Abstract

Purpose: There is an unmet need to develop prognostic biomarkers in post-neoadjuvant chemotherapy (NAC) muscle-invasive bladder cancer (MIBC) patients. We examine whether Ki-67 and PD-L1 expression can be used to guide adjuvant therapy. Methods: Tissue microarrays were constructed from 130 post-NAC radical cystectomy samples. Up to 5 cores per sample were included. Expressions of Ki-67 and PD-L1 were evaluated using immunohistochemistry (IHC). Results: Using a Cox regression model, positive Ki-67 expression in post-NAC radical cystectomy samples was associated with poorer overall survival (OS) (HR = 2.412, 95% CI, 1.076–5.408), independent of the pathological lymph node/N-stage. Positive Ki-67 expression was also associated with lack of tumor downstaging in a multivariable logistic regression model analysis (OR = 0.081, 95% CI, 0.014–0.464). PD-L1− and PD-L1+ expression was associated with a median OS of 49.8 months and 26.9 months, respectively, which did not reach statistical significance. Patients with Ki-67/PD-L1 double-negative tumors had a significantly longer median OS of 98.2 months versus 29.9 and 26.9 months in PD-L1−/Ki-67+ and PD-L1+/Ki-67+ tumors, respectively. Lack of tumor downstaging was significantly associated with positive Ki-67 and positive PD-L1 expression. Conclusion: Positive Ki-67 and PD-L1 expression in post-NAC radical cystectomy samples was associated with inferior OS and absence of tumor downstaging. IHC on Ki-67 and PD-L1 would help to select patients for adjuvant therapy in post-NAC muscle-invasive bladder cancer. [ABSTRACT FROM AUTHOR]

Abstract

Thermal decomposition of cyclohexane at temperatures up to 1310 K was performed using flash pyrolysis coupled with vacuum ultraviolet (118.2 nm) photoionization time-of-flight mass spectrometry. The experimental results revealed that the major initiation reaction of cyclohexane decomposition was C–C bond fission leading to the formation of 1,6-hexyl diradical. The 1,6-hexyl diradical could isomerize to 1-hexene and decompose into ˙C3H7 + ˙C3H5 and ˙C4H7 + ˙C2H5. The 1,6-hexyl diradical could also undergo direct dissociation; the C4H8 fragment via the 1,4-butyl diradical intermediate was observed, serving as evidence of the 1,6-hexyl diradical mechanism. Quantum chemistry calculations at UCCSD(T)/cc-pVDZ level of theory on the initial reaction pathways of cyclohexane were performed and found to be consistent with the experimental conclusions. Cyclohexyl radical was not observed as an initial intermediate in the pyrolysis. Benzene was produced from sequential H2 eliminations of cyclohexane at high temperatures. [ABSTRACT FROM AUTHOR]

Abstract

Methamphetamine (METH) is a commonly abused psychostimulant that can induce severe neurotoxicity. Cardiovascular injury caused by METH has recently gained increasing attention; however, the underlying mechanisms remain unclear. As autophagy has been shown to be associated with cell injury, the association between autophagy and METH-mediated cell apoptosis was investigated in the present study. METH treatment significantly increased the expression of two key autophagy proteins, i.e., Beclin-1 and LC3-II, in the cardiomyocyte cell line H9C2. Furthermore, according to western blot and flow cytometry analyses, METH contributed to cell injury and markedly enhanced cleaved-caspase 3 and PARP expression. In addition, the corresponding AKT-mTOR survival pathway axis was appeared deactivated. The autophagic activity was closely associated with METH-mediated cell injury because rapamycin, which is an autophagy inducer, markedly attenuated METH-induced cell injury, while 3-Methyladenine (3-MA), which is an autophagy inhibitor, and bafilomycinA1 (Baf-A1), which is a blocker of autophagosome-lysosome fusion, markedly exacerbated METH-induced cell injury. Notably, defective autophagosome-lysosome fusion might be partially involved in the METH-induced enhancement of LC3-II expression and cell injury. However, the underlying mechanisms require further investigation.

Abstract

Photo-predissociation dynamics of jet-cooled hydroxyl radical (OH) via several rovibrational levels (v′ = 2–4, N′ = 0–2, J′ = 0.5–2.5, F1) in the A2Σ+ state are studied using the high–n Rydberg-atom time-of-flight (HRTOF) technique. Spin–orbit branching fractions and angular distributions of the H(2S) + O(3PJ = 2,1,0) product channels are measured in the product translational energy distributions. The A2Σ+v′ = 2 and 3 states of OH predissociate predominantly via the single 14Σ− repulsive state, while the A2Σ+v′ = 4 state predissociates via the 14Σ−, 12Σ−, and 14Π repulsive states and quantum interferences among these dissociation pathways play an important role in the O(3PJ = 2,1,0) product fine-structure state distribution. The experimental O(3PJ) product spin–orbit branching ratios are in excellent agreement with the full quantum multichannel scattering calculations that include all the interactions in the crossing region, recoupling zone, and asymptotic zone [Parlant and Yarkony, J. Chem. Phys. 110, 363 (1999)]. The product angular distributions strongly depend on the rotational transitions and rotational levels in the A2Σ+ state. The measured anisotropy parameters are in reasonable agreement with predictions from the simulation programme (Betaofnu) [Kim et al., J. Chem. Phys.125, 133316 (2006)], using dissociation lifetime, excitation frequency, rotational level, and rotational constant. The predissociation time scales of OH A2Σ+ are determined to be 130 ± 30 ns and ≥ 14 ± 3 ps for the v′ = 2, N′ = 2, J′ = 2.5 and v′ = 4, N′ = 0, J′ = 0.5 levels, respectively. [ABSTRACT FROM AUTHOR]

Abstract

Ultraviolet (UV) photodissociation dynamics of jet-cooled phenyl radicals (C6H5 and C6D5) are studied in the photolysis wavelength region of 215-268 nm using high-n Rydberg atom time-of-flight and resonance enhanced multiphoton ionization techniques. The phenyl radicals are produced from 193-nm photolysis of chlorobenzene and bromobenzene precursors. The H-atom photofragment yield spectra have a broad peak centered around 235 nm and are in good agreement with the UV absorption spectra of phenyl. The H + C6H4 product translational energy distributions, P(ET)'s, peak near ∼7 kcal/mol, and the fraction of average translational energy in the total excess energy, , is in the range of 0.20-0.35 from 215 to 268 nm. The H-atom product angular distribution is isotropic. The dissociation rates are in the range of 107-108 s-1 with internal energy from 30 to 46 kcal/mol above the threshold of the lowest energy channel H + o-C6H4 (ortho-benzyne), comparable with the rates from the Rice-Ramsperger-Kassel-Marcus theory. The results from the fully deuterated phenyl radical are identical. The dissociation mechanism is consistent with production of H + o-C6H4, as the main channel from unimolecular decomposition of the ground electronic state phenyl radical following internal conversion of the electronically excited state. Abstract

The abrasion of materials leads to huge economic losses and resource waste. It is a common measure to protect substrates with organic coatings. To obtain a type of waterborne polyurethane coating with excellent abrasion resistance, 6,6′-(1,4-butanediyl)-bis-(1,3,5-triazine-2,4-diamine) (BBTDA) was synthesized and employed to prepare crosslinked waterborne polyurethane (CWPU). The introduction of BBTDA allows the construction of covalent bonding crosslinking and hydrogen bonding crosslinking networks, which contribute to improving the mechanical properties and wear resistance of the CWPU coatings. The effects of BBTDA content on the mechanical properties, thermal stability, water resistance and wear resistance of CWPU were researched. The test result indicated that the addition of BBTDA improved the mechanical properties and wear resistance of the CWPU coatings significantly. When the BBTDA content was 0.5 %, the tensile strength of the film reached 26 MPa, the water absorption decreased to 4.7 %, the hardness of the coating was 4H, and the abrasion value was only 0.4 mg. CWPU-0.5 coating offered the optimum combination property. • A crosslinked waterborne polyurethane coating with excellent abrasion resistance was prepared. • The synthesized crosslinker constructed a dual crosslinking network. • Synergistic effect of covalent crosslinking and hydrogen bonding crosslinking enhanced mechanical properties and abrasion resistance of the coatings.
