changes from blue-green to red as the environmental humidity increases. The wavelength of reflected light that is predicted by Bragg’s equation considering the effect of water absorption shows a good agreement with experimental results. [ABSTRACT FROM AUTHOR]
