401. International Monitoring System infrasound data products for atmospheric studies and civilian applications

Abstract

The International Monitoring System (IMS) was established in the late 1990s for verification of the Comprehensive Nuclear-Test-Ban Treaty (CTBT). Upon completion, 60 infrasound stations distributed over the globe will monitor the Earth's atmosphere for low-frequency pressure waves. In this study, we present advanced infrasound data products of the 53 currently certified IMS infrasound stations for atmospheric studies and civilian applications. For this purpose, 18 years of raw IMS infrasound waveform data (2003–2020) were reprocessed using the Progressive Multi-Channel Correlation (PMCC) method. A one-third octave frequency band configuration between 0.01 and 4 Hz was chosen to run this array-processing algorithm which detects coherent infrasound waves within the background noise. From the comprehensive detection lists, four products were derived for each of the certified 53 IMS infrasound stations. The four products cover different frequency ranges and are provided at the following different temporal resolutions: a very low-frequency set (0.02–0.07 Hz, 30 min; https://doi.org/10.25928/bgrseis_bblf-ifsd, Hupe et al., 2021a), two so-called microbarom frequency sets – covering both the lower (0.15–0.35 Hz, 15 min; https://doi.org/10.25928/bgrseis_mblf-ifsd, Hupe et al., 2021b) and a higher (0.45–0.65 Hz, 15 min; https://doi.org/10.25928/bgrseis_mbhf-ifsd, Hupe et al., 2021c) part – named after the dominant ambient noise of interacting ocean waves that are quasi-continuously detected at IMS stations, and observations with center frequencies of 1 to 3 Hz (5 min), called the high-frequency product (https://doi.org/10.25928/bgrseis_bbhf-ifsd, Hupe et al., 2021d). Within these frequency ranges and time windows, the dominant repetitive signal directions are summarized. Along with several detection parameters, calculated quantities for assessing the relative quality of the products are provided. The validity of the data products is demonstrated through example case studies of recent events that produced infrasound detected at IMS infrasound stations and through a global assessment and summary of the products. The four infrasound data products cover globally repeating infrasound sources such as ocean ambient noise or persistently active volcanoes, which have previously been suggested as sources for probing the winds in the middle atmosphere. Therefore, our infrasound data products open up the IMS observations also to user groups who do not have unconstrained access to IMS data or who are unfamiliar with infrasound data processing using the PMCC method. These types of data products could potentially serve as a basis for volcanic eruption early warning systems in the future.

Abstract

Decoy technology is a versatile and specific DNA oligonucleotide-based targeting strategy of pathogenic transcription factors (TFs). Chemical modifications of linear decoy oligonucleotides have been made to decrease nuclease sensitivity because of the presence of free ends but at the cost of new limitations that affect their use as therapeutic drugs. Although a short DNA minicircle is a phosphodiester nucleic acid without free ends, its potential therapeutic activity as a TF decoy oligonucleotide has not yet been investigated. Here we describe the in vitro and in vivo activity of formulated 95-bp minicircles bearing one or several STAT3 binding sequences in triple-negative breast cancer (TNBC). Minicircles bearing one STAT3 binding site interacted specifically with the active form of STAT3 and inhibited proliferation, induced apoptosis, slowed down cell cycle progression, and decreased STAT3 target gene expression in human and murine TNBC cells. Intratumoral injection of STAT3 minicircles inhibited tumor growth and metastasis in a murine model of TNBC. Increasing the number of STAT3 binding sites resulted in improved anticancer activity, opening the way for a TF multitargeting strategy. Our data provide the first demonstration of minicircles acting as STAT3 decoys and show that they could be an effective therapeutic drug for TNBC treatment.

Abstract

Abstract The lakeshore site of La Marmotta is one of the most important Early Neolithic sites of Mediterranean Europe. The site is famous for the exceptional preservation of organic materials, including numerous wooden artefacts related to navigation, agriculture, textile production, and basketry. This article presents interdisciplinary research on three of the most complete and well-preserved sickles recovered from the site, yet unpublished. All the components of the tools are analysed: the stone inserts, the wooden haft and the adhesive substances used to fix the stones inside the haft. Our innovative methodology combines use-wear and microtexture analysis of stone tools through confocal microscopy, taxonomical and technological analysis of wood, gas chromatography–mass spectrometry analysis of the adhesive substances, and pollen, non-pollen palynomorphs, and phytolith analysis of the remains incorporated within the adhesive. This multiproxy approach provides a significant insight into the life of these tools, from their production to their use and abandonment, providing evidence of the species of harvested plants and the conditions of the field during the harvesting.

Abstract

IntroductionRac-GTPases and their Rac-GEF activators play important roles in neutrophil-mediated host defence. These proteins control the adhesion molecules and cytoskeletal dynamics required for neutrophil recruitment to inflamed and infected organs, and the neutrophil effector responses that kill pathogens.MethodsHere, we used live cell TIRF-FRET imaging in neutrophils from Rac-FRET reporter mice with deficiencies in the Rac-GEFs Dock2, Tiam1 or Prex1/Vav1 to evaluate if these proteins activate spatiotemporally distinct pools of Rac, and to correlate patterns of Rac activity with the neutrophil responses they control.ResultsAll the GEFs were required for neutrophil adhesion, and Prex1/Vav1 were important during spreading and for the velocity of migration during chemotaxis. However, Dock2 emerged as the prominent regulator of neutrophil responses, as this GEF was required for neutrophil polarisation and random migration, for migration velocity during chemokinesis, for the likelihood to migrate and for the speed of migration and of turning during chemotaxis, as well as for rapid particle engulfment during phagocytosis. We identified characteristic spatiotemporal patterns of Rac activity generated by Dock2 which correlate with the importance of the Rac-GEF in these neutrophil responses. We also demonstrate a requirement for Dock2 in neutrophil recruitment during aseptic peritonitis.DiscussionCollectively, our data provide a first direct comparison of the pools of Rac activity generated by different types of Rac-GEFs, and identify Dock2 as a key regulator of polarisation, migration and phagocytosis in primary neutrophils.

Abstract

In this paper we present EMO-5 (“European Meteorological Observations”, spatial resolution of 5 km), a European high-resolution, (sub-)daily, multi-variable meteorological dataset built on historical and real-time observations obtained by integrating data from 18 964 ground weather stations, four high-resolution regional observational grids (i.e. CombiPrecip, ZAMG – INCA, EURO4M-APGD, and CarpatClim), and one global reanalysis (ERA-Interim/Land). EMO-5 includes the following at daily resolution: total precipitation, temperatures (minimum and maximum), wind speed, solar radiation, and water vapour pressure. In addition, EMO-5 also makes available 6-hourly precipitation and mean temperature data. The raw observations from the ground weather stations underwent a set of quality controls before SPHEREMAP and Yamamoto interpolation methods were applied in order to estimate for each 5×5 km grid cell the variable value and its affiliated uncertainty, respectively. The quality of the EMO-5 precipitation data was evaluated through (1) comparison with two regional high-resolution datasets (i.e. seNorge2 and seNorge2018), (2) analysis of 15 heavy precipitation events, and (3) examination of the interpolation uncertainty. Results show that EMO-5 successfully captured 80 % of the heavy precipitation events, and that it is of comparable quality to a regional high-resolution dataset. The availability of the uncertainty fields increases the transparency of the dataset and hence the possible usage. EMO-5 (version 1) covers the time period from 1990 to 2019, with a near real-time release of the latest gridded observations foreseen with version 2. Abstract

Abstract Background The purpose of SNAP Out Stigma (SOS) was to design and implement a community-based intervention to reduce HIV-related stigma for people living with HIV (PLWH) in the Deep South. This region is a subset of 9 states including Memphis, Tennessee (project site) driving the epidemic in the United States. The purpose of this paper is to explain how PLWH in the U.S. South used Photovoice to communicate stigmatizing lived experiences and contextualize their intersection with multi-level sources of support. Methods PLWH attended one-on-one and/or group sessions with other PLWH. In Session 1, PLWH received a project overview, met other participants, received a camera and camera training, completed a standardized internalized stigma scale, discussed experiences of internalized stigma, and were instructed to take 3–10 pictures that captured stigma. In Session 2, PLWH discussed the pictures and their meaning. In Session 3, PLWH expanded on what they shared in previous sessions in a one-on-one interview. Thematic analysis captured key patterns of how PLWH experienced stigma. Results Forty-seven PLWH attended Session 1 and were issued a camera. Of those, 35 completed sessions 2 and 3. Participants self-identified as cis man who has sex with men (n = 18), ciswoman (n = 5), transwoman (n = 10), and non-binary (n = 2). Four emergent themes intersecting with internalizations of stigma were identified including: medical, social support, church, and self. Conclusions The SOS intervention created a safe space for PLWH to share lived experiences of stigmatization. Photovoice facilitated discussion topics ranging from healing and recovery to overcoming factors of social determinants of HIV. We identified trauma-informed growth as an area of future programs for PLWH.

Abstract

Using the Horizon-AGN simulation data, we study the relative role of mergers and environmental effects in shaping the spin of early-type galaxies (ETGs) after $z \simeq 1$. We follow the spin evolution of 10,037 color-selected ETGs more massive than 10$^{10} \rm \, M_{\odot}$ that are divided into four groups: cluster centrals (3%), cluster satellites (33%), group centrals (5%), and field ETGs (59%). We find a strong mass dependence of the slow rotator fraction, $f_{\rm SR}$, and the mean spin of massive ETGs. Although we do not find a clear environmental dependence of $f_{\rm SR}$, a weak trend is seen in the mean value of spin parameter driven by the satellite ETGs as they gradually lose their spin as their environment becomes denser. Galaxy mergers appear to be the main cause of total spin changes in 94% of central ETGs of halos with $M_{vir} > 10^{12.5}\rm M_{\odot}$, but only 22% of satellite and field ETGs. We find that non-merger induced tidal perturbations better correlate with the galaxy spin-down in satellite ETGs than mergers. Given that the majority of ETGs are not central in dense environments, we conclude that non-merger tidal perturbation effects played a key role in the spin evolution of ETGs observed in the local ($z < 1$) universe., Comment: Accepted for publication in ApJ

Abstract

Using the two large cosmological hydrodynamical simulations, Horizon-AGN (H-AGN) and Horizon-noAGN (H-noAGN, no AGN feedback), we investigate how a typical sub-grid model for AGN feedback affects the evolution of the total density profiles (dark matter + stars) at the effective radius of massive early-type galaxies (M*>10^11 Msun). We have studied the dependencies of the mass-weighted density slope gamma'_tot with the effective radius, the galaxy mass and the host halo mass at z~0.3 and found that the inclusion of AGN feedbackalways leads to a much better agreement with observational values and trends. Our analysis suggests also that the inclusion of AGN feedback favours a strong correlation between gamma'_tot and the density slope of the dark matter component while, in the absence of AGN activity, gamma'_tot is rather strongly correlated with the density slope of the stellar component. Finally, we find that gamma'_tot derived from our samples of galaxies increases from z=2 to z=0,in good agreement with the expected observational trend. The derived slopes are slightly lower than in the data when AGN is included because the simulated galaxies tend to be too extended, especially the least massive ones. Abstract

Understanding the processes that drive the formation of black holes (BHs) is a key topic in observational cosmology. While the observed $M_{\mathrm{BH}}$--$M_{\mathrm{Bulge}}$ correlation in bulge-dominated galaxies is thought to be produced by major mergers, the existence of a $M_{\mathrm{BH}}$--$M_{\star}$ relation, across all galaxy morphological types, suggests that BHs may be largely built by secular processes. Recent evidence that bulge-less galaxies, which are unlikely to have had significant mergers, are offset from the $M_{\mathrm{BH}}$--$M_{\mathrm{Bulge}}$ relation, but lie on the $M_{\mathrm{BH}}$--$M_{\star}$ relation, has strengthened this hypothesis. Nevertheless, the small size and heterogeneity of current datasets, coupled with the difficulty in measuring precise BH masses, makes it challenging to address this issue using empirical studies alone. Here, we use Horizon-AGN, a cosmological hydrodynamical simulation to probe the role of mergers in BH growth over cosmic time. We show that (1) as suggested by observations, simulated bulge-less galaxies lie offset from the main $M_{\mathrm{BH}}$--$M_{\mathrm{Bulge}}$ relation, but on the $M_{\mathrm{BH}}$--$M_{\star}$ relation, (2) the positions of galaxies on the $M_{\mathrm{BH}}$--$M_{\star}$ relation are not affected by their merger histories and (3) only $\sim$35 per cent of the BH mass in today's massive galaxies is directly attributable to merging -- the majority ($\sim$65 per cent) of BH growth, therefore, takes place gradually, via secular processes, over cosmic time., Comment: 12 pages, 9 figures, accepted for publication in MNRAS

Abstract

Accurate cosmology from upcoming weak lensing surveys relies on knowledge of the total matter power spectrum at percent level at scales $k < 10$ $h$/Mpc, for which modelling the impact of baryonic physics is crucial. We compare measurements of the total matter power spectrum from the Horizon cosmological hydrodynamical simulations: a dark matter-only run, one with full baryonic physics, and another lacking Active Galactic Nuclei (AGN) feedback. Baryons cause a suppression of power at $k\simeq 10$ $h/$Mpc of $<15\%$ at $z=0$, and an enhancement of a factor of a few at smaller scales due to the more efficient cooling and star formation. The results are sensitive to the presence of the highest mass haloes in the simulation and the distribution of dark matter is also impacted up to a few percent. The redshift evolution of the effect is non-monotonic throughout $z=0-5$ due to an interplay between AGN feedback and gas pressure, and the growth of structure. We investigate the effectiveness of an analytic `baryonic correction model' in describing our results. We require a different redshift evolution and propose an alternative fitting function with $4$ free parameters that reproduces our results within $5\%$. Compared to other simulations, we find the impact of baryonic processes on the total matter power spectrum to be smaller at $z=0$. Correspondingly, our results suggest that AGN feedback is not strong enough in the simulation. Abstract

Grasslands' high diversity is threatened by land-use changes, such as nitrogen fertilization, leading to productive but low-richness, fast-growing plant communities. Bryophytes are a key component of grassland diversity and react strongly to land use. However, it is unclear whether land-use effects are direct or mediated by changes in vascular plants. Increases in vascular plant cover are likely to decrease bryophyte abundance through light competition. Whether changes in vascular plant composition and richness also play a role remains unclear. We sampled bryophytes in a factorial grassland experiment manipulating nitrogen fertilization, fungicide, species richness, and functional composition of vascular plants crossed with moderate disturbances by weeding. Disturbance increased bryophyte richness and modulated treatment effects. In contrast to previous studies reporting indirect negative fertilization effects via increasing vascular plant productivity and reduced light levels, nitrogen fertilization directly reduced bryophyte cover and species richness, possibly because of toxic effects. Low vascular plant richness and dominance of fast-growing species reduced bryophyte richness. This might be because of decreased structural and resource niche heterogeneity in species-poor communities. Our results highlight novel mechanisms by which land-use intensification can affect bryophytes and suggest that a loss of vascular plant richness might have cascading effects on other taxonomic groups. Abstract

Traumatic brain injury (TBI) is a risk factor for neurodegeneration, however little is known about how this kind of injury alters neuron subtypes. In this study, we follow neuronal populations over time after a single mild TBI (mTBI) to assess long ranging consequences of injury at the level of single, transcriptionally defined neuronal classes. We find that the stress-responsive Activating Transcription Factor 3 (ATF3) defines a population of cortical neurons after mTBI. Using an inducible reporter linked to ATF3, we genetically mark these damaged cells to track them over time. We find that a population in layer V undergoes cell death acutely after injury, while another in layer II/III survives long term and remains electrically active. To investigate the mechanism controlling layer V neuron death, we genetically silenced candidate stress response pathways. We found that the axon injury responsive dual leucine zipper kinase (DLK) is required for the layer V neuron death. This work provides a rationale for targeting the DLK signaling pathway as a therapeutic intervention for traumatic brain injury. Beyond this, our approach to track neurons after a mild, subclinical injury can inform our understanding of neuronal susceptibility to repeated impacts. This work shows that concussion, even without symptoms, causes neuron death, with ATF3-expressing layer V cortical neurons more vulnerable than those in layer II/III. Astrophysical black holes (BHs) have two fundamental properties: mass and spin. While the mass-evolution of BHs has been extensively studied, much less work has been done on predicting the distribution of BH spins. In this paper, we present the spin evolution for a sample of intermediate-mass and massive BHs from the NewHorizon simulation, which evolved BH spin across cosmic time in a full cosmological context through gas accretion, BH–BH mergers and BH feedback including jet spindown. As BHs grow, their spin evolution alternates between being dominated by gas accretion and BH mergers. Massive BHs are generally highly spinning. Accounting for the spin energy extracted through the Blandford–Znajek mechanism increases the scatter in BH spins, especially in the mass range |$10^{5}{-}10^{7}\,\rm M_\odot$|⁠ , where BHs had previously been predicted to be almost universally maximally spinning. We find no evidence for spin-down through efficient chaotic accretion. As a result of their high spin values, massive BHs have an average radiative efficiency of |$\lt \varepsilon _{\rm r}^{\rm thin}\gt \approx 0.19$|⁠. As BHs spend much of their time at low redshift with a radiatively inefficient thick disc, BHs in our sample remain hard to observe. Different observational methods probe different sub-populations of BHs, significantly influencing the observed distribution of spins. Generally, X-ray-based methods and higher luminosity cuts increase the average observed BH spin. When taking BH spin evolution into account, BHs inject, on average, between three times (in quasar mode) and eight times (in radio mode) as much feedback energy into their host galaxy as previously assumed.
