A palladium-catalyzed deuterated formylation of aryl sulfonium salts is prepared conveniently from readily available arenes, which enables the expedient synthesis of a series of structurally diverse C-1 deuterated aldehydes with 96%-99% deuterium incorporation. The easy to handle and cost-effective DCOONa provides a deuterium source, which can be introduced onto the formyl units with excellent selectivity under the palladium-catalytic redox neutral conditions. This catalytic route can accomplish the direct late-stage C-H functionalization of bioactive molecules and natural product derivatives assisted by C (sp 2 )-H thianthrenation. Moreover, on the basis of this practical approach, several deuterated drugs and analogues could be prepared with excellent levels of deuterium incorporation.

The simple, commercially available ZnBr 2 has been successfully employed as a highly efficient and chemoselective catalyst for the dehydrogenative borylation of terminal alkynes with HBpin under mild conditions. It shows a good tolerance toward various functional groups such as aryl, alkyl, heteroaryl, etc. The plausible reaction mechanism has been investigated based on the corresponding stoichiometric experiments and DFT calculations.

An unprecedented and expeditious tandem bisannulation of polyfluoroalkylated tetralones with benzamidines to access various fluoroalkyl tetracyclic [1,3]-diazepines through multiple C-N bond formation and C(sp 3 )-F bond cleavage is reported. The process features high regio-/chemoselectivities, broad substrate scope, good functional group tolerance, procedural simplicity, mild reaction conditions, and scale-up synthesis. Mechanistic studies showed that the distinctive fluorine effect of polyfluoroalkyl tetralone plays a vital role for the aza -tetracycle construction.

Five bioactive dehydroabietylamine Schiff-base derivatives (L 1 -L 5 ) had been synthesized from Dehydroabietylamine (L 0 ), and the complex Cu(L 1 ) 2 had been obtained from the compound L 1 and copper(II) acetate. Their activities against Hela (cervix), MCF-7 (breast), A549 (lung), HepG2 (liver) and HUVEC (umbilical vein, normal cell) in vitro were investigated. The toxicity of L 1 -L 5 and Cu(L 1 ) 2 was all lower than L 0 . For MCF-7 cell, L 1 , L 3 , L 4 , L 5 and Cu(L 1 ) 2 had higher antitumor activity than L 0 . The smallest IC 50 value was 2.58 μM of L 5 . For A549 cell, the IC 50 value of the compound L 4 was smaller than L 0 , which indicated that the compound L 4 had higher anti-A549 activity than L 0 . For HepG2 cell, the IC 50 value of L 4 (0.24 μM) and L 5 (0.14 μM) were much smaller than L 0 , which suggested L 4 and L 5 had higher anti-HepG2 activity. L 5 was 180 times more effective at inhibiting cultured HepG2 cells survival than normal cells, with average IC 50 values of 0.14 and 25.56 μM. Furthermore, L 0 , L 4 and L 5 contrasting with Doxorubicin had been measured with the ability to induce apoptosis. The well-defined heavy rare-earth ytterbium iodide complex 1 (L 2 YbI) has been successfully employed as an efficient catalyst for the hydroboration of a wide range of aldehydes and ketones with pinacolborane (HBpin) at room temperature. The protocol requires low catalyst loadings (0.1-0.5 mol %) and proceeds rapidly (>99% conversion in <10 min). Additionally, catalyst 1 shows a good functional group tolerance even toward the hydroxyl and amino moieties and displays chemoselective hydroboration of aldehydes over ketones under mild conditions.

Objective: BCRP is overexpressed in many tumors and mediates multidrug resistance in breast cancer. In this study, we determined the involvement of miR-302S in the development of drug resistance in breast cancer., Methods: The differential miRNA expression profiling in parental MCF-7 cells and its derivative mitoxantrone (MX)-resistant MCF-7 (MCF-7/MX) cells was determined by the microarray analysis. The levels of miR-302S family and BCRP mRNA expression were determined by using Quantitative Real-Time PCR. The targeting effect between the individuals of miR-302S and BCRP mRNA-3'UTR were detected by dual-luciferase reporter assay. Proteins of BCRP are represented by Western blot assay. Cell viability was assessed by MTS assay. Efflux capacity was evaluated using flow cytometry., Results: The miR-302S family including miR-302a, miR-302b, miR-302c, and miR-302d was significantly down-regulated in BCRP-overexpressing MCF-7/MX cells. Luciferase activity assay showed that miR-302 inhibited BCRP expression by targeting the 3'-untranslated region (UTR) of the BCRP mRNA. Overexpression of miR-302 increased intracellular accumulation of MX and sensitized breast cancer cells to MX. Furthermore, intratumoral injection of miR-302 potentiated the inhibitory effect of MX on tumor growth in mice transplanted with MCF-7/MX cells. Most importantly, miR-302S produced stronger effects than each individual member alone., Conclusions: These findings suggest that miR-302 inhibits BCRP expression via targeting the 3'-UTR of BCRP mRNA. miR-302 members may cooperatively downregulate BCRP expression to increase chemosensitivity of breast cancer cells. miR-302 gene cluster may be a potential target for reversing BCRP-mediated chemoresistance in breast cancer., (Copyright © 2016 Elsevier Inc. Wnt/β-catenin signaling pathway plays a major role in the cancer metastasis. Several microRNAs (miRNAs) are contributed to the inhibition of breast cancer metastasis. Here, we attempted to find novel targets and mechanisms of microRNA-100 (miR-100) in regulating the migration and invasion of breast cancer cells. In this study, we found that miR-100 expression was downregulated in human breast cancer tissues and cell lines. The overexpression of miR-100 inhibited the migration and invasion of MDA-MB-231 breast cancer cells. Inversely, the downregulation of miR-100 increased the migration and invasion of MCF-7 breast cancer cells. Furthermore, FZD-8, a receptor of Wnt/β-catenin signaling pathway, was demonstrated a direct target of miR-100. The overexpression of miR-100 decreased the expression levels not only FZD-8 but also the key components of Wnt/β-catenin pathway, including β-catenin, metalloproteniase-7 (MMP-7), T-cell factor-4 (TCF-4), and lymphoid enhancing factor-1 (LEF-1), and increased the protein expression levels of GSK-3β and p-GSK-3β in MDA-MB-231 cells, and the transfection of miR-100 inhibitor in MCF-7 cells showed the opposite effects. In addition, the expression of miR-100 was negatively correlated with the FZD-8 expression in human breast cancer tissues. Overall, these findings suggest that miR-100 suppresses the migration and invasion of breast cancer cells by targeting FZD-8 and inhibiting Wnt/β-catenin signaling pathway and manipulation of miR-100 may provide a promoting therapeutic strategy for cancer breast treatment.

The reactivity of two β-diketiminate coordinated magnesium(I) dimers, [LMgMgL], L=[(RNCMe)(2) CH](-) , R=C(6) H(3) iPr(2) -2,6 ((Dip) Nacnac(-) ) or mesityl ((Mes) Nacnac(-) ), towards a series of isonitriles and nitriles have been examined. Reactions with the isonitriles, RN≡C: (R=tBu or C(6) H(3) Me(2) -2,6 (Xyl)), led to reductive C-C couplings and the formation of [{((Dip) Nacnac)Mg}(2) {μ-(XylN=C-)(2) }] and [{((Mes) Nacnac)Mg}(2) {μ-(tBuN=C-)(2) }], or a reductive N-C cleavage and the generation of the magnesium cyanide complex, [{((Dip) Nacnac)Mg(μ-CN)}(3) ]. Reactions of the magnesium dimers with benzonitrile, PhC≡N, afforded the C-C-coupled products, [((Dip) Nacnac)Mg[μ-{N=C(Ph)-}(2) ]Mg(NCPh)((Dip) Nacnac)], and [{{((Mes) Nacnac)Mg}(2) [μ-{N=C(Ph)-}(2) ]}(2) ], whereas the reductive CC cleavage of tBuC≡N gave rise to a mixture of [((Dip) Nacnac)Mg(tBu)(NCtBu)] and [{((Dip) Nacnac)Mg(μ-CN)}(3) ]. In contrast, a combination of net nitrile isomerization and C-C coupling processes was involved in the reduction of Me(3) SiC≡N, which yielded [{((Dip) Nacnac)Mg}(2) {μ-(Me(3) SiN=C-)(2) }]. All new compounds were crystallographically and spectroscopically characterized. The outcomes of the reported reactions were found to be dependent upon both the steric bulk of the magnesium(I) reagent, and the nature of the isonitrile/nitrile substituent. This combined with a high degree of selectivity for the reactions, indicates that magnesium(I) dimers may find use by organic and organometallic chemists as viable alternatives to currently available reducing agents that are utilized for the reduction of unsaturated organic substrates., (Copyright © 2012 WILEY-VCH Verlag GmbH & Co.
