Heat stress negatively affects health and production in cows. Examining the cellular response to heat stress could reveal underlying protective molecular mechanisms associated with superior resilience and ultimately enable selection for more resilient cattle. This type of investigation is increasingly important as future predictions for the patterns of heat waves point to increases in frequency, severity, and duration. Cows identified as high immune responders based on High Immune Response technology (HIR) have lower disease occurrence compared to their average and low immune responder herd-mates. In this study, our goal was to identify epigenetic differences between high and low immune responder cows in response to heat stress. We examined genome-wide DNA methylation of blood mononuclear cells (BMCs) isolated from high and low cows, before and after in vitro heat stress. We identified differential methylation of promoter regions associated with a variety of biological processes including immune function, stress response, apoptosis, and cell signalling. The specific differentially methylated promoter regions differed between samples from high and low cows, and results revealed pathways associated with cellular protection during heat stress. The growing interest in the thermal comfort of dairy cows is justified, not only in countries located in tropical zones, but also in zones with temperate climate where high ambient temperatures are becoming a problem. The temperature-humidity index (THI) is a value representing the combined effect of air temperature and humidity associated with the level of heat stress. THI values lower than 72 mean that the cow's body is in favourable environmental conditions and is not subject to heat stress. At THI values of 75 to 78, the animal organism is under heat stress, but the mechanisms of thermoregulation still manage to cope, while at THI over 78 it is assumed that the stress is so high that it is impossible to maintain the thermoregulatory mechanisms or normal body temperature. This study compared physiological and productive parameters in ¾ Holstein × ¼ Gir dairy cows diagnosed or not with subclinical hypocalcemia (SCH) during early lactation. Nonlactating, multiparous cows (n = 32) were enrolled in this experiment 21 d prior to expected date of calving. Cows were maintained in a single pen with ad libitum access to corn silage before calving and received a limit-fed prepartum concentrate. Cow body weight (BW) and body condition score (BCS) were recorded weekly, and blood samples were collected on days −21, −14, −9, −6, and −3 relative to expected calving. After calving (day 0), cows were managed in a single pen with ad libitum access to a total mixed ration, and were milked twice daily. Cow BW and BCS were recorded upon calving and then weekly. Milk production was recorded daily and milk samples collected weekly until 30 d in milk (DIM). Blood was collected during the first 5 DIM, and at 6, 9, 16, 23, and 30 DIM. Cows were classified with SCH when mean total serum Ca during the first 5 DIM was ≤2.125 mmol/L. Cows diagnosed with SCH (n = 11) had less (P ≤ 0.04) mean BCS (2.85 vs. 3.07; SEM = 0.07) and less concentrations of serum insulin (0.396 vs. 0.738 ppmol/L; SEM = 0.115) and insulin-like growth factor I (35.9 vs. 57.9 ng/mL; SEM = 4.2), and these outcomes were noted since 21 d prior to expected calving. Cows diagnosed with SCH had greater (P < 0.01) serum concentrations of cortisol at calving (30.2 vs. 22.4 ng/mL; SEM = 2.0), serum haptoglobin at 3 and 6 DIM (0.453 vs. 0.280 mg/mL on day 3 and 0.352 vs. 0.142 mg/mL on day 6; SEM = 0.046), and tended (P = 0.09) to have greater mean concentrations of nonesterified fatty acids from calving to 30 DIM (0.368 vs. 0.304 μEq/L; SEM = 0.026). No differences were detected (P ≥ 0.41) for cow BW and milk production. Cows diagnosed with SCH had less (P = 0.05) mean concentrations of milk total solids (13.2 vs. 13.8 %; SEM = 0.21), tended to have less (P ≤ 0.10) mean concentrations of milk fat (4.34 vs. 4.81 %; SEM = 0.20), protein (3.31 vs. 3.45 %; SEM = 0.05), and lactose (4.45 vs. 4.55 %; SEM = 0.04), and had greater (P = 0.02) milk somatic cell count during the initial 14 DIM (504 vs. 140 cells/μL; SEM = 90). Collectively, Holstein × Gir cows diagnosed with SCH upon calving had altered periparturient physiological parameters denoting reduced energy nutritional, increased milk somatic cell count, and less concentration of milk components during early lactation compared with normocalcemic cows. The objective of this project was to develop and test a web-based virtual dairy herd to help students understand the structure and functioning of a dairy herd, and to promote active learning. At the beginning of the course, the instructor defines the profiles of herds to be assigned to students (e.g., herd size, production, diets, fertility). Each student has a unique herd and engages in decision-making for desired management practices in the herd. Modeled events are based on cow physiology and normal dairy herd management practices. Students' activities and decisions include heat detection, insemination, pregnancy diagnosis, dry-off, diet specifications, feeding groups, colostrum and milk-replacer feeding, weaning, treatment of diseases, and milk withdrawal from the tank if antibiotics are used, among others. The daily output provides information on technical indexes, economic performance, counters of incorrect decisions as feedback for students, and score. Time in class can be devoted to discussions of dairy management issues. Additional exercises based on students' own herds (e.g., calculating required space for cows, land for forage production, manure management) can also be implemented. Students' performance in the virtual dairy farm was monitored over 3 years. The average score (n = 326) was 87.8 ± 1.1 over 100 points, suggesting that self-learning with the virtual dairy farm was highly successful. At the end of each semester, students (n = 277) responded to a survey on the experience of working with the virtual dairy herd. Most students (>87%) agreed that the virtual dairy herd was more effective and motivating than traditional lectures and helped them understand dairy production better. In an unannounced test conducted at least 2 wk before the final exam, students (n = 207) were asked 14 questions on dairy cattle and 14 similar questions on other species taught in the same class through traditional lectures. A similar test on the same students (n = 142) was conducted in their fifth semester (2 years later). Results were better in dairy compared with other species questions in the first (9.6 vs. 3.7) and fifth (8.0 vs. 3.8) semesters. The virtual dairy herd is an effective tool for teaching introductory courses in dairy production. The effects of high stocking density during the dry period on dairy cow physiology, behaviour and welfare were investigated. Holstein Friesian cows (n = 48, calving over a seven month period) were dried-off 60 ± 4 d before their expected calving date, and allocated to either high (H) or low (L) stocking density groups. Cows were housed in cubicles from dry-off to 21 d before calving and then moved to straw yards until calving. In cubicle pens, H and L cows had 0.5 vs. 1.0 feed yokes/cow and 1.0 vs. 1.5 cubicles/cow, respectively, and in straw yards, 0.3 m vs. 0.6 m linear feed-face space and 6 m2 vs. 12 m2 lying space, respectively. Video observations of feeding behaviour during the 3 h after feed delivery (3 d/week) and agonistic interactions at the feed-face during peak feeding (2 d/week) were made. Daily lying proportion was measured using an accelerometer device throughout the dry period. Concentrations of faecal glucocorticoid metabolites (FGCM) at dry-off, d7 and d35 after dry-off and d21 and d7 before calving and the change in energy metabolites (glucose, NEFA, BHB) from dry-off to d7 before calving were measured. H cows were less likely to start feeding within 5 min of feed delivery and spent less time feeding compared to L cows, but they engaged in displacements more frequently and spent more time standing in the feed alley. Irrespective of the treatment groups, FGCM concentrations significantly increased from dry-off to d7 after dry-off and remained higher during the dry period. Stocking density did not affect daily lying proportion, energy metabolites during the dry period and milk yield during subsequent lactation. This study found that whilst high stocking density during the dry period increased competition at the feed-face and altered feeding behaviour, it did not affect stress responses, energy metabolism or subsequent milk yield. This experiment compared milk production, milk composition, and physiological responses in lactating dairy cows supplemented with or without a mixture of condensed tannins, encapsulated cinnamaldehyde, curcumin, capsaicin, and piperine. Thirty-six lactating, multiparous, pregnant ¾ Holstein × ¼ Gir cows were maintained in a single drylot pen with ad libitum access to water and a total-mixed ration and were milked twice daily (d –7 to 84). On d 0, cows were ranked by days in milk (86 ± 3 d), milk yield (27.8 ± 1.0 kg), body weight (BW; 584 ± 10 kg), and body condition score (BCS; 3.04 ± 0.06) and assigned to receive (SUPP; n = 18) or not (CON; n = 18) 30 g/cow daily (as-fed basis) of Actifor Pro (Delacon Biotechnik GmbH; Steyregg, Austria). From d 0 to 84, SUPP cows individually received (as-fed basis) 15 g of Actifor Pro mixed with 85 g of finely ground corn through self-locking headgates before each milking of the day. Each CON cow concurrently received 85 g (as-fed basis) of finely ground corn through self-locking headgates. Throughout the experimental period (d –7 to 84), cows from both treatments were administered 500 mg of sometribove zinc at 14-d intervals and were monitored daily for morbidity, including clinical mastitis. Individual milk production was recorded daily, whereas milk samples were collected weekly for analysis of milk composition. Cow BW, BCS, and blood samples were also collected weekly. Cows receiving SUPP gained more BCS (P = 0.05) and had greater (P = 0.04) milk yield during the experiment compared with CON cows (0.22 vs. 0.07 of BCS, SEM = 0.05; 29.5 vs. 27.9 kg/d, SEM = 0.5). Milk composition did not differ (P ≥ 0.15) between SUPP and CON cows; hence, SUPP cows also had greater (P ≤ 0.02) production of fat-corrected and energy-corrected milk. Incidence of clinical mastitis did not differ (P ≥ 0.49) between SUPP and CON cows. No treatment differences were also detected (P ≥ 0.21) for serum concentrations of glucose and serum urea N. Mean serum haptoglobin concentration during the experiment was greater (P = 0.05) in CON vs. SUPP cows. Cows receiving SUPP had less (P ≤ 0.04) serum cortisol concentrations on d 21 and 42, and greater (P ≤ 0.05) serum concentrations of insulin-like growth factor-I on d 7, 35, and 63 compared with CON cows (treatment × day interactions; P ≤ 0.02). Genetic mutations, including single nucleotide polymorphisms (SNPs), result in aberrant alternatively splicing of gene and involves in susceptibility of inflammatory diseases, including bovine mastitis. C-C motif chemokine ligand 5 (CCL5) is an immune-associated gene, but its alternative splicing (AS) mechanism of gene expression has not yet been understood. The present study identified the splice variant of CCL5 and the compared differential expression of various transcripts between healthy and mastitic mammary gland tissue from cows. A novel transcript lacking exon 2 with a deletion of 112 bp (referred to as CCL5-AS) was identified in the mammary gland. The expression of CCL5-AS was lower compared with CCL5-reference in the healthy and mastitic mammary tissues. A total of two novel SNPs (g.1647 C>T and g.1804 G>A) were identified in exon 2 of CCL5. Using the splicing mini-gene reporter assay in bovine mammary epithelial cells (MAC-T) and 293T cells, it was confirmed that the production of CCL5-AS was not caused by the two SNPs. The present findings suggested that alternative splicing is one of the mechanisms of CCL5 expression regulation and is involved in mastitis infection, but that genetic mutation was not responsible for the generation of the abnormal transcript of CCL5 in cows. Heat stress during late gestation in cattle negatively affects the performance of the dam and its calf. This brief exposure to an adverse environment before parturition affects the physiological responses, tissue development, metabolism, and immune function of the dam and her offspring, thereby limiting their productivity. During the dry period of a dairy cow, heat stress blunts mammary involution by attenuating mammary apoptosis and autophagic activity and reduces subsequent mammary cell proliferation, leading to impaired milk production in the next lactation. Dairy cows in early lactation that experience prepartum heat stress display reduced adipose tissue mobilization and lower degree of insulin resistance in peripheral tissues. Similar to mammary gland development, placental function is impaired by heat stress as evidenced by reduced secretion of placental hormones (e.g. estrone sulfate) in late gestation cows, which partly explains the reduced fetal growth rate and lighter birth weight of the calves. Compared with dairy calves born to dams that are exposed to evaporative cooling during summer, calves born to noncooled dry cows maintain lower BW until 1 yr of age, but display a stronger ability to absorb glucose during metabolic challenges postnatally. Immunity of the calves, both passive and cell-mediated immune function, is also impaired by prenatal heat stress, resulting in increased susceptibility of the calves to diseases in their postnatal life. In fact, dairy heifers born to heat-stressed dry cows without evaporative cooling have a greater chance leaving the herd before puberty compared with heifers born to dry cows provided with evaporative cooling (12.2% vs. 22.7%). Dairy heifers born to late-gestation heat-stressed dry cows have lower milk yield at maturity during their first and second lactations. Emerging evidence suggests that late-gestation heat stress alters the mammary gland microstructure of the heifers during the first lactation and exerts epigenetic alterations that might explain, in part, their impaired productivity. This study (1) investigated the effect of dietary transition at dry off on the behavior and physiology of Holstein dairy cows and (2) evaluated the efficacy of utilizing reductions in dietary nutrient density and milking frequency to mediate the dry-off process. Forty-eight cows were dried off over a 5-d period approximately 60 d before their expected calving dates. At the start of dry off (d 1), cows were randomly assigned to 1 of 2 total mixed rations that differed in nutrient density: higher nutrient density (HND; n = 24) and lower nutrient density (LND; n = 24). During dry off, cows were milked 1×/d on d 1, 2, and 3 and a final time on d 5. Milk yield, feeding behavior, rumination, lying behavior, and reticulorumen pH were monitored electronically and feed samples were collected to determine feed sorting. Blood samples, body condition scores, and body weights were taken periodically. Across treatments, milk yield was reduced by approximately 10 kg/d during dry off, with cows fed the LND diet producing 0.9 kg less milk at the time of dry off compared with cows fed the HND diet. Cows fed the LND diet consumed 2.2 kg less dry matter per day, fed at a slower rate, spent 15.7 min more feeding, and sorted to a greater extent for the smaller, more nutrient dense components of the diet compared with cows fed the HND diet. Despite sorting activity, cows fed the LND diet ruminated 3.3 min more per kilogram of dry matter consumed and had a mean reticulorumen pH 0.07 units higher compared with cows fed the HND diet. Following the start of dry off, cows spent approximately 5% more of their free time lying down. Dietary transition and dry off similarly affected the energy balance and inflammatory response of cows, although these parameters varied by day after the start of dry off. The results of this study indicate that the LND diet more effectively reduced milk production before dry off compared with the HND diet while not substantially altering cow physiology. The objective of the study was to analyse the effect of age at first calving (AFC) on first lactation milk yield, lifetime milk production and longevity of dairy cows. The study used SYMLEK data on the milk yield and culling of 111 857 Polish Holstein-Friesian cows from the active population in Pomerania and Kujawy, Poland. The cows first calved during 2000–2009 and were used or removed from the herds until the end of 2015. For each cow, calculations were made of first lactation milk yield, lifetime milk production, first lactation milk yield per milking day, lifetime milk production per day, as well as the lifespan (length of productive life, duration of rearing) and number of calvings. Based on the age at first calving (duration of the rearing period), the cows were grouped as follows: ≤22, 22.1–24.0, 24.1–26.0, 26.1–28.0, 28.1–30.0, 30.1–32.0 and >32 months. The GLM, CORR PEARSON and FREQ procedures from the SAS package were used in the statistical calculations. Considering first lactation milk yield, lifetime milk production and longevity, it is recommended that the cows should be first milked between 22.1 and 26.0 months of age. This is supported by the approximately 24% higher lifetime milk production in relation to the latest calving cows (P<0.01). Late commencement of the first milking (especially after 28 months) causes a considerable decrease in the first lactation milk yield and lifetime milk production, shortens the productive period, reduces the number of calvings, and increases culling rate due to low milk yield and udder diseases. Abstract Automation of the measurement of the physiological and behavioural parameters of livestock has become an important goal for both scientists and farmers. Accurate data and knowledge about farmed animals, especially in cattle breeding, are needed. Proper early diagnosis of a cow's health status in real time allows for preventing the development of infection, oestrus detection and leads to reduced environmental stress. Thus, it contributes to more effective herd management. Among the physiological parameters, body temperature and its fluctuations are key indicators of health and well-being in animals. Currently, along with the development of technical solutions and their implementation, increasingly more attention is being paid to the continuously measurement of body core and peripheral temperature in animals. Recently there has been an increased number of publications devoted to this subject. However, there is a need to systematise this knowledge as these studies have had different purposes, have been performed in various environmental conditions, and the measurements were taken using different methods and equipment. As such, the results obtained by the different authors often may not be comparable. For this reason, this paper has two main purposes: to present the most widely used continuous methods of peripheral and body core temperature measurement, and to show its references values which characterise the individual locations of the cattle body in thermoneutral ambient. An analysis of the professional publications regarding measurements of peripheral and deep body temperature led to the conclusion that these methods have high research and diagnostic potential. However, it is necessary to standardised research to enable better and more comparable results, including among others; different cattle groups, animal age, health and environmental conditions. Abstract In the present study we describe an outbreak where PVL positive MRSA belonging to spa -type t002 and multi-locus sequence type ST2659 persisted in a Swedish dairy herd for at least two years, despite efforts to hinder transmission between animals and between the farmer and his animals. This is the first description of persistence and spread of MRSA in a dairy herd in Sweden. Sampling of animals in the herd was initiated by the finding of MRSA in the farmer and was performed at eight occasions from November 2012 to September 2014. In total, MRSA was detected in 25 animals and in 16 of these MRSA was detected in milk samples. In addition, MRSA was also detected in bulk milk samples. Whole genome sequencing (WGS) of twelve isolates from farmer (n = 1), animals (n = 9) and bulk milk (n = 2) revealed high relatedness, implying a common source. MRSA may initially have been transmitted from humans to cows with further spread within the herd. WGS showed minor differences in one isolate (loss of phage ΦN315) which could indicate adaption of the strain to an animal host. [ABSTRACT FROM AUTHOR]

Intrapartum uterine torsion is a common cause of dystocia in dairy cows. To clarify the pathophysiology of this disease, the aim of the present study was to examine serum neuropeptide and cytokine concentrations of cows with intrapartum uterine torsion. Blood samples of 20 cows with uterine torsion, 36 healthy controls, and 15 intrapartum cows without uterine torsion were obtained. Concentrations for substance P (SP), vasoactive intestinal polypeptide (VIP), and interleukin-1β (IL-1β) were quantified by using commercially available ELISA kits. Significant differences between groups were observed for SP in cows with uterine torsion and cows calving normally ( P  <  0.01). Markedly greater SP concentrations were observed in calving cows than in cows with uterine torsion. Compared with healthy controls, there were greater SP concentrations during parturition ( P  <  0.01). No significant group differences were detected for VIP concentrations. Global differences were observed in IL-1β ( P  =  0.04). Large amounts of SP are released into the blood during parturition. Because SP is mainly present within the cervix and functions as a biomarker and mediator of pain, cows with uterine torsion are presumed to not experience as much pain as cows that are calving normally. Consistent with this, in cows with uterine torsion, there is a disturbance in the opening of the cervix, along with only mild signs of colic. Furthermore, significantly elevated IL-1β concentrations correlate with the inflammation that occurs in cases of uterine torsion. Summary: The relationship between in vitro rumen CH4 production of grass silages, using the gas production technique, and in vivo data obtained with the same cows and rations in respiration chambers was investigated. Silages were made from grass harvested in 2013 on May 6th, May 25th, July 1st and July 8th. The grass silages were used to formulate four different rations which were fed to 24 cows in early and late lactation, resulting in a slightly different dry matter intake (DMI; 16.5 kg/day vs. 15.4 kg/day). The experimental rations consisted of 70% grass silage, 10% maize silage, and 20% concentrates on a dry matter basis. Cows were adapted to the rations for 17 days before rumen fluid was collected via oesophageal tubing, and in vitro gas and CH4 production were analysed. In vitro total gas and CH4 production of the (ensiled) grass expressed as ml/g OM decreased with advancing maturity of the grass. The in vitro CH4 production after 48 hr of incubation expressed in ml/g OM did not correlate with the in vivo CH4 production expressed in g/kg organic matter intake or g/kg DMI (R2 = .00–.18, p ≥ .287). The differences in CH4 emission per unit of intake observed in vivo were rather small between the different rations, which also contributed to the observed poor relationship. Utilizing stepwise multiple regression improved the correlation only slightly. In vitro gas and CH4 production varied based on whether donor cows were previously adapted to the respective ration or not, suggesting that careful adaption to the experimental diet should be envisaged in in vitro gas and CH4 production experiments. The aim of the present study was to examine the effect of 28 days of dietary difructose anhydride (DFA) III supplementation on calcium (Ca) metabolism in late-lactation dairy cows. Twenty-four multiparous pregnant Holstein cows were divided into two groups. The DFA group was fed total mixed ration (TMR) supplemented with 40 g of DFA III, and the control group was fed TMR only. The replenishment of bone Ca reserves was evaluated by measuring bone mineral density (BMD) and blood biochemical bone markers. Serum Ca concentrations, urinary Ca-to-creatinine (Cre) (Ca/Cre) ratios, and milk Ca concentrations were also analyzed. The BMD of the 4th caudal vertebra in the DFA group was higher than in the control group on day 28. With respect to bone markers, the ratios of undercarboxylated osteocalcin (ucOC) to osteocalcin (OC) in the DFA group were significantly lower than those in the control group on days 21 and 28. Milk Ca concentrations in the DFA group were also higher than those in the control group on days 14, 21, and 28, whereas serum Ca concentrations and urinary Ca/Cre ratios were unchanged in both groups. These results suggest that dietary supplementation with DFA III increased BMD and decreased serum ucOC/OC ratios in late-lactation dairy cows; this indicates that the replenishment of bone Ca reserves may be enhanced by dietary DFA III supplementation. Aim: The current study aimed to compare the impact of heat stress (HS) on some physiological functions and blood oxidative stress biomarkers between dry dairy crossbred (Balady X Friesian) cows and buffaloes during Egyptian summer season (July-September). Materials and Methods: A total of 26 healthy animals were equally used in the in the current study. The criterion for cows and buffaloes selection and the management conditions were similar. A total mixed ration to meet the animal's requirements was used, and dry matter intake (DMI) was calculated. Ambient temperature, relative humidity, temperature humidity index (THI), respiratory rate, and rectal temperature (RT) were daily recorded. Meanwhile, live body weight and body condition score were weekly recorded. Blood samples were collected bi-weekly, and plasma samples were harvested for malondialdehyde (MDA) content and enzymatic antioxidants such as glutathione peroxidase, superoxide dismutase, and catalase activities determinations throughout the experimental period (8 weeks - prepartum). Results: The results confirmed, the HS condition, as the THI values ranged from 79.74 to 90.4 throughout the experimental period. In both species, HS increased RT and decreased DMI (<10.5 kg/day and 9.5 kg/day in cows and buffaloes, respectively). Buffaloes seemed to be more affected by the hostile environmental condition of this study compared with their respective cows. Buffaloes had recorded up to 1 °C increase in their RTs in most of the point's period compared to cows. There was a continuous increase in MDA values (194.7 and 208.4 nmol/gHb in buffaloes and cows, respectively, 2 weeks prepartum) as the animals come close to parturition with moderate decrements for the enzymatic antioxidant activities in both cows and buffaloes. We assessed dairy cows’ body postures while they were performing different stationary activities in a loose housing system and then used the variation within and between individuals to identify potential connections between specific postures and the valence and arousal dimensions of emotion. We observed 72 individuals within a single milking herd focusing on their ear, neck and tail positions while they were: feeding from individual roughage bins, being brushed by a mechanical rotating brush and queuing to enter a single automatic milking system. Cows showed different ear, neck and tail postures depending on the situation. When combined, their body posture during feeding was ears back up and neck down, with tail wags directed towards the body, during queuing their ears were mainly axial and forward, their neck below the horizontal and the tail hanging stationary, and during brushing their ears were backwards and asymmetric, the neck horizontal and the tail wagging vigorously. We then placed these findings about cow body posture during routine activities into an arousal/valence framework used in animal emotion research (dimensional model of core affect). In this way we generate a priori predictions of how the positions of the ears, neck and tail of cows may change in other situations, previously demonstrated to vary in valence and arousal. We propose that this new methodology, with its different steps of integration, could contribute to the identification and validation of behavioural (postural) indicators of how positively or negatively cows experience other activities, or situations, and how calm or aroused they are. Historically sulfonated steroids were primarily considered as inactive metabolites destined for elimination. However, more recently they have been increasingly recognized as precursors for the production of bioactive steroids in target tissues and as functional molecules without preceding hydrolysis. In order to comprehensively characterize their occurrence in cyclic cows and their formation and hydrolysis in bovine ovarian steroidogenesis, ovaries from cyclic cows were screened for the expression of oestrogen sulfotransferase (SULTE1) and steroid sulfatase (STS) by Western blot and immunohistochemistry. Moreover, a broad spectrum of 13 sulfonated steroids was measured applying liquid chromatography-tandem mass spectrometry (LC–MS/MS) in blood samples collected from three cycling heifers during defined stages of the ovarian cycle and in fluid obtained from ovarian follicles of different size. SULT1E1 was undetectable in ovarian tissues. For STS only a weak immunostaining was found predominantly in granulosa cells of larger follicles. However, no specific band occurred in Western blot. In blood, concentrations of all sulfonated steroids investigated were below the limit of quantification (LOQ). In follicular fluid, only cholesterol sulfate was measured in considerable concentrations (328.3 ± 63.8 ng/ml). However, the role of cholesterol sulfate in bovine follicular steroidogenesis remains unclear as concentrations were obviously unrelated to follicular size. The remaining sulfonated steroids investigated were undetectable or only slightly exceeded LOQ in a minor proportion of samples. Rumen microbial biohydrogenation (BH) of unsaturated fatty acids (UFA) has been extensively studied in vitro; however, in vitro BH pathways, rates, and extents may not parallel those in vivo. The objective was to develop an assay to assess in vivo rates, pathways, and extent of BH of oleic (OA), linoleic (LA), and α-linolenic (ALA) acids. Each UFA was characterized in a separate experiment, each using 4 ruminally cannulated lactating Holstein cows. A single bolus consisting of 200 g of a UFA-oil [experiment 1 (EXP1): 87% OA sunflower, experiment 2 (EXP2): 70% LA safflower, and experiment 3 (EXP3): 54% ALA flaxseed] and 12 g of heptadecanoic acid (C17:0) was mixed into the rumen through the fistula. Rumen digesta was collected at −1, −0.25, 0.1, 0.25, 0.5, 0.75, 1, 1.5, 2, 3, 4, and 6 h relative to the bolus. Overall, the triglyceride boluses increased total fatty acids (FA) in the rumen from 3.9 (standard deviation = ±1.4) to 7.3% (±1.4) of rumen dry matter and enriched C17:0 from 0.4 (±0.1) to 2.5% (±0.5) of FA. The bolus enriched OA from 8.9 (±1.0) to 30.1% (±4.6) of FA in EXP1, LA from 11.1 (±1.8) to 35.9% (±5.0) of FA in EXP2, and ALA from 2.1 (±0.1) to 19.8% (±4.3) of FA in EXP3. The disappearances of C17:0, OA, LA, and ALA were fit to a single exponential decay model. The first-order rate of C17:0 rumen disappearance (turnover) was 9.1, 6.9, and 5.2%/h in EXP1, EXP2, and EXP3, respectively, and was used as a marker of FA passage. The rate of total rumen turnover of OA was 54.1%/h, LA was 60.5%/h, and ALA was 93.0%/h in EXP1, EXP2, and EXP3, respectively. Rumen concentration of all 3 UFA reached prebolus concentrations within 4 h. The calculated extent of lipolysis and initial isomerization was 85.6% for OA, 89.8% for LA, and 94.7% for ALA in EXP1, EXP2, and EXP3, respectively. Assuming that BH equals total disappearance minus passage, the rates of lipolysis and initial isomerization were 45.0, 53.6, and 87.8%/h for OA, LA, and ALA in EXP1, EXP2, and EXP3, respectively. Analysis of the data using compartmental modeling showed that the normal BH pathways proposed in the literature explained 46.0, 37.3, and 49.8% of the BH of OA, LA, and ALA in EXP1, EXP2, and EXP3, respectively. Based on the model, BH of trans C18:1 FA was the rate-limiting step to complete BH. Importantly, oils were provided as triglycerides and the reported rates represent the rate of lipolysis and BH. In conclusion, the rate of ruminal BH of OA, LA, and ALA was higher than that commonly observed in vitro, but the extent of BH was near expected values. Abstract: When European Union regulations restricted the use of estrogenic compounds in food‐producing animals, refined hormonal protocols were no longer applicable for anovulatory cows. However, Ovsynch and its adaptations are routinely and uniformly applied to all cows regardless of ovarian function. To evaluate their efficacy on anovulatory cows, 143, 147 and 144 anovulatory cows received Ovsynch, Presynch and G6G protocols, respectively. In comparison, 150 cyclic cows were bred without using a synchronized protocol. Results showed that cows in the Presynch group had luteolysis responding to the last prostaglandin F2α (PGF2α) injection greater than the Ovsynch group. The serous progesterone levels at the first gonadotropin‐releasing hormone of Ovsych and the last PGF2α injection was greater in the G6G group than the other two hormonal treatment groups. Concentrations of Ca2+ and total protein in cervical mucus in all three hormone‐treated groups before artificial insemination (AI) were significantly different from the controls. The G6G group obtained a greater pregnancy rate compared with Ovsynch and Presynch, but significantly less than the controls. For open cows in the Ovsynch group, estrus rate within 24 days after the first AI was significantly less than the controls. This study aimed to evaluate the effect of supplementing biotin (BIO), nicotinamide (NAM) and their interaction on feed intake, milk production, and energy metabolism of transition dairy cows. Forty multiparous Holsteins were assigned randomly within block to 1 of the following 4 treatments: control (no supplementation), 30 mg/d BIO, 45 g/d NAM (ruminally unprotected), and 30 mg/d BIO + 45 g/d NAM. Cows were fed the same basal diet and received the supplements in drenches daily from about d −14 to 35 relative to parturition. Feed intake, energy balance and calf birth weight were not affected by BIO supplementation whereas NAM supplementation tended to increase prepartum dry matter intake and decrease body weight loss. Tendencies of BIO × NAM × week interaction were observed on energy balance during prepartum and entire period. The interaction of BIO × NAM was found on milk lactose percentage and fat yield. Supplementation of BIO resulted in increased concentration of BIO in plasma and decreased triglyceride concentration. The BIO × week interaction effects were detected for triglyceride, very-low-density lipoprotein cholesterol and low-density lipoprotein cholesterol concentrations. Supplementation of NAM increased plasma NAM and glucose while decreased nonesterified fatty acid, β-hydroxybutyric acid, triglyceride, high-density lipoprotein cholesterol and low-density lipoprotein concentrations. The BIO × NAM × week interaction effect was observed on nonesterified fatty acid concentration during entire period. Blood insulin and total cholesterol concentrations were not affected. Supplementing BIO and NAM in a combination did not show additional benefits above those observed for BIO or NAM supplementation alone for most of the variables. In conclusion, drenching ruminally unprotected NAM at a high dose (45 g/d) is effective to provide utilizable NAM and elicit beneficial effects on the energy metabolism of transition dairy cows. The objective of this study was to evaluate the safety of a standardized Macleaya cordata Extract Product (MCEP) containing the quaternary benzophenanthridine alkaloids, sanguinarine and chelerythrine, when fed to dairy cows. Thirty-six dairy cows were randomized into three groups with twelve cows/treatment in two replica pens for each treatment group: control (C) without MCEP added to feed, treatment 1 ( SANG-1000) with MCEP added to feed at 1,000 mg/animal/day (1.5 mg/kg bw/day) and treatment 2 ( SANG-10000) with MCEP added to feed at 10,000 mg/animal/day (15.5 mg MCEP/kg bw/day). After two weeks of acclimation, animals were observed for an 84-day experimental period, with body weight, feed intake and milk production measured daily. Milk composition was analysed every two weeks. Haematological analyses were performed on Day 0 and Day 84, and clinical chemistry analyses were performed on Day 84 of the study. There was no statistically significant difference ( p > .10) among the three groups on body condition score, milk production or milk composition over the study period. There were no significant differences in body weight gain or feed consumption among the three groups. Animals in the SANG-10000 group had significantly higher mean corpuscular volume ( MCV) than the C group ( p < .1) and lower mean corpuscular haemoglobin concentration ( MCHC) than the SANG-1000 group ( p < .1). Concentrations of sanguinarine and chelerythrine in milk samples collected on Day 84 were below the detection limit ( LOD) as measured by high-performance liquid chromatography-mass spectrometry ( HPLC- MS/ MS). In conclusion, this study presents compelling data supporting the hypothesis that the test product MCEP, when included in the TMR at up to 10,000 mg/animal/day (15.5 mg MCEP/kg bw/day), is well tolerated by dairy cows. The study was conducted during the regular milk control of cows from the Bulgarian Brown Cattle (BBC) breed from the Agricultural Institute - Stara Zagora herd for the months of July and October 2014. Animals were divided into two groups according to the daily milk level - Elite group with a daily milking density of over 20 kg and a First group with a milk density of up to 20 kg. The ration consists of a total mix ration that was provided in the feeding racks. The milk urea content was determined in the laboratory of the Agricultural Institute - Stara Zagora by the Angelov, Ibrisimov and Milashki's method (1999). Milk samples for analysis were taken during the morning milking of the cows and studied immediately. The results of the survey were statistically processed with the STATISTICA for Windows software package. The study provides the opportunity to supplement information on the impact of protein and energy levels on the ration for maximizing the use of protein feeds effectively and limiting nitrogen losses in the environment. Concentration of crude protein above 16% in the dairy cow diet with sufficient energy for microbial growth, is associated with poor utilization of the protein - increase in urea level in milk above 20.0 mg/dl. Low levels of urea in milk - below 6.0 mg/dl are associated with insufficient amount of microorganism-accessible rumen protein (PBR), although the availability of enough energy in the diet. Two experiments were conducted with the aim of determining the effect of equine chorionic gonadotropin (eCG) administration on day 14 after insemination on ovarian response and pregnancy establishment in postpartum anestrous beef cows. In both experiments, cows were subjected to a progesterone- and estradiol-based treatment for fixed-time artificial insemination (FTAI) and were randomly allocated into 4 groups to receive or not receive eCG (400 IU) at the time of device removal and/or at 14 d after FTAI. In experiment 1, from day 14 to 22, daily ultrasonographic determinations were performed to monitor ovarian dynamics, and blood was collected to determine hormone concentrations in 60 cows. In experiment 2, confirmation of pregnancy was performed at 30 and 60 d after FTAI in 1,060 anestrous cows assigned to the same experimental design. Cows that received eCG on day 14 after FTAI showed increases in corpus luteum area ( P < 0.01), follicle diameter ( P < 0.05), serum progesterone concentrations ( P < 0.01), and estradiol-17β concentrations ( P < 0.01), compared with cows that did not receive eCG on day 14. Pregnancy rate on day 30 was greater in those cows that received both eCG treatments (ie, at device removal and 14 d after insemination) than in those that did not receive eCG treatment ( P < 0.05). In conclusion, eCG administered on day 14 after FTAI increases serum progesterone concentrations during the critical period of pregnancy in anestrous cows, and this second eCG treatment seems to have a positive effect on achieving pregnancy. The objective of this work was to evaluate proliferation and apoptosis in the bovine ovary in a model of follicular persistence induced by low levels of progesterone to detect incipient changes during cystic ovarian disease development on the expected day of ovulation (day 0) and after 5, 10, and 15 days of follicular persistence. We analyzed cell proliferation by evaluating the expression of Ki-67 and apoptosis by evaluating caspase-3, BAX, and BCL2 expression. Proliferation was similar in the granulosa and theca cells of antral follicles in the P0 group (treated with progesterone up to the expected day of ovulation) and in the control group. A decrease in cell proliferation was detected after 5 days of persistence (P5) in relation to P0 (p < 0.05). Similar changes were found in the granulosa cells of the persistent follicles in relation to the control group (p < 0.05). Caspase-3 expression was similar in granulosa cells of antral follicles at early stages of persistence, with an increase after 15 days of persistence (p < 0.05). In the granulosa cells of group P10 (10 days of persistence), caspase-3 expression was reduced relative to that of antral follicles from the control group (p < 0.05). BCL2 expression was higher in granulosa cells of the persistent follicles of group P0 relative to the control follicles, with no changes in BAX expression, which was increased in persistent follicles of group P15 (p < 0.05). Similar results were observed in theca cells at initial stages of persistence. The results show that, initially, proliferation is maintained with low apoptosis and an increase in cell survival. There is little information on methane (CH 4 ) emissions from non-lactating pregnant dairy cattle when grazing. To address this issue, enteric CH 4 emissions were estimated from a total of 68 non-lactating pregnant Holstein-Friesian dairy cattle during three successive years (23, 22 and 23 cows in year 1, 2 and 3, respectively). Cows used in these studies ranged from 19 to 69 days prior to their actual calving date when measurements commenced. Methane emissions were measured using the sulphur hexafluoride (SF 6 ) technique over six consecutive 24-h periods, with measurements commencing 28 August, 16 September and 15 September during years 1–3, respectively. Cows were given access to fresh grazing on the first and fourth day of the measurement period, but were not offered a concentrate supplement. Across the three years of the experiment mean CH 4 emissions were 216 g/cow/day and 25.2 g/kg dry matter intake. Emission increased with increasing lactation number, from 200 (1st lactation) to 228 (> 2nd lactation) g/day. Relationships between individual cow body weight and methane emissions (R 2 = 0.17) and estimates of intakes for individual cows and CH 4 emissions (R 2 = 0.21) were poor. [ABSTRACT FROM AUTHOR]

The effects of 50% or 100% of a herbage diet of plantain on milk production and urinary nitrogen (N) concentration were measured in two experiments for late (autumn 2015) and early (spring 2015) lactation dairy cows. Three groups of 12 mixed age Friesian × Jersey dairy cows were offered a perennial ryegrass-white clover pasture, or pure plantain or 50% perennial ryegrass-white clover and 50% pure plantain by ground area (50–50 pasture–plantain). Urine N concentration was lower in both experiments (P< .001) for plantain (2.4 and 2.2 g N/L) and 50–50 pasture–plantain (3.6 and 3.4 g N/L) than pasture (5.4 and 4.7 g N/L). Cows on plantain or 50–50 pasture-plantain produced at least as much milk as those on pasture in both experiments. Plantain may offer environmental benefits to dairy systems by reducing the N concentration of urine deposited on the soil from grazing cows. [ABSTRACT FROM PUBLISHER]

Background: Lactation is extremely important for dairy cows; however, the understanding of the underlying metabolic mechanisms is very limited. This study was conducted to investigate the inherent metabolic patterns during lactation using the overall biofluid metabolomics and the metabolic differences from non-lactation periods, as determined using partial tissue-metabolomics. We analyzed the metabolomic profiles of four biofluids (rumen fluid, serum, milk and urine) and their relationships in six mid-lactation Holstein cows and compared their mammary gland (MG) metabolomic profiles with those of six non-lactating cows by using gas chromatographytime of flight/mass spectrometry. Results: In total, 33 metabolites were shared among the four biofluids, and 274 metabolites were identified in the MG tissues. The sub-clusters of the hierarchical clustering analysis revealed that the rumen fluid and serum metabolomics profiles were grouped together and highly correlated but were separate from those for milk. Urine had the most different profile compared to the other three biofluids. Creatine was identified as the most different metabolite among the four biofluids (VIP = 1.537). Five metabolic pathways, including gluconeogenesis, pyruvate metabolism, the tricarboxylic acid cycle (TCA cycle), glycerolipid metabolism, and aspartate metabolism, showed the most functional enrichment among the four biofluids (false discovery rate < 0.05, fold enrichment >2). Clear discriminations were observed in the MG metabolomics profiles between the lactating and non-lactating cows, with 54 metabolites having a significantly higher abundance (P < 0.05, VIP > 1) in the lactation group. Lactobionic acid, citric acid, orotic acid and oxamide were extracted by the S-plot as potential biomarkers of the metabolic difference between lactation and non-lactation. The TCA cycle, glyoxylate and dicarboxylate metabolism, glutamate metabolism and glycine metabolism were determined to be pathways that were significantly impacted (P < 0.01, impact value >0.1) in the lactation group. Among them, the TCA cycle was the most up-regulated pathway (P < 0.0001), with 7 of the 10 related metabolites increased in the MG tissues of the lactating cows. Conclusions: The overall biofluid and MG tissue metabolic mechanisms in the lactating cows were interpreted in this study. Our findings are the first to provide an integrated insight and a better understanding of the metabolic mechanism of lactation, which is beneficial for developing regulated strategies to improve the metabolic status of lactating dairy cows. A direct comparison of cow and goat performance and milk fatty acid (FA) responses to diets known to induce milk fat depression in the bovine has suggested interspecies differences in rumen and mammary lipid metabolism. Thus, this study was conducted to infer some potential mechanisms responsible for the differences in mammary lipogenesis due to diet and ruminant species. To meet this objective, 12 cows and 15 goats were fed a basal diet (control), a similar diet supplemented with 2.2% fish oil (FO), or a diet containing 5.3% sunflower oil and additional starch (+38%; SOS) according to a 3 × 3 Latin square design with 26-d experimental periods. Milk yield, milk composition, FA profile, and FA secretion were measured. On the last day of each period, the mRNA abundance of 19 key genes in mammary metabolism or the enzyme activity or both were measured in mammary tissue sampled by biopsy or at slaughter or both. The results show significant differences in the response of cows and goats to the dietary treatments. In cows, milk fat content and yield were lowered by FO and SOS (−31%), whereas only FO decreased milk fat content in goats (−21%) compared with the control. In cows and to a lesser extent in goats, FO and SOS decreased the secretion of C16 FA output (mmol/kg of BW). However, SOS increased the secretion of >C16 FA in goats. These changes in milk fat content and FA secretion were not associated with modifications in mammary expression or the activity of 19 proteins involved in the major lipogenic pathways. This absence of variation may be attributable to posttranscriptional regulation for these genes or related to the time of sampling of the mammary tissue relative to the previous meal and milking. Otherwise, the abundances of 15 mRNA among the 19 encoding for genes involved in lipid metabolism in the mammary gland were different among species, with 9 more abundant in cows (FASN, FADS1, SCD1, GPD1, LALBA, SREBF1, LXRA, PPARA, and PPARG1) and 6 more abundant in goats (G6PD, GPAM, SCD5, XDH, CSN2, and SP1). Similarly, a significant effect of the species was observed in the 4 enzyme activities measured; glycerol-3-phosphate dehydrogenase and malic enzyme were higher in cows, and FA synthase and glucose-6-phosphate dehydrogenase activities were higher in goats. In conclusion, the differences between cow and goat performance and milk FA responses to the FO and SOS treatments were not related to changes in the measured mammary lipogenic gene expression. Furthermore, the data provide evidence that the major mammary lipogenic pathways differ between the caprine and the bovine, whose biological significance remains to be unraveled. Eight multiparous periparturient Holstein cows fitted with ruminal cannula were used in a split plot design to evaluate the effects of monensin on plasma glucose metabolism. Diets were top-dressed daily with 0 mg/cow of monensin (control) or 300 mg/cow of monensin (MON) both pre- and postpartum. Plasma glucose kinetic parameters on d −13 ± 2.0 and 19 ± 1.6 relative to parturition were determined by using stable isotopes. Na-1-13C3-Propionate (labeled propionate) was infused into the rumen to measure glucose synthesis originating from ruminal propionate, and U-13C-glucose (labeled glucose) was injected into the jugular vein to determine total glucose kinetics. A sampling period of 480 min following labeled glucose injection was implemented. A compartmental analysis was employed to determine steady state glucose kinetic parameters. To develop a steady state glucose model, the Windows version of SAAM software (WinSAAM) was used. A 4-compartment model was adequate to comprehensively describe plasma glucose metabolism. The main model compartments consisted of propionate and plasma glucose. The time frame of the 480-min sampling period post-tracer glucose infusion allowed accurate quantification of glucose metabolism. The model estimated that glucose input from sources other than ruminal propionate decreased with MON, from 2.26 to 1.09 g/min postpartum. Gluconeogenesis, expressed as the propionate contribution to the plasma glucose pool, increased in cows fed MON (22 vs. 31%), whereas glucose oxidation, expressed as the glucose disposal rate, significantly decreased (1.67 vs. 0.92 g/min). In conclusion, MON may improve the energy status of transition cows by (1) improving the efficiency of propionate to produce glucose and (2) decreasing glucose oxidation in body tissues. This study aimed to evaluate the effect of dietary integration of dried olive pomace (DOP), a by-product of olive oil separation, on nutritional and aromatic properties of milk and cheese. Twenty dairy cows were divided into 2 groups that were balanced for milk yield, parity, and days in milk. The control group was fed a conventional diet (20 kg of dry matter/head per day), whereas the experimental group (EG) received the conventional diet supplemented with DOP as 10% of dry matter. During the trial, milk yield was recorded and the samples of milk, cheese, total mixed rations, and DOP were collected and analyzed to determine the chemical–nutritional composition and aromatic profile. Atherogenic and thrombogenic indices were calculated on the basis of the fatty acid (FA) profile of milk and cheese. Data were analyzed according to the mixed model for milk yield and chemical composition, including cows nested within treatment as a random effect, whereas the general linear model was used for the analysis of cheese parameters. Differences were assessed by Tukey's test. The EG diet had a lower content of palmitic, stearic, and linoleic acids and a higher level of oleic acid compared with the control. Dietary DOP integration did not affect milk yield and composition with the exception of protein content, which was greater in EG and significantly affected by diet and period. Instead, period was found to be significant for fat and casein in both groups. Dietary supplementation with DOP modified the FA profile of milk and cheese. There was a decrease in short- and medium-chain FA, but significance was achieved only for palmitic acid. The stearic, isomer trans of oleic (in particular vaccenic acid), oleic, and isomer trans of linoleic acids significantly increased. Monounsaturated FA increased in EG milk and cheese and saturated FA were significantly lower, whereas no difference was marked between the groups regarding level of polyunsaturated FA. Supplementation with DOP reduced atherogenic and thrombogenic indices and increased conjugated linoleic acid in both milk and cheese. The free fatty acids, ketones, lactones, esters, and phenylalanine catabolites were increased in raw milk, whereas only leucine metabolism was affected by diet in pasteurized milk cheese at both 1 and 30 d of ripening. Despite its low NDF digestibility, sugarcane is an option for feeding dairy cattle in tropical regions. We evaluated the effect of sugarcane silages inoculated with Lactobacillus hilgardii CCMA 0170 (LH; an epiphytic bacteria isolated from sugarcane) or with L. buchneri NCIMB 40788 (LB; a commercial strain isolated from temperate grasses) on dairy cow performance and feeding behavior. The microbial inoculums were previously grown in the laboratory to obtain 5 log cfu/g of fresh forage. Nine tons of each inoculated silage and a noninoculated control silage (CON) were harvested from the same field and stored for at least 35 d in experimental 20 × 2.1 × 0.4 m bunker silos. Fifteen Holstein cows in late lactation (336 ± 175 days in milk at the start of the experiment) received the treatments in five 3 × 3 Latin squares with 21-d periods. The diets contained 20% of DM of sugarcane silage and 41% of DM of corn silage. Milk yield was increased from 18.0 kg/d for CON to18.8 kg/d for LH, but LB did not elicit a detectable increase in milk yield (18.1 kg/d). The daily yields of fat, protein, lactose, and total solids were increased by LH. Daily DMI and total tract apparent digestibility of nutrients did not differ among treatments. Both inoculated silages reduced acetate and increased butyrate proportions in ruminal VFA, but only LH silage reduced the acetate to propionate ratio (3.0 vs. 3.3). First meal duration was shorter for CON compared to LH and LB. The proportion of daily intake between 0700 and 1300 h tended to be increased, and the proportion between 1900 and 0700 h was reduced by LH. The inoculation of sugarcane silage with Lactobacillus hilgardii affected rumen fermentation profile and feeding behavior of late lactation dairy cows, increasing the yield of milk solids. The study objective was to evaluate the effects of reducing dietary starch content in fresh cow diets while maintaining NDF levels by substituting barley grain (BG), corn silage (CS), or both with beet pulp (BP) on nutrient digestion, ruminal fermentation, DMI, lactation performance, meal patterns, chewing activity, and sorting behavior. Thirty-six multiparous cows were randomly assigned to 1 of 4 experimental diets from calving to 21 d in lactation. Experimental diets were a high-starch diet with ground BG (CO; 24.9% starch; 0% BP) and 3 lowstarch diets: the CO with BP substituted for ground BG (BB; 19.6% starch; 7% BP), the CO with BP substituted for CS (BC; 20.6% starch; 12% BP), or the CO with BP substituted for CS and ground BG (BCB; 20.3% starch; 12% BP). The CO, BB, BC, and BCB contained 33.4, 34.4, 32.9, and 33.7% NDF, respectively, and 23.1, 22.0, 18.2, and 19.2% forage NDF, respectively. There was no effect of diet on total tract nutrient digestibility (P > 0.20), ruminal pH (P = 0.49), or total VFA (P = 0.39). However, diets affected molar proportions of propionate and acetate (P ≤ 0.01). Relative to CO cows, the molar propionate percentage was less in BB cows (P < 0.01) and tended to be greater in BC cows (P = 0.10), whereas the molar percentage of acetate was less in BC cows than in BB (P < 0.01) and CO cows (P = 0.02). Relative to CO cows (16.50 kg/d), DMI was greater for BC (17.70 kg/d; P < 0.01) and BCB cows (17.50 kg/d; P = 0.01), but it was less in BB cows (15.60 kg/d; P = 0.02). Similar to DMI results, milk yields tended to be greater for BC (37.89 kg/d; P = 0.08) and BCB cows (37.81 kg/d; P = 0.09) compared with CO cows (35.41 kg/d), but BB cows (33.05 kg/d) tended to produce less milk than CO cows (P = 0.1). Milk fat content tended to be less in BC (P = 0.08) and BCB cows (P = 0.10) than in CO cows. There was no effect of diet on eating and rumination patterns (P ≥ 0.18), except shortened meal intervals for BC and BCB cows compared with CO cows (P = 0.02). Dietary treatments did not affect chewing activities per day or bout (P = 0.50), but BC and BCB cows spent less time on chewing activities per kilogram DM (P < 0.01) and NDF intake (P < 0.01) compared with CO cows. Cows fed the BC and BCB sorted for long particles (>19 mm; P < 0.01) but against particles < 1.18 mm (P < 0.01). In the present study, propionate did not seem to play a central role in feed intake regulation of fresh cows, because BB cows had depressed DMI and the lowest molar proportion of propionate. California summers are hot, compromising the welfare and productivity of dairy cows. To minimize negative efects, producers use shade, fans and sprayed water. However, little is known about how those heat abatement strategies are provided in commercial conditions, nor their efectiveness. Ten dairies with drylots, a common housing system in California, were assessed for strategies provided, and the cows' responses to heat load were observed for 3 days in the afternoon. Dairies were diverse in all aspects. Shade varied in terms of placement (at corral and feed bunk or at corral only) and amount (28 to 74 square feet, or 2.6 to 6.9 square meters, per cow). The quantity of water used to spray cows ranged from 0 to 6.8 gallons (0 to 25.6 liters) per hour per cow. Across dairies, there was a range in the cows' shade use (47% to 98% of herd) and feeding activity (7% to 33% of herd). Respiration rates ranged from 65 (normal) to 95 breaths per minute (very hot) and were positively related to inactivity. This paper presents the results of a research study which had the objective of investigating the effect of a sprinkler system coupled with forced ventilation on the heat stress of dairy cows bred in a free stall barn without paddock. To this aim, an experiment was carried out inside a free-stall dairy house equipped with two different cooling systems: a fogging system associated with forced ventilation in the resting area and a sprinkler system associated with forced ventilation in the feeding alley. The experiment regarded two adjacent pens of the barn and was constituted by three different trials carried out in the following periods: 27th June - 7th July (P1), 25th July - 4th August (P2), 24th August - 3rd September (P3). The experimental protocol of each trial required that the treatment group was housed in one pen where the two cooling systems were always activated following an established timetable, whereas the control group was housed in the other pen, where the sprinkler system associated with forced ventilation was always deactivated. Climatic parameters were measured inside each pen of the barn and outside. Then, thermal humidity index (THI) was calculated. Rectal temperature and respiration rate of a sample of dairy cows were monitored each day during the three periods considered (P1, P2 and P3). During the three trials the cows of both groups were subjected to climatic conditions that resulted in average daily THI values between 72.8 and 74.7, corresponding to mild or moderate heat stress. However, during daytime, air temperature and relative humidity reached values corresponding to a severe heat stress, as attested by the maximum THI values that were higher than or very close to 80. Furthermore, it was observed that the sprinklers do not influence the microclimatic conditions. However, the physiological parameters values of the treatment group were always significantly lower than the corresponding ones of the control group. Specifically, the system especially influenced the respiration rate that, in the treatment group, was close to 50 breath/min, while in the control group it reached 70 breath/min. The sprinkler system had more limited effects on rectal temperature that, however, in the treatment group was significantly lower than in the control group (38.7°C to 38.8°C vs 39.1°C to 39.4°C) in all the three periods of the experiment. Using a model to generate experimental groups with different manifestations of post-partum (p.p.) fat mobilization and ketogenesis, the effects of a dietary and a medical intervention on biochemical and haematological parameters, antibody titre, leucocytes subsets and function of transition cows were examined. In total, 60 German Holstein cows were allocated 6 weeks antepartum (a.p.) to 3 high-body condition score ( BCS) groups ( BCS 3.95) and 1 low- BCS group ( LC, BCS 2.77). High- BCS cows received a monensin controlled-release capsule ( HC/ MO) or a blend of essential oils ( HC/ EO) or formed a control group ( HC). Parameters were evaluated in 3 periods (day (d) −42 until calving, 1 until 14 days in milk ( DIM), 15 until 56 DIM). Over the course of trial, various parameters were influenced by period with greatest variability next to calving. White blood cell count was higher in the HC (8.42 × 103/ μl) and HC/ EO (8.38 × 103/ μl) groups than in the HC/ MO group (6.81 × 103/ μl) considering the whole trial. Supplementation of monensin decreased aspartate aminotransferase in comparison with the HC group similar to LC treatment. Bilirubin concentration was nearly doubled in all high- BCS cows in period 2. In period 3, essential oils increased γ-glutamyltransferase (80.4 Units/l) in comparison with all other groups and glutamine dehydrogenase (61 Units/l) in comparison with the LC (19 Units/l) and the HC/ MO group (18 Units/l). Results suggest that parameters were generally characterized by a high variability around calving. Based on biochemical characteristics, it appeared that the HC cows seemed to have compromised hepatocyte integrity when compared to the LC cows. Herbage feeding with only little input of concentrates plays an important role in milk production in grassland dominated countries like Switzerland. The objective was to investigate the effects of a solely herbage-based diet and level of milk production on performance, and variables related to the metabolic, endocrine and inflammatory status to estimate the stress imposed on dairy cows. Twenty-five multiparous Holstein cows were divided into a control (C+, n = 13) and a treatment group (C−, n = 12), according to their previous lactation yield (4679-10 808 kg) from week 3 ante partum until week 8 post-partum (p.p.). While C+ received fresh herbage plus additional concentrate, no concentrate was fed to C− throughout the experiment. Within C+ and C−, the median of the preceding lactation yields (7752 kg) was used to split cows into a high ( HYC+, HYC−)- and low-yielding ( LYC+, LYC−) groups. Throughout the study, HYC+ had a higher milk yield (35.9 kg/d) compared to the other subgroups (27.2-31.7 kg/d, p < 0.05). Plasma glucose (3.51 vs. 3.72 mmol/l) and IGF-1 (66.0 vs. 78.9ng/ mL) concentrations were lower in HYC−/ LYC− compared to HYC+/ LYC+ cows (p < 0.05). Plasma FFA and BHBA concentrations were dramatically elevated in HYC− (1.1 and 1.6 mmol/l) compared to all other subgroups (mean values: 0.5 and 0.6 mmol/l, p < 0.05). Saliva cortisol, plasma concentrations of serum amyloid A ( SAA), haptoglobin (Hp), beta-endorphin ( BE) and activity of alkaline phosphatase ( AP) were not different between C+ and C−. In conclusion, herbage-fed high-yielding cows without supplementary concentrate experienced a high metabolic load resulting in a reduced performance compared to cows of similar potential fed accordingly. Low-yielding cows performed well without concentrate supplementation. Interestingly, the selected markers for inflammation and stress such as cortisol, Hp, SAA, BE and AP gave no indication for the metabolic load being translated into compromised well-being. From their time of birth until their first lactation, dairy heifers incur management, health, and feed expenses while not producing milk. Much effort has been made to estimate optimal ages of first calving (AFC) for cows to reduce these costs, which can be as high as $2.50 per day, and ensure that animals are productive earlier in life. To identify AFC for 3 dairy cattle breeds (Holstein, Jersey, and Brown Swiss) that maximizes production, we retrieved phenotypic records for more than 14 million cows calving between 1997 and 2015 from the US national dairy database. The mean AFC for Holstein and Jersey has decreased by 2.4 and 2.7 mo, respectively, since 2006. When comparing the association of AFC with production and fertility traits, we found that decreased AFC was correlated with greater fertility and higher milk yield for all but the earliest group (18 to 20 mo). We also identified an unfavorable correlation of lower AFC with increasing stillbirth rates in Holstein (0.047 least squares means compared with a baseline of 24 mo) and Brown Swiss (0.062 least squares means). Finally, we identified favorable genetic correlations of lower AFC with lifetime net merit, heifer conception rate, cow conception rate, and daughter pregnancy rate in Holstein and Jersey cattle, and favorable correlations for net merit and heifer conception rate in Brown Swiss. To maximize lifetime production and reduce the effects of AFC on stillbirth, the AFC that maximizes production for Holstein and Brown Swiss is 21 to 22 mo, and for Jersey it is 20 to 21 mo. However, the effect of AFC on stillbirth reduces the benefits of calving at very young ages. Calculated genomic predicted transmitting ability for AFC showed an improvement in reliability of 20 percentage points in genomic young bulls compared with parent averages in Holstein, suggesting that genomic testing can improve selection for this trait. The liver functionality index (LFI) represents an assessment of transition cow metabolic health by measuring changes in biomarkers associated with liver plasma protein synthesis (albumin), lipoprotein synthesis (cholesterol), and heme catabolism (bilirubin). The present analysis was conducted to determine the role of peripartal rumen-protected Met or choline (CHOL) supplementation on LFI groupings, and to assess relationships with performance, inflammation, oxidative stress status, and plasma AA profiles. A cohort of 40 multiparous Holstein cows that were part of a randomized complete block design with 2 × 2 factorial arrangement of Met (Smartamine M, Adisseo NA, Alpharetta, GA) and CHOL (ReaShure, Balchem Inc., New Hampton, NY) level (with or without) were used. From -21 d to calving, cows received the same close-up diet and were assigned randomly to each treatment. From calving to 30 d, cows were on the same postpartal diet and continued to receive the same treatments until 30 d. Addition of Met was adjusted daily at 0.08% dry matter of diet and CHOL was fed at 60 g/cow per day. Liver (-10, 7, 20, and 30 d) and blood (-10, 4, 8, 20, and 30 d) samples were harvested for biomarker analyses. Cows were ranked retrospectively and assigned to low (LLFI, LFI <0) and high (HLFI, LFI >0) LFI groups regardless of Met or CHOL supplementation. Compared with cows in LLFI, close-up and lactation DMI, milk yield, milk fat yield, and milk protein yield were greater in HLFI cows. As expected, cows in LLFI had lower plasma cholesterol and albumin but greater bilirubin concentrations around parturition. Plasma haptoglobin concentration was also lower in HLFI cows, but plasma paraoxonase and hepatic total and reduced hepatic glutathione concentrations were greater. Although higher concentrations of His, Met, and Trp, as well as a tendency for greater Ile, were observed in HLFI cows, overall essential AA concentrations did not differ with LFI status. In contrast, overall concentrations of nonessential AA were greater in HLFI cows due to greater circulating concentrations of Ala, Asn, Gln, Pro, and Ser. Similarly, overall concentrations of total AA and total sulfur-containing compounds were greater in cows with HLFI. Feeding Met compared with CHOL led to a tendency for more cows classified as HLFI. Overall, results support the broader application of the LFI in the management of transition cows. In that context, the fact that precalving concentrations of compounds such as reduced glutathione, total sulfurcontaining compounds, Met, Tau, and homocysteine differed between HLFI and LLFI independent of Met or CHOL feeding also underscores their potential for monitoring cows that might be at a greater risk of developing health problems after calving. Heat stress in dairy cows during the dry period impairs milk yield in the next lactation. Feeding Omni-Gen-AF (OG; Phibro Animal Health Corp., Teaneck, NJ) to lactating cows during heat stress may increase dry matter intake (DMI) and lowers respiration rate (RR) and rectal temperature (RT), but the effects in dry cows are not known. We hypothesized that OG supplementation before, during, and after the dry period (approximately 160 d total) would overcome the effects of heat stress and improve cow performance in the next lactation. Cows were randomly assigned to OG or control (placebo) treatments for the last 60 d in milk (DIM), based on mature-equivalent milk yield in the previous lactation. Cows were dried off 45 d before expected calving and randomly assigned to heat stress (HT) or cooling (CL) treatments. Thus, cows received dietary supplementation during late lactation before they were exposed to either CL or HT. After dry-off, treatment groups included heat stress with placebo (HT, only shade, 56 g/d of placebo, n = 17), HT with OG supplementation (HTOG, 56 g/d of OG, n = 19), cooling with placebo (CL, shade, fans, and soakers, 56 g/d of placebo, n = 16), and CL with OG supplementation (CLOG, 56 g/d of OG, n = 11). After parturition, all cows were kept under the same CL system and management, and all cows continued to receive OG or control treatment until 60 DIM. Cooling cows during the dry period reduced afternoon RT (CL vs. HT; 38.9 ± 0.05 vs. 39.3 ± 0.05°C) and RR (CL vs. HT; 45 ± 1.6 vs. 77 ± 1.6 breaths/min). Respiration rate was also decreased by OG supplementation under HT conditions (HTOG vs. HT; 69.7 ± 1.6 vs. 77.2 ± 1.6 breaths/min). An interaction was observed between OG supplementation and HT; HTOG cows tended to have lower morning RT compared with HT cows. During the dry period, OG reduced DMI relative to control cows. Birth weight was greater in calves from CL cows (CL vs. HT; 40.6 ± 1.09 vs. 38.7 ± 1.09 kg). No differences were detected among treatments in hematocrit, total protein, and body condition score. Cows offered CLOG, CL, and HTOG treatments had greater body weight during the dry period (794.9 ± 17.9, 746.8 ± 16.7, and 762.9 ± 14.9 kg, respectively) than HT cows (720 ± 16.2 kg). Gestation length was approximately 4 d longer for CL cows compared with HT cows. Cows offered CLOG, CL, and HTOG treatments produced more milk (41.3 ± 1.6, 40.7 ± 1.6, and 40.5 ± 1.6 kg/d, respectively) than HT treatment (35.9 ± 1.6 kg/d). Body weight after parturition and DMI were evaluated up to 60 DIM and averaged 661.5 ± 15.8 and 19.4 ± 0.7 kg/d, respectively, with no differences observed among treatments. These results confirm that exposure of dry cows to heat stress negatively affects milk yield in the subsequent lactation. This experiment was conducted to compare conventional (CON; 21 d) and shortened (SH; 10 d) close-up period, and evaluate the effect of shortened close-up period combined with feeding different metabolizable protein (MP) levels on dry matter (DM) intake, metabolic status, and performance of dairy cows. Forty-eight multiparous Holstein cows with similar parity, body weight (BW), and previous lactation milk yield were divided into 2 groups. The first group (n = 24) received the far-off diet from -60 to -21 d (CON), and the second group (n = 24) received same far-off diet from -60 to -10 d (SH) relative to expected parturition. Cows were then moved to individual stalls and randomly allocated to 1 of 3 close-up diets: low MP diet (LMP; MP = 79 g/kg of DM), medium MP diet (MMP; MP = 101 g/kg of DM), or high MP diet (HMP; MP = 118 g/ kg of DM). Treatments were used in a 2 × 3 factorial arrangement with 2 lengths of close-up period (CON and SH) and 3 levels of MP (LMP, MMP, and HMP). All diets were fed for ad libitum intake during the closeup period. After calving, all cows received the same fresh cow diet. We found no interaction between closeup period length and MP levels for traits, except for postpartum serum fatty acids and β-hydroxybutyrate (BHB). The concentrations of postpartum serum fatty acids and BHB were higher on LMP than MMP and HMP diets in SH group. The cows of the SH group tended to produce less colostrum in the first milking than cows in CON group. The length of close-up period did not affect pre- and postpartum DM intake or energy balance of cows during the last week of prepartum, but cows of the CON group had greater BW changes during the last 3 wk before parturition than cows in SH group. Cows fed MMP and HMP diets consumed 1.2 and 1 kg more DM than for those fed LMP prepartum, respectively. The concentrations of prepartum BHB and Ca were higher for SH cows than CON group cows. Except for blood urea N concentration, no other blood metabolite in prepartum was affected by dietary MP. We found no effects of close-up period length or MP levels in the close-up diet on urinary pH, purine derivative excretion, and microbial N flow. Postpartum, milk yield was not affected by close-up period length, but cows in CON group tended to have higher 4% fatcorrected milk yield, had higher milk fat content and yield, had greater BW and body condition score loss, and higher energy negative balance than cows in the SH group. Cows fed MMP diet ate 1.8 kg more DM and yielded 3.37 kg more milk than those fed the LMP diet. Milk fat, protein, and lactose content, milk urea N, and somatic cell count were not affected by MP levels, but the yield of milk protein and lactose were higher on MMP diet than on LMP diet. Concentrations of postpartum serum fatty acids and BHB were decreased by shortening the close-up period length, but glucose, cholesterol, and triglyceride were similar between closeup groups. During the postpartum period, serum fatty acids, BHB, aminotransferase, and Ca concentrations were decreased by increasing the MP levels in the closeup diet. It appears from this data set that multiparous cows will benefit from a shortened close-up period, and feeding a moderate MP diet could improve DM intake, milk yield, and metabolic status of periparturient dairy cows. The modulation of milk fat nutritional quality through fish oil supplementation seems to be largely explained by the action of n-3 very long chain polyunsaturated fatty acids (PUFA) on ruminal biohydrogenation (BH) of C18 fatty acids (FA). However, relationships among this action, disappearance of those PUFA in the rumen, and potential detrimental consequences on ruminal fermentation remain uncertain. This study compared the effect of 20:5n-3 (eicosapentaenoic acid; EPA), 22:5n-3 (docosapentaenoic acid; DPA), and 22:6n-3 (docosahexaenoic acid; DHA) on rumen fermentation and BH of C18 FA and was conducted simultaneously in cows and sheep to provide novel insights into interspecies differences. The trial was performed in vitro using batch cultures of rumen microorganisms with inocula collected from cannulated cows and ewes. The PUFA were added at a dose of 2% incubated dry matter, and treatment effects on ruminal C18 FA concentrations, PUFA disappearances, and fermentation parameters (gas production, ammonia and volatile FA concentrations, and dry matter and neutral detergent fiber disappearances) were examined after 24 h of incubation. A principal component analysis suggested that responses to PUFA treatments explained most of the variability; those of ruminant species were of lower relevance. Overall, EPA and DHA were equally effective for inhibiting the saturation of trans-11 18:1 to 18:0 and had a similar influence on ruminal fermentation in cows and sheep (e.g., reductions in gas production and acetate:propionate ratio). Nevertheless, DHA further promoted alternative BH pathways that lead to trans-10 18:1 accumulation, and EPA seemed to have specific effects on 18:3n-3 metabolism. Only minor variations attributable to DPA were observed in the studied parameters, suggesting a low contribution of this FA to the action of marine lipids. Although most changes due to the added PUFA were comparable in bovine and ovine, there were also relevant specificities, such as a stronger inhibition of 18:0 formation in cows and a greater increase in 18:3n-3 metabolites in sheep. No direct relationship between in vitro disappearance of the incubated PUFA and effect on BH (in particular, inhibition of the last step) was found in either cows or ewes, calling into question a putative link between extent of disappearance and toxicity for microbiota. Conversely, an association between the influence of these PUFA on ruminal lipid metabolism and fermentation may exist in both species. In vivo verification of these findings would be advisable. Neospora caninum is a protozoan parasite with a preference for cattle and dogs as hosts. When N. caninum infection occurs in cattle it induces abortion, bovine neosporosis being a main cause of abortion worldwide. In dairy cattle, the economic burden of neosporosis-associated abortion is so great that it might results in closure of a farm. However, not all infected cows abort and it is not yet understood why this occurs. At present there is no effective treatment or vaccine. This review provides insights on how immune response against the parasite determines protection or contribution to abortion. Aspects on markers of risk of abortion are also discussed. Humoral immune responses are not protective against N. caninum but seropositivity and antibody level can be good markers for a diagnosis of bovine neosporosis and its associated abortion risk. In addition, humoral mechanisms against N. caninum infection and abortion differ in pure-breed and cross-breed pregnant dairy and beef cattle. Concentrations of Pregnancy Associated glycoprotein -2 (PAG-2) can also be used to predict abortion. A partially protective immune response encompasses increased IFN-γ expression, which has to be counterbalanced by other cytokines such as IL-12 and IL-10, especially towards the end of pregnancy. Although IFN-γ is required to limit parasite proliferation a critical threshold of the IFN-γ response is also required to limit adverse effects on pregnancy. The ruminal microbiome, comprising large numbers of bacteria, ciliate protozoa, archaea and fungi, responds to diet and dietary additives in a complex way. The aim of this study was to investigate the benefits of increasing the depth of the community analysis in describing and explaining responses to dietary changes. Quantitative PCR, ssu rRNA amplicon based taxa composition, diversity and co-occurrence network analyses were applied to ruminal digesta samples obtained from four multiparous Nordic Red dairy cows fitted with rumen cannulae. The cows received diets with forage:concentrate ratio either 35:65 (diet H) or 65:35 (L), supplemented or not with sunflower oil (SO) (0 or 50 g/kg diet dry matter), supplied in a 4 × 4 Latin square design with a 2 × 2 factorial arrangement of treatments and four 35-day periods. Digesta samples were collected on days 22 and 24 and combined. QPCR provided a broad picture in which a large fall in the abundance of fungi was seen with SO in the H but not the L diet. Amplicon sequencing showed higher community diversity indices in L as compared to H diets and revealed diet specific taxa abundance changes, highlighting large differences in protozoal and fungal composition. Methanobrevibacter ruminantium and Mbb. gottschalkii dominated archaeal communities, and their abundance correlated negatively with each other. Co-occurrence network analysis provided evidence that no microbial domain played a more central role in network formation, that some minor-abundance taxa were at nodes of highest centrality, and that microbial interactions were diet specific. This study evaluated the effect of a once-used progesterone (P4) intravaginal device (PRID) associated with four different shortened P4-based estrus synchronization (ES) protocols on estrous response (ER) and pregnancy per AI (P/AI) in cyclic and acyclic lactating dairy cows. Cows (n=465) were randomly assigned to one of the following protocols: 1) 2PGG, cows were given a PRID-Delta and 100 μg GnRH i.m. at PRID insertion (day 0). The PRID was left for 5 d, and 25 mg of dinoprost (PGF2α) i.m. given twice at PRID removal and 24 h later; 2) 2PGGe, same treatments as 2PGG plus 500 IU of eCG i.m. at PRID removal; 3) 2PGe, same treatments as 2PGGe, except GnRH was not given at PRID insertion; 4) PGe, same treatments as 2PGe, except PGF was only given at PRID removal. A total of 258 cows received a new PRID-Delta containing 1.55 g of P4, whereas 207 cows received a once-used PRID. Estrus was determined from P4 device removal until 96 h after using an automated heat detection system. Cows in estrus were given a second GnRH at AI and those without signs of estrus by 96 h after PRID removal were given GnRH and timed-AI (TA I). All inseminations were performed by one technician with commercially available frozen-thawed semen. Ultrasonography was performed at initiation of protocol and 28-34 days post AI to determine cyclicity and pregnancy status, respectively. Cows receiving once-used P4 devices had greater ER than cows receiving a new device (59.9 vs. 50.0; P=0.029), but P/AI did not differ between P4 devices, respectively (P>0.1; 40.6 vs 40.7%). Cyclic cows were less likely to display estrus than acyclic cows by a factor of 0.66 (P=0.036). Cows subjected to the 2PGe (2.41; P<0.01) protocol were more likely to display estrus than cows subjected to the 2PGG, whereas cows subjected to the PGe protocol did not differ (0.94; P=0.8) from those in the 2PGG group. Despite differences in ER, neither cyclicity nor estrus synchronization protocol affected P/AI (overall 40.6%). In summary, cyclic cows, those given a new P4 device and those subjected to either 2PGG or PGe protocol had reduced ER. However, all the factors examined had no significant effect on P/AI. Our objective was to compare the ranking of dairy cows according to their methane (CH4) emissions as measured by a respiration chamber (RC) technique and the GreenFeed (GF) technique during 3 periods in second lactation. Two-day CH4 measurements in a RC performed in wk 3, 14, and 42 of lactation were flanked by GF measurements for 20 (period 1 [P1]), 35 (period 2 [P2]), and 35 (period 3 [P3]) days, respectively, before and after RC measurement. This gave the total duration of CH4 measurements using the GF system of 40, 70, and 70 d for P1, P2, and P3, respectively. Mean daily CH4 production (g/d) of the 8 dairy cows was 346, 439, and 430 using the RC technique and 338, 378, and 416 using the GF system during P1, P2, and P3, respectively. Average daily CH4 production determined by the GF technique was 2.4, 13.8, and 3.2% lower in P1, P2, and P3, respectively. Methane normalized to DMI continuously increased from P1 to P3 when measured in a RC, whereas it was lowest during P2 when measured by the GF method. Ranking of the cows according to CH4 production, CH4/energy-corrected milk yield (ECM; CH4/ECM), and CH4/DMI differed between periods no matter which method was used. Cluster analysis including all 3 periods, however, identified the same cows with the highest and lowest CH4 production determined either by the RC technique or the GF system. In conclusion, multiple CH4 measurements at different stages of lactation are necessary for reliable discrimination of highest and lowest CH4 emitting cows and the GF system may be used to discriminate the extremes.
