Genes encoding VQ motif-containing (VQ) transcriptional regulators and WRKY transcription factors can participate separately or jointly in plant growth, development, and abiotic and biotic stress responses. In this study, 222 VQ and 645 WRKY genes were identified in six Prunus species. Based on phylogenetic tree topologies, the VQ and WRKY genes were classified into 13 and 32 clades, respectively. Therefore, at least 13 VQ gene copies and 32 WRKY gene copies were present in the genome of the common ancestor of the six Prunus species. Similar small Ks value peaks for the VQ and WRKY genes suggest that the two gene families underwent recent duplications in the six studied species. The majority of the Ka/Ks ratios were less than 1, implying that most of the VQ and WRKY genes had undergone purifying selection. Pi values were significantly higher in the VQ genes than in the WRKY genes, and the VQ genes therefore exhibited greater nucleotide diversity in the six species. Forty-one of the Prunus VQ genes were predicted to interact with 44 of the WRKY genes, and the expression levels of some predicted VQ-WRKY interacting pairs were significantly correlated. Differential expression patterns of the VQ and WRKY genes suggested that some might be involved in regulating aphid resistance in P. persica and fruit development in P. avium.

Met tyrosine kinase, a receptor for a hepatocyte growth factor (HGF), plays a critical role in tumor growth, metastasis, and drug resistance. Mitochondria are highly dynamic and undergo fission and fusion to maintain a functional mitochondrial network. Dysregulated mitochondrial dynamics are responsible for the progression and metastasis of many cancers. Here, using structured illumination microscopy (SIM) and high spatial and temporal resolution live cell imaging, we identified mitochondrial trafficking of receptor tyrosine kinase Met. The contacts between activated Met kinase and mitochondria formed dramatically, and an intact HGF/Met axis was necessary for dysregulated mitochondrial fission and cancer cell movements. Mechanically, we found that Met directly phosphorylated outer mitochondrial membrane protein Fis1 at Tyr38 (Fis1 pY38). Fis1 pY38 promoted mitochondrial fission by recruiting the mitochondrial fission GTPase dynamin-related protein-1 (Drp1) to mitochondria. Fragmented mitochondria fueled actin filament remodeling and lamellipodia or invadopodia formation to facilitate cell metastasis in hepatocellular carcinoma (HCC) cells both in vitro and in vivo. These findings reveal a novel and noncanonical pathway of Met receptor tyrosine kinase in the regulation of mitochondrial activities, which may provide a therapeutic target for metastatic HCC.

Paraphoma radicina is the causal pathogen of alfalfa root rot. Identification of P. radicina using the traditional morphological methods is difficult and time-consuming. Therefore, there is a need to develop rapid molecular techniques that can detect and identify P. radicina. This study employed conventional and real-time polymerase chain reaction (PCR) assays using a primer pair designed from the pathogen’s internal transcribed spacer (ITS) region to detect the P. radicina levels in alfalfa plants. Accordingly, a 363 bp fragment of P. radicina DNA was amplified using the primers Par-F and Par-R. Conventional PCR could detect as little as 1.2 pg/μL, but real-time PCR was more sensitive and could amplify as low as 12 fg/μL. The two PCR methods detected P. radicina from 15 alfalfa field plants, among which nine had typical P. radicina symptoms, while the other six did not display the typical symptoms. These results show that the PCR assays were more sensitive in detecting P. radicina than the traditional isolation methods. Moreover, relative qPCR, employing the comparative threshold cycle (Ct) method, was reliable for quantifying P. radicina DNA from the infected alfalfa root tissues. The P. radicina DNA quantities detected by real-time PCR assay had high correlations with the disease severity index (DSI) rating of alfalfa cultivars. This study is the first report of P. radicina identification by conventional and real-time PCR assays. The assays might be useful in detecting P. radicina in alfalfa field plants and determining the levels of P. radicina DNA in different alfalfa cultivars for pathogen resistance evaluation.

A lack of efficient diagnostic tools for early and noninvasive diagnosis of breast cancer has restricted the clinical treatment effect. This problem might be addressed by the combination of aggregation-induced emission (AIE) fluorescence imaging and positron emission tomography (PET) with the dual advantages of high resolution and easy operation, and unlimited penetration and high sensitivity. Here, a mitochondria-targeted AIE luminogen (AIEgen) radiolabeled with 18 F was developed through a two-step radiochemical reaction by virtue of a prosthetic group. The obtained 18/19 F-Bz-CP imaging probe was examined by in vitro cell uptake and cell proliferation inhibition in two breast cancer cell lines, showing that the probe can efficiently target and locate in the mitochondria through the analysis of fluorescence imaging and PET simultaneously. Additionally, the probe can induce cancer cell apoptosis with the half maximal inhibitory concentration (IC50) of 4.8 μM for MCF-7 cells and 7.2 μM for T47D cells, indicating its potential application for breast cancer therapy.

We recently reported that intraperitoneal injection of 7,8-dihydroxyflavone (7,8-DHF), a brain-derived neurotrophic factor-mimicking small compound, could attenuate alcohol-related behaviors in a two-bottle choice ethanol consumption procedure (IA2BC) in rats via tropomyosin receptor kinase B in the ventral tegmental area (VTA), which is closely related to alcohol use disorder. However, the detailed mechanisms underlying the regulation of 7,8-DHF on alcohol drinking behavior remain elusive. In this study, we determined the role of nitric oxide (NO), a pleiotropic signaling molecule, in the VTA in the action of 7,8-DHF upon alcohol drinking behavior. Intermittent alcohol exposure led to the overexpression of NO in the VTA, especially 72 h after withdrawal from four weeks of ethanol exposure in IA2BC rats. A higher amount of alcohol intake was also found at the same time point, consistent with the overexpression of NO in the VTA. Microinjection of NG-Nitro-l-Arginine Methyl Ester, (NO synthase inhibitor) or 2-4-carboxyphenyl-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (NO scavenger) into the VTA inhibited alcohol intake, whereas application of S-Nitroso-N-acetyl-DL-penicillamine (SNAP, the NO donor) in the VTA further enhanced alcohol consumption in IA2BC rats. Interestingly, either 1H-[1,2,4]oxadiazolo[4,3,-a]quinoxalin-1-one (a sGC inhibitor) or KT5823 [a selective protein kinase G (PKG) inhibitor] blocked NO's enhancing effect on ethanol intake. Intraperitoneal injection of 7,8-DHF reduced the overexpression of NO; SNAP microinjected into the VTA reversed the inhibitory effects of 7,8-DHF on alcohol consumption. Our findings suggest that NO-cGMP-PKG might be involved in regulation of 7,8-DHF on alcohol consumption in IA2BC rats.

The discovery of band convergence has opened an effective avenue for significantly enhancing thermoelectric performance of SnTe, while alloying CdTe in SnTe is evidenced efficient for improving the valley degeneracy. However, the thermoelectric transport properties are limited due to the low solubility of CdTe in SnTe (∼3%). Inspired by the improvement of dimensionless figure of merit zT in Cu or Se-doped SnTe, investigating the effect of Cu2Se on the electronic and phonon transport properties of SnTe-CdTe alloys is highly desired. Traditionally, improving the quality factor can trigger an increase of the potential of a compound for higher zT, which is of importance for design of thermoelectric materials. Here, alloyed 3% Cu2Se in SnTe-3%CdTe system enables an increased peak zT, which is attributed by the optimization of electronic performance (∼21 μW cm−1 K-2 at 800 K), as well as the decreased lattice thermal conductivity owing to the enhanced mass and strain fluctuations. More importantly, alloying Cu2Se not only improves the quality factor from ∼0.25 to ∼0.45, resulting in a higher maximum potential zT, but also effectively preserves the Fermi energy in a relative optimized level. The current findings demonstrate the role of Cu2Se for manipulating thermoelectrics in SnTe.

A nano-CuO/silica sol wood preservative was obtained by dispersing CuO nanoparticles in propylene glycol and silica sol. Scanning electron microscopy, Fourier transform infrared spectroscopy, X-ray diffraction analysis, thermogravimetric analysis, and compressive tests were conducted to investigate the effects of different post-treatments, i.e., steaming at 100 °C and freezing at -30 °C, on the variations in microstructure, mechanical, physical, and thermal stability properties of the preservative-impregnated wood. The results revealed that the mechanical properties, water resistance, and thermal stability of the impregnated specimens were greatly ameliorated. The steaming treatment resulted in a more uniform and dense distribution of the preservative in the blocks. The steaming treatment performed better in terms of enhancing the compressive strength of the specimens, while the freezing treatment was more effective in improving the thermal stability of the specimens. Both the steaming and freezing treatments can considerably improve the water resistance of the specimens. The different post-treatments retain the basic properties of the wood; however, they differ in the improved wood properties and provide a basis for their selection in the industrial production of nano-preservatives.

Sevoflurane (Sev) is a commonly used anesthetic in hospitals that can cause neurotoxicity. Postoperative cognitive dysfunction (POCD) is a common clinical problem induced by some anesthetics. However, the exact mechanism of neurotoxicity induced by Sev is unclear. Here we studied a new mechanism of POCD induced by Sev. We treated 15-month-old mice with 2% Sev for 6 hours, and we had found that Sev causes POCD. Using isobaric tags for relative and absolute quantitation (iTRAQ), we found that the transporter and the metabolism of carbohydrates and inorganic ions were involved in the cognitive impairment induced by Sev. Using synchrotron radiation micro-X-ray fluorescence (μ-XRF), we showed that Sev caused the iron overload in the brain of 15-month-old mice. Subsequently, excessive iron led to oxidative stress and impaired mitochondrial function that further led to glucose metabolism disorder and reduced ATP production by regulating the expression of key enzyme genes or proteins including G6Pase, Pck1, and Cs. Meanwhile, Sev also inhibited the oxygen consumption rate and glucose absorption by downregulating the expression of glucose transporter 1 in cerebral vascular endothelial cells. The cross-dysfunction of iron and glucose metabolism caused the apoptosis in the cortex and hippocampus through Bcl2/Bax pathway. In conclusion, the data here showed a new mechanism that Sev caused apoptosis by cross-dysregulation of iron and glucose metabolism and induced energy stress in mice. Maintaining iron and glucose metabolism homeostasis may play an important role in cognitive impairment induced by Sev.

Photoresponsive materials with high photoresponse sensitivity and photofatigue resistance are indispensable for applications, such as textiles and sensors fields. Herein, we synthesized photoresponsive polyamide 6 (PA6) with spiropyran (SP) as molecular component, by incorporating spiropyran-containing diisocyanate activator into anionic ring-opening polymerization of e-caprolactam. To improve photosensitivity and reduce response time, the corresponding nanofibrous membranes were obtained by electrospinning. The reversibility of the isomerization of SP resulted in colored nanofibrous membrane patterns, which can be written, erased, and rewritten, using UV light and visible light. It revealed that photosensitivity of nanofibrous membranes were further improved, when appropriate polyethylene glycol was covalently embedded into PA6 segments to increase the chain flexibility. Photoresponse reversibility and photofatigue resistance displayed that UV–vis absorbance intensity of SP-embedded PA6 nanofibrous membrane decreased just 13% after 10 cycles of UV light irradiation, while its absorbance intensity in UV–vis spectra after visible light irradiation has negligible decrease. For better understanding the structure, electronic characteristics and UV–vis absorbance peak of SP-embedded PA6, theoretical calculations were also carried out. Finally, UV absorption of transparent and flexible poly(dimethyl siloxane) (PDMS)/photoresponsive nanofibers composite film was assessed. The composite film was photochromic, and possessed excellent property for blocking ultraviolet radiation. This work explores a strategy for synthesizing photoresponsive SP-embedded polyamide 6 by incorporating spiropyran-containing activator into anionic ring-opening polymerization of e-caprolactam. SP-embedded PA6 nanofibrous membrane with excellent photosensitivity and photofatigue resistance was electrospun. The isomerization reversibility of spiropyran-merocyanine (SP-MC) resulted in colored patterns which can be written, erased, and rewritten using UV light and visible light. Transparent photochromic composite film for UV absorption and protection was also developed.

Breast cancer (BC) is the most common malignancy with high morbidity and mortality in females worldwide. Emerging evidence indicates that transferrin receptor 1 (TfR1) plays vital roles in regulating cellular iron import. However, the distinct role of TfR1 in BC remains elusive. TfR1 expression was investigated using the TCGA, GEO, TIMER, UALCAN and Oncomine databases. The prognostic potential of TfR1 was evaluated by Kaplan-Meier (KM) plotter and univariate and multivariate Cox regression analyses. Moreover, Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG) and gene set enrichment analysis (GSEA) were used to explore the molecular mechanism of TfR1. The potential link between TfR1 expression and infiltrating abundances of immune cells was examined through the TIMER and CIBERSORT algorithm. The expression of TfR1 was dramatically upregulated in BC tissues. Increased TfR1 expression and decreased methylation levels of TfR1 were strongly correlated with multiple clinicopathological parameters. Elevated TfR1 expression was associated with a poor survival rate in BC patients. The nomogram model further confirmed that TfR1 could act as an independent prognostic biomarker in BC. The results of GO, KEGG and GSEA revealed that TfR1 was closely correlated with multiple signaling pathways and immune responses. Additionally, TfR1 was positively associated with the infiltration abundances of six major immune cells, including CD4+ T cells, CD8+ T cells, B cells, neutrophils, macrophages, and dendritic cells in BC. Interestingly, TfR1 influenced prognosis partially through immune infiltration. These comprehensive bioinformatics analyses suggest that TfR1 is a new independent prognostic biomarker and a potential target for immunotherapy in BC.

Propensity score matching (PSM) was performed to reduce bias due to confounding variables. The primary outcome of the study was overall survival (OS). The secondary outcomes were progression-free survival (PFS), objective response rate (ORR), disease control rate (DCR), and adverse events (AEs), respectively.Results: Sixty-four patients were enrolled in the study, among which 24 and 40 received TACE+IT and IT, respectively. The PSM cohort included 24 patients receiving TACE+IT (TACE+IT group) and 24 patients receiving IT (IT group) alone. During a median follow-up of 23 months, patients in TACE+IT group had significantly longer OS (median, 17.3 vs 11.8 months, P = 0.023), better ORR (41.7% vs 12.5%, P = 0.023) and DCR (79.2% vs 50.0%, P = 0.035) than those in the IT group, whereas a non-significant trend in PFS (median, 7.4 vs 6.7 months, P = 0.23) was observed. According to multivariable cox regression analysis, it was found that treatment modality was the only independent risk factor for OS (HR = 0.404, 95% CI = 0.179– 0.911, P < 0.05). There were no remarkable differences in AEs associated with ICIs and TKIs between the two groups, with the exception of gastrointestinal reaction.Conclusion: TACE combined with ICIs plus TKIs significantly improved OS, ORR, and DCR and showed a relatively longer PFS trend over ICIs combined with TKIs for advanced HCC.Keywords: hepatocellular carcinoma, transarterial chemoembolization, immune checkpoint inhibitors, tyrosine kinase inhibitors

Ceruloplasmin (CP) plays an important role in maintaining iron homeostasis. Cp gene knockout (Cp-/-) mice develop a neurodegenerative disease with aging and show iron accumulation in the brain. However, iron deficiency has also been observed in 3 M Cp-/- mice. The use of systemic Cp gene knockout is insufficient to reveal specific functions for CP in the central nervous system. Considering recent discoveries that astrocytes synthetize the majority of brain CP, we generated astrocyte conditional Cp knockout (CpGfapcKO) mice, and found that iron contents decreased in the cerebral cortex and hippocampus of young (6 M) and old (18 M) CpGfapcKO mice. Further experiments revealed that 6 M CpGfapcKO mice exhibited impaired learning and memory function, while 18 M CpGfapcKO mice exhibited improved learning and memory function. Our study demonstrates that astrocytic Cp deletion blocks brain iron influx through the blood-brain-barrier, with concomitantly increased iron levels in brain microvascular endothelial cells, resulting in brain iron deficiency and down-regulation of ferritin levels in neurons, astrocytes, microglia and oligodendrocytes. At the young age, the synapse density, synapse-related protein levels, 5-hydroxytryptamine and norepinephrine, hippocampal neurogenesis and myelin formation were all decreased in CpGfapcKO mice. These changes affected learning and memory impairment in young CpGfapcKO mice. In old CpGfapcKO mice, iron accumulation with aging was attenuated, and was accompanied by the alleviation of the ROS-MAPK-apoptosis pathway, Tau phosphorylation and β-amyloid aggregation, thus delaying age-related memory decline. Overall, our results demonstrate that astrocytic Cp deletion has divergent effects on learning and memory function via different regulatory mechanisms induced by decreased iron contents in the brain of mice, which may present strategies for the prevention and treatment of dementia.

Surface-enhanced Raman scattering (SERS) spectroscopy analysis has long been the central task of nanoscience and nanotechnology to realize the ultrasensitive recognition/quantitation applications. Recently, the blooming of artificial intelligence algorithms provides an edge tool to revolutionize the spectroscopy analysis, especially for multiple substances analysis and large-scale data handling. In this study, a single-model multi-tasks deep learning network is proposed to simultaneously achieve the qualitative recognition and quantitative analysis of SERS spectroscopy. The SERS spectra of two kinds of hypoglycemic drugs (PHE, ROS) and the corresponding mixtures are collected, respectively, with the concentration grade from 10−4 M to 10−8 M. Based on the SERS spectroscopy dataset, the loss functions and hyperparameters of the multi-tasks classifications model are optimized, and the recognition accuracies are tested by simulation experiments. It is demonstrated that the accuracy rates of qualitative and quantitative analysis can reach up to 99.0% and 98.4%, respectively. Moreover, the practical feasibility of this multi-tasks model is demonstrated by using it to achieve qualitative and quantitative analysis of PHE and ROS in complex serum matrix. Overall, this single-model multi-tasks deep learning network shows significant potential for the recognition and quantitation of SERS spectroscopy, which provides the algorithmic and experimental basis for large-scale and multiple substances SERS spectra analysis.

Purpose: To investigate whether the combination scheme of deep learning score (DL-score) and radiomics can improve preoperative diagnosis in the presence of micropapillary/ solid (MPP/SOL) patterns in lung adenocarcinoma (ADC). Material and Methods: A retrospective cohort of 514 confirmed pathologically lung ADC in 512 patients after surgery was enrolled. The clinicoradiographic model (model 1) and radiomics model (model2) were developed with logistic regression. The deep learning model (model3) was constructed based on the deep learning score (DL-score). The combine model(model4) was based on DL-score and R-score and clincoradiographic variables. The performance of these models was evaluated with area under the receiver operating characteristic curve (AUC) and compared using DeLong's test. The prediction nomogram was plotted and clinical utility was depicted with decision curve. Results: The performance of model1, model2, model3 and model4 was supported by AUCs of 0.848, 0.896,0.906, 0.921 in the Testing set, respectively. These models existed statistical significance (model4 vs model3, P=0.016; model4 vs model1, P=0.009, respectively). The decision curve analysis (DCA) demonstrated that model 4 predicting the lung ADC with MPP/SOL structure would be more beneficial than the model 1 but comparable with the model2 and model3. Conclusion: The combined model can improve preoperative diagnosis in the presence of MPP/SOL pattern in lung ADC in clinical practice.

Interferon regulatory factor (IRF) 3 and IRF7 are master regulators of type I interferon (IFN-I)-dependent antiviral innate immunity. Upon viral infection, a positive feedback loop is formed, wherein IRF7 promotes further induction of IFN-I in the later stage. Thus, it is critical to maintain a suitably low level of IRF7 to avoid the hyperproduction of IFN-I. In this study, we find that early expression of IFN-I-dependent STAT1 promotes the expression of XAF1 and that XAF1 is associated specifically with IRF7 and inhibits the activity of XIAP. XAF1-knockout and XIAP-transgenic mice display resistance to viral infection, and this resistance is accompanied by increases in IFN-I production and IRF7 stability. Mechanistically, we find that the XAF1-XIAP axis controls the activity of KLHL22, an adaptor of the BTB-CUL3-RBX1 E3 ligase complex through a ubiquitin-dependent pathway. CUL3-KLHL22 directly targets IRF7 and catalyzes its K48-linked ubiquitination and proteasomal degradation. These findings reveal unexpected functions of the XAF1-XIAP axis and KLHL22 in the regulation of IRF7 stability and highlight an important target for antiviral innate immunity.

Background: This study aimed to evaluate whether a large paraumbilical vein (L-PUV) was independently associated with the occurrence of overt hepatic encephalopathy (OHE) after the implantation of a transjugular intrahepatic portosystemic shunt (TIPS). Methods: This bi-center retrospective study included patients with cirrhotic variceal bleeding treated with a TIPS between December 2015 and June 2021. An L-PUV was defined in line with the following criteria: cross-sectional areas > 83 square millimeters, diameter ≥ 8 mm, or greater than half of the diameter of the main portal vein. The primary outcome was the 2-year OHE rate, and secondary outcomes included the 2-year mortality, all-cause rebleeding rate, and shunt dysfunction rate. Results: After 1:2 propensity score matching, a total of 27 patients with an L-PUV and 54 patients without any SPSS (control group) were included. Patients with an L-PUV had significantly higher 2-year OHE rates compared with the control group (51.9% vs. 25.9%, HR = 2.301, 95%CI 1.094–4.839, p = 0.028) and similar rates of 2-year mortality (14.8% vs. 11.1%, HR = 1.497, 95%CI 0.422–5.314, p = 0.532), as well as variceal rebleeding (11.1% vs. 13.0%, HR = 0.860, 95%CI 0.222–3.327, p = 0.827). Liver function parameters were similar in both groups during the follow-up, with a tendency toward higher shunt patency in the L-PUV group (p = 0.067). Multivariate analysis indicated that having an L-PUV (HR = 2.127, 95%CI 1.050–4.682, p = 0.037) was the only independent risk factor for the incidence of 2-year OHE. Conclusions: Having an L-PUV was associated with an increased risk of OHE after a TIPS. Prophylaxis management should be considered during clinical management.

Vascular calcification (VC) is an important hallmark of cardiovascular disease, the osteo-/chondrocyte phenotype differentiation of vascular smooth muscle cells (VSMCs) is the main cause of vascular calcification. Accumulating evidence shows that mitochondrial dysfunction may ultimately be more detrimental in the VSMCs calcification. Mitochondrial participate in essential cellular functions, including energy production, metabolism, redox homeostasis regulation, intracellular calcium homeostasis, apoptosis, and signal transduction. Mitochondrial dysfunction under pathological conditions results in mitochondrial reactive oxygen species (ROS) generation and metabolic disorders, which further lead to abnormal phenotypic differentiation of VSMCs. In this review, we summarize existing studies targeting mitochondria as a treatment for VC, and focus on VSMCs, highlighting recent progress in determining the roles of mitochondrial processes in regulating the phenotype transition of VSMCs, including mitochondrial biogenesis, mitochondrial dynamics, mitophagy, mitochondrial energy metabolism, and mitochondria/ER interactions. Along these lines, the impact of mitochondrial homeostasis on VC is discussed.

Nuclear receptor 4A1 (NR4A1), a member of the NR4A subfamily, acts as a gene regulator in a wide range of signaling pathways and responses to human diseases. Here, we provided a brief overview of the current functions of NR4A1 in human diseases and the factors involved in its function. A deeper understanding of these mechanisms can potentially improve drug development and disease therapy.

Mitogenomes of most flowering plants evolve slowly in sequence, but rapidly in structure. The rearrangements in structure are mainly caused by repeat-mediated recombination. However, patterns of repeat-mediated recombination vary substantially among plants, and to provide a comprehensive picture, characterization of repeat-mediated recombination should extend to more plant species, including parasitic plants with a distinct heterotrophic lifestyle. Here we assembled the mitogenome of the holoparasitic plant Aeginetia indica (Orobanchaceae) using Illumina sequencing reads. The mitogenome was assembled into a circular chromosome of 420,362 bp, 18,734 bp longer than that of another individual of A. indica which was assembled before as a linear molecule. Synteny analysis between the two mitogenomes revealed numerous rearrangements, unique regions of each individual and 0.2% sequence divergence in their syntenic regions. The A. indica mitogenome contains a gene content typical of flowering plants (33 protein-coding, 3 rRNA, and 17 tRNA genes). Repetitive sequences >30 bp in size totals 57,060 bp, representing 13.6% of the mitogenome. We examined recombination mediated by repeats >100 bp in size and found highly active recombination for all the repeats, including a very large repeat of ~16 kb. Recombination between these repeats can form much smaller subgenomic circular chromosomes, which may lead to rapid replication of mitochondrial DNA and thus be advantageous for A. indica with a parasitic lifestyle. In addition, unlike some other parasitic plants, A. indica shows no evidence for horizontal gene transfer of protein-coding genes in its mitogenome.

Dendrobium polysaccharide exhibits multiple biological activities, such as immune regulation, antioxidation, and antitumor. However, its resistance to viral infection by stimulating immunity is rarely reported. In this study, we explored the effect and mechanism of DVP-1, a novel polysaccharide from Dendrobium devonianum, in the activation of immunity. After being activated by DVP-1, the ability of mice to prevent H1N1 influenza virus infection was investigated. Results of immune regulation showed that DVP-1 significantly improved the immune organ index, lymphocyte proliferation, and mRNA expression level of cytokines, such as IL-1β, IL-4, IL-6, and TNF-α in the spleen. Immunohistochemical results showed that DVP-1 obviously promoted the mucosal immunity in the jejunum tissue. In addition, the expression levels of TLR4, MyD88, and TRAF6 and the phosphorylation levels of TAK1, Erk, JNK, and NF-κB in the spleen were upregulated by DVP-1. The virus infection results showed that the weight loss of mice slowed down, the survival rate increased, the organ index of the lung reduced, and the virus content in the lung decreased after DVP-1 activated immunity. By activating immunity with DVP-1, the production of inflammatory cells and inflammatory factors in BALF, and alveolar as well as peribronchiolar inflammation could be prevented. The results manifested that DVP-1 could resist H1N1 influenza virus infection by activating immunity through the TLR4/MyD88/NF-κB pathway.

ObjectivesThe aim of the current study is to explore the association between extended adjuvant endocrine treatment and prognosis of women with hormone receptor-positive (HR+) early breast cancer.MethodsDatabases including PubMed, Web of Science, Embase and the Cochrane Library databases were electronically searched to identify randomized controlled trials (RCTs) that reported extended endocrine therapy for women with HR+ early breast cancer. The retrieval time was limited from inception to September 2022. Two reviewers independently screened literature, extracted data, and assessed risk bias of included studies. Meta-analysis was performed by using R software Version 4.1.2 and STATA Version 12.0.ResultsA total of 15 RCTs involving 29497 cases were included. The overall analysis showed that compared with the control, extended adjuvant endocrine therapy increased disease-free survival (DFS) (HR=0.814, 95% CI: 0.720-0.922, 95% PI: 0.556-1.194), overall survival (OS) (HR=0.885, 95% CI: 0.822-0.953, 95% PI: 0.771-1.035), relapse-free survival (RFS) (HR=0.833, 95% CI: 0.747-0.927, 95% PI: 0.575-1.159), distant metastatic-free survival (DMFS) (HR=0.824, 95% CI: 0.694-0.979, 95% PI: 0.300-2.089) and reduced new breast cancer cumulative incidence (NBCCI) (HR=0.484, 95% CI: 0.403-0.583, 95% PI: 0.359-0.654). For adverse events, extended adjuvant endocrine treatment was associated with a significantly higher risk of bone fracture (RR=1.446, 95% CI: 1.208-1.730, 95% PI: 1.154-1.854) and osteoporosis (RR=1.377, 95% CI: 1.018-1.862, 95% PI: 0.347-5.456).ConclusionOur study showed that extended adjuvant endocrine therapy increased DFS, OS, RFS, DMFS, the incidence of bone fracture and osteoporosis, and reduced NBCCI.Systematic Review Registrationhttps://www.crd.york.ac.uk/prospero, identifier (CRD42022351295)

Objective: This study aimed to determine the effective dose 50% (ED50) value of remifentanil in inhibiting coughing during extubation in children with snoring.Methods: The subjects were children who scored a grade I in the American Society of Anesthesiology (ASA) metric and who were undergoing tonsillectomy (with or without adenoidectomy) under general anesthesia. Using Dixon’s up-and-down sequential method, the initial infusion rate of remifentanil was 0.06 μg/kg/min, and the difference between the infusion rates of the two adjacent groups was 0.01 μg/kg/min. If a child had no cough response during extubation, the infusion rate for the next child was reduced by 0.01 μg/kg/min. If that child had cough response, the infusion rate for the next child was increased by 0.01 μg/kg/min, and the test was terminated when seven pairs of children with positive-negative alternating results were obtained. The ED50 value and its 95% confidence interval (CI) were calculated by probit regression. The times for extubation, awakening, agitation, and respiratory complications after extubation were compared between the two groups.Results: 1) The ED50 value of a continuous infusion of remifentanil required to inhibit the cough response of children during extubation was 0.042 μg/kg/min, and the 95% confidence interval was 0.025–0.062 μg/kg/min. 2) The total dosage and infusion rate of remifentanil in the cough suppression group were higher than those in the cough group (p < 0.05), but the differences in the times for extubating and awakening between the two groups were not statistically significant (p > 0.05). 3) There was no correlation between the infusion rate of remifentanil and the time for extubating and awakening in the cough suppression group; the r values were 0.13 and 0.12, respectively, and p > 0.05. 4) The differences in postoperative respiratory complications between the two groups were not statistically significant (p > 0.05).Conclusion: The ED50 value of a continuous infusion of remifentanil required to inhibit the cough response of children during extubation after tonsillectomy (with or without adenoidectomy) was 0.042 μg/kg/min, and a low-dose infusion of remifentanil does not affect the times for awakening and extubating in children.

Soil crust is a normal natural phenomenon with different water hydrophilicity and repellency due to different formation mechanism, thus affecting soil hydraulic characteristics and hydrological cycle. In this study, we measured water repellent characteristics of physical and biological crusts under different vegetations in the field using water drop penetration time (WDPT). The surface morphology of crusts was observed using scanning electron microscopy, and the infiltration characteristics of crusts and their non-crust soils (control) was evaluated with micro-infiltration device. The results showed that: 1) The average WDPT of physical crusts and the control soils was 3.3 s and 0.9 s, respectively, indicating that both were hydrophilic. The average WDPT of biological crusts ranged from 20.9 s to 140.9 s, which was 2.8 to 19 times that of control, and that under土壤结皮是一种常见的自然现象,但由于结皮形成机制的不同,会产生不同的亲水性和斥水性,从而影响土壤的水力学特征与水文循环。本研究利用水滴穿透时间法测定了野外不同植被下物理结皮和生物结皮的斥水特征,利用扫描电镜观测了结皮的表面形态,并用微型入渗装置测定了结皮及其对照土壤的入渗特征。结果表明: 1)物理结皮的平均水滴穿透时间(WDPT)为3.3 s,对照为0.9 s,表现为亲水性;生物结皮的平均WDPT介于20.9～140.9 s,是无结皮的2.8～19倍,其中君迁子和刺槐林下的生物结皮平均WDPT分别为134.5和140.9 s。2)与对照相比,物理结皮累积入渗量、平均入渗速率和吸湿力分别降低了0～4.3%、3.5%～5.1%和27.2%～90.1%,生物结皮分别降低了0～25%、1.4%～28.2%和36.0%～84.9%。3)无论是否存在结皮,利用Philip模型拟合处理入渗数据均存在“曲棍球状”曲线;曲线上斥水性停止时间(WRCT)之前,点源微入渗以水平方向上的扩散为主,WRCT点以后以垂直方向上的入渗为主,土壤结皮的形成延长了该转折点的形成时间。综上,物理结皮是无机矿质颗粒堵塞了表层土壤,不影响斥水性的变化;生物结皮表现为斥水性有机物对土壤结构的影响,增强了其斥水性。物理结皮和生物结皮均会降低土壤的累积入渗量和平均入渗速率,但物理结皮主要影响土壤的吸湿力,对稳定入渗速率影响不大;生物结皮不仅降低了土壤吸湿力,还增加了稳定入渗速率。.
