Abstract

Neural progenitor cells (NPCs) are multipotent cells that can self-renew and differentiate into neurons and glial cells. However, mechanisms that control their fate decisions are poorly understood. Here, we show that Smek1, a regulatory subunit of the serine/threonine protein phosphatase PP4, promotes neuronal differentiation and suppresses the proliferative capacity of NPCs. We identify the cell polarity protein Par3, a negative regulator of neuronal differentiation, as a Smek1 substrate and demonstrate that Smek1 suppresses its activity. We also show that Smek1, which is predominantly nuclear in NPCs, is excluded from the nucleus during mitosis, allowing it to interact with cortical/cytoplasmic Par3 and mediate its dephosphorylation by the catalytic subunit PP4c. These results identify the PP4/Smek1 complex as a key regulator of neurogenesis.

Abstract

Over the last decades, epidemiology and functional studies have started to reveal a pivotal role of vitamin D in both type 1 and type 2 diabetes pathogenesis. Acting through the vitamin D receptor (VDR), vitamin D regulates insulin secretion in pancreatic islets and insulin sensitivity in multiple peripheral metabolic organs. In vitro studies and both T1D and T2D animal models showed that vitamin D can improve glucose homeostasis by enhancing insulin secretion, reducing inflammation, reducing autoimmunity, preserving beta cell mass, and sensitizing insulin action. Conversely, vitamin D deficiency has been shown relevant in increasing T1D and T2D incidence. While clinical trials testing the hypothesis that vitamin D improves glycemia in T2D have shown conflicting results, subgroup and meta-analyses support the idea that raising serum vitamin D levels may reduce the progression from prediabetes to T2D. In this review, we summarize current knowledge on the molecular mechanisms of vitamin D in insulin secretion, insulin sensitivity, and immunity, as well as the observational and interventional human studies investigating the use of vitamin D as a treatment for diabetes.

Abstract

In this study, the plasma oxidation effect in tin-oxide (SnOx) thin film was investigated. And on this basis, we fabricated n-type thin-film transistors (TFTs) using the SnOx thin film with the plasma oxidation by experiments. By adjusting the processing time of the oxygen plasma treatment (OPT), the optimized SnOx TFT device exhibited an extremely high field-effect mobility of 87.6 cm2 V−1 s−1, a desirable on-to-off current ratio of 1.9 × 104 and a threshold voltage of − 0.9 V. Furthermore, we investigated the origin of the performance enhancements in the n-type SnOx TFTs with the optimized OPT by introducing the density of states (DOS) modeling in TCAD simulation. The numerical simulation indicated that the attributes of donor-like Gaussian defect states (oxygen vacancies) were modified in overall DOS due to the plasma oxidation effect. These present results show that the SnOx TFT treated by oxygen plasma has great promise in the future high-performance flat panel display industries.

Abstract

In the current study, heterostructured Ag@AgCl@AgI nanotubes were synthesized via an anion-exchange reaction between Ag@AgCl nanotubes and I− anions. Moreover, their photocatalytic performance towards the decontamination of dye contaminants were examined. These prepared photocatalysts were accordingly characterized using high-resolution scanning electron microscopy (HR-SEM), XRPD, XPS, field-emission transmission electron microscopy (FE-TEM), and photoluminescence (PL) spectroscopy. The structural identification analyses confirmed that the photocatalysts had nanotubular morphology. Compared to polyhedral Ag@AgI nanocomposites, the amalgamation of I− ions into the Ag@AgCl framework extraordinarily improved the photocatalytic performance. The synthesized Ag@AgCl@AgI nanotubes can also be reused due to their high stability. Under visible-light irradiation, >91.8% of Rhodamine B (RhB) was degraded within 45 min using the catalyst AI–2 (Cl−/I− = 1:1). The radical trap experiments indicated that the photodegradation of RhB was driven by the participation of superoxide radical and the action of holes. Due to the surface plasmon resonance effect, the photocatalyst AI–2 demonstrated higher photocatalytic performance, enhanced surface area and photoelectron transfer, which is also ascribed to its unique heterostructured configuration.

427. A Risk-Factor Model for Antineoplastic Drug-Induced Serious Adverse Events in Cancer Inpatients: A Retrospective Study Based on the Global Trigger Tool and Machine Learning

Abstract

The objective of this study was to apply a machine learning method to evaluate the risk factors associated with serious adverse events (SAEs) and predict the occurrence of SAEs in cancer inpatients using antineoplastic drugs. A retrospective review of the medical records of 499 patients diagnosed with cancer admitted between January 1 and December 31, 2017, was performed. First, the Global Trigger Tool (GTT) was used to actively monitor adverse drug events (ADEs) and SAEs caused by antineoplastic drugs and take the number of positive triggers as an intermediate variable. Subsequently, risk factors with statistical significance were selected by univariate analysis and least absolute shrinkage and selection operator (LASSO) analysis. Finally, using the risk factors after the LASSO analysis as covariates, a nomogram based on a logistic model, extreme gradient boosting (XGBoost), categorical boosting (CatBoost), adaptive boosting (AdaBoost), light-gradient-boosting machine (LightGBM), random forest (RF), gradient-boosting decision tree (GBDT), decision tree (DT), and ensemble model based on seven algorithms were used to establish the prediction models. A series of indicators such as the area under the ROC curve (AUROC) and the area under the PR curve (AUPR) was used to evaluate the model performance. A total of 94 SAE patients were identified in our samples. Risk factors of SAEs were the number of triggers, length of stay, age, number of combined drugs, ADEs occurred in previous chemotherapy, and sex. In the test cohort, a nomogram based on the logistic model owns the AUROC of 0.799 and owns the AUPR of 0.527. The GBDT has the best predicting abilities (AUROC = 0.832 and AUPR = 0.557) among the eight machine learning models and was better than the nomogram and was chosen to establish the prediction webpage. This study provides a novel method to accurately predict SAE occurrence in cancer inpatients.

Abstract

Transit Timing Variation (TTV) of hot Jupiters provides direct observational evidence of planet tidal dissipation. Detecting tidal dissipation through TTV needs high precision transit timings and long timing baselines. In this work, we predict and discuss the potential scientific contribution of SiTian Survey in detecting and analyzing exoplanet TTV. We develop a tidal dissipation detection pipeline for SiTian Survey that aims at time-domain astronomy with 72 1-meter optical telescopes. The pipeline includes the modules of light curve deblending, transit timing obtaining, and TTV modeling. SiTian is capable to detect more than 25,000 exoplanets among which we expect $\sim$50 sources showing evidence of tidal dissipation. We present detection and analysis of tidal dissipating targets, based on simulated SiTian light curves of XO-3b and WASP-161b. The transit light curve modeling gives consistent results within 1$\sigma$ to input values of simulated light curves. Also, the parameter uncertainties predicted by Monte-Carlo Markov Chain are consistent with the distribution obtained from simulating and modeling the light curve 1000 times. The timing precision of SiTian observations is $\sim$ 0.5 minutes with one transit visit. We show that differences between TTV origins, e.g., tidal dissipation, apsidal precession, multiple planets, would be significant, considering the timing precision and baseline. The detection rate of tidal dissipating hot Jupiters would answer a crucial question of whether the planet migrates at an early formation stage or random stages due to perturbations, e.g., planet scattering, secular interaction. Abstract

Adversarial attacks on deep learning-based models pose a significant threat to the current AI infrastructure. Among them, Trojan attacks are the hardest to defend against. In this paper, we first introduce a variation of the Badnet kind of attacks that introduces Trojan backdoors to multiple target classes and allows triggers to be placed anywhere in the image. The former makes it more potent and the latter makes it extremely easy to carry out the attack in the physical space. The state-of-the-art Trojan detection methods fail with this threat model. To defend against this attack, we first introduce a trigger reverse-engineering mechanism that uses multiple images to recover a variety of potential triggers. We then propose a detection mechanism by measuring the transferability of such recovered triggers. A Trojan trigger will have very high transferability i.e. they make other images also go to the same class. We study many practical advantages of our attack method and then demonstrate the detection performance using a variety of image datasets. The experimental results show the superior detection performance of our method over the state-of-the-arts.

Abstract

Highlights: A triple functional additive with a trace amount (1 mM) was proposed to protect Zn anodes. The additive lowers the hydrogen evolution reaction potential, encourages the formation of an in situ solid electrolyte interphase and shields the "tip effect" Dendrite free Zn deposition and highly reversible Zn plating/stripping behaviors were realized by the triple protections Although their cost-effectiveness and intrinsic safety, aqueous zinc-ion batteries suffer from notorious side reactions including hydrogen evolution reaction, Zn corrosion and passivation, and Zn dendrite formation on the anode. Despite numerous strategies to alleviate these side reactions have been demonstrated, they can only provide limited performance improvement from a single aspect. Herein, a triple-functional additive with trace amounts, ammonium hydroxide, was demonstrated to comprehensively protect zinc anodes. The results show that the shift of electrolyte pH from 4.1 to 5.2 lowers the HER potential and encourages the in situ formation of a uniform ZHS-based solid electrolyte interphase on Zn anodes. Moreover, cationic NH4+ can preferentially adsorb on the Zn anode surface to shield the "tip effect" and homogenize the electric field. Benefitting from this comprehensive protection, dendrite-free Zn deposition and highly reversible Zn plating/stripping behaviors were realized. Besides, improved electrochemical performances can also be achieved in Zn//MnO2 full cells by taking the advantages of this triple-functional additive. Abstract

The development of flexible electrodes with high mass loading and efficient electron/ion transport is of great significance but still remains the challenge of innovating suitable electrode structures for high energy density application. Herein, for the first time, lignosulfonate‐derived N/S‐co‐doped graphene‐like carbon is in situ formed within an interface engineered cellulose textile through a sacrificial template method. Both experimental and theoretical calculations disclose that the formed pomegranate‐like structure with continuous conductive pathways and porous characteristics allows sufficient ion/electron transport throughout the entire structures. As a result, the obtained flexible electrode delivers a remarkable integrated capacitance of 6534 mF cm−2 (335.1 F g−1) and a superior stability at an industrially applicable mass loading of 19.5 mg cm−2. A pseudocapacitive cathode with ultrahigh capacitance of 7000 mF cm−2 can also be obtained based on the same electrode structure engineering. The as‐assembled asymmetric supercapacitor achieves a high areal capacitance of 3625 mF cm−2, and a maximum energy density of 1.06 mWh cm−2, outperforms most of other reported high‐loading supercapacitors. This synthesis method and structural engineering strategy can provide materials design concepts and a wide range of applications in the fields of energy storage beyond supercapacitors.
