402. Regulation and control roles of the basal ganglia in the development of absence epileptiform activities.

Abstract

Absence epileptiform activities are traditionally considered to be primarily induced by abnormal interactions between the cortical and thalamic neurons, which form the thalamocortical circuit in the brain. The basal ganglia, as an organizational unit in the brain, has close input and output relationships with the thalamocortical circuit. Although several studies report that the basal ganglia may participate in controlling and regulating absence epileptiform activities, to date, there have been no studies regarding whether the basal ganglia directly cause absence epileptiform activities. In this paper, we built a basal ganglia-corticothalamic network model to determine the role of basal ganglia in this disease. We determined that absence epileptiform activities might be directly induced by abnormal coupling strengths on certain pivotal pathways in the basal ganglia. These epileptiform activities can be well controlled by the coupling strengths of three major pathways that project from the thalamocortical network to the basal ganglia. The results implied that the substantia nigra pars compacta (SNc) can be considered to be the effective treatment target area for inhibiting epileptiform activities, which supports the observations of previous studies. Particularly, as a major contribution of this paper, we determined that the final presentation position of the epileptic slow spike waves is not limited to the cerebral cortex; these waves may additionally appear in the thalamus, striatal medium spiny neurons, striatal fast spiking interneuron, the SNc, subthalamic nucleus, substantia nigra pars reticulata and globus pallidus pars externa. In addition, consistent with several previous studies, the delay in the network was observed to be a critical factor for inducing transitions between different types of absence epileptiform activities. Abstract

Background: Durvalumab has shown meaningful clinical activity in patients with metastatic urothelial carcinoma (mUC) in Study 1108 (NCT01693562). An important focus in treatment is health-related quality of life (HRQOL). Here, patient-reported outcomes (PROs) from Study 1108 and their relationship with inflammatory biomarkers are explored.Methods: Disease-related symptoms, functioning, and HRQOL were assessed with the Functional Assessment of Cancer Therapy-Bladder (FACT-Bl) and the European Organisation for Research and Treatment of Cancer (EORTC) Quality of Life Questionnaire Core 30 (QLQ-C30). Relationships between PRO improvements and the best changes in the tumor size, albumin level, and neutrophil-lymphocyte ratio (NLR) were assessed with Spearman correlation analysis.Results: The mean FACT-Bl total score improved from 107.5 (standard deviation [SD], 23.0) at the baseline to 115.4 (SD, 22.6) on day 113, with similar increases found for the Trial Outcome Index (TOI) and Bladder Cancer Subscale (BLCS) scores. The mean FACT-Bl total scores improved over time, and the FACT-Bl TOI scores significantly improved by day 113 (P < .05). The mean EORTC QLQ-C30 Global Health Status/Quality of Life score improved from 57.1 (SD, 24.8) at the baseline to 69.0 (SD, 21.4) on day 113; the functional scale and symptom scores (day 113) were higher than the baseline scores (P < .05) for EORTC Social Functioning. The FACT-Bl total, BLCS, and TOI scores improved in 32.6%, 34.9%, and 32.6% of the patients by day 113; 26.3% to 37.8% of the patients exhibited improvements in EORTC QLQ-C30 functional scores. The best tumor shrinkage and posttreatment improvements in serum albumin and NLR correlated with increases in FACT-Bl total, TOI, and BLCS scores and in EORTC Physical Functioning and Role Functioning scores (P < .05).Conclusions: Durvalumab was associated with improvements in disease-related symptoms, functioning, and HRQOL in patients with mUC. 404. Thermal decomposition of 1-hexene by flash pyrolysis: A study of initial decomposition mechanism.

Abstract

Thermal decomposition of 1-hexene at temperatures 295-1410 K was conducted using a flash pyrolysis micro-reactor coupled to laser-based vacuum ultraviolet photoionization time-of-flight mass spectrometer (VUV-PI-TOFMS). The decomposition mechanism of 1-hexene was developed with the help of theoretical calculation performed at the MRCI/cc-pvtz//CASSCF/6–31+ G (d,p) level. The γ -scission and diradical retro-ene reactions were determined as the main initial decomposition reactions in the temperature range 990-1240 K. Two diradical retro-ene reaction channels, 1,5-diradical and 1,6-diradical reactions, were proposed in order to interpret the appearance of the C 4 H 8 species. The 1,5-diradical retro-ene reaction involved a 1,5-diradical intermediate that subsequently decomposed via C C β -scissions to the C2, C3 and C4 products. The 1,6-diradical retro-ene reaction proceeded via a 1,6-diradical intermediate and C C β -scissions to produce the C2 and C4 species. The proposed diradical retro-ene mechanism was evidenced indirectly by the early product distribution of 1-hexene pyrolysis in a flow reactor at 1173 K determined by synchrotron radiation VUV-PI-TOFMS. Abstract

Ultraviolet (UV) photodissociation dynamics of jet-cooled phenyl radicals (C6H5 and C6D5) are studied in the photolysis wavelength region of 215-268 nm using high-n Rydberg atom time-of-flight and resonance enhanced multiphoton ionization techniques. The phenyl radicals are produced from 193-nm photolysis of chlorobenzene and bromobenzene precursors. The H-atom photofragment yield spectra have a broad peak centered around 235 nm and are in good agreement with the UV absorption spectra of phenyl. The H + C6H4 product translational energy distributions, P(ET)'s, peak near ∼7 kcal/mol, and the fraction of average translational energy in the total excess energy, , is in the range of 0.20-0.35 from 215 to 268 nm. The H-atom product angular distribution is isotropic. The dissociation rates are in the range of 107-108 s-1 with internal energy from 30 to 46 kcal/mol above the threshold of the lowest energy channel H + o-C6H4 (ortho-benzyne), comparable with the rates from the Rice-Ramsperger-Kassel-Marcus theory. The results from the fully deuterated phenyl radical are identical. The dissociation mechanism is consistent with production of H + o-C6H4, as the main channel from unimolecular decomposition of the ground electronic state phenyl radical following internal conversion of the electronically excited state. Abstract

Photo-predissociation dynamics of rotational states (N[sup ′]) of OH (A [sup 2]Σ[sup +],ν[sup ′]=3 and 4) are studied using high-n Rydberg atom time-of-flight technique. Spin–orbit branching fractions of O([sup 3]P[sub J=2,1,0]) are measured: 0.676±0.010:0.138±0.013:0.186±0.017 for N[sup ′]=0 in ν[sup ′]=3 and 0.873±0.026:0.102±0.028:0.025±0.005 for N[sup ′]=0 in ν[sup ′]=4, in excellent agreement with the recent theory. While OH A [sup 2]Σ[sup +] ν[sup ′]=3 predissociates predominantly via a single [sup 4]Σ[sup -] state, ν[sup ′]=4 decays via the [sup 4]Σ[sup -], [sup 2]Σ[sup -], and [sup 4]Π states, and interferences of these dissociation pathways strongly influence the O([sup 3]P[sub J]) fine-structure distributions. Abstract

H-atom channels in the photodissociation of jet-cooled ethyl radical (C[sub 2]H[sub 5]) via the A˜ [sup 2]A[sup ′](3s) state are studied near 245 nm by using the high-n Rydberg-atom time-of-flight technique. Bimodal product translational energy release and energy-dependent angular distribution suggest two dissociation pathways. A slow (∼0.35) and isotropic channel corresponds to unimolecular dissociation of the radical, presumably after internal conversion. A previously unobserved fast (∼0.78) and anisotropic (β=0.5±0.1) channel is consistent with direct H-atom scission via a nonclassical H-bridged transition state from the 3s state to yield H+C[sub 2]H[sub 4](X˜ [sup 1]A[sub g]). The fast/slow branching ratio is ∼0.2. Site-selective loss of the β hydrogen atom is confirmed by using the partially-deuterated CH[sub 3]CD[sub 2] radical. © 2001 American Institute of Physics. Abstract

The effects of grinding process on the extraction of polysaccharide from Coprinus comatus were investigated. The extracts named as CCPF and CCPP were obtained from the fragments and the powder of the C. comatus fruiting bodies, respectively. The comparison on the chemical analysis of CCPF and CCPP showed that reducing particle size could dramatically improve the extract yield and content of polysaccharide fractions, which resulted from the increasing of high molecular weight (Mw) fractions. The high Mw fraction CCPP-1 was isolated using the ethanol precipitating method. Its molecular weight was estimated to be 2.44×107 g/mol by high-performance size exclusion chromatography-multi angle laser light scattering (HPSEC-MALLS). CCPP-1 took compact conformation in aqueous solution. Structural analysis showed that CCPP-1 was α- d -(1→4)-glucan with branches at C-6 consisting of non-reducing terminal approximately every fourteen residues. The hypoglycemic activity study revealed that CCPP-1 had no effect on reducing blood sugar. The crude polysaccharide extracts CCPF showed significant hypoglycemic activity.

437. Estimating methane emissions in California’s urban and rural regions using multi-tower observations

Abstract

We present an analysis of methane (CH4) emissions using atmospheric observations from 36 thirteen sites in California during June 2013 – May 2014. A hierarchical Bayesian inversion 37 method is used to estimate CH4 emissions for spatial regions (0.3° pixels for major regions) by 38 comparing measured CH4 mixing ratios with transport model (WRF-STILT) predictions based 39 on seasonally varying California-specific CH4 prior emission models. The transport model is 40 assessed using a combination of meteorological and carbon monoxide (CO) measurements 41 coupled with the gridded California Air Resources Board (CARB) carbon monoxide (CO) 42 emission inventory. Hierarchical Bayesian inversion suggests that state annual anthropogenic 43 CH4 emissions are 2.42 ± 0.49 Tg CH4/yr (at 95% confidence, including transport bias 44 uncertainty), higher (1.2 - 1.8 times) than the CARB current inventory (1.64 Tg CH4/yr in 2013). 45 We note that the estimated CH4 emissions drop to 1.0 - 1.6 times the CARB inventory if we 46 correct for the 10% median CH4 emissions assuming the bias in CO analysis is applicable to 47 CH4. The CH4 emissions from the Central Valley and urban regions (San Francisco Bay and 48 South Coast Air Basins) account for ~58% and 26% of the total posterior emissions, 49 respectively. This study suggests that the livestock sector is likely the major contributor to the 50 state total CH4 emissions, in agreement with CARB’s inventory. Attribution to source sectors for 51 sub-regions of California using additional trace gas species would further improve the 52 quantification of California’s CH4 emissions and mitigation efforts towards the California Global 53 Warming Solutions Act of 2006 (AB-32).

Abstract

Background: To determine whether combining brachytherapy with immunotherapy is safe in prostate cancer (PCa) and provides synergistic effects, we performed a Phase I/II trial on the feasibility, safety, and benefit of concurrent delivery of anti-PD-1 (nivolumab) with high-dose-rate (HDR) brachytherapy and androgen deprivation therapy (ADT) in patients with Grade Group 5 (GG5) PCa. Methods: Eligible patients were aged 18 years or older with diagnosis of GG5 PCa. Patients received ADT, nivolumab every two weeks for four cycles, with two cycles prior to first HDR, and two more cycles prior to second HDR, followed by external beam radiotherapy. The primary endpoint was to determine safety and feasibility. This Phase I/II trial is registered with ClinicalTrials.gov (NCT03543189). Results: Between September 2018 and June 2019, six patients were enrolled for the Phase I safety lead-in with a minimum observation period of 3 months after nivolumab administration. Overall, nivolumab was well tolerated in combination with ADT and HDR treatment. One patient experienced a grade 3 dose-limiting toxicity (elevated Alanine aminotransferase and Aspartate aminotransferase) after the second cycle of nivolumab. Three patients (50%) demonstrated early response with no residual tumor detected in ≥4 of 6 cores on biopsy post-nivolumab (4 cycles) and 1-month post–HDR. Increase in CD8+ and FOXP3+/CD4+ T cells in tissues, and CD4+ effector T cells in peripheral blood were observed in early responders. Conclusion: Combination of nivolumab with ADT and HDR is well tolerated and associated with evidence of increased immune infiltration and antitumor activity.
